Objective:To analyze the expression of heat shock protein 90α (HSP90α) in pancreatic cancer tissues and its potential diagnostic value for pancreatic cancer. Methods:A retrospective study was conducted on surgical specimens and clinical data from 99 patients with pancreatic cancer who were treated at Qingdao Municipal Hospital from January 2018 to May 2023, including 58 males and 41 females, with the age of (63.5±23.5) years. Among them, 44 patients (44.4%) were used for pathological examination and prognostic analysis, while 55 patients (55.6%) were tested for plasma HSP90α levels to evaluate its diagnostic efficacy for pancreatic cancer. Blood samples from 119 healthy individuals undergoing routine physical examinations at the same hospital during the same period were collected, including 74 males and 45 females, with the age of (50.5±25.5) years, and plasma HSP90α levels were measured. Immunohistochemistry (IHC) was performed to detect the expression of HSP90α in cancerous and adjacent non-cancerous tissues. Receiver operating characteristic (ROC) curve analysis was used to assess the diagnostic performance of HSP90α, carcinoembryonic antigen (CEA), carbohydrate antigen 19-9 (CA19-9), and carbohydrate antigen 125 (CA125) for pancreatic cancer. Survival analysis was conducted using the Kaplan-Meier method, and the log-rank test was used to compare survival rates. The correlation between HSP90α positive expression in cancer tissues and mutant p53 positive expression was analyzed using Spearman correlation analysis. Results:Immunohistochemical analysis showed that the positive expression rate of HSP90α in pancreatic cancer tissues was 81.8%(36/44), higher than that in adjacent non-cancerous tissues 13.6%(6/44)( χ2=19.82, P<0.01). HSP90α positive expression in pancreatic cancer tissues was positively correlated with mutant p53 positive expression (correlation coefficient was 0.57, P<0.001). The median plasma HSP90α level in the pancreatic cancer group ( n=55) was 83.30 (48.30, 212.00) μg/L, which was significantly higher than that in the normal control group ( n=119), with a median of 37.00 (29.20, 43.50) μg/L, showing a statistically significant difference ( Z=-7.34, P<0.001). The area under the ROC curve (AUC) for plasma HSP90α in diagnosing pancreatic cancer was 0.85 (95% CI: 0.77-0.92), with an optimal cutoff value of 53.52 μg/L, yielding a sensitivity of 69.1% (38/55) and a specificity of 98.3% (117/119). The AUC for HSP90α in diagnosing pancreatic cancer was higher than that of CEA, CA19-9, and CA125. In the immunohistochemical analysis of cancer tissues, the one-year cumulative survival rate of the HSP90α-negative group ( n=8) was 87.5%, which was significantly higher than that of the HSP90α-positive group ( n=36), which was 18.8%( χ2=12.74, P<0.001). Conclusions:HSP90α is highly expressed in pancreatic cancer tissues, which is positively correlated with mutant p53 positive expression. Patients with positive HSP90α expression have a poorer prognosis. HSP90α demonstrates good diagnostic performance for pancreatic cancer and holds potential for clinical application.

Objective:To study the epidemiological characteristics of brucellosis (hereinafter referred to as brucellosis) among non-occupational population in Jinan City, and to provide a basis for formulating further prevention and control measures. Methods:From "the Infectious Disease Monitoring System of the China Center for Disease Control and Prevention", Center for Disease Control and Prevention of Jinan City and its subordinate counties, the epidemiological data and case investigation data of brucellosis in Jinan from 2012 to 2021 were collected and statistically analyzed using descriptive epidemiological methods. Results:From 2012 to 2021, a total of 453 cases of brucellosis were reported in the non-occupational population of Jinan City, accounting for 20.21% (453/2 241) of the total number of cases. The proportion of brucellosis cases in the non-occupational population showed an increasing trend year by year (χ 2tend = 25.50, P < 0.001). Among the non-occupational population, there were 317 males and 136 females, with a gender ratio of 2.33∶1.00; the age was (46.34 ± 17.72) years old, with a minimum age of 1 year and a maximum age of 86 years. The majority of the affected population were farmers, accounting for 71.74% (325/453). The peak onset time was April and May. The main routes of infection were through the digestive tract and respiratory tract, accounting for 36.87% (167/453) and 33.77% (153/453), respectively. The clinical symptoms were mainly fever, muscle soreness, fatigue, and hyperhidrosis. The diagnosis time was 20.0 (7.5, 37.5) d, and the sensitivity of city and district general hospitals to detect and diagnose brucellosis was relatively low. Conclusions:The incidence rate of brucellosis among non-occupational population in Jinan is increasing year by year. The high incidence season is spring, and the high incidence population is male young and middle-aged farmers. Digestive and respiratory transmission are the main transmission routes, with atypical clinical symptoms and a long diagnosis time. The next step is to strengthen the promotion of brucellosis prevention and control knowledge and the training of medical personnel in brucellosis diagnosis.

A triple-transgenic (tyrosine hydroxylase/dopamine decarboxylase/GTP cyclohydrolase 1, TH/DDC/GCH1) bone marrow mesenchymal stem cell line (BMSCs) capable of stably synthesizing dopamine (DA) transmitters were established to provide experimental evidence for the clinical treatment of Parkinson's disease (PD) by using this cell line. The DA-BMSCs cell line that could stably synthesize and secrete DA transmitters was established by using the triple transgenic recombinant lentivirus. The triple transgenes (TH/DDC/GCH1) expression in DA-BMSCs was detected using reverse transcription-polymerase chain reaction (RT-PCR), Western blotting, and immunofluorescence. Moreover, the secretion of DA was tested by enzyme-linked immunosorbent assay (ELISA) and high-performance liquid chromatography (HPLC). Chromosome G-banding analysis was used to detect the genetic stability of DA-BMSCs. Subsequently, the DA-BMSCs were stereotactically transplanted into the right medial forebrain bundle (MFB) of Parkinson's rat models to detect their survival and differentiation in the intracerebral microenvironment of PD rats. Apomorphine (APO)-induced rotation test was used to detect the improvement of motor dysfunction in PD rat models with cell transplantation. The TH, DDC and GCH1 were expressed stably and efficiently in the DA-BMSCs cell line, but not expressed in the normal rat BMSCs. The concentration of DA in the cell culture supernatant of the triple transgenic group (DA-BMSCs) and the LV-TH group was extremely significantly higher than that of the standard BMSCs control group (P < 0.000 1). After passage, DA-BMSCs stably produced DA. Karyotype G-banding analysis showed that the vast majority of DA-BMSCs maintained normal diploid karyotypes (94.5%). Moreover, after 4 weeks of transplantation into the brain of PD rats, DA-BMSCs significantly improved the movement disorder of PD rat models, survived in a large amount in the brain microenvironment, differentiated into TH-positive and GFAP-positive cells, and upregulated the DA level in the injured area of the brain. The triple-transgenic DA-BMSCs cell line that stably produced DA, survived in large numbers, and differentiated in the rat brain was successfully established, laying a foundation for the treatment of PD using engineered culture and transplantation of DA-BMSCs.

Objective:
To investigate the hot spots and epidemiologic characteristics of hand, foot and mouth disease (HFMD) in Jinan municipality from 2009 to 2016.
Methods:
Disease reports of HFMD in Jinan from 2009-2016 were collected and analyzed with ArcGis 10.2 to show the hot spot in different villages and towns, as well as clustering analysis and descriptive epidemiology to show epidemiologic characteristics.
Results:
A total of 89 486 HFMD cases were reported and the reported annual incidence rate was 160.94/100000 during the 7-year period, which increased year by year, and within the whole city, each county was at a higher epidemic level; the curve of incidence is unimodal and the incidence peak occurred mostly between May and August, especially in June; 115 severe cases were reported and the ratio was 0.13%. Of the reported cases, 81.51% were between 1 to 4 years old; 60.36 % were children living scattered. The hot spots were like a circle surrounding the core areas, showing a tendency of increase; the proportion of EV71, CVA16 and other enteroviruses were 33.67%、37.22%and 29.09%, respectively, and they appeared in turn, but severe cases were mostly affected by EV71.
Conclusions
The HFMD in Jinan is in a highly prevalent level, with low ratio of severe cases. Seasonal(high in summer) and unimodal; more common among children between 1 to 4 years old, living scatted and in urban and rural linking areas, with the tendency of increasing of hot spots; prevalent pathogens appear in turn.
