1.Research progress on the mechanisms of traditional Chinese medicine in improving premature ovarian insufficiency

Premature ovarian insufficiency (POI) is a reproductive endocrine disorder that severely affects female reproductive health. However, there remains a lack of clinically effective and safe therapeutic options to restore ovarian function. Traditional Chinese medicine (TCM), with its advantages of multi-component and multi-target holistic regulation, has shown unique potential in the prevention and treatment of POI. Among TCM monomers, curcumin, resveratrol, and icariin exert antioxidant, anti-inflammatory, and anti-ovarian granulosa cell apoptosis effects by regulating signaling pathways such as nuclear factor-erythroid 2-related factor 2/heme oxygenase-1, phosphoinositide 3-kinase/protein kinase B, and mitogen-activated protein kinase. Single TCM herbs, including American ginseng, Cuscutae Semen, and Cistanches Herba, can regulate sex hormone levels, and alleviate ovarian oxidative stress and immune damage. TCM compound formulas such as Yijing decoction, Siwu decoction, Zuogui pill, and Kuntai capsule modulate multiple signaling pathways, including vascular endothelial growth factor/vascular endothelial growth factor receptor-2, phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin, and silence information regulator 1/p53, thereby promoting angiogenesis, inhibiting mitochondrial apoptosis, and regulating autophagy and ferroptosis, which ultimately delay follicular atresia and improve ovarian reserve function. Further studies are suggested to apply artificial intelligence to deeply explore the underlying mechanisms and carry out high-quality clinical trials, so as to promote the standardized application of TCM in the prevention and treatment of POI.
2.Association analysis of endometrial microbiome characteristics in RIF patients with subsequent transplantation outcomes

Objective To explore the correlation between the characteristics of the endometrial microbiome in patients with recurrent implantation failure(RIF)and the subsequent transplantation outcomes.Methods A total of 438 RIF patients underwent embryo transfer again in our hospital were retrospective selection.According to the pregnancy status of the patients 8 weeks after embryo transfer,patients were divided into the pregnancy group(n=85)and the non-pregnancy group(n=353).The clinical data,characteristics and composition diversity of the endometrial microbiome were compared between the two groups.Binary Logistic regression was used to analyze the influencing factors,and the receiver operating characteristic(ROC)was employed to predict the efficacy of the influencing factors.The interaction between the above-mentioned risk factors and Shannon index in the subsequent transplantation outcomes of RIF patients was further analyzed.Results The levels of basal estradiol(E2),fasting insulin(Fins),total cholesterol(TC)and triglycerides(TG)were significantly higher in the non-pregnant group than those in the pregnant group(P<0.05).Meanwhile,there were significant differences in the composition of endometrial microorganisms between the two groups(P<0.05).Among them,the abundance of Fusobacterium phylum,the abundance of Bacillus genus and the α-diversity(Chao1,Shannon,Simpson)index all showed significant differences(all P<0.05).Binary Logistic regression multivariate analysis showed that the abundance of Fusobacterium phylum increased(OR=1.628,95%CI:1.416-1.841),the abundance of Bacillus genus decreased(OR=0.725,95%CI:0.557-0.934)and E2 increased(OR=1.654,95%CI:1.343-1.965).The elevated insulin(OR=1.691,95%CI:1.393-1.980)and decreased Shannon index(OR=0.388,95%CI:0.075-0.697)were independent risk factors for failure after subsequent transplantation.ROC curve analysis showed that the area under the curve(AUC)of the Shannon index was 0.836(95%CI:0.782-0.890),with the highest predictive efficacy.There was significant interaction among the subgroups of Fusobacteria,Bacillus genus,E2 and insulin in Shannon index on the subsequent transplantation outcomes of patients with RIF.Conclusion The independent risk factors for subsequent transplantation failure in RIF patients can be used as sensitive indicators to predict the subsequent transplantation outcomes of RIF patients,and Shannon index has a higher clinical predictive value.

4.IMMUNOHISTOCHEMICAL STUDY ON GLIAL CELL LINE-DERIVED NEUROTROPHIC FACTOR IN THE LUMBAR SEGMENT OF THE SPINAL CORD IN POSTNATAL RATS

In order to explore the effect of glial cell line derived neurotrophic factor (GDNF) on the motor neurons of the spinal cord, the present study investigated the localization of GDNF in the lumbar segment of the spinal cord on postnatal 1、3、7、10、14 and 28 days in rats with immunohistochemical staining method. The results showed that the positive GDNF immunoreactivity was present in the motor neurons of the ventral horn on postnatal 1、7 and 28 days, the motor neurons on postnatal 3 days were also weakly stained in rats,and some neurons of the dorsal horn and some glial cells had positive immunoreactivity. Furthermore, GDNF was found in the glial cell of the dorsal root in all groups of the postnatal spinal cord. The present results suggested that GDNF might play important roles in the development and function of the motor neurons of the spinal cord. More experiments should be done to explain why there is no positive immunoreactivity in the motor neurons between postnatal 7 and 28 days.
