1.Study on Quality Evaluation of Classical Formula-Linggui Zhugan Decoction Based on UPLC Fingerprint Chromatogram Combined with Chemometric Analysis

Objective To establish the UPLC fingerprint chromatogram combined with chemometric analysis for the quality evaluation of classical formula Linggui Zhugan Decoction.Methods SHIMADZU Shim-Pack GIST C18 column(100 mm×2.1 mm,2.0 μm)was used with acetonitrile-0.1%phosphoric acid aqueous solution as mobile phase,gradient elution;flow rate was 0.2 mL/min;the detection wavelength was 266 nm for the first 30 minutes and 235 nm for the last 36 minutes;the column temperature was 30℃.The UPLC fingerprint of Linggui Zhugan Decoction was established by Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012.130723 version),and the common peak was determined and the similarity evaluation was carried out.Based on the peak area determination results of the common peak of the fingerprint,the quality of different batches of Linggui Zhugan Decoction was evaluated by chemometrics such as clustering analysis and principal component analysis.Results A total of 24 common peaks were confirmed and 14 components were identified by using reference substances.The similarity of 10 batches of Linggui Zhugan Decoction samples was greater than 0.950,which could be divided into two categories by chemometrics,and the principal component 1-4 were the main factors affecting its quality evaluation.OPLS-DA identified 6 differential markers.Conclusion The fingerprint research method established in the study is simple,reliable and reproducible.Through the method of fingerprint combined with chemometrics analysis,the differences between Linggui Zhugan Decoction from different origins of medicinal materials are identified,which provides a reference for the internal quality evaluation of Linggui Zhugan Decoction.
2.Effect of Maxing Shigantang and Supplemented Guominjian Decoction on Symptoms and Inflammatory Cytokine Levels in Induced Sputum of Children with Cough Variant Asthma

ObjectiveTo investigate effect of Maxing Shigantang and supplemented Guominjian decoction on symptoms and levels of inflammatory cytokines in induced sputum of children with cough variant asthma (CVA). MethodA total of 118 CVA children who were treated in our hospital from January 2020 to January 2021 were enrolled and randomized into the control group and the observation group with the random number table method. Control group received routine western medicine and the observation group was treated by routine western medicine, Maxing Shigantang, and supplemented Guominjian decoction. In the one-month follow-up, the scores of cough and accompanying symptoms, levels of inflammatory cytokines [interleukin-10 (IL-10), interleukin-5 (IL-5), tumor necrosis factor-α (TNF-α), neutrophil, eosinophil] in induced sputum, pulmonary function parameters [forced vital capacity (FVC), forced expiratory volume in one second (FEV1), FEV1/FVC], and treatment outcomes were compared between the two groups. Moreover, the frequency of acute asthma attacks during the three-month follow-up was also compared. ResultNo cases dropped out from this study. After treatment, the scores of cough and accompanying symptoms were decreased in both groups (P<0.05) and were lower in observation group than in control group (P<0.05). After treatment, FVC, FEV1, and FEV1/FVC were raised in both groups and were higher in observation group than in control group (P<0.05). The increase in the level of IL-10 along with the decrease in levels of IL-5, TNF-α, neutrophil, and eosinophil in induced sputum was found in both groups after treatment (P<0.05), and observation group had higher level of IL-10 and lower levels of IL-5, TNF-α, neutrophil, and eosinophil than the control group (P<0.05). The effective rate was 86.44% (51/59) in observation group, which was higher than the 69.49% (41/59) in control group (χ2=4.933, P<0.05). No serious adverse reaction occurred in either group. The frequency of acute asthma attacks during the three-month follow-up was (1.09±0.18) in observation group, which was lower than the (2.83±049) in the control group (P<0.05). ConclusionRoutine western medicine combined with Maxing Shigantang and supplemented Guominjian decoction can effectively and safely alleviate the airway inflammatory responses, control the clinical symptoms, improve pulmonary function, and reduce the frequency of acute recurrence in the treatment of CVA children.

Crohn′s disease (CD) is a chronic, progressive and destructive inflammatory disease affecting the entire digestive tract. Changes in the intestinal microbiota, particularly a decrease in gut microbiome diversity, are thought to be associated with chronic intestinal inflammation of CD. As for the mechanism of antibiotics in CD treatment, some scholars believe that antibiotics can affect the course of disease by reducing the concentration of intestinal bacteria and changing the composition of intestinal microbiota. Different antibiotics, including ciprofloxacin, metronidazole and rifaximin, have been proved to have certain therapeutic effect on some patients with CD in clinical practice, but there are still limitations in the use of antibiotics.In this review, the relationship between intestinal flora and the incidence of CD and the application of antibiotics in CD were reviewed, providing reference and help for the standardized application of antibiotics in CD.
4.Trend on Mortality of Gastric Cancer and Cause-eliminated Life Expectancy from 1987 to 2019 in Harbin

Objective To analyze the mortality characteristics and trends and the cause-eliminated life expectancy of gastric cancer in Harbin City from 1987 to 2019. Methods Mortality data of residents with gastric cancer from 1987 to 2019 in Harbin was analyzed to describe the mortality characteristics and trends of gastric cancer. Abridged life table and cause-eliminated life table were applied to calculate life expectancy and cause-eliminated life expectancy. Average annual percentage change (AAPC) was calculated with Joinpoint 4.2 software to evaluate the trends of mortality and cause-eliminated life expectancy of gastric cancer. Results From 1987 to 2019, the crude mortality, ASMRC and ASMRW and the truncated rate (35-64) were 14.3/105, 10.9/105, 10.9/105, and 13.5/105, respectively. 5.Best evidence summary for the management of injection site pain in subcutaneous patients

Objective:To retrieve, evaluate and summarize the relevant evidence of subcutaneous injection in patients to reduce pain at the injection site, so as to provide reference for clinical practice.Methods:According to the evidence pyramid "6S" model, all evidence on subcutaneous injection and injection site pain, including guidelines, expert consensus, summary of evidence, clinical decision, systematic review, etc were retrieved from domestic and international guideline networks and databases. The search period was from database establishment to October 2, 2022. The literature quality evaluation and evidence grading system of Joanna Briggs Institute (JBI) Evidence-based Health Care Center was used to evaluate the literature quality and classify the evidence level.Results:A total of 12 articles were included, including 2 expert consensus, 6 systematic reviews, 2 best evidence summaries and 2 clinical guidelines. A total of 21 pieces of best evidence were summarized from 8 aspects, including pre-subcutaneous injection assessment, injection site, injection needle, injection position, injection method, precautions, personnel training and health education.Conclusions:This study summarized a comprehensive and practical subcutaneous injection method. Clinical practitioners can use evidence to administer subcutaneous injection to patients, reduce pain and improve patients ′ comfort.
6.The effect of modified ginseng and schisandra decoction on the efficacy and inflammatory indicators of children with lobar pneumonia(lung spleen deficiency syndrome)caused by mycoplasma pneumonia infection

Objective To explore the clinical efficacy of ginseng schisandra decoction in children with lobar pneumonia(lung spleen deficiency syndrome)induced by mycoplasma pneumoniae infection and improvement of inflammatory indicators.Method 84 children with lobar pneumonia(lung spleen deficiency syndrome)caused by mycoplasma infection admitted to the First Clinical Hospital of Jilin Academy of Traditional Chinese Medicine from January 2020 to July 2022 were selected and randomly divided into two groups with 42 cases each.The conventional group was treated according to the traditional treatment plan,while the combined group was treated with ginseng and schisandra decoction in addition to the traditional treatment plan.After 7 days,compare the differences in traditional Chinese medicine syndrome scores,clinical efficacy,inflammatory indicators,and total incidence of adverse reactions between the two groups of children.Result The results showed that the traditional Chinese medicine syndrome score and inflammatory indicators in the combined group were lower than those in the conventional group(P<0.05),and the clinical efficacy level distribution was better than that in the conventional group(P<0.05).The total effective rate was higher than that in the conventional group(P<0.05),and there was no difference in the total incidence of adverse reactions between the two groups(P>0.05).Conclusion In the treatment of children with lobar pneumonia(lung spleen deficiency syndrome)caused by Mycoplasma pneumoniae infection,the modified Ginseng Wuweizi decoction has a positive therapeutic effect and good safety,and is worth promoting.
7.Epidemiological investigation and phylogenetic analysis of Classical Swine Fever virus in Yunnan province from 2015 to 2021

Background:
Classical swine fever virus (CSFV), the causative agent of classical swine fever (CFS), is a highly contagious disease that poses a serious threat to Chinese pig populations.
Objectives:
Many provinces of China, such as Shandong, Henan, Hebei, Heilongjiang, and Liaoning provinces, have reported epidemics of CSFV, while the references to the epidemic of CSFV in Yunnan province are rare. This study examined the epidemic characteristics of the CSFV in Yunnan province.
Methods:
In this study, 326 tissue samples were collected from different regions in Yunnan province from 2015 to 2021. A reverse transcription-polymerase chain reaction (RT-PCR), sequences analysis, and phylogenetic analysis were performed for the pathogenic detection and analysis of these 326 clinical specimens.
Results:
Approximately 3.37% (11/326) of specimens tested positive for the CSFV by RTPCR, which is lower than that of other regions of China. Sequence analysis of the partial E2 sequences of eleven CSFV strains showed that they shared 89.0–100.0% nucleotide (nt) and 95.0–100.0% amino acid (aa) homology, respectively. Phylogenetic analysis showed that these novel isolates belonged to the subgenotypes 2.1c and 2.1d, with subgenotype 2.1c being predominant.
Conclusions
The CSFV was sporadic in China’s Yunnan province from 2015 to 2021. Both 2.1c and 2.1d subgenotypes were found in this region, but 2.1c was dominant.
8.Analysis on Difference of Medicinal Properties Between Atractylodes lancea and A. chinensis Based on Correlation of Energy Metabolism and Dryness

ObjectiveTo explore the correlation between dryness and energy metabolism of Atractylodis Rhizoma, and to analyze the difference of medicinal properties between Atractylodes lancea and A. chinensis. MethodA total of 110 healthy SD rats were randomly divided into 11 groups, including normal group, volatile oil of A. lancea 1-5 group (S1-S5 group, doses of 447, 473, 442, 489, 496 mg·kg-1) and volatile oil of A. chinensis 1-5 group (N1-N5 group, doses of 197, 118, 281, 222, 185 mg·kg-1), the administration volume was 0.01 mL·g-1 with intragastric administration for 21 days. Dryness effect of A. lancea and A. chinensis on rats was evaluated by comparing the body weight, drinking water volume, urine volume, whole blood viscosity and pathological sections of submandibular gland stained with hematoxylin-eosin (HE). The expression of aquaporin 2 (AQP2) in rat kidney was measured by immunohistochemistry, the mRNA expressions of cytochrome C oxidase subunit 7A2 (COX7A2) and succinate dehydrogenase (SDH) complex subunit D (SDHD) in liver tissue were detected by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). The contents of SDH, lactate dehydrogenase (LDH) and sodium ion-potassium ion-adenosine triphosphatase (Na+-K+-ATPase) in rat plasma were determined by colorimetry. The quality of A. lancea and A. chinensis was evaluated by coefficient of variation method, and Pearson correlation coefficient was used to analyze the correlation between dryness and energy metabolism. ResultCompared with the normal group, the amounts of drinking water and urine in volatile oil of A. lancea group and volatile oil of A. chinensis group increased, and the submandibular gland acini atrophied, the whole blood viscosity of rats in the volatile oil of A. lancea group increased significantly (P<0.01), the expression levels of COX7A2 and SDHD mRNA, the activities of SDH, LDH and Na+-K+-ATPase increased significantly (P<0.01), and the expression of AQP2 in kidney decreased significantly (P<0.01). Compared with the normal group, the expression level of COX7A2 mRNA, SDH activity and whole blood viscosity in the volatile oil of A. chinensis group increased (P<0.05, P<0.01), the AQP2 and SDH mRNA expression levels, LDH and Na+-K+-ATPase activities had no significant difference. The comprehensive score analysis of each index showed that the effect of volatile oil of A. lancea on dryness and energy metabolism was stronger than that of volatile oil of A. chinensis, and there was a positive correlation between dryness index and energy metabolism index. ConclusionThe two indexes show that medicinal properties of A. lancea is stronger than that of A. chinensis, and energy metabolism is closely related to the dryness of Atractylodis Rhizoma. It is suggested that it is reasonable to evaluate the dryness effect of Atractylodis Rhizoma from the perspective of energy metabolism, which can further enrich the evaluation indexes of medicinal properties.
9.Anti-tumor Effect of Ligustrazine Alone and in Combination with Western Medicine: A Review

With serious environmental problems and the aging of population, the incidence of cancers worldwide has increased dramatically. For modern medicine, cancer is an incurable malignant disease with a high metastasis rate and strong cell proliferation and invasion ability. At present, clinical anti-cancer therapies mainly include chemotherapy and radiotherapy, but most chemotherapy drugs can cause serious side effects. Therefore, Chinese medicine with high safety has been widely used in anti-cancer treatment, which can inhibit the proliferation of cancer cells, improve the quality of life, and prolong life. Ligustrazine is a pyrazine alkaloid isolated and purified from Chuanxiong Rhizoma, which has pharmacological effects, such as anti-fibrosis, anti-oxidation, and anti-tumor. In recent years, ligustrazine, as an index active component of Chuanxiong Rhizoma alkaloids, has gradually become one of the hot spots in clinical medical research due to its pharmacological actions, and its anti-tumor effect has attracted more attention from researchers. The present study mainly investigated the anti-cancer effect of ligustrazine from the induction of tumor cell apoptosis and autophagy, inhibition of cell proliferation and migration, reversal of multidrug resistance of cancer cells, and inhibition of angiogenesis, and the anti-tumor efficacy was observed in various malignancies such as gastric cancer, lung cancer, rectal cancer, bladder cancer, breast cancer, and ovarian cancer. In addition, ligustrazine can be combined with cisplatin, paclitaxel, podophyllotoxin, chalcone, curcumin, and other drugs to exert an anti-tumor effect. This study conducted a literature review based on the anti-tumor mechanism of ligustrazine to provide a reference for the treatment of clinical malignancies.
10.Awareness of early childhood sex education and influencing factors among rural kindergarten teachers in Sichuan

Objective:
To investigate the awareness of sexuality education knowledge in kindergarten teachers in rural areas of Sichuan and its influencing factors, and to provide a scientific basis for sex education capacity improvement among rural teachers.
Methods:
With multi stage stratified cluster sampling method, 162 teachers selected from 16 rural kindergartens in rural areas of Sichuan Province to conduct questionnaires related to early childhood sex education.
Results:
The awereness of early childhood sex education foreskin care in rural areas in Sichuan was 73.8%. Knowledge regarding foreskin care(58.8%)", "the function of the family includes rest and entertainment functions(65.6%)", "girls need to clean the reproductive truets from the front to back (68.8%)". Multivariate non conditional Logistic regression analysis results showed that teaching experience>4 years(OR=4.16, 95%CI=1.74-9.98), notice of young children with sexual related behaviors(OR=2.97, 95%CI=1.19-7.40), the number of sources of sexual knowledge sources ≥5(OR=3.39, 95%CI=1.42-8.13) were associated with high awareness of preschool sex education knowledge.
Conclusion
The preschool sex education knowledge awareness among Sichuan rural kindergarten teachers needs to be improved. A variety of sex education channels should be constructed to encourage teachers, especially those who are newly employed, to acquire more relevant knowledge, pay attention to children’s sex related behaviors, and improve their ability in sex education.
