1.A new model constructed using biochemical indices for predicting cardiac diastolic function in patients with type 2 diabetes mellitus

Objective To establish a new model for predicting cardiac function in patients with type 2 diabetes mellitus(T2DM)by using biochemical indicators,in order to find the intrinsic factors affecting cardiac function in T2DM patients.Methods A total of 361 patients with T2DM admitted to the Department of Endocrinology of The First Affiliated Hospital of Zhengzhou University from January 2023 to January 2024 were retrospectively selected.They were divided into T2DM group(n=117)and decreased ventricular diastolic function group(n=244)according to cardiac function.General characteristics and biochemical indicators were collected in the two groups,as well as echocardiography.Logistic regression was used to construct a new prediction model.The area under the receiver operating characteristic(ROC)curve(AUC)was used to evaluate the accuracy of the model,and the Hosmer-Lemeshow goodness-of-fit test was used to evaluate the calibration of the model.Results Age,DM duration,TC,urea,serum creatinine(Scr),25(OH)D3 and HDL-C were higher,while TG,triglyceride and glucose product index,serum uric acid(SUA)and glomerular filtration rate(eGFR)were lower in the ventricular diastolic function decline group than in the T2DM group(P<0.05).Logistic regression analysis showed that the duration of DM,urea,Scr,SUA and eGFR were the influencing factors for cardiac diastolic function decline inpatients with T2DM,and the prediction model was constructed as follows:y=7.054+0.097×ln DM duration+0.209×ln urea-0.203×ln Scr-0.003×ln SUA-0.052×ln eGFR,AUC was 0.832,sensitivity was 0.713,specificity was 0.803,cut-off value was 0.69,and the model fitted well and had certain predictive value.Conclusions The decline of cardiac diastolic function in patients with T2DM is related to DM duration,urea,SUA,Scr and eGFR.The new model has a good predictive effect on cardiac function in patients with T2DM and provides clinical value.

3.Decision-making experience of patients with abdominoplasty: a qualitative study

Objective:To explore the decision-making experience of patients undergoing abdominoplasty in our country.Methods:A semi-structured interview was conducted with patients suffering from postpartum abdominal wall laxity who were treated at Union Hospital, Tongji Medical College, Huazhong University of Science and Technology from June to August 2023. The data were sort and analyzed using the Clolaizzi 7-step analysis method for thematic description.Results:A total of 15 patients, aged 29-53 years, were included in the study. 11 had a college education or higher, while 4 had less than a college education.Thirteen were married and two were divorced. According to the interview data, four themes were extracted: (1) negative perceptual experiences; (2) decision-making dilemmas; (3) peer support; (4) a physician-led decision-making model. According to the interview, the main decision-making dilemma faced by patients with postpartum abdominal wall laxity was lack of information and family support, and the support they get was mostly from information exchange among patients, and they had insufficient cognition of surgical knowledge, improvement degree and risk, and low participation in clinical decision making.Conclusion:Patients with postpartum abdominal wall laxity in China have an urgent psychological need for abdominoplasty, but there are many factors that have adverse effects on their decision-making experience.
4.Effects and mechanism of Setaria italica extract on improving sleep in insomnia mice

OBJECTIVE To investigate the effects of Setaria italica extract on improving insomnia model mice and to explore its potential mechanisms. METHODS The mice were randomly assigned into blank group, model group, positive control group (diazepam, 2.6 mg/kg), and S. italica extract low-dose, medium-dose and high-dose groups (1.2, 2.4, 4.8 g/kg), with 10 mice in each group. Except for the blank group, all other groups received intraperitoneal injection of para-chlorophenylalanine (PCPA) to establish the insomnia model. After modeling, the blank group and model group were given a constant volume of normal saline intragastrically, and administration groups were given relevant medicine intragastrically, with a volume of 0.01 mL/g, once a day, for 7 consecutive days. After the administration, the open-field test was conducted to observe the praxiological changes of mice, and to determine the levels of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HTAA) in the hippocampal tissue, as well as the contents of 5-HT, brain-derived neurotrophic factor (BDNF), interleukin-2 (IL-2), IL-6, B-cell lymphoma-2 (Bcl- 2), and Bcl-2-associated X protein (Bax) in the serum. The expression of phosphoinositide 3-kinase/protein kinase B/nuclear factor- κB (PI3K/Akt/NF-κB) signaling pathway related protein was determined in the hippocampus of mice. RESULTS Compared with the model group, the total exercise time of mice in S. italica extract high-dose group was significantly prolonged, but the total rest time was significantly shortened (P<0.01); the number of standing times and modification times were significantly reduced (P< 0.01). The contents of 5-HT, BDNF, and Bcl-2 in serum, and Bcl-2/Bax were significantly increased, while the contents of IL-2, IL-6, and Bax were significantly reduced (P<0.05 or P< 0.01). The content of 5-HTAA in the hippocampal tissue and 202104010910029);the phosphorylation levels of PI3K and Akt proteins were increased significantly, while the phosphorylation level of NF-κB p65 protein was decreased significantly (P<0.05).CONCLUSIONS High-dose of S. italica extract demonstrates significant therapeutic effects on insomnia in mice, and the mechanism of which may be associated with the regulation of PI3K/Akt/NF-κB signaling pathway.

6.Progress in Biological Functions of NBAS Gene and Related Diseases

Neuroblastoma-amplified sequence (NBAS) is a highly conserved gene firstly found in neuroblastoma, targeting on the human chromosome 2 p24.3 and encodes a protein that is a subunit of the Syntaxin 18 complex. The functions of NBAS include the involvement in transport of Golgi-to-Endoplasmic Reticulum retrograde and degradation of nonsense-mediated mRNA. NBAS gene is widely expressed in more than 30 tissues, suggesting that it may play an important role in human body. In 2010 and 2015, NBAS was identified successively as the pathogenic gene of SOPH syndrome and fever-related liver failure respectively. Recent studies shows that NBAS gene mutations can involve immune system and cause immune deficiency. In this paper, we review the biological function of NBAS, NBAS gene mutation-related diseases and their pathogenesis in recent years.
7. Study on the Specific Complexation of GMDTC and Metal Ion

Objective:
Study the response of GMDTC to cadmium ions and metal ions in vivo to determine whether GMDTC are specifically complexed with cadmium ions to provide a reference for the safety and dfficacy of GMDTC.
Methods:
Complexometric titration, HPLC and HPLC-MS were applied to research the complexation reaction of GMDTC and various metal ions. The molecular ion peak of GMDTC, GMDTC-Cd complex and GMDTC-Pb complex also detected by LC-MS. Additionally, the initial structure was determined by DFT simulation method.
Results:
Results of complexometric titration and HPLC detection showed that GMDTC characteristic absorption peak area was proportional to the concentration of itself and there was no color change and peak time change when the GMDTC mixed with Ca2+, Fe2+, Mg2+, Zn2+. However, the color changed to black transition when the GMDTC mixed with Cu2+ and the color changed from yellow precipitate to light yellow transparent transition when GMDTC mix with Hg2+. Moreover, the peak area as well as the retention time has changed a lot which indicated that a chemical reaction has already happened. When the GMDTC mixed with Cd2+ and Pb2+, the color has changed from pale yellow to colorless transparent and the peak area of GMDTC has increased a lot. Finally, the GMDTC-Cd complex ratio both of which are 2:1 were calculated based on the results of LC-MS instrument and atomic calculations.
Conclusion
The specific cadmium chelating agent GMDTC can not react with the Ca2+, Fe2+, Mg2+, Zn2+, but it can react chemically with Cu2+ and Hg2+, even specific complex with Pb2+ and Cd2+.
