Cysteine protease inhibitor SN(Cystatin SN , CST1), a member of the type 2 cystatins superfamily, regulates tumor biological activities by modulating cysteine protease activity. CST1 is abnormally expressed in digestive system neoplasms, such as esophageal cancer, gastric cancer, hepatocellular carcinoma, colorectal cancer, and pancreatic cancer. CST1 influences tumor cell proliferation, migration, and epithelial-mesenchymal transition (EMT) through signaling pathways including Wnt, STAT3, and PI3K-AKT. Its expression levels significantly correlated with tumor stage, metastasis, and patient prognosis, suggesting its potiential as a novel biomarker for early diagnosis, therapeutic evaluation, and prognosis in digestive system neoplasms. This article systematically reviews the molecular characteristics of CST1, its mechanisms of action in digestive system neoplasms, clinical significance, and explores its potential as a therapeutic target.

Objective To explore the impactof Lysosome-Associated Membrane Protein 3(LAMP3)on theproliferation, migration and angiogenesis of PC-3 cells. Methods LAMP3 expression in normal prostate epithelial cells and prostate cancer bone metastasis cells was detected using western blot and RT-PCR.Stable LAMP3-silenced PC-3 cells were constructed, and the effects of LAMP3 on proliferation, invasion, and migration of PC-3 cells were assessed using CCK8,scratch assay, and transwell assay, respectively.ELISA and angiogenesis assays were employed to examine the expression of VEGF and MMP9,as well as angiogenesis of HUVEC cells induced by PC-3 cells. Finally,WB and RT-PCR were used to detect the expression of VEGF,AKT/p-AKT.Results Our findings showed that the expression level of LAMP3 was significantly higherin prostate cellsthan in normal prostate epithelial cells, especially in PC-3 cells(P<0.05).We also found that silencing LAMP3 could inhibit the proliferation, migration and invasion of PC-3 cells, along with the expression of VEGF and MMP9 and the PC-3 cells-induced angiogenesis, and these results were statistically significant(P<0.05).Furthermore,LAMP3 downregulated the expression of VEGF and AKT/p-AKT in PC-3 cells. Conclusion LAMP3 can affect the proliferation, migrationand angiogenesis of PC-3 cells through the regulation of VEGF/AKT pathway.Thus,LAMP3 might be a potential thera-peutic target for prostate cancer bone metastasis.

Objective To investigate the effect of chronic psychological stress on testicular cell senescence in rats and the role of glucocorticoid receptor(GR)in the process. Methods Thirty-two male SD rats(4~6 weeks old, weighing 120~140 g)were randomly assigned into dimethyl sulfoxide(DMSO)group,mifepristone(RU486)group,chronic unpredictable mild stress(CUMS)group, and CUMS+RU486 group.DMSO of 0.5 ml/(kg·d)was injected subcutaneously into the rats of DMSO group. After the rats of the CUMS group were exposed to CUMS,mifepristone of 1 mg/(kg·d)was given to the rats of the RU486 group and CUMS+RU486 group. All the above 4 groups were inflicted for 42 consecutive days. After modeling, the psychological stress status of rats was evaluated with animal behavioral experiments.Computer-aided sperm analysis(CASA)system was used to detect sperm density.ELISA was employed to measure the serum levels of corticosterone(CORT),sex hormones and inflammation factors. The contents of malondialdehyde(MDA),H2O2,NAD+/NADH and ATP in the testicular tissues were tested with corresponding reagent kits.Western blotting was utilized to detect protein levels of[3-gal,p16,and GR in testicular tissue, and transmission electron microscopy was used to observe mitochondrial structure of spermatogenic cells. Results Compared with the DMSO group, the sperm density, serum levels of testosterone(T),luteinizing hormone(LH)and follicle stimulating hormone(FSH)were decreased while those of CORT and estradiol(E2)were increased in the CUMS group(all P<0.001).Serum pro-inflammatory factors IL-6 and TNF-α concentrations were increased while anti-inflammatory factor IL-10 concentration was decreased in the CUMS group than the DMSO group(all P<0.05).And,ATP content and NAD+/NADH ratio were decreased while MDA and H2O2 levels were elevated in testicular tissues in the CUMS group(all P<0.05).Moreover,the protein levels of testicular cell aging markers β-gal,p16 and GR were significantly enhanced in the CUMS group(all P<0.01),accompanied by swelling and vacuolization of mitochondrial structures in spermatogenic cells.RU486 treatment effectively improved sperm density and increased CORT level and down-regulated the testicularβ-gal,p16 and GR protein level(all P<0.05).Conclusion Chronic psychological stress induces testicular damage in rats, presenting declines in sperm density and CORT level, which is possibly associated with GR-mediated cell senescence.

Objective To explore a new microsurgical method for reconstruction of finger pulp defect. Methods From May 2008 to May 2009,10 male patients with finger pulp defect were treated in our hospital, aged from 18 to 38 years (average,26 years). The index finger was involved in 6 patients, the middle finger in 3 patients and the ring finger in 1 patient. All finger pulp defects were caused by machine injury. The defect sizes ranged from 1.5 cm×2.0 cm to 2.0 cm×3.0 cm.Six patients suffered from single skin defect, and 4 patinets suffered from skin defect combined with nail bed laceration and distal phalangeal fractures. All patients were performed emergency operations. The defects were reconstructed by using arterialized venous flap with microsurgical suture of the dorsal branch of the proper digital nerve. The fractures were fixed by Kirschner wires. The donor area was covered with skin grafts. Results All flaps survived completely. The fractures healed 8 to 10 weeks postoperatively. All patients were followed up for 4 to 6 months, all flaps presented satisfactory appearance, normal texture, with no pigmentation at the last followup. The static 2-point discrimination of the flaps ranged from 8 to 12mm.All injured fingers obtained good recovery of flexion and extension of the distal interphalangeal joints. The nails of the fingers with laceration of nail bed grew smoothly. The nail bed with laceration grew smoothly, and some new nails could be seen. The skin grafts applied to the donor area survived completely. Conclusion The arterialized venous flap with suture of the dorsal branch of the proper digital nerve is a good method for reconstruction of finger pulp defect, which had the following advantages:slight donor injury, low anesthesia risk, simple operative technique, and satisfactory postoperative function and appearance.
