1.The predictive value of serum YKL-40 and GDF-15 levels for clinical outcomes in patients with Parkinson disease

Objective:To explore the predictive value of human cartilage glycoprotein 39 (YKL-40) and growth differentiation factor 15 (GDF-15) for clinical outcomes of patients with Parkinson disease (PD).Methods:A total of 109 patients with PD admitted to Xinxiang Central Hospital from February 2021 to February 2023 were selected and treated with regular anti-PD medications for 4 weeks, with dosage appropriately adjusted according to clinical status and individual response.Clinical outcomes were evaluated after 2 months of treatment, and the predictive value of serum YKL-40 and GDF-15 at admission for clinical outcomes was analyzed.Data were analyzed by independent sample t-test, χ2 test and Logistic regression using SPSS 26.0. Results:PD patients with poor outcomes exhibited higher serum levels of YKL-40((3.18±0.67)mg/L, (2.34±0.41)mg/L) and GDF-15((457.82±142.83)pg/mL, (282.95±105.96)pg/mL) than those with good outcomes, and the differences were statistically significant ( t=8.082, 7.349, both P<0.05).Logistic regression analysis showed that elevated serum levels of YKL-40( B=0.664, OR=1.943, 95% CI=1.237-3.052) and GDF-15( B=0.185, OR=1.787, 95% CI=1.145-2.789) both influenced the clinical outcomes of PD patients(both P<0.05).Serum YKL-40 combined with GDF-15 demonstrated a predictive sensitivity of 87.23%, specificity of 90.32%, and AUC of 0.927(95% CI=0.861-0.968) for clinical outcomes in PD patients.The AUC was significantly higher than that achieved by either indicator alone (YKL-40: AUC (95% CI)=0.722 (0.628-0.804); GDF-15: AUC (95% CI)=0.797 (0.709-0.868)). Conclusion:The elevated levels of YKL-40 and GDF-15 in PD patients are associated with clinical outcomes, which may be the potential markers for predicting clinical outcomes of patients with PD.

3.Effect of Curcumin on Non-Small Cell Lung Cancer A549 Cells

Objective To study the inhibitory effect of curcumin on the proliferation,migration and invasion of non-small cell lung cancer cell A549,and to discuss further if it is closely related to the expression of c-Jun N-terminal kinase (JNK) and relative protein p38.Methods A549 cells were cultured by conventional method,and then treated with different concentration of curcumin (10,20,40,80 μmol · L-1).The proliferation,migration and invasion of A549 cells were measured by real-time cellular analysis (RTCA).The expression levels of JNK,p-JNK,p38 and P-p38 were detected by real-time PCR and Western blotting.Results Curcumin showed an antiproliferation effect against A549 cells with IC50 =40 μmol · L-1,and curcumin exhibited obviously inhibitory effect on the migration and invasion of A549 cells.Additionally,compared with control group,curcumin suppressed the expression of JNK and p38 at the gene level,and significantly inhibited the expression of JNK,P-JNK,p38 and p38 (P<0.05) at the protein level.Conclusion These results demonstrated that curcumin can inhibit the proliferation,migration and invasion of A549 cells via reducing the level of JNK,p38 phosphorylation,and blocking JNK signal transduction pathway.
4.Improving Effect of Curcumin Analogue H8 on Glucose and Lipid Metabolism Disorder in db/db Mice

Objective To investigate the influence of curcumin and its analogue H8 on glucose and lipid metabolism disorder in db/db mice. Methods The type 2 diabetes mouse model (db/db mice) was intragastrically administrated with curcumin and analogue H8 for 8 weeks.The blood biochemical indexes were measured.The expression of PEPCK and G6Pase mRNA was detected by real-time PCR in liver tissues.The expression of PEPCK and G6Pase protein was detected by Western blotting. Results Curcumin analogue H8 reduced blood glucose and lipids in db/db mice (P<0.01) and improved liver function related enzymes significantly.The levels of PEPCK and G6Pase mRNA in db/db mice were significantly decreased (P<0.01) and the expression levels of PEPCK and G6Pase protein were significantly decreased(P<0.01). Conclusion Curcumin analogue H8 improves the glucose and lipid metabolism disorder in db/db mice,and it is related to inhibiting the expression of PEPCK and G6Pase gene and protein.
5.Comparison of the decompression effect of static air cushion at different intervals on skin in bedridden patient

Objective To explore the effect of static air cushion at different intervals on preventing from pressure ulcers in bedridden patient .Methods One hundred patients in the department of orthopedic were chosen through the convenience sampling and self-controlled.The patients were supine , and the computer pressure sensor ( Xsensor manometry carpet ) was tiled under the body of patients .The average pressure peaks in the two lateral heel and sacrococcygeal were respectively measured after 2 h of lying ordinary mattress and 2, 3, 4 h of lying static air cushion , and the integrity of skin and the incidence of pressure ulcer were monitored during patients’ stay in bed.Results After 2 h of lying ordinary mattress , the cases of I stage pressure ulcer were 3 cases including 1 case of left heel and 2 cases of sacrococcygeal , and the incidence rate of pressure ulcer was 3%.After 2, 3, 4 h of lying static air cushion, the incidence of pressure ulcer in patients was not found , and the integrity rate of skin was 100%.After 2 h of lying ordinary mattress and static air cushion , the average pressure peaks were respectively (185.03 ±11.34), (69.13 ±9.64) mmHg in the left heel, and were (176.03 ±18.64), (76.40 ±11.52) mmHg in the right heel, and were (253.03 ±28.64), (100.23 ± 18.36)mmHg in the sacrococcygeal, and the differences were statistically significant (t =7.126,6.287, 4.235, respectively;P<0.05).No differences were found in the change of average pressure peaks in the two lateral heel and sacrococcygeal after 2, 3, 4 h of lying static air cushion ( F =5.863, 3.128, 4.852, respectively;P>0.05).Conclusions Static air mattress can effectively reduce vertical pressure on the surface of pressed parts in the bedridden patients , and the interval time of turning over can be prolonged to 4 h every time, and which can reduce the patients ’ pain caused by frequently turning over but also reduce the nurses ’ labor intensity .
6.Value of renal parenchymal volume and thickness by non-contrast spiral CT in evaluating differential glomerular filtration rate for chronic obstructed kidneys

Objective To evaluate the value of renal parenchymal volume and thickness by non-contrast spiral CT in evaluating the differential glomerular filtration rate (GFR) for chronic obstructed kidneys, and to compare the correlations between the two morphologic indices of renal parenchyma and the GFR for chronic obstructed kidneys. Methods Seventy-one patients who had a diagnosis of unilateral chronic upper urinary tract obstruction were included in this analysis. (1) The renal parenchymal volume was mea-sured by non-contrast spiral CT. Both kidneys were scanned by non-contrast spiral CT. The renal parenchymal area of each section was marked manually. Renal parenchymal volume was calculated as the sum of renal parenchymal area multiplied by the width of each section. The volume percentage of obstructed kidney (%CTvol) was also calculated. (2) Renal parenchymal thickness was measured on the first and last non-contrast CT image levels from the anterior, posterior and lateral locations of the kidney that clearly contained the collecting system. The mean of these measurements was defined as the renal parenchymal thickness. The differential renal parenchymal thickness of the obstructed kidney (%CTt) was defined as the percentage of the obstructed renal parenchymal thickness to the total renal parenchymal thickness for both kidneys. GFR was determined with 99Tcm-DTPA dynamic imaging system by Gates method. The differential GFR for obstructed kidney (%GFR) was the GFR percentage of obstructed kidney to the total GFR for both kidneys. The Pearson relation test was carried out between the %CTvol, %CTt and the %GFR respectively. Results %CTvol and %CTt correlated well with %GFR in chronic obstructed kidneys among the 71 test group patients. Pearson correlation coefficient r was 0.80 (t=11.20, P<0.05) and 0.66 (t=7.24, P<0.05), respectively. The linear correlation equation respectively was %GFR=0.05+0.80×%CTvol (F=125.48, P<0.05) and %GFR=0.12+0.66×%CTt (F=52.36, P<0.05). Conclusions Renal parenchymal volume and thickness by non-contrast spiral CT might be used as clinical practical parameters to evaluate the differential GFR for chronic obstructed kidneys. Renal parenchymal volume is more accurate than renal parenchymal thickness.
7.The management for 51 cases of ureteral obstruction caused by pelvic malignancy

Objective To discuss the effect and treatment methods of ureteral obstruction caused by primary or metastatic pelvic malignancy. Methods Clinical data of 51 cases with ureteral obstruction caused By pelvic malignancy were reviewed retrospectively. The treatment included traditional open operation in 17 cases in which 3 cases with uretero-uwtero, 9 cases with uretero-bladder anastomotic stoma and 5 cases with cutaneous ureterostomy, retrograde stenting in 19 cases (11 cases with unilateral ureter, 8 cases with bilateral ureter), pereutaneous nephrostomy in 15 cases. Results The median follow-up time was 21 months (range 6 to 72 months ). Three months after operation, uhrasonograph or intravenous urography(IVU)showed that 39 (76.5%) cases had nomal renal function, 12 (23.5%) cases had hydronephrosis relief and renal function improvement. No stricture in the uretero-uretero or uretero-bladder anastomotic stoma was recorded. Conclusions Appropriate treatment is dependent upon the accurate identification of the underlying pathological process and it can relieve ureteral obstruction and increase the quality of life for ureteral obstruction caused by pelvic malignancy.
