Diabetic nephropathy (DKD), as a common microvascular complication of diabetes mellitus (DM), is a major cause of end-stage renal disease (ESRD). Its clinical manifestations include increased urinary protein excretion, thickening of the glomerular basement membrane, and renal tubulointerstitial fibrosis. The pathogenesis of DKD is complex and involves multiple factors, including disordered glucose metabolism, hemodynamic alterations, and oxidative stress. Although modern medical approaches can alleviate certain symptoms, they still have limitations such as insufficient therapeutic targeting and prominent adverse effects. The transforming growth factor-β/Smad (TGF-β/Smad) signaling pathway is not only a tissue fibrosis pathway that has attracted considerable attention in recent years, but also regulates multiple protein molecules, including the glomerular podocyte slit diaphragm protein Podocin, interleukin-1β (IL-1β), and superoxide dismutase (SOD), thereby participating in various pathological processes and ultimately mediating renal injury. Flavonoid compounds, owing to their sustained pharmacological effects, broad spectrum of action, and high safety profile, have become ideal candidates for targeted therapy research in DKD. Existing studies have shown that these compounds can exert inhibitory effects on renal fibrosis, alleviate inflammatory responses, protect podocytes, and reduce oxidative stress by regulating the interactions between the TGF-β/Smad signaling pathway and the aforementioned protein molecules, thereby maintaining renal structure and function, reducing proteinuria, and significantly improving DKD lesions. This review briefly outlines the composition and functions of the TGF-β/Smad signaling pathway, elucidates the mechanisms by which this pathway regulates DKD, and focuses on summarizing major studies from the past decade on flavonoid-based interventions in DKD through targeted inhibition of the TGF-β/Smad signaling pathway. Furthermore, it discusses the considerable therapeutic potential of flavonoids in the treatment of this disease, aiming to provide a scientific basis for future clinical prevention and treatment of DKD and to promote the development of targeted drugs.

Morphological and molecular identification reached the same result as water mites(Hydrachnidiae, Hydracrina). COI gene sequence showed 94% similarity with the closest species Arrenurus truncatellus. Conlusions Literature review suggests water mites are host-specific parasitism of mosquito species and herein with the first record of Arrenurus sp. parasiting on An. Messeae in the most high-latitude region globally.
3.Evaluation of high-throughput detection technology for ubiquitination signals based on ThUBD.
Chinese Journal of Biotechnology 2025;41(8):3301-3310
Ubiquitination is one of the most widely distributed, structurally diverse, and functionally important post-translational modifications for proteins in eukaryotic cells. At present, the methods for detecting ubiquitination signals mainly include immunological detection based on specific antibodies, mass spectrometry, and detection based on ubiquitin-binding domain (UBD), which together constitute a tool library for studying ubiquitination signals. Our team has previously developed a high-throughput detection technology based on an artificial tandem hybrid ubiquitin-binding domain (ThUBD), which achieves universal and highly sensitive detection of all polyubiquitin chain modification signals. This study aims to evaluate the specificity and range of ThUBD-coated multi-well plates in detecting ubiquitination signals and verify the reliability and practicality of these plates in practical applications. We then used this technology to analyze the complex and diverse ubiquitination signals in different biological samples such as cells, tissues, and urine and detect ubiquitination signals in different mass ranges. The results showed that this technology had strong universality and good specificity, and it can accurately identify ubiquitinated proteins from non-ubiquitinated proteins and achieve accurate quantification. This study provides a sensitive, specific, rapid, and efficient analytical technology for the high-throughput detection of ubiquitination signals.

In the 1950s and 1960s,Japan's implementation of policies prioritizing economic develop-ment caused a lack of effective supervision over the discharge of industrial wastewater and exhaust gases, which led to the occurrence of the"Four Major Pollution Diseases",including Minamata disease, causing serious social and public health problems. To more effectively address public nuisances and pro-vide compensation to victims, the Japanese government gradually established an environmental damage compensation system with administrative relief characteristics since the 1970s.Through long-term prac-tice and system optimization, this system has evolved into a mature institutional framework with a clear division of labor and efficient collaboration. This paper systematically reviews the development process of Japan's environmental damage compensation system and deeply analyzes its legal frame-work and supporting policies, aiming to provide useful references for the construction and improve-ment of China's environmental damage compensation system.Meanwhile, through the case analysis of Minamata disease, the paper explores the specific mechanisms and effects in the compensation practices, further revealing the system's operational characteristics and implications, and providing a reference ba-sis for the construction of China's environmental governance legal system.

Protein-protein interactions play an extremely important role in the biochemical functions of cells, and in-depth analysis of protein interactions is the key to understanding cellular life activities. In this study,we systematically mined the interacting proteins of Golgi protein 73(GP73)using classical immunoprecipitation combined with mass spectrometry, and sought to further analyze the molecular func-tion of GP73.Hepatocellular carcinoma cell line HepG2 was selected, and a stable cell line overexpress-ing GP73-3Flag was constructed using lentiviral infection technology.A total of 78 high-confidence GP73 interacting proteins were identified by immunoprecipitation coupled with mass spectrometry.Bioinformat-ics analyses suggested that GP73 interacted with nearly 40 cytosolic proteins and participated in the bio-logical processes of RNA transport, splicing, and translation.Further immunofluorescence and cytosolic protein isolation experiments confirmed the cytosolic localization of GP73 in a variety of tumor cells.Based on the 78 interacting proteins,we further screened protein interaction networks related to mRNA splicing and verified the existence of interactions between GP73 and seven proteins, including HNRN-PH3,SMN1,RBM14,andNCBP1,by co-immunoprecipitation experiments. In addition, minigene spli-cing assay results indicated that GP73 inhibited the splicing efficiency of pre-mRNA by cells. This study contributes to the expansion of knowledge regarding the function of GP73 and aids in elucidating its criti-cal role in cell biology and its potential association with diseases.

The xylitol dehydrogenase(XDH)is a crucial enzyme involved in the xylose utilization in pentose-catabolizing yeasts and fungi. In addition to producing xylulose,XDH can also be employed to develop a biosensor for monitoring xylitol concentration. In this study, the gene encoding the thermophilic fungus Talaromyces emersonii XDH(TeXDH)was heterologously expressed in Escherichia coli BL21(DE3)at 16 ℃ in the soluble form.Recombinant TeXDH with high purity was purified by using a Ni-NTA affinity column.Size-exclusion chromatography and SDS-PAGE analysis demonstrated that the puri-fied recombinant TeXDH exists as a native trimer with a molecular mass of approximately 116 kD,and is composed of three identical subunits, each with a molecular weight of around 39 kD.The TeXDH strictly preferred NAD+as a coenzyme to NADP+.The optimal temperature and pH of the TeXDH were 40 ℃and 10.0,respectively.After EDTA treatment, the enzyme activity of TeXDH decreased to 43.26%of the initial enzyme activity, while the divalent metal ions Mg2+or Ca2+could recover the enzyme activity of TeXDH,reaching 103.32%and 110.69%of the initial enzyme activity, respectively, making them the optimal divalent metal ion cofactors for TeXDH enzyme. However, the divalent metal ions of Mn2+,Ni2+,Cu2+,Zn2+,Co2+,and Cd2+significantly inhibited the activity of TeXDH.ICP-MS and molecular doc-king studies revealed that 1 mol/L of TeXDH bound 2 mol/L Zn2+ions and 1 mol/L Mg2+ion.Further-more,TeXDH exhibited a high specificity for xylitol, laying the foundation for the development of future xylitol biosensors.
