1.Effects of Ziziphi Spinosae Semen-Albiziae Flos on the IRE1α/ASK1/JNK Pathway of Endoplasmic Reticulum Stress in Hippocampus of Depression Model Rats

Objective Based on the endoplasmic reticulum stress(ERS)inositol-requiring enzyme-1α(IRE1α)/apoptosis signal-regulated kinase 1(ASK1)/c-Jun N-terminal kinase(JNK)pathway to investigate the intervention effect of Ziziphi Spinosae Semen(ZSS)-Albiziae Flos(AF)on the depression model of rats,which were prepared by solitary rearing combined with chronic unpredictable mild stress(CUMS).Methods A total of 144 rats were randomly divided into normal group,model group,high-,medium-and low-dose groups of ZSS-AF,and Venlafaxine group.In addition to the normal group,the rats in other groups were isolated for 21 days combined with CUMS to prepare the depression model.The behavioral changes of rats were observed by open field test and Morris water maze test.The ultrastructural changes of hippocampal neurons were observed by transmission electron microscope.TUNEL staining was used to observe the apoptosis of nerve cells in hippocampus.The protein expression levels of IRE1α,phosphorylated(P)-IRE1α,ASK1,P-ASK1,JNK,P-JNK,B cell lymphoma-2(Bcl-2),Bcl-2 associated X protein(Bax)and cysteme aspartate specific protease-3(Caspase-3)in hippocampus were detected by Western Blot.The mRNA expression levels of IRE1α,ASK1,JNK,Bax,Bcl-2 and Caspase-3 in hippocampus were detected by Real-Time PCR.Results Compared with the normal group,the scores of horizontal movement and vertical movement in the open field test of rats in the model group were decreased(P<0.01).In the water maze test,the escape latency was increased and the number of crossing the original platform was decreased(P<0.01).The number of hippocampal apoptosis was increased(P<0.01).The protein expression levels of P-IRE1 α/IRE1 α,P-ASK1/ASK1,P-JNK/JNK,Bax,Caspase-3 and mRNA expressions of IRE1α,ASK1,JNK,Bax,Caspase-3 in hippocampus were increased,while the protein and mRNA expression levels of Bcl-2 were decreased(P<0.01).Compared with model group,the scores of horizontal movement and vertical movement in the open field test of rats in each dose ZSS-AF groups and Venlafaxine group were increased(P<0.01).In the water maze test,the escape latency was decreased and the times of crossing the original platform was increased(P<0.01).The number of hippocampal apoptosis was decreased(P<0.01).The mRNA expression levels of P-IRE1α/IRE1α,P-ASK1/ASK1,P-JNK/JNK,Bax,Caspase-3 protein and IRE1α,ASK1,JNK,Bax,Caspase-3 in hippocampus were decreased,while the protein and mRNA expression levels of Bcl-2 were increased(P<0.05,P<0.01).The effect of medium-dose ZSS-AF group was better than that of high-and low-dose groups.Conclusion ZSS-AF may play an antidepressant role by regulating IRE1α/ASK1/JNK pathway of endoplasmic reticulum stress.
2.Esophageal squamous cell carcinoma exosomal miR-181b-5p promotes tumor associated macrophage polarization by inhibiting PTEN

Objective:To investigate the effect and mechanism of esophageal squamous cell carcinoma exosomal miR-181b-5p to the polarization of M2 macrophages.Methods:Extracted and identified exosomes from esophageal squamous cell carcinoma,and detected the expression of miR-181b-5p in esophageal squamous cell carcinoma cells and their exosomes by qRT-PCR.M0 type macro-phages were divided into PBS group,HEEC exo group,Eca-109 exo group,miR-NC exo group,miR-181b-5p exo group,miR-NC group,miR-181b-5p mimic group,si-NC group,si-PTEN group,miR-181b-5p exo+PTEN group.qRT-PCR was used to detect the expressions of CD163,CD206,iNOS and TNF-α in each group.The targeting relationship between miR-181b-5p and PTEN were veri-fied by double luciferase reporter gene experiment.Results:miR-181b-5p was significantly overexpressed in esophageal squamous cell carcinoma cells TE-13,TE-12,TE-10,Eca-109,KYSE30 and their exosomes(P<0.001).Compared with miR-NC group,the expression of CD163 and CD206 in cells were significantly upregulated in the miR-181b-5p mimic group,as well as the expressions of iNOS and TNF-α were significantly downregulated(P<0.001).The results of double luciferase reporter genes showed that PTEN was the target gene of miR-181b-5p.Compared with si-NC group,the expressions of CD163 and CD206 in cells were significantly upregu-lated in the si-PTEN group,as well as the expressions of iNOS and TNF-α were significantly downregulated(P<0.001).Compared with PBS group,the expressions of CD163 and CD206 in cells were significantly upregulated in the Eca-109 exo group,as well as the expressions of iNOS and TNF-α were significantly downregulated(P<0.001).Compared with miR-NC exo group,the expressions of CD163 and CD206 in cells were significantly upregulated in the miR-181b-5p exo group,as well as the expressions of iNOS and TNF-α were significantly downregulated(P<0.001).Compared with miR-181b-5p exo group,the expressions of CD163 and CD206 in cells were significantly downregulated in the miR-181b-5p exo+PTEN group,as well as the expressions of iNOS and TNF-α were significantly upregulated(P<0.001).Conclusion:Exosomal miR-181b-5p inhibits PTEN expressions to promote M2 macrophage polarization.
3.Meta-synthesis of qualitative research on the psychological experience of non-suicidal self-injury in adolescents

Objective:To systematically integrate qualitative research on the psychological experience of non-suicidal self-injury (NSSI) in adolescents.Methods:A comprehensive literature search was conducted in databases including China National Knowledge Infrastructure, WanFang data, VIP, China Biology Medicine disc, PubMed, Web of Science, Embase, and Cochrane Library for qualitative studies on the psychological experience of NSSI in adolescents and the search period covered the inception of the databases until June 27, 2023. The methodological quality of the included studies was assessed using the Joanna Briggs Institute Critical Appraisal Checklist for Qualitative Research (2016 edition). The results were integrated using the Meta-synthesis method.Results:A total of 15 studies were included, comprising ten phenomenological studies, one descriptive qualitative study, one mixed-method study, one narrative study, one exploratory study, and one grounded theory study. Thirty-one findings were extracted and synthesized into ten categories, resulting in three main themes: triggers of adolescent NSSI, NSSI as a coping strategy, and the internal conflict and struggle of adolescents engaging in NSSI.Conclusions:Adolescents engaging in NSSI experience complex psychological and sensory responses. Healthcare professionals should closely monitor adolescents' emotional changes, enhance psychological assessments, and guide them in coping with negative emotions effectively to improve their physical and mental health.
4.Security of newborn fathers and its related factors

Objective:To understand the security status of newborn fathers and analyze its related factors.Methods:A total of 256 fathers(aged 22-49 years old)of newborns born within 1 week were select-ed.They were assessed with the self-designed general information questionnaire,Parents'Postnatal Sense of Securi-ty(father version),Emotional Inhibition Scale,and Marriage Satisfaction Subscale of Enrich Marital Inventory.Results:The average score of security was(36.0±4.4).Multiple linear regression analysis showed that the security scores were positively correlated with residence area(β=0.35),education level(β=0.25),preterm birth(β=0.11)and the marital satisfaction scores(β=0.25);and were negatively associated with daily crying time(β=-0.28)and the EIS scores(β=-0.18).Conclusion:The sense of security of postpartum fathers is moderate,and it is associated with residence area,education level,preterm birth,marital satisfaction,daily crying time and emotional inhibition.

Trauma-associated bone infection is one of the most serious infectious diseases after surgery, characterized by high incidence, complex pathogenic microorganisms, therapeutic difficulty and poor prognosis. Therefore, early accurate identification of pathogenic microorganisms and sensitivity of related antibiotics is very important to the success of surgery as well as of local and systemic antibiotic support therapies. As traditional detection methods have such shortcomings as complex items, numerous indicators, slow speed, a low positive rate, and inability to detect rare pathogenic microorganisms, there is an urgent need for better detection methods to identify pathogenic microorganisms of bone infection. In recent years, polymerase chain reaction (PCR) and metagenomic next generation sequencing (mNGS) have been used in clinical diagnosis of a variety of infectious diseases because of their advantages in accuracy, speed, efficiency, and detection range of pathogenic microorganisms over traditional cultures. PCR and mNGS can also be used as effective tools to diagnose trauma-associated bone infection. This review explores the advantages, disadvantages, development trends and guiding significance of these two techniques in the diagnosis of trauma-associated bone infection.

7.Relationship Between the Migration of Endogenous Neural Stem Cells and the Pattern of Change in Immune Cell Phenotypes in the Microenvironment After Intracerebral Hemorrhage in Rats

Objective Intracerebral hemorrhage(ICH),the second most common type of stroke,can cause long-lasting disability in the afflicted patients.The study was conducted to examine the patterns of change in endogenous neural stem cells(eNSCs)and in the regenerative microenvironment after ICH,to observe the relationship between the migration of eNSCs and the pattern of change in the polarization state of immune cells in the microenvironment,and provide a research basis for research on clinical nerve repair.Methods The collagenase injection method was used for modeling.The ICH model was induced in adult female Sprague-Dawley(SD)rats by injecting type Ⅶ collagenase(2 U)into the brain tissue of rats.All the experimental rats weighed 280-300 g.In order to simulate the ICU at different time points,including the acute phase(within 1 week),subacute phase(1-3 weeks),and the chronic phase(over 3 weeks),brain tissues were harvested at 3 day post injection(3 DPI),10 DPI,20 DPI,and 30 DPI to evaluate the modeling effect.Immunofluorescence staining of the brain tissue sections was performed with DCX antibody to observe the pattern of change in the migration of eNSCs in the brain tissue at different time points.Immunofluorescence staining of brain tissue sections was performed with CD206 antibody and CD86 antibody for respective observation of the pattern of change in pro-inflammatory(M1-type)and anti-inflammatory(M2-type)immune cells in the regenerative microenvironment of the brain tissue after ICM.Results Spontaneous ICH was successfully induced by injecting type Ⅶ collagenase into the brain tissue of SD rats.The volume of the hematoma formed started to gradually increase at 3 DPI and reached its maximum at 10 DPI.After that,the hematoma was gradually absorbed and was completely absorbed by 30 DPI.Analysis of the pattern of changes in eNSCs in the brain tissue showed that a small number of eNSCs were activated at 3 DPI,but very soon their number started to decrease.By 10 DPI,eNSCs gradually began to increase.A large number of eNSCs migrated to the hemorrhage site at 20 DPI.Then the number of eNSCs decreased significantly at 30 DPI(P<0.01).Analysis of the immune microenvironment of the brain tissue showed that pro-inflammatory(M1 type)immune cells increased significantly at 10 and 20 DPI(P<0.01)and decreased at 30 DPI.Anti-inflammatory(M2 type)immune cells began to increase gradually at 3 DPI,decreased significantly at 20 DPI(P<0.05),and then showed an increase at 30 DPI.Conclusion After ICH in rats,eNSCs migrating toward the site of ICH first increase and then decrease.The immune microenvironment demonstrates a pattern of change in which inflammation is suppressed at first,then promoted,and finally suppressed again.Inflammation may have a stimulatory effect on the migration of eNSCs,but excessive inflammatory activation has an inhibitory effect on the differentiation and further activation of eNSCs.After ICH,the early stage of repair and protection(10 d)and the subacute phase(20 d)may provide the best opportunities for intervention.
8.Recombinant expression of Japanese encephalitis virus non-structural protein NS1 gene and its reaction with Flavivirus antigen and antibody

Abstract: Objective To elucidate the antigenic antibody reaction of recombinant expression of non-structural protein 1 (NS1) of Japanese encephalitis (JE) virus with various mosquito-borne flaviviruses, including JE virus, and the antigenic antibody reaction of serum samples of patients infected with JE virus in acute stage. Methods In this study, Escherichia coli prokaryotic expression vector (pET) system was used to recombinant express Japanese encephalitis virus NS1 gene. Western Blot assay was performed to detect the antibody responses of the recombinantly expressed protein against a variety of mosquito-transmitted flaviviruses, including JE virus, as well as antigen-antibody reactions of serum from patients with acute JE virus infection. Results The NS1 gene expression product of JE virus (P3 strain) was in the form of an inclusion body, and the denatured and renatured expression product was displayed as a single band in the denatured gel (polyacrylamide gel electrophoresis, PAGE), with a molecular weight of about 45 000. The results of further antigen-antibody analysis showed that the antigen/antibody hybridization reaction of the expression product with polyclonal or monoclonal antibody of JE virus (mosquito isolates, encephalitis isolates) and serum samples of patients with acute JE virus infection could be completely consistent. The recombinant product showed negative antigen/antibody hybridization reactions with mosquito-transmitted flaviviruses, such as dengue virus and yellow fever virus polyclonal antibodies, but positive reactions with polyclonal antibodies to West Nile virus and Murray Valley encephalitis virus. Conclusions In this study, the recombinant expression of the NS1 protein of JE virus was successfully obtained, and the antigen/antibody reaction between the recombinant protein and samples of patients infected with mosquito-borne flavivirus and JE virus was analyzed. The study results provide important basic data for elucidating the antigen-antibody reaction between the NS1 protein of JE virus and mosquito-borne flavivirus. The recombinant expression protein obtained in this study provides an important material basis for further research on the function of JE virus NS1 protein.
9.Analysis of the current situation of defeat in men undergoing IVF-ET therapy and its influencing factors

Objective:To investigate the defeat of men undergoing in vitro fertilization and embryo transfer (IVF-ET) and analyze its influencing factors. Methods:A total of 202 male patients undergoing IVF-ET in Reproductive Medicine Hospital in Changsha from May 2021 to September 2021 were selected by random sampling. General data questionnaire, Chinese version of defeat scale, social support scale, Olson marital quality questionnaire (marital satisfaction, communication), self-rating depression scale, mental resilience scale were used.Results:The defeat score of 202 men in IVF-ET was 16.25±9.93. Marital status, marital quality, education, communication, social support, depression and resilience were the main influencing factors of male defeat after IVF-ET (all P<0.05). Conclusion:Male who have received IVF-ET treatment also have a certain degree of defeat. While paying attention to the psychological status of women who have received IVF-ET, we should also pay attention to the psychological health of the male who have high defeat in IVF-ET treatment and carry out targeted psychological intervention measures to alleviate their defeat, which will be beneficial to the treatment outcome.
