1.IL-17D regulates recruitment of lung NK cells and promotion of Astragalus membranaceus

Objective:To investigate the role of IL-17D to regulate the recruitment of lung NK cells and the effects of Astragalus membranaceus on IL-17D.Methods:The lung metastasis of tumor model was established by injecting B16 melanoma cells through caudal vein.After being treated with IL-17D or Astragalus membranaceus respectively,the expression of IL-17D and NK cells in lung was detected by flow cytometry and RT-PCR.Results:The production of IL-17D and the content of NK cells in the lung tumor me-tastasis model were dramatically decreased.After treatment with IL-17D,the content of NK cells in lung was significantly increased and the lung tumor foci were decreased.Along with up-regulated recruitments of NK cells,CXCL9 and IL-15,which support NK cell recruitments and maintain survival,were significantly increased.Astragalus inhibited lung tumor developments in lung by increasing the IL-17D expression and up-regulated the lung NK cell content.Conclusion: IL-17D can regulate the lung recruitment of NK cells.Astragalus mongholicus can enhance the capacity of NK cell recruitment and promote anti-tumor immune effects by up-regulating the expression of IL-17D.
2.Comparison of two gastric cancer screening schemes in a high-risk population.

OBJECTIVETo evaluate the effects of two gastric cancer screening schemes for early detection of gastric cancer in a high-risk population.
METHODSA cluster random sampling method was used to select local residents aged 40-69 years from Linqu County, Shandong Province. "Serum pepsinogen initial screening combined with further endoscopic examination (PG scheme)" and "direct endoscopic examination (endoscopy scheme)" were conducted. The associations between screening schemes and detection rates of gastric cancer, and early gastric cancer/high-grade intraepithelial neoplasia were evaluated by unconditional logistic regression analysis.
RESULTSOverall, 3654 and 2290 participants completed PG and endoscopy schemes, respectively. A total of 11 (0.30%) cases of gastric cancer and 10 (0.27%) cases of high-grade intraepithelial neoplasia were detected by PG scheme, of which 7 (0.19%) cases were early gastric cancer. While, 19 (0.83%) cases of gastric cancer and 10 (0.44%) cases of high-grade intraepithelial neoplasia were detected by endoscopy scheme, with 12 (0.52%) cases of early gastric cancer. Compared with the PG scheme, the endoscopy scheme had a significantly higher detection rates of gastric cancer (OR = 2.83, 95%CI 1.34-5.98), and early gastric cancer/high-grade intraepithelial neoplasia (OR = 2.12, 95%CI 1.12-4.02).
CONCLUSIONSThe endoscopy scheme is more effective in the detection of gastric cancer in a high-risk population, particularly for early gastric cancer/high-grade intraepithelial neoplasia than the PG scheme.

3.Immune changes in type 1 diabetes animal model after syngeneic bone marrow transplantation.

Syngeneic bone marrow transplantation (syn-BMT), as a novel therapy for type 1 diabetes (T1D), has been used more and more widely. This study was aimed to detect the changes of peripheral CD4(+) T lymphocytes, CD8(+) T lymphocytes, CD4(+)/CD8(+) T lymphocytes and NK cells before and after T1D mice were treated with syn-BMT, and to investigate the effects of these cells in T1D and the effects of syn-BMT-inducing immunotolerance. T1D mouse model was established by multiple low dose streptozotocin injection, the syn-BMT was performed on 10 day after the onset of diabetes. The T1D model mice were divided into group of diabetic mice treated with syn-BMT and group of diabetic control mice (DC), 6 normal C57BL/6J mice were regarded as normal control group (NC). On 30 day after syn-BMT, peripheral proportion of CD4(+) T lymphocytes, CD8(+) T lymphocytes, CD4(+)/CD8(+) T lymphocytes and NK cells were detected by flow cytometry. These cells of normal control mice (NC), diabetes control mice (DC) and diabetes mice treated by syn-BMT were also detected. Blood glucose level in three groups was assayed during the whole observation period. The results showed that syn-BMT could reduce blood glucose level of T1D mice to near normal (p > 0.05). Hematopoietic reconstitution happened in a month. The proportion of peripheral CD4(+) T lymphocytes, CD4(+)/CD8(+) T lymphocytes, NK cells all increased in new-onset diabetic mice (p < 0.01), while the proportion of peripheral CD8(+) T lymphocytes decreased (p < 0.01). On 30 day after T1D mice were treated with syn-BMT, the proportion of peripheral CD4(+) T lymphocytes was significantly lower than that in DC mice (p < 0.01), but still higher than NC (p < 0.05). The proportion of CD8(+) T lymphocytes was higher than that in DC and NC mice (p < 0.01). The ratio of CD4(+)/CD8(+) T lymphocytes and proportion of NK cells were both obviously lower than that in DC and NC mice (p < 0.01). It is concluded that the syn-BMT can reverse hyperglycemia and immune disorder in diabetic mice. On early period of diabetes onset, the proportions of CD4(+) T lymphocytes and NK cells, the ratio of CD4(+)/CD8(+) T lymphocytes increase, while proportion of CD8(+) T lymphocytes decreases in peripheral blood which mye be associated with onset of diabetes.

4.The effect of inhalable titanium dioxide on the oxidative stress among occupational population.

OBJECTIVETo investigate the inhalable titanium dioxide exposure level and make an assessment of its oxidative effect on occupational exposed population.
METHODSA total of 7 workers occupationally exposing to inhalable titanium dioxide were recruited into the study. The basic information and occupational history were collected by interview, while their blood sample (10 ml for each subject) were collected before and after the investigation, respectively. Pre- and post-work shift urine samples (60 ml for each subject) were collected for 29 days consecutively. The daily personal titanium dioxide exposure level, temperature and relative humidity were detected too. Urinary 8-hydroxy-deoxyguanosine (8-OHdG) and serum high-sensitivity C-reactive protein (hs-CRP) were detected by ELISA and latex immunoturbidimetric assay, respectively.
RESULTSThe mean concentration of air inhalable titanium dioxide was (1.194 ± 1.015) mg/m(3). Serum hs-CRP level before and after the investigation was (1.13 ± 1.08), (1.33 ± 1.01) mg/L, respectively. No statistical significance was observed between hs-CRP level before and after the investigation (t = -0.848, P = 0.425). Pre- and post-work shift urinary 8-OHdG was (3.51 ± 1.39), (3.65 ± 1.06) µmol/mol Cr, respectively. A positive correlation was found between the concentration of inhalable titanium dioxide and the changes of 8-OHdG level (r = 0.192, t = 2.09, P = 0.039). Linear mixed-effect models, adjusted by work shift, years of employment, age, body mass index, smoking status, temperature and relative humidity, showed no significant exposure-respond trend between the inhalable titanium dioxide concentration and 8-OHdG level (β = 0.288, t = 1.940, P = 0.055).
CONCLUSIONOur findings do not support the potential link between occupationally exposure to inhalable titanium dioxide and high induction of DNA oxidative stress.
