1.Expression and research progress of mucins in ocular surface diseases

Mucins are a class of large, highly glycosylated proteins that play a critical role in maintaining ocular surface homeostasis, tear film stability and the ocular surface barrier. This article provides a systematic review of recent advances in clinical and basic research on mucins, summarizing their role in maintaining ocular surface health and their involvement in the onset and progression of ocular surface diseases. It also reviews current methods for detecting mucins and therapeutic strategies targeting mucins. Particular emphasis is placed on examining changes in the mucin expression profile in different subtypes of dry eye, ocular surface diseases and following ocular surface surgery, with the aim of providing guidance for the diagnosis, assessment and treatment of mucin-related ocular surface diseases.
2.Advances in the treatment of primary membranous nephropathy

Primary membranous nephropathy (PMN) is one of the common causes of nephrotic syndrome in adults and is characterized by the deposition of immune complexes, resulting in thickening of basement membrane. With the identification of more autoantibodies, the understanding of the pathogenesis of PMN has expanded. Currently, supportive therapy and immunosuppressive therapy are the primary treatments for PMN, which mainly consists of cyclophosphamide, calcineurin inhibitors, rituximab, and so on. With the advent and application of an increasing number of biological agents, a subset of refractory or drug-resistant PMN patients can be effectively treated and achieve remission. This article reviews recent advances in the management of PMN. It covers pretreatment assessment, immunosuppressive therapy, complement- targeted therapy, and anticoagulant therapy and deliberates on the management of rituximab- resistant patients. The review aims to provide clinicians with an up-to-date basis for individualized clinical decision-making.
3.Preparation of SpaD polyclonal antibody against Corynebacterium striatum and its inhibitory effect on biofilm formation ability of Corynebacterium striatum strains with strong biofilm production abilities

Objective:To prepare the polyclonal antibody against pilus assembly protein(SpaD),a biofilm formation related protein of Corynebacterium striatum(C.striatum),and to evaluate its inhibitory effect on biofilm formation by strong biofilm-producing strains of C.striatum along with its potential application value.Methods:A total of 117 strains of C.striatum isolated from clinical specimens of hospitalized patients at Affiliated Hospital of Inner Mongolia Medical University from 2011 to 2021 were selected as the study subjects.The biofilm formation ability of each strain was detected by crystal violet staining assay.Real-time fluorescence quantitative PCR(RT-qPCR)method was used to examine the distribution of SpaD protein encoding gene spaD in strong biofilm-producing strains of C.striatum.The strong biofilm-producing strains of C.striatum were divided into control group,and proteinase K group,and cystol violet staning method was used to detect the inhibitory effect of proteinaskon the biofilm formation abilitys of the strong biofilm-producing strains of C.striatum.The recombinant SpaD protein was constructed using protein recombination technology and the was divided into control group and 5 and 10 mg·L-1 SpaD recombinant proterin groups,and cystol violet staning method was used to detect the inhibitory effect of SpaD recombinant protein.The rabbit anti-SpaD polyclonal antibodies were subsequently obtained through animal immunization,the experiment was divided into control group and 1∶400,1∶200,and 1∶100 SpaD polyclonal antibody groups,the inhibitory effect of SpaD polyclonal antibodies on biofilm and crystal violet staining method was used to detect abilities of strong biofilm-producing strains of C.striatum.Results:The Crystal violet staining results revealed that strong,moderate,and weak biofilm-producing strains accounted for 47.9%(56/117),29.0%(34/117),and 23.1%(27/117),respectively.The RT-qPCR results showed that all the strong biofilm-producing strains carried the spaD gene.Compared with control group,the biofilm formation abilities of 13 strong biofilm-producing C.striatum strains in both 5 and 10 mg·L-1 SpaD recombinant protein groups were significantly decreased(P<0.05).Compared with control group,the biofilm formation abilities in 61.5%(8/13)and 7.7%(1/13)of strong biofilm-producing C.striatum strains in 1∶100 and 1∶200 SpaD polyclonal antibody groups were decreased,respectively(P<0.05).Conclusions:The spaD gene is highly expressed in strong biofilm-producing clinical strains of C.striatum.Anti-SpaD polyclonal antibodies significantly inhibits biofilm formation in these clinical isolates,demonstrating a inhibitory effect as a manner of concentration-dependent.SpaD can be a promising novel target for therapeutic intervention against biofilm-producing C.striatum infections.

5.Association between incidence of fatal intracerebral hemorrhagic stroke and fine particulate air pollution.

OBJECTIVE:
Few studies investigating associations between fine particulate air pollution and hemorrhagic stroke have considered subtypes. Additionally, less is known about the modification of such association by factors measured at the individual level. We aimed to investigate the risk of fatal intracerebral hemorrhage (ICH) incidence in case of PM (particles ≤ 2.5 μm in aerodynamic diameter) exposure.
METHODS:
Data on incidence of fatal ICH from 1 June 2012 to 31 May 2014 were extracted from the acute stroke mortality database in Shanghai Municipal Center for Disease Control and Prevention (SCDC). We used the time-stratified case-crossover approach to assess the association between daily concentrations of PM and fatal ICH incidence in Shanghai, China.
RESULTS:
A total of 5286 fatal ICH cases occurred during our study period. The averaged concentration of PM was 77.45 μg/m. The incidence of fatal ICH was significantly associated with PM concentration. Substantial differences were observed among subjects with diabetes compared with those without; following the increase of PM in lag2, the OR (95% CI) for subjects with diabetes was 1.26 (1.09-1.46) versus 1.05 (0.98-1.12) for those without. We did not find evidence of effect modification by hypertension and cigarette smoking.
CONCLUSIONS
Fatal ICH incidence was associated with PM exposure. Our results also suggested that diabetes may increase the risk for ICH incidence in relation to PM.

6.Content and Dissolution Determination of Berberine Hydrochloride in Fengshi Qutong Capsule by HPLC

OBJECTIVE:To establish a method for content and dissolution determination of berberine hydrochloride in Fengshi qutong capsule. METHODS:HPLC was conducted to determine the content of berberine hydrochloride in the preparation:the column was Waters Xbridge C18 with mobile phase of acetonitrile-0.05 mol/L sodium hydrogen(45:55,V/V)at a flow rate of 0.5 ml/min,de-tection wavelength was 345 nm,column temperature was 30 ℃,and the injection volume was 10 μl. Rotating basket method was used to determine the dissolution of berberine hydrochloride. RESULTS:The linear range of berberine hydrochloride was 1.06-74.2 μg/ml(r=0.9999);RSDs of precision, stability and reproducibility tests were lower than 1%;recovery was 96.8%-100.6%(RSD=1.6%,n=9). The dissolution of samples in 30 min was higher than 80%. CONCLUSIONS:The method is simple and stable with good reproducibility,and can be used for content and dissolution determination of berberine hydrochloride in Fengshi qutong capsule.
