1.Application of flipped classroom in endocrine system integrated course for students from 4+4 clinical medicine pilot class

Objective To evaluate the application of flipped classroom(FC)approach in endocrine system integrated course for students from 4+4 clinical medicine pilot class at Peking Union Medical College(PUMC).Methods The study included the students of 4+4 clinical medicine pilot class grades 2019-2023 in PUMC.The students of grades 2019-2021(n=77)served as the control receiving traditional teaching method,while the students of grades 2022 and 2023(n=76)were selected as the experimental group,which were taught by FC approach.The selected teaching content is thyroid theme.The scoring rates of thyroid related questions in the final exam were as-sessed and a questionnaire survey was conducted to evaluate teaching satisfaction and effectiveness.Results The scoring rates of experimental group were significantly higher as compared to that of control group(P<0.05).Over 90%of the students in the experimental group strongly satisfied or satisfied with the teaching content arrangement,design form,classroom atmosphere,teacher-student interaction of FC and expressed willingness to continue with this methodology.In addition,over 90%of the students strongly agreed or agreed that FC stimulated learning inter-est,improved self-learning ability,strengthened the connection between theory and clinical practice,inspired clini-cal reasoning,enhanced the abilities to analyze and solve problems,and cultivated communication and teamwork skills.Conclusions The application of FC approach in endocrine system integrated course achieved excellent teaching outcomes with high satisfaction of the students.
2.Effects of carboxyamidotriazole on the activation and degranulation of RBL-2H3 cells

Objective To explore the anti-infection mechanism of carboxyamidotriazole (CAI) through studying the effects of CAI on the proliferation, apoptosis and degranulation of RBL-2H3 mass cells.Methods Compound 48/80 (C48/80) was used to induce the model of activation and degranulation in RBL-2H3 cells.The morphological change of cell degranulation was observed by neutral red staining.The release levels of histamine and β-hexosaminidase were measured by ELISA method and chromogenic assay, respectively.The cell activity was determined by CCK-8 method.And cell apoptosis was detected by Hoechst 33342 fluorescent staining.Results Compared with the control group, 10, 20, 40 μmol/L CAI inhibited C48/80-induced degranulation of RBL-2H3 cells in different degrees.CAI (20, 40 μmol/L) reduced the histamine release (P<0.01), and CAI (40 μmol/L) decreased the β-hexosaminidase release (P<0.01).In addition, the viability and apoptosis of RBL-2H3 cells were not affected at the concentrations of CAI used.Conclusions CAI can effectively inhibit the activation and degranulation of RBL-2H3 mast cells, and this effect is not through cytotoxicity.The anti-infection effect of CAI may partially due to the down-regulation of mast cell activity.
3.Inhibitory effect of CAI on iNOS expression and NF-κB activation degradation in rat peritoneal macrophages

Objective This study is designed to explore the regulation of carboxyamidotriazole(CAI)on inflammatory factors in vitro and its underlying mechanisms.Methods Peritoneal macrophages were incubated with different concentrations(5~40 μmol/L),and then cell viabilities were evaluated by MTT assay.Cells were pretreated with CAI(5~40 μmol/L),LPS was then added,and celia were incubated for 18 h.NO and TNF-α levels were determined with Griess reagent and ELISA kit,respectively.The iNOS expression and NF-κB activation were detected by Western blot method.Results The rat peritoneal macrophage viability was not affected at the concentrations of CAI used.CAI(5~40 μmol/L)was found to reduce NO(P<0.01,P<0.001)and TNF-α production(P<0.05,P < 0.01)in a dose-dependent manner.CAI was also found to inhibit the LPS-induced expression of iNOS and degradation of IκBα in rat peritoneal macrophages.Conclusion The findings from the present study suggest that CAI has suppressive effect on iNOS expression,and this inhibitory effect was found to be associated with NF-κB inactivation via the blockade of IκBα phosphorylation.
4.Clinical study of health promotion on the prognosis of patients with acute myocardial infarction combined with diabetes

Objective To study on the effect of health promotion on the prognosis of patients with acute myocardial infarction(AMI)combined with diabetes.Methods 104 patients of AMI complicated with diabetes were:randomly divided into the control and the experimental groups with 52 cases in each group.Two groups were conferred fixed medicine and other clinical interventions as well as programmed education during their hospitalization.After discharge.the experimental group continued to implement health promotion for six months'follow-up.After six months,the fasting blood glucose,postprandial blood glucose,triglyceride,cholesterol,clinic cost,level of knowledge and behavior were compared between the two groups after six months.Results after six months,there was significant difference between two groups in blood glucose,postprandial blood glucose,triglyceride,cholesterol clinic cost,level of knowledge and behavior(P<0.05).Rate of readmission and mortality rate were not stanstically different(P>0.05).Conclusions Health promotion can improve capital clinical index of the patients,decrease the cost and promote the life quality of patients.
