1.Effect of Pricking-Cupping Therapy on Endotoxin-Induced Fever in Rabbits Based on Mitochondrial Energy Metabolism

OBJECTIVE To observe the fever-reducing effect of pricking-cupping method on endotoxin-induced fever in rab-bits,and to further explore the fever-reducing mechanism of pricking-cupping method from the perspective of skeletal muscle mito-chondrial energy metabolism.METHODS A total of 72 rabbits were randomly divided into blank group,model group,Western medi-cine group,pricking group,cupping group and pricking-cupping group,with 12 rabbits in each group.The rabbit model of endotoxin-in-duced fever was reproduced and the corresponding therapeutic intervention was carried out.The body temperature of rabbits was recor-ded continuously for 6 h.The mitochondrial structure was observed by transmission electron microscopy.Oxygraph-2k system was used to detect mitochondrial function.ATP content in thigh muscle tissue of rabbits in each group was detected.The expression of uncoupling protein 2(UCP2)and mitochondrial fusion protein 2(Mfn2)in thigh muscle of rabbits was detected by Wes automated western blot quantitative analysis system.RESULTS The model was successfully replicated,and the body temperature of rabbits in the four treatment groups was lower than that in the model group.The skeletal muscle mitochondrial morphology of rabbits in the blank group was intact,and the mitochondrial morphology was closest to normal after the treatment of pricking-cupping.Moreover,the oxygen con-sumption of mitochondrial CII OXPHOS stage in the pricking-cupping group was significantly higher than that in the model group.Compared with the model group,the ATP content in the muscle tissue of rabbits in the pricking-cupping group was significantly increased,Mfn2 protein expression was significantly increased,and UCP2 protein expression was significantly decreased.CONCLU-SION The pricking-cupping method has a good fever-reducing effect on endotoxin-induced fever in rabbits,and its mechanism may be related to inhibiting UCP2 protein expression in skeletal muscle,promoting Mfn2 protein expression,promoting the oxidative phos-phorylation process in skeletal muscle mitochondria,and increasing ATP synthesis in mitochondria.

3.Effects of total flavone of oldenlandia diffusa on the proliferation and apoptosis of hepatocellular carcinoma stem cell

【Objective】 To investigate the effects of total flavone of oldenlandia diffusa (FOD) on the proliferation and apoptosis of hepatocellular carcinoma (HCC) stem cells sorted from Huh7. 【Methods】 Human HCC cell lines Huh7 was cultured in vitro; CD133 positive (CD133+) stem cells in Huh7 cell line were sorted by flow cytometry, and stem cell markers such as Nanog, Oct4 and Sox2 were tested by Western blotting. CD133+-Huh7 was stimulated by different concentrations (0 μg/mL, 50 μg/mL, 100 μg/mL and 400 μg/mL) of FOD for different time (24 h, 48 h, 72 h and 96 h). CCK8 and plate cell cloning assay were used to detect the effect of FOD on CD133+-Huh7 proliferation while Annexin V-PE/7-AAD was used to detect the effect of FOD on CD133+-Huh7 apoptosis. Western blotting was used to detect the protein expressions of protein 53 (P53), factor associated suicide-Fas-associating protein with a novel death domain (Fas-FADD), B-cell lymphoma-2 (Bcl-2), Cleaved-Caspase3, and Bcl-2 associated X protein (Bax). 【Results】 More than 95% of stem cells were purified for further experiments. Cell proliferation of CD133+-Huh7 was significantly inhibited by FOD, with the significant suppression at the concentration of 100 μg/mL for 72 h compared with negative control group (P<0.05). The apoptosis rate was significantly upregulated than that in the negative control group (P<0.05). The protein expression of Bcl2 decreased while Bax and Cleaved-Caspae3 increased via FAS/FADDD and P53 axis. 【Conclusion】 FOD can significantly inhibit the proliferation and promote the apoptosis of CD133+-Huh7.
4.Effect of miR200a and SIRT1 expression on epithelial cell apoptosis in age-related cataract lens epithelial cells

Objective:To investigate the effects of miR200a and SIRT1 expression on epithelial cell apoptosis in age-related cataract lens epithelial cells.Methods:30 anterior capsule samples of age-related cataract patients were enrolled from the ophthalmology outpatient department of Beichen Hospital affiliated with Nankai University from January to August 2022 as the cataract group and another 30 normal anterior capsule samples of corneal transplantation were enrolled during the same period as the control group. Real -time fluorescence quantitative PCR (RT-qPCR) was used to detect the expression levels of miR200a and SIRT1 genes in the cataract group and control group. The oxidative stress level divided human lens epithelial cells SRA01/04 into a negative control group, miR200a overexpression group, and miR200a inhibition group. The negative control group, miR200a overexpression plasmid, and miR200a inhibitor group were transfected, respectively. Cell apoptosis rate and oxidative stress level were detected by flow cytometry, and Western Blot was used to detect the expression of SIRT1 protein and Caspase-3, Bcl-2, and Bcl-2 related X (Bax) proteins containing cysteine in each group of cells. The targeting relationship between miR200a and SIRT1 was validated using dual luciferase reporter gene detection. Results:Compared with the control group, the expression level of miR200a in the cataract group was significantly increased ( P < 0.01), while the expression level of SIRT1 was significantly reduced ( P < 0.01). Compared with the control group, the levels of SOD, GSH, and GSH-Px in the cataract group were significantly reduced (all P < 0.01), and the MDA level was significantly higher than those in the control group ( P < 0.01). The apoptosis rate of the miR200a overexpression group was significantly higher than that of the negative control group ( P < 0.05), and the apoptosis rate of the miR200a inhibition group was significantly lower than that of the negative control group, the differences were statistically significant ( P < 0.05). The level of SIRT1 protein in the miR200a overexpression group was significantly lower than that in the negative control group ( P < 0.05), and the level of SIRT1 protein in cells of the miR200a inhibition group was significantly higher than that of the negative control group ( P < 0.05). Compared with the negative control group, the miR200a overexpression group showed a significant increase in Caspase-3 activity and Bax protein (all P < 0.05), while the Bcl-2 protein level was significantly reduced ( P < 0.05). The miR200a inhibition group showed a significant decrease in Caspase-3 activity and Bax protein (all P < 0.05), while the Bcl-2 protein level was significantly increased ( P < 0.05). The dual luciferase experiment showed that the overexpression group of miR-200a can significantly inhibit the luciferase activity of the wild-type SIRT1 plasmid, which is the target gene of miR-200a. Conclusions:miR220a is highly expressed in the capsule of lens of patients with age-related cataracts, and its mechanism may be that miR220a promotes apoptosis of lens epithelial cells by targeting the expression of SIRT1.
5.Clinical efficacy of Atezolizumab and Bevacizumab in the treatment of initially borderline resectable advanced liver cancer

Conversion therapy has become the core in the treatment of borderline resectable or unresectable liver cancer, which provides resectable opportunities for more advanced liver cancer patients. In accordance with the first-choice treatment regimen recommended by the guidelines, the authors reported a successful case of Atezolizumab and Bevacizumab (T+A regimen) conversion therapy. The patient with initially borderline resectable advanced liver cancer was performed liver segment resection sucessfully after conversion therapy, and non-tumor recurrence was observed at postoperative 9 months. Postoperative pathological examination showed combined hepatocellular-cholangiocarcinoma, which also indicated the important value of T+A regimen in the conversion therapy of combined hepatocellular-cholangiocarcinoma.
6.The correlation study of left ventricular systolic function calculated by automated cardiac motion quantification

Objective To explore the correlation of left ventricular systolic function calculated by automated cardiac motion quantification (aCMQ) and three-dimensional quantitative analysis (3DQA). Methods According to LVEF by 3DQA,patients were divided into abnormal cardiac function group(LVEF<50%)and normal cardiac function group(LVEF≥50%).Dynamic images from two chamber view(AP2), four chamber view(AP4)and three chamber view(AP3)of left ventricular long axis were acquired from 32 patients with abnormal cardiac function and 119 normal subjects.AP2 longitudinal strain (AP2LS),AP4 longitudinal strain (AP4LS) and AP3 longitudinal strain (AP3LS) as well as the left ventricular global longitudinal strain (LVGLS) were measured by aCMQ. While left ventricular end diastolic volume (LVEDV),left ventricular end systolic volume (LVESV) and LVEF were derived through aCMQ automatically calculated region of interest (ROI) driven automation.The correlation of left ventricular systolic function indexes obtained by aCMQ and 3DQA were compared.Results No significant differences were found between the two groups in gender,body mass index(BMI) and age (P > 0.05).LVEF in abnormal group by 3QDA and aCMQ were much lower than those in normal group (P < 0.01).The absolute values of AP2LS,AP4LS,AP3LS and LVGLS in abnormal group were significantly lower than those in normal group (P < 0.01).LVEF by aCMQ in abnormal group was positively correlated with LVEF by 3QDA methods(r =0.91 ,P <0.01);LVEF by aCMQ in normal group was positively correlated with LVEF by 3QDA (r = 0.73,P < 0.01).The left ventricular global longitudinal strain (LVGLS) measured by aCMQ was negatively correlated with LVEF by 3QDA(r = -0.815,P < 0.01).LVEF measured by aCMQ and 3DQA showed high inter-observer and intra-observer agreements in Bland-Altman charts.Conclusions aCMQ has preferable repeatability.Comparing with the traditional measurement method,LVEF measured by aCMQ has higher correlation with that measured by 3QDA.aCMQ can be a new and relatively accurate method to evaluate the left ventricular systolic function.
7.Clinical value of fetal intelligent navigation echocardiography (5D Heart) in the display of key diagnostic elements in basic fetal echocardiographic views

Objective To evaluate the clinical value of fetal intelligent navigation echocardiography (5D Heart) in the display of key diagnostic elements in basic fetal echocardiographic views.Methods Using volume probe,3D volume datasets of the 209 normal singleton fetuses in the second and third trimesters were acquired from a four-chamber view.After processing the datasets by using 5D Heart,9 cardiac diagnostic planes were acquired and then the image qualities of key diagnosis elements of the heart and great vessels were graded by 3 doctors with different experiences of performing fetal echocardiography.Results Two hundred and sixty-two volume datasets were acquired from 209 normal fetuses,including 53 volume datasets collected when cardiac apex was in different directions,satisfactory datasets failed to be acquired in 27 fetuses because of the maternal obesity,the artifacts of fetal movement,or other influencing factors.Two hundred and thirty-one volume datasets of 182 fetuses could be used for 5D Heart analyzing and displaying,and the displaying rate of 9 standard diagnostic views was up to 88.2%.No significant difference was found when comparing the grading results of one observer in different periods of time,the grading results between two experienced doctors and the grading results among the experienced doctor and one relatively inexperienced doctor (P > 0.05).Conclusions 5D Heart has high reliability and repeatability in obtaining fetal echocardiographic diagnosis views and displaying fetal elemental diagnostic information.
8.Z-scores in evaluation of aortic and pulmonary diameter growth in normal fetuses of second and late trimesters

Objective To explore the application value of Z-scores in evaluating growth of aorta (AO) and pulmonary artery (PA) diameter.Methods AO and PA diameters in 332 normal fetuses were measured twice using fetal echocardiography.Z-scores of AO and PA were obtained based on a simple linear equation using gestational age (GA),biparietal diameter (BPD) and femur length (FL) as independent variables.The difference of Z-scores calculated by twice examinations was evaluated.Results The gestation ages of the first and the second examinations of fetal echocardiography were (25.82± 2.27) weeks (range 18-35 weeks) and (30.61± 2.28) weeks (range 23-39 weeks).There were statistical significant differences of AO and PA diameters between the first and the second examinations (both P<0.05).AO and PA diameters increased with the growth of GA in all fetuses obtained using two examinations,whereas there was no obvious change of Zscores of AO and PA with increasing GA,BPD and FL in individual pregnant woman.No statistical significant difference was found for Z-scores of AO and PA between the first and the second examinations (both P>0.05).Conclusion The growth of AO and PA diameters are positively correlated with the growth of biological parameters in normal fetuses,while Z-scores of AO and PA remain constant in second and late trimesters.

10.Value of fetal intelligent navigation echocardiography (5D heart) and Virtual Intelligent Sonographer Assistance (VIS-Assistance ?) in the display of key diagnostic elements in basic fetal echocardiographic views

Objective To evaluate the value of fetal intelligent navigation echocardiography ( 5D Heart) and Virtual Intelligent Sonographer Assistance ( VIS-Assistance ? ) in the display of key diagnostic elements in basic fetal echocardiographic views . Methods The heart volumes data of 209 normal fetuses were collected and then the offline analysis was conducted . Processing of the built-in VIS-Assistance ? for each diagnosis plane was optimized . The display success rate and image quality difference of diagnosis images and diagnosis elements before and after the VIS-Assistance ? processing as well as the repeatability between observers were analyzed and compared . Results Totally 262 fetal heart volume data were collected from 209 fetuses ,and 231 volume data of 182 cases could be used for the analysis ,with the acquiring success rate of 88 .2% . For the display rate of 8 diagnosis sections of the 231 fetal heart volume data before and after VIS-Assistance ? processing ,abdominal transverse section at stomach level showed no significant difference ,while the other diagnostic sections showed significant differences ( all P < 0 .05) ,and the display rate after the processing was higher than that before the processing . For the image quality scores of diagnosis sections before and after VIS-Assistance ? processing ,abdominal transverse section at stomach level showed no significant differences ,while other diagnostic sections showed significant differences ( all P < 0 .01) ,and the image quality scores after the processing were higher than those before the processing . According to Bland-Altman analysis ,95% credibility interval of the total scores of 8 sections measured by different observers before and after VIS-Assistance ? were ( - 0 .17/ + 0 .18 ) and ( - 0 .17/ + 0 .18 ) , respectively ,6 .06% (14/231) and 3 .89% (9/231) of the points were outside the 95% credibility interval , and the rest had high consistency . Conclusions 5D Heart and VIS-Assistance ? technology can significantly improve the display rate of diagnostic section and image quality of element structures , and has high reliability and repeatability in obtaining fetal cardiac diagnosis sections and displaying fetal elemental diagnostic information .
