Acute lung injury (ALI), as a common clinical emergency, is pulmonary edema and diffuse lung infiltration caused by inflammation. The lack of non-invasive alert strategy, resulting in failure to carry out preventive treatment, means high mortality and poor prognosis. Stimulator of interferon genes (STING) is a key molecular biomarker of innate immunity in response to inflammation, but there is still a lack of STING-targeted strategy. In this study, a novel STING-targeted PET tracer, [¹⁸F]FBTA, was labeled with high radiochemical yield (79.7 ± 4.3%) and molar activity (32.5 ± 2.9 GBq/μmol). We confirmed that [¹⁸F]FBTA has a strong STING binding affinity (K_d = 26.86 ± 6.79 nmol/L) and can be used for PET imaging in ALI mice to alert early lung inflammation and to assess the efficacy of drug therapy. Our STING-targeted strategy also reveals that [¹⁸F]FBTA can trace ALI before reaching the computed tomography (CT) diagnostic criteria, and demonstrates its better specificity and distribution than [¹⁸F]fluorodeoxyglucose ([¹⁸F]FDG).

Objective: The relationship between serum uric acid (SUA) levels and glycemic indices, including plasma glucose (FPG), 2-hour postload glucose (2h-PG), and glycated hemoglobin (HbA1c), remains inconclusive. We aimed to explore the associations between glycemic indices and SUA levels in the general Chinese population. Methods: The current study was a cross-sectional analysis using the first follow-up survey data from The China Cardiometabolic Disease and Cancer Cohort Study. A total of 105,922 community-dwelling adults aged ≥ 40 years underwent the oral glucose tolerance test and uric acid assessment. The nonlinear relationships between glycemic indices and SUA levels were explored using generalized additive models. Results: A total of 30,941 men and 62,361 women were eligible for the current analysis. Generalized additive models verified the inverted U-shaped association between glycemic indices and SUA levels, but with different inflection points in men and women. The thresholds for FPG, 2h-PG, and HbA1c for men and women were 6.5/8.0 mmol/L, 11.0/14.0 mmol/L, and 6.1/6.5, respectively (SUA levels increased with increasing glycemic indices before the inflection points and then eventually decreased with further increases in the glycemic indices). Conclusion An inverted U-shaped association was observed between major glycemic indices and uric acid levels in both sexes, while the inflection points were reached earlier in men than in women.

Excess accumulation of white adipose tissue (WAT) causes obesity which is an imbalance between energy intake and energy expenditure. Obesity is a serious concern because it has been the leading causes of death worldwide, including diabetes, stroke, heart disease and cancer. Therefore, uncovering the mechanism of obesity and discovering anti-obesity drugs are crucial to prevent obesity and its complications. Browning, inducing white adipose tissue to brown or beige (brite) fat which is brown-like fat emerging in WAT, becomes an appealing therapeutic strategy for obesity and metabolic disorders. Due to lack of efficacy or intolerable side-effects, the clinical trials that promote brown adipose tissue (BAT) thermogenesis and browning of WAT have not been successful in humans. Obviously, more specific means still need to be developed to activate browning of white adipose tissue. In this review, we summarized seven kinds of natural products (alkaloids, flavonoids, terpenoids, long chain fatty acids, phenolic acids, else and extract) promoting white adipose tissue browning which can ameliorate the metabolic disorders, including obesity, dislipidemia, insulin resistance and diabetes. Since natural products are important drug sources and the browning property plays a significant role in not only obesity treatment but also in type 2 diabetes (T2DM) improvement, natural products of inducing browning may be an irreplaceable drug discovery orientation for obesity, diabetes and even other metabolic disorders.

Hyperglycemia is the dominant phenotype of diabetes and the main contributor of diabetic complications. Puerarin is widely used in cardiovascular diseases and diabetic vascular complications. However, little is known about its direct effects on diabetes. The aim of our study is to investigate its antidiabetic effect in vivo and in vitro, and explore the underlying mechanism. We used type I diabetic mice induced by streptozotocin to observe the effects of puerarin on glucose metabolism. In addition, oxidative stress and hepatic mitochondrial respiratory activity were evaluated in type I diabetic mice. In vitro, glucose consumption in HepG2 cells was assayed along with the qPCR detection of glucogenesis genes expression. Moreover, ATP production was examined and phosphorylation of AMPK was determined using Western blot. Finally, the molecular docking was performed to predict the potential interaction of puerarin with AMPK utilizing program LibDock of Discovery Studio 2018 software. The results showed that puerarin improved HepG2 glucose consumption and upregulated the glucogenesis related genes expression. Also, puerarin lowered fasting and fed blood glucose with improvement of glucose tolerance in type I diabetic mice. Further mechanism investigation showed that puerarin suppressed oxidative stress and improved hepatic mitochondrial respiratory function with enhancing ATP production and activating phosphorylation of AMPK. Docking study showed that puerarin interacted with AMPK activate site and enhancing phosphorylation. Taken together, these findings indicated that puerarin exhibited the hypoglycemic effect through attenuating oxidative stress and improving mitochondrial function via AMPK regulation, which may serve as a potential therapeutic option for diabetes treatment.

OBJECTIVETo investigate the role of donor human milk in the prevention of nosocomial infection in very low birth weight infants. MeETHODS: A total of 105 hospitalized preterm infants with a very low birth weight were enrolled. They were classified into mother's own milk feeding group, donor human milk feeding group, and preterm formula feeding group, with 35 infants in each group. The three groups were compared in terms of incidence rates of nosocomial infection, necrotizing enterocolitis, and feeding intolerance, time to full enteral feeding, and early growth indices.

RESULTSCompared with the preterm formula feeding group, the donor human milk feeding group and the mother's own milk feeding group had significantly lower incidence rates of nosocomial infection and necrotizing enterocolitis and shorter time to full enteral feeding (P<0.05). There were no significant differences in head circumference, body length, and weight growth velocity among the three groups.

CONCLUSIONSDonor human milk can be used in case of a lack of mother's own milk and may help to reduce nosocomial infection.

Objective To investigate the expression of RUNX3 mRNA and protein in hepatic cell carcinoma (HCC)and surrounding normal tissue,to analyze the relationship between RUNX3 expression and clinical pathological parameters.Methods Reverse transcription polymerase chain reaction ( RT-PCR ) and immunohistochemistry(IHC) were performed to detect the expression of RUNX3 mRNA and protein in HCC and surrounding normal tissue respectively,and their relationship with clinical pathological parameters were analyzed.Results The relative expression value of RUNX3 mRNA found in 51 cases HCC was 0.4509 ± 0.0963,and that did in 51 cases surrounding normal tissue was 0.9147 ±0.0222.The difference of RUNX3 mRNA expression between two kinds of samples was statistically significant( t =33.6087,P ＜ 0.001 ).The positives rate of RUNX3 protein expression found in 51 cases HCC tissue was 49.02％ (25/51)and that did in 51 cases surrounding normal tissue was 82.35％ (42/51).The difference of RUNX3 protein expression between two kinds of samples was statistically significant ( x2 =12.5706,P ＜ 0.005 ).The difference of RUNX3 mRNA and protein expression in some clinical pathological parameters involving differentiation degree,invasion,cancer thrombus and diversion in liver were statistically significant (P ＜ 0.05 ).However that were not in another clinical pathological parameters involving gender,cancer diameter,cancer location as well as hemorrhage and necrosis of cancer,histotype ( P ＞ 0.05 ).Conclusion The expression of RUNX3 mRNA and protein in HCC were significantly lower than that in surrounding normal tissue. The lower expression of runx3 protein in the HCC probably plays an important role in the tumorigenesis and development of HCC.The RUNX3 gene may be an anti-oncogene of HCC.

Objective To investigate knowledge requirements of hemodialysis patients in induction phase and the perception of nurses and compare differences between them,then provide a theory basis for improving patients' quality of life and developing health education.Methods A questionnaire was designed to investigate 63patients of hemodialysis in induction phase and 45 nurses of Hemopurification center about knowledge and perception.Results 82.54% of hemodialysis patients hope to receive health education related hemodialysis knowledge.There were striking differences on expenses of hemodialysis,protection of vascular access,diet instruction,complication of hemodialysis and its treatment and the interval of hemodialysis.On the effect of hemodialysis,the highest requirement was patients and nurse' s knowledge.Conclusions Hemodialysis patients'knowledge requirement is comparably higher.It also suggested that the main way in health education should facus on communication between doctors and nurses.

Objective To explore the influence of practical teaching in community in health education on the nurses' critical thinking ability in higher vocational school.Methods The contrast group was to fulfill practical teaching according to teaching plans.On weekends,the experimental group organizeed nurses to create wall newspapers about health education,carry out health education consultation,check residents' health responsibly and create family healthy files etc.The assessment of critical thinking ability was measured with Chinese Critical Thinking Disposition Inventory,CTDI-CV.Results There was a distinct difference about their critical thinking ability between the two groups(P ＜ 0.01).5 items showed clear differences among 7 excepts the ability of analysis and desire for knowledge.Conclusions Pratical teaching in community in health education can improve nurses' ability of critical thinking effectively.[Key words ] Health education;Community; Practical teaching; Nurse of higher vocationalschool;Critical thinking ability
