1.Analysis of Alleviating Effect of Calcium Cyanamide on Replanting Problems of Rehmannia glutinosa

ObjectiveTo investigate the alleviating effect of calcium cyanamide (CaCN2) soil fumigation on replanting problems of Rehmannia glutinosa. MethodsNewly soil (NP) was used as the control group, while three treatment groups were established: replanted soil (RP), newly soil treated with CaCN2 (120 g·m², tillage depth 25 cm) (NPCC), and replanted soil treated with CaCN2 (RPCC). R. glutinosa was cultivated in all groups. At harvest, the tuber agronomic traits (number of enlarged roots, maximum root diameter, fresh weight, dry weight) were measured. The content of catalpol and rehmannioside D was quantified by ultra-high-performance liquid chromatography (UPLC) to evaluate medicinal quality. Rhizosphere soil available nutrients and enzyme activities were analyzed by assay kits. The community structure and composition of fungi and bacteria in rhizosphere soil were assessed via internal transcribed spacer 2 (ITS2) sequencing and 16S rDNA sequencing, respectively. ResultsCompared with NP, the RP group showed obviously reduced in tuber agronomic traits and quality indicators (P0.05). However, the RPCC group showed significant improvement in agronomic traits and a notable increase in rehmannioside D content compared to RP (P0.05). The contents of available phosphorus and potassium in RPCC and NP groups were obviously lower than those in RP (P0.05). The polyphenol oxidase soil (S-PPO) activity in RP was obviously lower than in NP (P0.05), while sucrose soil (S-SC), acid phosphatase soil (S-ACP), and S-PPO activities in RPCC were obviously higher than in RP (P0.05). Microbial richness and diversity in RP were obviously higher than in NP (P0.05), whereas no significant differences were observed between the RPCC and NP. The relative abundances of fungal genera Nectria, Myrothecium, Tomentella, and bacterial genus Skermanella were obviousl lower in RPCC and NP than in RP (P0.05). Correlation analysis that S-ACP activity was positively correlated with the content of rehmannioside D (P0.05). Fungal genera Engyodontium and Alternaria, and bacterial genera Pir4 lineage, Pirellula, Methyloversatilis, Brevundimonas, Ralstonia, and Acidibacter were obviously positively correlated with tuber dry weight (P0.05). Conversely, fungal genera Pseudaleuria, Nectria, Haematonectria, Ceratobasidium, and bacterial genera Streptomyces, Skermanella, RB41, Gemmatimonas, and Bacillus were obviously negatively correlated with dry weight (P0.05). The fungal genus Alternaria and bacterial genera Brevundimonas, Ralstonia, Acidibacter, and Dongia showed positive correlations with medicinal quality of R.glutinosa tuber, while fungal genera Pseudaleuria, Nectria, Stachybotrys, Fusarium, Gibberella, Ceratobasidium, and bacterial genera Sphingomonas, Skermanella, RB41, Gemmatimonas, and Bacillus were obviously negatively correlated (P0.05). ConclusionCaCN2 soil fumigation can significantly improve enzyme activities in replanted Rehmannia rhizosphere soil, enhance the utilization of available nutrients, reshape microbial community structure of replanted R.glutinosa at the family and genus level, and notably improve tuber agronomic traits and medicinal quality. This study provides a novel approach to alleviating replanting problems and offers insights for the integrated development of standardized cultivation techniques, including soil disinfection, nutrient-targeted regulation, and microbial inoculant application.

In recent years, with the advancement of nanotechnology, manganese-based nanomaterials have shown potential in various aspects of early breast cancer diagnosis, drug delivery, and tumor treatment. Compared to other nanomaterials, manganese-based nanomaterials exhibit excellent biocompatibility and have become a significant focus in the research of breast cancer diagnosis and treatment.

Glioblastoma (GBM) is the most common and aggressive malignant brain tumor in adults and is poorly controlled. Previous studies have shown that both macrophages and angiogenesis play significant roles in GBM progression, and co-targeting of CSF1R and VEGFR is likely to be an effective strategy for GBM treatment. Therefore, this study developed a novel and selective inhibitor of CSF1R and VEGFR, SYHA1813, possessing potent antitumor activity against GBM. SYHA1813 inhibited VEGFR and CSF1R kinase activities with high potency and selectivity and thus blocked the cell viability of HUVECs and macrophages and exhibited anti-angiogenetic effects both in vitro and in vivo. SYHA1813 also displayed potent in vivo antitumor activity against GBM in immune-competent and immune-deficient mouse models, including temozolomide (TMZ) insensitive tumors. Notably, SYHA1813 could penetrate the blood-brain barrier (BBB) and prolong the survival time of mice bearing intracranial GBM xenografts. Moreover, SYHA1813 treatment resulted in a synergistic antitumor efficacy in combination with the PD-1 antibody. As a clinical proof of concept, SYHA1813 achieved confirmed responses in patients with recurrent GBM in an ongoing first-in-human phase I trial. The data of this study support the rationale for an ongoing phase I clinical study (ChiCTR2100045380).
5.Comparison Between Integrated and Traditional Decoction Pieces of Pinelliae Rhizoma Praeparatum cum Zingibere et Alumine Based on Quality and Pharmacological Effect

ObjectiveTo compare the consistency and difference between the integrated decoction pieces (IDP) and traditional decoction pieces (TDP) of Pinelliae Rhizoma Praeparatum cum Zingibere et Alumine (PRPZA) in the component content and pharmacological effect, so as to explain the rationality of the integrated production of PRPZA. MethodThin layer chromatography (TLC), extract determination, ultraviolet spectrophotometry and high performance liquid chromatography (HPLC) were used to analyze and determine the TLC identification, the contents of water-soluble extract, alum residue, needle-like calcium oxalate crystal, protein, total alkaloids, polysaccharides and three nucleosides (inosine, guanosine and adenosine) of IDP and TDP of PRPZA, and the statistical comparison was made. The anti-inflammatory effect and irritation of IDP and TDP of PRPZA were compared by xylene-induced mouse auricle swelling test and rabbit conjunctival irritation test. ResultCompared with the TDP of PRPZA, the contents of alum residue, needle-like calcium oxalate crystal, protein, polysaccharides, inosine, guanosine and the total amount of three nucleosides in the IDP of PRPZA decreased by 16.95%, 21.27%, 23.78%, 4.74%, 52.12%, 0.24% and 26.04%, the contents of water-soluble extract, total alkaloids and adenosine increased by 7.62%, 114.83% and 125.42%, respectively. IDP and TDP of PRPZA had obvious inhibitory effects on ear edema in mice, but there was no significant difference between them, indicating that the anti-inflammatory effect was consistent. The two decoction pieces of PRPZA had no irritation to the conjunctiva of rabbits, and the difference was not statistically significant between them, suggesting that the safety was similar. ConclusionThere is a certain difference in the component content between the IDP and the TDP of PRPZA, but their anti-inflammatory and irritant effects are similar, and the quality of IDP is slightly better than TDP, which provides a reference for the industrial production and clinical application of the IDP of PRPZA.
6.Rapid Determination of Total Phenylethanoid Glycosides and Total Iridoid Glycosides in Rehmannia glutinosa by Near Infrared Spectroscopy

OBJECTIVE: To establish a method for rapid determination of total phenylethanoid glycosides and total iridoid glycosides in the root of Rehmannia glutinosa. METHODS: The contents of total phenylethanoid glycosides and total iridoid glycosides in medicinal material samples were determined by UV spectrophotometry. Quantitative model of total phenylethanoid glycosides and total iridoid glycosides in medicinal samples was established by NIRS-PLS method. The optimal pretreatment spectra were multivariate scattering correction combined with first derivative method, standard normalization combined with first derivative method. The optimum spectral ranged from 6 703.35-11 065.54 cm-1 and 3 999.63-9 102.36 cm-1. The optimum principal factor number were 10 and 7. RESULTS: The content determination of total phenylethanoid glycosides and total iridoid glycosides in medicinal material samples was proved to meet the requirements by methodological experience. The internal cross validation determination coefficients of total phenylethanoid glycosides and total iridoid glycosides were 0.998 2 and 0.980 9. The correction of root mean square error was 0.032 7 and 0.186 0. The root mean square error of prediction were 0.035 5 and 0.035 1. The root mean square error of cross validation were 0.256 9 and 0.574 3. The predicted values of total phenylethanol glycosides and total iridoid glycosides were 0.268%-1.636% and 3.424%-6.978%, respectively; the determination value of them were 0.299%-1.629% and 3.431%-6.952%, respectively; the absolute deviations were -0.042%-0.067% and -0.111%-0.088%, respectively;the relative deviations were -0.819%-0.076%、-2.257%-1.672%, respectively;There was no statistical significance between predicted values and measured values (P>0.05). CONCLUSIONS: The method is accurate and simple. The method can be used for the rapid determination of total phenylethanoid glycosides and total iridoid glycosides in different germplasms of R. glutinosa.
7.Effects of ADRB2(rs1042713)Gene Polymorphism on Therapeutic Efficacy of Anticholinergic Drugs in the Treatment of Refractory Asthma Pediatric Patients

OBJECTIVE: To study the effects of ADRB2 (rs1042713) gene polymorphism on therapeutic efficacy of anticholinergic drug in the treatment for refractory asthma pediatric patients. METHODS: 171 children with refractory asthma were selected from outpatient department of Kunming Children’s Hospital during Nov. 2016 to Jul. 2019. The distribution of ADRB2 (rs1042713) genotype, the clinical efficacy [asthma control test (C-ACT) score, FEV1, FVC, PEF, maximal mid-expiratory flow (MMEF)] of anticholinergic drug were analyzed statistically; the response of different genotypes to the use of anticholinergic drug were also analyzed statistically. RESULTS: 148 of 171 refractory asthmatics pediatric patients were administered anticholinergic drug, among them 50 of the 71 AA genotype and 36 of the 77 GA genotype responded to anticholinergic drug treatment. Statistical analysis showed that 71 children with AA refractory asthma had improved C-ACT score, FEV1, FVC, PEF and MMEF, there was statistical significance, compared with GA genotype (P<0.05); the response rate of the AA genotype to anticholinergic drugs was 2.71 times that of the GA genotype [OR=2.71, 95%CI (1.38, 5.34), P=0.005]. CONCLUSIONS: The detection of ADRB2 (rs1042713) gene polymorphism has some guiding significance in the treatment of refractory asthma with anticholinergic drugs, and the response of AA genotype is better.
8.Practice of ADRB2, GLCCIl, FCER2 Gene Detection in Individualized Medication of 2 Children with Refractory Asthma

OBJECTIVE: To study the value of ADRB2, GLCCI1, FCER2 gene detection in individualized medication of children with refractory asthma.METHODS: Clinical pharmacists participated in therapy for 2 cases of refractory asthma, and comprehensively analyzed risk factors as its pathogenic factors (allergens and pathogens of respiratory infections), lung function indexes and family history. It was suggested to conduct anti-asthmatic drugs gene [p2-adrenergic receptor (ADRB2), glucocorticoid induced transcriptional 1 gene (GLCCI1), low affinity IgE receptor (FCER2)] testing. According to detection results, the suggestions were put forward such as increasing the dose of Glucocorticoid for inhalation, stopping β2 receptor agonist, additionally using anticholinergic drug. RESULTS: The clinical physicians adopted the suggestions of clinical pharmacists. After optimizing refractory asthma therapy plan according to the results of gene testing and clinical factors, 2 patients were stable and the number of seizures decreased significanthy. CONCLUSIONS: Gene test can provide evidence for the formulation of individualized therapy in asthma children.
9.Investigation of the effects induced by 1 400 W on nasopharyngeal carcinoma cell and the mechanism

Objective To investigate the effect induced by specific inducible nitric oxide synthase (iNOS) inhibitor 1 400 W on nasopharyngeal carcinoma CNE-2 cells lines and its mechanism.Methods CNE-2 cells were treated by different concentrations of 1 400 W,diamminedichloroplatinum (DDP),and both chemicals.Cell counting kit-8 (CCK-8) was used to examine the viability of cells.Quantitative realtime polymerase chain reaction (qRT-PCR) was applied to detect the iNOS mRNA expression.Results The expression of iNOS mRNA was down-regulated by 1400W and was positively correlated with inhibitionrate of cell proliferation.1 400 W inhibits proliferation of CNE-2 cell in a concentration-dependent manner.The proliferation inhibition rate of CNE-2 cells treated by 1 400 W combined with DDP was not enhanced.Conclusions Specific inducible nitric oxide synthase inhibitor 1 400 W can exerts anti-tumor effect though inhibiting the expression of iNOS mRNA;The mechanism of chemosensitization induced by iNOS inhibitor on CNE-2 cells may be closely related level of down-regulation of iNOS expression.
10.The detecting value of CTCs in peripheral venous blood of patients with advanced metastatic breast cancer

Objective To detect the value of circulating tumor cells(CTCs)in peripheral venous blood in patients with metastatic breast cancer. Methods 50 female patients with advanced advanced metastatic breast can-cer hospitalized in our hospital from May 2016 to December 2016 were enrolled in the research. Patients were divid-ed into oligometastases group and extensive metastasis group through multi-department comprehensive analysis andimaging diagnosis. 10 cases with early stage breast cancer were selected randomly. 10 healthy female volunteers were recruited as control group. After obtaining written informed consent from research subjects ,7.5 mL peripheral blood was drew from patients and volunteers prior to starting a new line of chemotherapy ,surgery or other treat-ment. CTCs counts from Blood samples were detected density gradient centrifugation associate with flow cytometry. Results The base line was formulated as CTCs≥5/7.5 mL positive and CTCs<5/7.5 mL negative. By comparing the positive expression of CTCs in early and advanced metastatic breast cancer(Pa = 0.01,P < 0.05),positive CTCs was associated with advanced metastatic breast cancer. Comparision of the positive expression of CTCs between oligometastases group and the extensive transfer group showed significant difference in the CTCs count be-tween the two groups(Pb = 0.018,P < 0.05). In the corresponding period,no positive CTCs was detected in all healthy volunteers. Conclusion CTCs count was associated with metastatic breast cancer. There was a correlation between tumor metastasis and CTCs count (the more widely metastasis ,the higher the detection rate of CTCs). CTCs may be of relevant value in the diagnosis and treatment ,and prognosis evaluation of metastatic breast cancer.
