1.The correlation analysis of PM2.5 environmental factors in Tianmen city with clinical indicators of pediatric snoring accompanied by allergic rhinitis

OBJECTIVE To evaluate the impact of environmental factors in Tianmen on clinical indicators of pediatric snoring with allergic rhinitis and to analyze their correlations.This study aims to deepen the understanding of the relationship between environmental factors and pediatric respiratory diseases,and to provide scientific evidence for future prevention and treatment strategies.METHODS A retrospective study was conducted based on the clinical data of 3 071 pediatric patients with snoring admitted to Tianmen First People's Hospital from January 1,2021,to December 31,2023.The 95th percentile values of PM2.5,SO?,NO?,PM10,and CO,as well as the 90th percentile value of O?,in Tianmen during the same period were collected.Correlation analysis and binary logistic regression were used to investigate the association between environmental factors and pediatric snoring with allergic rhinitis.RESULTS Pearson correlation analysis revealed positive correlations among SO?,NO?,PM10,PM2.5,and CO,with the strongest correlation between PM2.5 and PM10(r=0.862,P<0.001).In contrast,O? was negatively correlated with other pollutants,with the most significant negative correlation observed between CO and O?(r=-0.505,P<0.001).Seasonal distribution showed that pollutant levels were significantly lower in summer than in winter.Among the 3 071 patients,840 cases were complicated with allergic rhinitis.Significant differences were observed between the allergic rhinitis and non-allergic rhinitis groups in terms of PM2.5 levels,length of hospital stay,total cost,drug cost,antibiotic cost,year of treatment,and treatment modalities(P<0.05).Logistic regression indicated that PM2.5 was an independent risk factor for pediatric snoring with allergic rhinitis(OR=1.013,95%CI:1.010-1.017,P<0.05),and this remained significant after adjusting for multiple clinical variables(OR=1.010,95%CI:1.007-1.014,P<0.05).ROC curve analysis showed that the combined diagnostic model using PM2.5 and total cost had the highest diagnostic performance(AUC=0.642,95%CI:0.620-0.664),which was superior to PM2.5 alone(AUC=0.636)or total cost alone(AUC=0.623).CONCLUSION Pediatric snoring with allergic rhinitis is closely related to environmental factors,especially PM2.5,which has a significant impact.These findings may provide a scientific basis for targeted environmental interventions and treatment strategies to improve the quality of life in affected children.
2.Effect of Astragaloside Ⅳ on Proliferation, Migration, and Invasion of Colorectal Cancer HCT116 Cells

ObjectiveTo investigate effect of astragaloside Ⅳ on the proliferation, migration, and invasion of colorectal cancer HCT116 cells and the underlying molecular mechanism. MethodColorectal cancer HCT116 cells were classified into blank group (DMSO) and low-dose (15.7 mg·L-1), medium-dose (31.4 mg·L-1), and high-dose (62.8 mg·L-1) astragaloside Ⅳ groups. After drug treatment, the morphological changes of HCT116 cells were observed under an inverted microscope. Cell viability was detected by cell counting kit-8 (CCK-8) assay, and the migration and invasion of cells were detected based on scratch assay and Transwell assay. The expression of cyclin-dependent kinase inhibitor (p21), CyclinD1, B-cell lymphoma-2 (Bcl-2), and Bcl-2-associated X protein (Bax) in the cells was examined by Western blot. ResultCompared with the blank group, cells in the three astragaloside Ⅳ groups demonstrated slow growth, low density, inconsistent morphology, nuclear shrinkage, degradation of cytoplasm, and high death rate. Moreover, cell viability decreased in a concentration-dependent manner in the astragaloside Ⅳ groups. Cell migration and invasion were inhibited (P<0.05, P<0.01), and the inhibition rate was in positive correlation with the concentration of the astragaloside Ⅳ. The expression of pro-apoptotic protein Bax in low-dose, medium-dose and high-dose astragaloside Ⅳ groups increased gradually in a concentration-dependent manner, while the expression of p21, CyclinD1 and anti-apoptotic protein Bcl-2 decreased gradually in a concentration-dependent manner compared with those in the blank group (P<0.05, P<0.01). ConclusionAstragaloside Ⅳ can suppress the proliferation, migration, and invasion of colorectal cancer HCT116 cells and promote the apoptosis, thus inhibiting the occurrence and development of colorectal cancer.
3.In vitro comparison of transdermal characteristics of xiaoyu ointment and xiaoyu plaster.

OBJECTIVEThrough the comparison of Xiaoyu ointment and xiaoyu plaster by in vitro transdermal demonstrate, to demonstrate the scientificity and feasibility of reformed formulation.
METHODThe improved Franz diffusion cells and in vitro rabbit skin were used in vitro penetration experiment with emodin as an indicator of penetration rate quantitated by HPLC.
RESULTThe cumulative penetration rate of emodin in Xiaoyu ointment fit the model of Weibull distribution, while the cumulative penetration rate of emodin in Xiaoyu plaster fit the model of Density equation. Take emodin as an index,the transdermal rate in Xiaoyu plaster was 1.93 times as Xiaoyu ointment, and the total penetrated amount was 2.84 times as Xiaoyu ointment. The results showed that the emodin of xiaoyu plaster reserved in the skin were 3.95 times more than the ointment.
CONCLUSIONThe penetration rate, total penetrated amount and the reserves in the skin of Xiaoyu plaster were better than the ointment, and the transdermal dosage form was better than the original form.
