1.The status,problems,and countermeasures of clinical diagnosis and treatment of Chinese medicine enabled by artificial intelligence

Traditional Chinese medicine has been paid more and more attention in the development of modern healthcare,and its clinical diagnosis and treatment have broad prospects and enormous potential.However,the current traditional Chinese medicine diagnosis and treatment model have serious shortcomings in service capacity and,diagnosis,and treatment effect.The rapid development of big data and artificial intelli-gence technology provides an opportunity for the iterative upgrade of traditional Chinese medicine diagnosis and treatment models.This article reviewed the current situation of artificial intelligence empowering tradi-tional Chinese medicine clinical diagnosis and treatment,clarified the problems and challenges faced by artifi-cial intelligence technology in data integration,data quality,and data analysis in traditional Chinese medicine clinical diagnosis and treatment,and proposed to empower from the aspects of disciplinary integration,data quality optimization,data privacy protection,and promotion and application,so as to provide reference for im-proving the effectiveness of traditional Chinese medicine clinical diagnosis and treatment.
2.Comprehensive evaluation and analysis of laboratory resource allocation in 14 blood stations based on entropy weight -TOPSIS method

【Objective】 To investigate the resource allocation status of blood testing laboratories in 14 blood stations in Gansu Province, explore the impact of differences in basic conditions on the comprehensive testing ability of laboratories, so as to promote the homogenization and standardization of blood screening capacity in blood stations in Gansu and improve blood safety and effectivenes. 【Methods】 An evaluation index system of laboratory resource allocation was constructed and a question-naire was designed. The data of human resources, infrastructure and key equipment of 14 blood stations were collected. The entropy weight -TOPSIS method was used to evaluate and rank the resource allocation of 14 blood stations. 【Results】 In the comprehensive evaluation of blood testing laboratory resource allocation in 14 blood stations in Gansu, the top three were laboratories A, B and I, and the last three were laboratories G, M and J. On the whole, the main issue was unreasonable structure of human resources: most laboratories had unreasonable age structure; except for Laboratory A, there was no personnel with bachelor's degree or above in laboratories; most laboratories had not established a team with intermediate professional titles. In terms of infrastructure, the size of seven laboratories could not meet the needs of modern laboratory testing, and all eight blood stations had no spare nucleic acid laboratories nor a mutual spare laboratory with other blood stations As for the key equipment, 5 laboratories had no automatic blood grouping diagnostic instrument, 5 laboratories only had one set of enzyme immunoassay detection system, 3 laboratories had no spare equipment for the key equipment, which means if the equipment failure could not be repaired in time, the release of results would be affected. 【Conclusion】 There were significant differences in human resources, infrastructure and key equipment of blood testing laboratories in 14 blood stations in Gansu, which had a great impact on laboratory testing capacity and subsequent development. It is suggested that governments at all levels and health administrative departments optimize the input of laboratory resource allocation according to the blood collection volume of blood stations to gradually narrow the differences in resource distribution between different regions, improve the degree of laboratory automation and optimize the personnel structure, so as to build high-quality and efficient blood testing laboratories and ensure the safety of clinical blood use.
3.Analysis of the status quo and influencing factors of self-management ability in young and middle-aged patients with gout

Objective:To explore the status quo of self-management ability in young and middle-aged patients with gout and analyze its influencing factors.Methods:Totally 225 patients with gout admitted in the Department of Rheumatology and the Department of Endocrinology at a ClassⅢ Grade A hospital in Jilin Province from September 2020 to March 2021 were selected by convenient sampling and investigated with the general information questionnaire, Gout Patient Self-Management Assessment Scale, and Medical Coping Modes Questionnaire.Results:The total score of self-management ability in young and middle-aged patients with gout was (127.29±22.42) , including (41.15±7.38) in disease treatment management, (37.41±7.20) in diet management, (27.42±5.62) in lifestyle management, and (21.31±5.06) in psychosocial management. The results of multiple linear regression analysis showed that the education level, course of illness, smoking, facing and avoiding coping styles were the influencing factors of the self-management ability of gout patients ( P<0.05) . Conclusions:The self-management ability of young and middle-aged gout patients is the best in disease treatment management and the worst in psychosocial management in all dimensions, but the overall level is at a medium level and needs to be improved. Clinical medical staff should take targeted health education strategies based on its influencing factors to improve the self-management level of disease in young and middle-aged gout patients.
4.Downregulation of microRNA-188-5p suppresses proliferative and invasive abilities of cutaneous squamous cell carcinoma cells by regulating the PTEN/Akt pathway

Objective:To determine the expression of microRNA-188-5p (miR-188-5p) in cutaneous squamous cell carcinoma (CSCC) tissues and cells, and to assess the effect of its downregulation on the proliferation and invasion of CSCC cells.Methods:From November 2012 to October 2018, 50 surgically resected CSCC tissue specimens and 50 paracancerous normal skin tissue specimens were collected from the First Affiliated Hospital of Xinxiang Medical College in Henan Province. Real-time fluorescence-based quantitative PCR (qPCR) was employed to determine the expression of miR-188-5p in CSCC tissues, paracancerous normal skin tissues, CSCC cell lines SCL-1, A431 and HSC-5, and a human immortalized keratinocyte line HaCaT. Cultured A431 and HSC-5 cells were both divided into 2 groups: miR-188-5p inhibitor group and negative control group, which were transfected with a miR-188-5p inhibitor and its negative control respectively. Then, qPCR was performed to determine the relative expression level of miR-188-5p (expressed as 2 -△△Ct), and cell counting kit-8 (CCK8) and Transwell assays were conducted to assess cellular proliferative activity and invasive ability respectively in the above groups. Dual-luciferase reporter assay was performed to investigate interactions between miR-188-5p and phosphatase and tensin homologue deleted on chromosome 10 (PTEN), and Western blot analysis to determine the protein expression of PTEN, total Akt (t-Akt) and phosphorylated Akt (p-Akt). Two independent samples were compared by using t test. Results:The relative expression level of miR-188-5p was significantly higher in the CSCC tissues (5.213 ± 3.138) than in the paracancerous normal skin tissues (1.010 ± 0.364, t = 9.187, P < 0.001), and significantly higher in the SCL-1, A431 and HSC-5 cells (3.858 ± 0.163, 7.068 ± 0.262 and 4.572 ± 0.413, respectively) than in the HaCaT cells (1.079 ± 0.300, t = 17.890, 21.110 and 8.737, respectively, all P < 0.05). Compared with the negative control group, the miR-188-5p inhibitor group showed significantly decreased miR-188-5p expression in both A431 and HSC-5 cells (both P < 0.01), and decreased proliferative activity and invasive ability of both A431 and HSC-5 cells (all P < 0.05). Dual-luciferase reporter assay showed that the downregulation of miR-188-5p significantly increased the expression of PTEN, but inhibited the expression of p-Akt in A431 and HSC-5 cells. Conclusion:MiR-188-5p is highly expressed in CSCC tissues and cells, and the downregulation of miR-188-5p may inhibit the proliferative activity and invasive ability of CSCC cells by regulating the PTEN/Akt pathway.
5.Expression of Erbin in esophageal squamous cell carcinoma and its relationship with patient prognosis

To explore the significance of Erbin expression in esophageal squamous cell carcinoma (ESCC) and its relation-ship with patient prognosis. Methods: Erbin expression in a tissue chip containing samples from 299 cases were examined using immu-nohistochemistry; in addition, the relationship between Erbin expression and clinical-pathological parameters and patient survival time were also analyzed. Cox regression was used to predict the risk of clinical-pathological parameters. The mRNA and protein expres-sion of Erbin was also determined in 25 cases with paired ESCC and normal tissues through polymerase chain reaction (PCR) and West-ern blot. Results: The expression of Erbin protein and mRNA in ESCC were significantly higher than those of normal esophageal epithe-lium adjacent to cancer (55.2% vs. 0, P<0.05). The high expression of Erbin in ESCC was closely related to TNM stage (64.9% vs. 47.3%, P=0.002) and lymph node metastasis (65.5% vs. 45.0%, P<0.001). Moreover, the high expression of Erbin in ESCC was closely related to poor prognosis (P<0.05). Conclusions: Erbin expression was increased in ESCC and was closely related to poor patient prognosis, which suggested that Erbin may be an important biomarker for the prognosis of cancer patients.
6.Effect of downregulation of microRNA-373 expression on cell cycle and apoptosis of a cutaneous squamous cell carcinoma cell line A431

Objective To evaluate the effect of downregulation of microRNA (miR)-373 expression on cell cycle and apoptosis of a cutaneous squamous cell carcinoma (CSCC) cell line A431.Methods A431 cells at exponential growth phase were classified into 3 groups:miR-373 inhibitor group and negative control group transfected with miR-373 inhibitor and negative control miRNA respectively,and untreated group receiving no treatment.At 48 hours after the transfection,real-time PCR was performed to determine the expression of miR-373 in the above 3 groups,cell counting kit-8 (CCK-8) assay to evaluate the effect of downregulated expression of miR-373 on the proliferation of A431 cells,flow cytometry to investigate the distribution of cell cycle and changes in apoptosis of A431 cells in different treatment groups,and colorimetric analysis to detect the changes in caspase-3 activity in different treatment groups.Statistical analysis was carried out with SPSS 17.0 software by using two-sample t test for the comparison between two groups,one-way analysis of variance (ANOVA) for the comparison among 3 groups,and least significant difference (LSD)-t test for multiple comparisons.Results The expression of miR-373 was significantly lower in the miR-373 inhibitor group (0.120 ± 0.036) than in the untreated group (1.002 ± 0.022) and negative control group (1.037 ± 0.028,LSD-t =36.21,34.83,respectively,both P < 0.001).At 48,72 and 96 hours,the miR-373 inhibitor group showed significantly decreased proliferative activity of A375 cells compared with the untreated group and negative control group (F =10.805,13.720 and 30.907 respectively,P =0.038,0.010 and 0.001 respectively).The proportion of A375 cells in G0/G1 phase was significantly higher in the miR-373 inhibitor group (64.69% ± 1.18%) than in the untreated group (52.74% ± 0.66%,t =15.51,P < 0.001) and negative control group (53.80% ± 0.80%,t =13.24,P < 0.001).The proportion of total apoptotic cells and activity of caspase-3 in the miR-373 inhibitor group were 22.69% ± 1.24% and 1.238 ± 0.057 respectively,which were significantly higher than those in the untreated group (9.62% ± 1.14%,0.413 ± 0.028 respectively,both P < 0.001)and negative control group (9.66% ± 0.97%,0.437 ± 0.036 respectively,both P < 0.001).Conclusion MiR-373 may play an important role in the regulation of cell cycle and induction of apoptosis of the CSCC cell line A431.
7.Expression of microRNA-373 in cutaneous squamous cell carcinoma and its effect on cell invasion

Objective To investigate the expression of microRNA-373 (miR-373) in cutaneous squamous cell carcinoma (CSCC) tissues and cells,and to explore its effects on cell invasion.Methods Real-time PCR was performed to determine the expression of miR-373 in CSCC tissues and paralesional normal skin tissues,as well as in CSCC cell lines (A431 and SCL-1) and HaCaT cells.A431 cells were divided into 4 groups:miR-373 mimic group,miR-373 inhibitor group and negative control group which were transfected with miR-373 mimic,miR-373 inhibitor and negative control miRNA respectively,and untreated group receiving no treatment.Cell invasion assay was performed to evaluate effects of miR-373 downregulation on cell invasion.Western blot analysis was conducted to assess effects of miR-373 downregulation on the protein expression of matrix metalloproteinase-2 (MMP-2) and MMP-9.Results Expression of miR-373 was significantly higher in the CSCC tissues (2.465 ± 0.218) than in the paralesional normal skin tissues (1.000 ± 0.000,P < 0.05),and higher in SCL-1 cells (1.864 ± 0.178) and A431 cells (2.919 ± 0.277) than in HaCaT cells (1.000 ± 0.000,P < 0.05).Most notably,miR-373 expression was also markedly higher in metastatic CSCC tissues than in non-metastatic CSCC tissues (3.323 ± 0.344 vs.1.914 ± 0.161,t =4.158,P =0.000 4).Compared with the untreated group and negative control group,the miR-373 mimic group showed significantly increased miR-373 expression and invasive ability,while the miR-373 inhibitor group showed markedly decreased miR-373 expression and invasive ability (all P < 0.05).Conclusion MiR-373 downregulation can significantly suppress the invasion of A431 cells,and obviously decrease the protein expression of MMP-2 and MMP-9.
8.Delta-catenin protein reduces apoptosis of lung cancer cells via inhibiting the activity of JNK pathway

Background and purpose:As a member of the catenin family, Delta-catenin protein could promote proliferation and invasion of tumor cells, but the accurate mechanism of Delta-catenin promoting cell proliferation is not clear. In the present study, we illustrated that Delta-catenin’s effect on cell apoptosis and their relationship with mitogen-activated protein kinase (MAPK) signaling pathway, and the possible mechanism was also explored for Delta-catenin promoting invasion and proliferation of tumor cells. Methods:The alterations of p38 and c-jun N-terminal rinasel JNK protein activity were detected in SPC and SK lung cancer cell lines with Delta-catenin overexpression or not, by Western blot method. At the same time, the apoptotic number of tumor cells was also examined by FCM method. Furthermore, the number of invasive tumor cells was examined by Matrigel invasive experiment. Results:Compared with untreated group and empty vector group, the activity of p38 protein was unchanged in lung cancer cell lines with Delta-catenin overexpressed (P>0.05), but the activity of JNK protein was decreased signiifcantly (P<0.05),meanwhile, apoptotic proportion of tumor cells were also reduced (P<0.05), and invasive ability of tumor cells was enhanced signiifcantly (P<0.05). Conclusion:Delta-catenin probably decreases apoptosis number of lung cancer cells via inhibiting the activity of JNK pathway, and then promotes invasive ability of tumor cells.
9.Downregulation of DEK induces cell apoptosis via inhibition of NF-κB signaling pathway in gastric carcinoma SGC-7901 cells

[ ABSTRACT] AIM:To investigate the effect of DEK downregulation on the apoptosis of gastric carcinoma SGC-7901 cells, and to explore its associations with NF-κB signaling pathway and apoptosis related proteins.METHODS:SGC-7901 cells with different treatments were divided into 3 groups including untreated group, control siRNA group and DEK siRNA group.The expression of DEK at mRNA and protein levels in the SGC-7901 cells was detected by real-time PCR and Western blot.The cell apoptosis was examined by flow cytometry.Furthermore, the activities of caspase-3 and caspase-9 in the SGC-7901 cells were investigated by Caspase-Glo?-3/9 kit.Finally, the expression of key regulatory pro-tein p65 of NF-κB signaling pathway and apoptosis-related proteins Bcl-2 and Bax in the SGC-7901 cells was investigated by Western blot.RESULTS:Compared with untreated group and control siRNA group, the expression of DEK at mRNA and protein levels was significantly downregulated in DEK siRNA group (P<0.05).In addition, the ratios of early phase apoptosis and total apoptosis in DEK siRNA group were markedly higher than those in untreated group and control siRNA group (P<0.05).Most notably, the decrease in p65 and Bcl-2 proteins, increase in Bax protein and the increases of caspase-3 and caspase-9 activities were observed in DEK siRNA group.CONCLUSION:Downregulation of DEK mediates cell apoptosis of gastric carcinoma may be tightly associated with NF-κB signaling pathway.
10.Clinical and imaging efficacy observation of closed reduction and plaster splint fixation in the treatment of ge-rontal patients with distal radius fractures

Objective To discuss the clinical efficacy of closed reduction and plaster splint fixation ( CRPSF) in the treatment of gerontal patients with distal radius fractures ( DRF) .Methods 76 elderly patients with DRF who treated by CRPSF were selected .According to AO classification of fractures ,the patients were divided into three groups,the A group had 27 cases;B group had 26 cases,C group had 23 cases.The treatment effect was evalua-ted by analyzing the follow-up data,the corresponding imaging measurement parameters and clinical scores .Results All patients had 12-month clinical and imaging follow-up.In the last follow-up, the arm-shoulder-hand dysfunction score and palmar angle ,ulnar deviation angle of A and B group were significantly better than those of C group , the differences were statistically significant(all P<0.05),the difference between A and B group was not statistically sig-nificant (P>0.05).In the last follow-up,the satisfaction score of C group was slightly lower than that of the A or B group,but had no statistically significant difference (P>0.05).Conclusion CRPSF in the treatment of gerontal pa-tients with DRF has good function and imaging effects ,and the improvement level has a certain relationship with the degree of fracture,but has no significant impact on the patients'satisfaction.
