1.Detection of vocal fold nodules in children based on multi-band cepstral features of sustained vowels

Objective To propose an effective objective acoustic voice assessment method for detecting vocal fold nodules in children.Methods Continuous vowel sounds/a/from 48 children with vocal fold nodules and 40 healthy chil-dren were analyzed using multi-band cepstral analysis.Thirteen Mel-frequency cepstral coefficients(MFCC),five cepstral peak values,and six cepstral distances were extracted as subband features.An independent sample t-test was conducted to compare the acoustic feature parameters between the two groups.Features with statistically significant differences were fur-ther analyzed using the receiver operating characteristic(ROC)curve to identify the optimal acoustic features with the high-est discriminative power for detecting vocal fold nodule disorders.Results The MFCC2,MFCC3,MFCC5,MFCC11,MFCC12,DQP,EP1,and EP2 feature values were significantly higher in the vocal fold nodule group compared to the nor-mal children group(P<0.05,P<0.001),while MFCC1,MFCC6,MFCC8,MFCC13,and EEP were significantly lower(P<0.05).ROC curve analysis of these features showed that the combined ROC curve area under the curve(AUC)for MFCC1,MFCC2,MFCC3,MFCC5,MFCC6,MFCC8,MFCC11,MFCC12,MFCC13,DQP,EP1,EP2,and EEP was 0.98.The individual AUCs for MFCC1,MFCC2,MFCC3,MFCC5,MFCC6,MFCC8,MFCC11,MFCC12,DQP,and EP2 were all greater than 0.7,indicating a certain level of accuracy.Among these,MFCC2 and MFCC3 had AUCs of 0.85 and 0.87,respectively,indicating that these features have high diagnostic value for vocal fold nodule-related voice seg-ments in children.Conclusion The specific combination of acoustic parameters derived from multiband cepstral features of sustained vowels,including Mel-frequency cepstral coefficients(MFCC1,MFCC2,MFCC3,MFCC5,MFCC6,MFCC8,MFCC11,MFCC12,MFCC13)and cepstral peak values(DQP,EP1,EP2,EEP),can enhance the accuracy of detecting vocal fold nodule-related voice disorders in children.Particularly,MFCC2 and MFCC3 demonstrate high sensitivity and specificity,enabling more accurate identification of vocal fold nodule-related voice disorders in children.

3.The contrast-enhanced T1WI radiomics for predicting pathological grade in rectal adenocarcinoma

Objective To investigate the feasibility of using contrast-enhanced T1WI radiomics in predicting the pathological grade in rectal adenocarcinoma.Methods The MRI and pathological data of 127 patients with rectal adenocarcinoma were analyzed retrospectively.ITK-SNAP software was used to manually draw region of interest(ROI)in rectal cancer on axial T,WI enhanced images.The radiomics features were extracted by the Pyradiomics software from ROI.The task was divided into two parts:task 1("high & non-high"group)predicted the high-differentiation and moderate/low-differentiation of the tumor;task 2("moderate & low"group)predicted the tumor's moderate-differentiation and low-differentiation in"non-high"group.Maximum relevance and minimum redundancy(mRMR)method was used to screen features.The five methods including least absolute shrinkage and selection operator(LASSO),logistic regression(LR),naive Bayes(NB),random forest(RF),and support vector machine(SVM)were used to build the models,and the efficiency of each model was evaluated and compared.Results In task 1,the area under the curve(AUC)of five methods were 0.86,0.90,0.59,1.00,0.99 in the training cohort and 0.71,0.62,0.53,0.67,0.64 in the testing cohort.In task 2,the AUC of five methods in the training cohort were 0.93,0.85,0.67,0.92,0.89,and in the testing cohort were 0.86,0.80,0.50,0.78,0.71.The models constructed by LASSO in both tasks were the dominant models,the AUC of the fusion model in the testing cohort which combined with age,gender and the dominant Radiomics score(Radscore)was 0.80[95%confidence interval(CI)0.63-0.96]in task 1,and the accuracy,sensitivity and specificity were 78.94%,77.78%,and 79.31%respectively.They were 0.89(95%CI 0.74-1.00),90.00%,95.65%,and 71.43%,respectively in task 2.The calibration curves showed that the fusion models had a good goodness of fit.Conclusion Based on the establishment of two dichotomous models,the radiomics based on the contrast-enhanced T1 WI is feasible in predicting the high,moderate and low differentiation degree of rectal adenocarcinoma.

Morita therapy has been bom for more than 100 years.Inpatient Morita therapy is highly oper-able and easy to master.It can improve many refractory neuroses through four-stage treatment.But more neuroses are treated in outpatient clinics,and Morita therapy cannot be used in hospitalized patients.Therefore,the formula-tion of expert opinions on outpatient operations is particularly important.This paper is based on domestic and for-eign references,and after many discussions by domestic Morita therapy experts,and then drew up the first version of the expert opinions on operation of outpatient Morita therapy.Meanwhile the operation rule of Morita therapy in three stages of outpatient treatment was formulated:in the etiological analysis stage,under the theoretical guidance of Morita therapy,analyze the pathogenic factors,to improve treatment compliance and reduce resistance;during the operating stage,guide patients to engage in constructive and meaningful actions,realizing the achievement of letting nature take its course principle;in the cultivating character and enriching life stage,pay attention to positive infor-mation,expanding the scope and content of actions,improving the ability to adapt to complex life,and preventing recurrence caused by insufficient abilities.It will lay a foundation for the promotion of Morita therapy in domestic outpatient clinics,so that more patients with neurosis and other psychological diseases could receive characteristic Morita therapy treatment in outpatient clinics.
5.ABO blood group screening results among blood donors in Guangzhou from 2021 to 2022

【Objective】 To investigate the distribution of ABO and RhD blood groups among voluntary blood donors in Guangzhou, in order to ensure clinical blood safety and better serve blood donors. 【Methods】 Routine ABO and RhD blood group screening tests were carried out among voluntary blood donors from January 2021 to December 2022. The composition ratio of ABO blood group was statistically analyzed. The samples with discrepancy between forward and reverse blood grouping and negative RhD blood group samples were further verified by serological test to analyze the ABO subtypes and the reasons for missed detection. 【Results】 A total of 749 123 blood samples were screened from January 2021 to December 2022, and 513 291 samples were collected after excluding repeat blood donors, with the ABO blood groups as 208 126(40.55%) of O type, 138 859(27.05%) of A type, 130 987(25.52%) of B type and 35 319(6.88%) of AB type. The screening results showed discrepancy between forward and reverse blood grouping in 506 samples, of which 58 were with weak/non-erythrocyte reaction, 16 with erythrocyte reaction, 215 with weak/non-serum reaction, and 217 with serum reaction. Further serological test indicated that 44 samples were ABO subtypes, among which 13 were subtype A, 26 subtype B, 5 subtype AB and 3 B (A) and 14 Bombay-like blood group. Objectives:To determine the expressions of decoy receptors (DcR1 and DcR2) of TRAIL in carcinoma of endometrium. Methods:The expressions of DcR1 and DcR2 in endometrium tissues from 13 carcinoma of endometrium and 7 normal endometrium were detected by immunohistochemical staining.Results: The expressions of DcR1 and DcR2 in carcinoma of endometrium were much lower than in normal endometrium. Conclusions:The decreasing of DcR1 and DcR2 in carcinoma of endometrium may be concerned with its pathogenesis, which may be related to the prevention of endometrium from carcinomatous change.
7.Preparation and storage of coltivirus antigen and its application in detection of coltivirus antibodies in serum from patients.

OBJECTIVETo prepare purified and concentrated coltivirus high titer antigen in order to further detect antibodies against coltivirus in serum sample of patients.
METHODSThe coltivirus in C6/36 cells was cultured and harvested at different time, and the titer was titrated. The virus was purified and concentrated by polyethylene glycol (PEG), and stored at -20 degrees and 4 degrees, with and without glycerol, respectively, then the titer of coltivirus antigen was tested by indirect ELISA. By using the antigen, coltivirus antibodies in serum samples from both suspected Japanese encephalitis (JE) and viral encephalitis (VE) patients were detected.
RESULTSThe highest titer of coltivirus was found at 3-4 weeks of culturing. The antigen titer could be maintained at least for 6 months, especially antigen with glycerol either at 4 degrees or at -30 degrees even for two years. Totally 1141 serum samples from patients diagnosed clinically as JE and VE were tested. The results showed that 130 samples were coltivirus IgM antibody positive and the average positive rate was 11.4% (130/1141). Among 41 samples of paired-serum from patients in Guangzhou Children's Hospital, 9 samples were positive, the positive rate was 22.0% (9/41) in which 5 samples were diagnosed clinically as VE.
CONCLUSIONSStable and purified coltivirus antigen was obtained in order to test coltivirus antibodies as well as development of kits. Coltivirus probably can cause summer-autumn encephalitis in China.

8.Identification of the Peptides that Bind to PTA1 mAb(LeoA1) by Phage Displayed Library

Aim To screen peptides binding specifically to anti-human PTA1mAbs from a random twelve-peptide phage-disp-layed library. Methods Series of PTA1mAbs(LeoA1、 1B11、 C9、 2D1、 2E9、 2G8、 2H2 and E8)were purified using protein-A affinity column. PTA1mAbs which could bind PTA1-Fc fusion protein binding to its ligand were confirmed by flow cytometry, and then used as target to screen phage library. After three rounds of affinity screening, the peptide sequences of positive phage clones were determined and analyzed. Results LeoA1 could block PTA1-Fc fusion protein binding to its ligand. 13 phages which could bind specifically to LeoA1 were isolated from phage library and further confirmed by ELISA. Conserved motifs were found among the sequences of the peptides. Conclusion It was shown that the conserved motifs were candidafe regions binding to PTA1 ligand,which is important to identify functional epitopes for seeking ligand of PTA1 and further investigation of biological function of PTA1.

Objective:To investigate the impact of alanyl-glutamine(Ala-Gln) on outcome in radiation enteritis rats.Methods: Male SD rats(n=70)were separated randomly into four groups: control group(n=10),AR+pseudosurgery group(n=20),AR+TPN group(n=20) and AR+TPN+Ala-Gln group(n=20).Rats were observed for mortality,changs of body weight,villous hight and area,the bcteriral translocation(BT)in mesenteric lymph nodes(MLNs),liver,spleen and peritoneal cavity.Serum TNF-? and sIL-2R level were determined by sandwich ELISA.Results: When Ala-Gln was administered in radiation enteritis rat,the mortality,body weight loss and bacterial translocation were decreaded,the villous hight and area was increased and the TNF-? and sIL-2R levels were reduced.Conclusion: Parenteral Ala-Gln nutrition can improves the results of radiation enteritis rats.
10.Transforming growth factor ? inhibits the cytokine-mediated induction of cytotoxic T cell from murine thymus.

Murine thymocytes can be induced to proliferate and differentiate into cytotoxic effector cells when stimulated with Con A and provided with various cytokines. We have analysed the requirements for a range of cytokines in this system and, i n particular, the role of transforming growth factor ? (TGF-P) in regulating the cellular response to optimal concentration of the cytokines in combination. It was found that Con A+IL-2 (?)ed to marked proliferation and t hi s was augmented with IL-6, TGF-? inhibited the prolifera tion induced by IL-2+IL-6, the presence of TGF-?from the beginning of culture inhibited the development of cytotoxic effector cells and such effects were dose-related and could be reversed with antiserum to TGF-? but not. by a range of cytoki nes including IL-2, IL-4, IL-5, IL-6, TNF-a or ?-IFN. Phenotypic analysis indicated that TGF-? preferentially inhibited the expression of IL-2 receptor and the growth of ly2~+ cells.
