1.An improved method for the establishment of an autistic mouse model with down-regulation of PTEN gene

Objective :
To establish a PTEN gene-downregulated ASD juvenile rat model (VPA-ADV) through combined application of valproic acid (VPA) and PTEN adenovirus (ADV),then to compare the newly constructed animal model with two traditional autistic animal models of VPA and AD,and ultimately to prove the advantages of this model in animal model establishment of acupuncture treatment for ASD.
Methods :
Wista rats at 12.5 days of gestation were randomly divided into 2 groups.VPA rats were given 600 mg/kg of normal saline (NS) intraperitoneally.Weight,eye opening time and tail deformity were recorded.The newborn mice in NS group were randomly divided into three groups (10 rats in each group):normal (NS) group,virus (ADV) group and virus-negative control (ADV-NC) group;VPA group (20 young rats) was randomly divided into 2 groups (10 rats in each group):valproic acid (VPA) group and valproic acid-binding virus interference (VPA+ADV) group.The body weight,tail length,curved tail,geotaxis text time and skeletal deformity of 5 groups of young rats after birth,the neurobehavioral behavior of 21-day-old rats,and the myelin structure of brain tissue under electron microscope were observed,the expression levels of PTEN,PI3K,AKT,GSK3β and MBP were detected by immunohistochemistry,Western blot and q-PCR.
Results:
Compared with the NS group,the VPA group had significantly increased malformation rate,slow weight gain,slow tail length growth,and long negative geotaxis reflex time (P<0.05).Compared with the NS group,the weight gain,tail length growth and negative geotaxis reflex time of the three model groups were slower (P<0.05),and the performance of VPA+ADV model was the most significant.There were significant differences in the time of crossing the central grid,the number of crossing the edge grid,and the time of crossing the edge grid in the open field test between the three groups and the NS group (P<0.05).In the selfgrooming experiment,the number of interactions in VPA-ADV model was the least (P<0.05),and the number of digging and self-grooming was the most (P<0.05).In the three-box social experiment,VPA-ADV model had the shortest average number of entries into the social box and the shortest residence time (P<0.05),the time of finding the platform in the water maze experiment was the longest,and the number of times crossing the third quadrant was the least (P<0.05).The structure of the myelin sheath layer in the corpus callosum was observed by transmission electron microscopy.The structure of the NS group was clear and complete.Compared with the NS group,the myelin structure of the ADV-NC group was similar,and the myelin structure of the three model groups was stratified and broken,and there were pathological changes and myelin damage in the ASD.Among them,the myelin sheath of the VPA-ADV model was thickened,stratified,and severe visible disintegration.The results of immunohistochemistry,Western blot and q-PCR showed that the expression of PTEN in VPA+ADV model was down-regulated by about 50%,which was the most obvious.The expression of AKT and MBP increased,and the expression of GSK3β decreased (P<0.05).However,the results of q-PCR showed that the expression of PI3K-mRNA in the three model groups significantly increased (P<0.05),and the change of VPA+ADV model was the most significant.
Conclusion
The novel PTEN-ASD animal model established by VPA+ADV method is observed to have significant pathological changes that are typical of the manifestation of ASD myelin dysplasia and is determined to have better results than the two traditional autistic animal models.
2.IC4R-2.0:Rice Genome Reannotation Using Massive RNA-seq Data

Genome reannotation aims for complete and accurate characterization of gene models and thus is of critical significance for in-depth exploration of gene function. Although the availability of massive RNA-seq data provides great opportunities for gene model refinement, few efforts have been made to adopt these precious data in rice genome reannotation. Here we reannotate the rice (Oryza sativa L. ssp. japonica) genome based on integration of large-scale RNA-seq data and release a new annotation system IC4R-2.0. In general, IC4R-2.0 significantly improves the completeness of gene structure, identifies a number of novel genes, and integrates a variety of functional annota-tions. Furthermore, long non-coding RNAs (lncRNAs) and circular RNAs (circRNAs) are system-atically characterized in the rice genome. Performance evaluation shows that compared to previous annotation systems, IC4R-2.0 achieves higher integrity and quality, primarily attributable to mas-sive RNA-seq data applied in genome annotation. Consequently, we incorporate the improvedannotations into the Information Commons for Rice (IC4R), a database integrating multiple omics data of rice, and accordingly update IC4R by providing more user-friendly web interfaces and implementing a series of practical online tools. Together, the updated IC4R, which is equipped with the improved annotations, bears great promise for comparative and functional genomic studies in rice and other monocotyledonous species. 3.Expression of interleukin-33 and vascular endothelial growth factor-C in gastric cancer and its clinical significance

Objective To investigate the expression of interleukin-33(IL-33)and vascular endothelial growth factor C(VEGF-C)in gastric cancer tissues and serum,and to explore the relationship between these two indicators and gastric cancer lymph node metastasis.Methods The levels of IL-33 and VEGF-C in the tissues of gastric mucosa and serum were detected by immunohistochemical SP method and enzyme-linked immunosorbent assay(ELISA)in 98 patients with gastric cancer and 36 healthy subjects.Results The expression rates of IL-33 and VEGF-C in gastric cancer were 67.35%and 74.49%,which were significantly higher than the rates in normal gastric tissue(47.22%and 61.11%).The difference was statistically significant(P<0.01).The expression of IL-33 and VEGF-C was correlated with the degree of tumor differentiation,tissue infiltration,lymph node metastasis,distant metastasis and clinical stage(P<0.05).The positive rates of IL-33 and VEGF-C in gastric cancer lymph node metastasis group were higher than those in non-lymph node metastasis group(P<0.05).The serum concentrations of IL-33 and VEGF-C in patients with gastric cancer were(50.24±13.08)pg/mL and(210.73±58.35)pg/mL,respectively,which were higher than those in healthy control group(P<0.05);the expressions of serum concentration of IL-33 and VEGF-C in the cases with lymph node metastasis were higher than those without lymph node metastasis and the difference was statistically significant(P<0.05).Conclusion High levels of IL-33 in gastric carcinoma patients might induce the secretion of VEGF-C,promote lymph node metastasis,and be applied as an important index of the appraisal to the prognosis of gastric cancer.
4.Protective action of Semen Ziziphi Spinosaeand on acute myocardial ischemia

Objective To study the protective action of Semen Ziziphi Spiosaeand on acute myocardial ischemia and its effecte on infarcted area. Methods Left anterior descending(LAD) of coronary artery was ligated to cause the big rat's acute myocardial ischemia, and electrocardiogram (ECG) was used to appraise the effect of sour jujube kernel total soap on the myocardial infarcted area. Results Compared with control group, the experimental group treated with sour jujube kernel total soap glucosidal showed significantly smaller infarcted area; and ECG showed the heart rhythm, S-T piece and T wave value were significantly decreased in the experimental group. Conclusion Sour jujube kernel total soap glucosidal can reduce the myocardial infarcted area caused by ligature of LAD, so as to protect the cardiac muscle.
5.Sedative and hypnotic effects of oil of spine date seeds from different extraction technologies in mice

Objective To study the sedative and hypnotic effects of oil of spine date seeds by different extraction technologies(compression and supercritical CO2 extraction,OSDSE and OSDSS) in mice.Equivalence test was used to test the effects.Methods Spontaneous activity and tolerance were observed by photoelectric counting method after long-term intragastric administration of OSDSE and OSDSS in mice.The sleeping time and delitescence induced by above threshold dose of pentobarbital sodium and sleeping number induced by below threshold dose of pentobarbital sodium were utilized to observe the hypnotic effect of OSDSE and OSDSS in mice.Diazepam was applied as a positive control drug.Results OSDSE and OSDSS could obviously reduce spontaneous activity,increase sleeping time and sleeping number in mice,while the sleeping delitescence was shortened.All the effects induced by OSDSE and OSDSS were equivalent.The inhibitory effect of spontaneous activity was enhanced following repeated administration,while the effect of diazepam was reduced.Conclusion Both oils of spine date seeds can induce sedative and hypnotic effects without tolerance.They have equivalent effects on sedation and hypnosis.
