1.Clinical efficacy of drugs combined with hyperbaric oxygen in the treatment of patients with postherpetic neuralgia and its effect on the expression of brain-derive neurotrophic factor

Objective To explore the clinical efficacy of drugs combined with hyperbaric oxygen (HBO) in the treatment of patients with postherpetic neuralgia and its effect on the expression of brain-derive neurotrophic factor (BDNF).Methods One hundred patients with postherpetic neuralgia who were treated in the hospital from January 2015 to June 2018 were selected as the research subjects,and were randomly divided into the control group and the observation group,each consisting of 50 patients.The control group was treated with carbamazepine tablets in addition to routine treatment,while the observation group was supplemented with HBO in addition to the treatment received by the control group.Then,the scores of visual analogue scale (VAS) and patients' global impression of change (PGIC),the expression level of BDNF and rate of adverse reactions at admission and 3 months after treatment were compared between the patients of the 2 groups.Results After 3 courses of treatment,the VAS scores in the patients of the 2 groups all decreased significantly,with the scores of the observation group all being lower than those of the control group,and statistical significance could be noted when comparisons were made between the 2 groups (P < O.O1).Total efficacy of the observation group (84.0%) was obviously higher than that of the control group (58.0%),with statistical significance(P <0.01).The serum levels of BDNF in the patients of the 2 groups were markedly elevated,with that of the observation group being higher than that of the control group,also with statistical significance(P <0.O1).The rate of adverse reactions for the patients of the observation group (8.0%)being lower than that of the control group(16.0%),but without statistical significance(P > 0.05).Conclusion Carbamazepine tablets combined with HBO could effectively improve clinical symptoms,decrease the seriousness of pain,increase BDNF level and enhance treatment efficacy.For this reason,it was worth further clinical promotion.

3.Effects of hyperbaric oxygen on the expressions of endothelin-1, endothelial nitric oxide synthase and endothelin A receptor in cerebral circulation blood vessel in the rabbit model of cerebral ischemia

Objective To investigate the effects of hyperbaric oxygen (HBO on the expressions of endothelin-1 (ET-1),endothelial nitric oxide synthase (eNOS)and endothelin A receptor(ETAR) in cerebral circulation blood vessel in the rabbit model of cerebral ischemia.Methods Forty healthy adult New Zealand rabbits were randomly divided into 2 groups;the surgical group (n =30) and the control group (n =10).The rabbits in the surgical group were further divided into the HBO + cerebral ischemia group (n =15) and the simple cerebral ischemia group (n =15) in accordance with different treatment methods.In the surgical group,cerebral ischemia model was established by thread embolus method.The HBO + cerebral ischemia group was exposed to HBO at a pressure of 0.22 MPa(2.2 ATA)for 60 minutes,one session a day,for a succession of 3 days.In the sham surgical group,neck vessel was only separated and the incision was soon sutured.The animals in the sham surgical group and the cerebral ischemia group did not receive HBO exposure,had normal diet and left there intact for 3 days.Three days later,all the animals were sacrificed,and then immunohistochemical staining was performed by using ET-1,ETAR and eNOS antibodies,and observations were made on the expressions of ET-1,eNOS and ETAR in the internal carotid artery and basilar artery of the 3 groups.Results Experimental results indicated that there were expressions of ET-1,eNOS and ETAR in the internal carotid artery and basilar artery after HBO exposure.As compared with those of the sham surgical group and the cerebral ischemia group,the expression levels of ET-1 and ETAR in the internal carotid artery (51.6 ± 2.1,53.4 ± 1.5) of the HBO + cerebral ischemia group were obviously higher than those in the basilar artery (20.3 ± 2.6,24.3 ± 1.7),and eNOS expression level in the basilar artery (52.1 ± 2.3) was significantly higher than that in the internal carotid artery(22.3 ± 1.2).Conclusions HBO exposure could enhance ET-1 and ETAR expression levels in the internal carotid artery,and could improve eNOS expression in basilar artery,resulting in anterior circulation contraction and posterior circulation dilation.

5.Influence of Portulaca extracts on TNF-αand IL-4 in skins of rats with acute ec-zema

Objective:To investigate the influence and mechanism of Portulaca extracts on Tumor Necrosis Factor -α(TNF-α), Interleukin-4(IL-4) in skins of rats with atopic eczema (AE).Methods:40 SD rats were randomly divided into normal control group , model group,Cortex Phellodendri group and Porulaca group.2,4-dinitrochlorobenzene was used on the right back of rat for making atopic eczema.21 days later , all animals were sacrificed , compared the weight of the same size of the eczema and healthy skin.The levels of TNF-αand IL-4 in rats skins were detected by immunohistochemical method.Results:The right back of rats in model group appeared obvious erythema ,erosion and leakage ,which meant the modeling succeeded.Compared with model group ,the swelling degree of Cortex Phellodendri group and Purslane group were significantly decreased ( P<0.01 ) ,compared with Cortex Phellodendri group ,the swelling degree of Purslane group were less than them ( P<0.05 ).The levels of TNF-α, in rats skins of normal control group , model group,Cortex Phellodendri group and Portulaca group were (6 652.66±1 190.94 ),(19 927.10±5 494.21 ), (7 515.13±877.66 ), (6 809.93±1 385.54).The levels of IL-4 were (5 378.44±1 685.01),(26 334.89±3 993.48),(7 814.84±1 751.38),(8 246.57± 975.08 ).Compared with model group ,the levels of TNF-α,IL-4 in rats skins of Cortex Phellodendri group and Purslane group were sig-nificantly decreased(P<0.01).Compared with positive drug group,the levels of TNF-α,IL-4 in rats skins of Purslane group were a little decreased(P<0.05).Conclusion:Purslane extracts has effect to treat atopic eczema ,its possible mechanism is by erducing the proinflammatory factor ,and moderating the anti-inflammatory factor and the ratio between them.
