1.Correlation of FRMD6 and CLDN9 expression with clinicopathological features and prognosis in pancreatic cancer patients

Objective To investigate the expression of FERM domain-containing protein 6(FRMD6)and claudin 9(CLDN9)in pancreatic cancer(PC)and their correlation with clinicopathological features and prog-nosis.Methods A total of 95 patients with PC admitted to the hospital from September 2019 to September 2021 were selected as the research objects,and the cancer tissues and adjacent tissues that were resected dur-ing the operation of PC patients were collected.The expressions of FRMD6 and CLDN9 were detected by im-munohistochemistry.Kaplan-Meier survival curve and Log Rank x2 test were used to analyze the survival of PC patients.Multivariate Cox regression analysis was used to analyze the factors affecting the prognosis of PC patients.Results Compared with adjacent tissues,the positive expression rate of FRMD6 in PC tissues was significantly decreased(P<0.05).The proportions of TNM stage Ⅲ and lymph node metastasis in patients with FRMD6 positive expression were significantly lower than those in patients with FRMD6 negative expres-sion(P<0.05).Compared with the adjacent tissues,the positive expression rate of CLDN9 in PC tissues was significantly increased(P<0.05).The proportion of TNM stage Ⅲ and lymph node metastasis in patients with positive expression of CLDN9 was significantly higher than that in patients with negative expression of CLDN9(P<0.05).The 3-year overall survival rate of patients with FRMD6 positive expression(48.00%,12/25)was significantly higher than that of patients with FRMD6 negative expression(8.57%,6/70),and the difference was statistically significant(Log Rank x2=17.944,P<0.001).The 3-year overall survival rate of patients with positive CLDN9 expression(7.35%,5/68)was significantly lower than that of patients with negative CLDN9 expression(48.15%,13/27),and the difference was statistically significant(Log Rank x2=25.159,P<0.001).The positive expression of CLDN9,TNM stage Ⅲ,poor differentiation,and lymph node metastasis were all risk factors for the death of PC patients(P<0.05),and the positive expression of FRMD6 was a protective factor(P<0.05).Conclusion The expression of FRMD6 is low and CLDN9 is high in PC tissues.The expression of FRMD6 and CLDN9 is closely related to the clinicopathological characteristics of PC patients,and they are prognostic factors of PC patients.
2.Application of bionano optical mapping for the diagnosis of a 16p11.2-p12.2 microdeletion.

OBJECTIVE:
To delineate chromosomal aberration caused by structural chromosomal abnormalities with bionano optical mapping.
METHODS:
Chromosomal karyotyping, bionano optical mapping and copy number variation sequencing (CNV-seq) were used to delineate the chromosomal aberration carried by a patient.
RESULTS:
The patient was found to have an anomalous chromosome 16 by karyotyping analysis, which was verified by bionano optical mapping and CNV-seq as loss of heterozygosity at 16p11.2-p12.2.
CONCLUSION
Bionano optical mapping has provided a novel tool for the detection and diagnosis of structural chromosomal aberrations.
3.PCMRI study of cerebral blood flow dynamics in chronic cerebral ischemia

Objective To measure middle cerebral artery (MCA) hemodynamic parameters of chronic cerebral ischemia in middle aged and elderly people with phase contrast magnetic resonance imaging (PCMRI),and to explore cerebral blood flow dynamic pattern in different degrees of chronic cerebral ischemia patients.Methods Middle aged and elderly people underwent conventional MRI scan,and were divided into 12 cases of the normal group,16 cases of mild cerebral ischemia group and 13 cases of obvious cerebral ischemia group.All groups underwent bilateral MCA PCMRI.Bilateral MCA hemodynamic parameters and phase-velocity curve in a heartbeat cycle were obtained by QFLOW analysis software on the workstation for post-processing.Bilateral MCA vascular area,peak velocity (PV),mean velocity (MV) and mean flow (MF) were recorded.The differences of bilateral MCA hemodynamic parameters among the three groups were compared by SPSS17.0 software.Results Bilateral MCA vascular area,MV,MF and right MCA PV among three groups were statistically significant by one-Way ANOVA (P<0.05).Compared with normal group,systolic peak of bilateral MCA phase-velocity curve was low,dull,widen,and the connection between the diastolic peak and systolic peak became flattened in chronic cerebral ischemia patients.Conclusion PCMRI can reflect the differences of the cerebral blood flow dynamic change pattern between different degrees of chronic cerebral ischemia.
4.Analysis of risk factors that affect type 2 diabetic arrhythmia

Objective To investigate risk factors that affect type 2 diabetic cardiac arrhythmia.Methods Clinical data of 292 diabetic patients who were performed examination of low limb vascular were analyzed retrospectively.Based on electrocardiogram,the patients were divided into cardiac arrhythmia group and non-cardiac arrhythmia group with case-control study,and the clinical data and evaluation indexes of lower extremity vascular disease were compared between two groups.Results Data comparison between two groups indicated age [(59 ± 14) years vs (54 ± 12) years],symptoms of low limbs [40.7% vs 29.0%] had statistical significance (P < 0.05).Glycosylated hemoglobin,hypertension history,results of color Doppler ultrasound for lower limbs artery,ankle-brachial index,toe-brachial index,transcutaneous oxygen pressure,heat-controlled laser Doppler between two groups had no statistical significance (P > 0.05).Age,gender,history of high blood pressure,symptoms of low limbs,and heat-controlled laser Doppler were analyzed with multiple logistic regression,which indicated that age was risk factor of type 2 diabetic arrhythmia.Conclusions Age is correlated with type 2 diabetic arrhythmia,and evaluation indexes of lower limb vascular disease have no significant correlation with type 2 diabetic arrhythmia.
