1.Clinical Values of Combined Detection of SerumCystatin C, β2-Microglobulin, and Urine Transferrin in Diagnosing Early Primary Glomerulonephritis

Despite primary glomerulonephritis (PGN) being a leading cause of chronic kidney disease and end-stage renal disease, specific and sensitive biomarkers for the early detection and monitoring of this condition are lacking. We evaluated the value of the combined detection of serum cystatin C (CYSC), β2-microglobulin (β2-MG), and urine transferrin (TRF) for diagnosing early-stage PGN. From May 2021 to May 2023, we enrolled 105 patients in our hospital as the observation group and 50 healthy volunteers as the control group. Their serum expression levels of CYSC, β2-MG, and TRF were evaluated. We plotted separate ROC curves and calculated the area under the curve (AUC) values of CYSC, β2-MG, and TRF to assess their diagnostic performance in PGN. The levels of CYSC, β2-MG, and TRF were significantly higher (P < 0.05) in the observation group than in the healthy control group. CYSC, β2-MG, and TRF were expressed at significantly higher levels in G2, G3a, and G3b of PGN than in G1. The combined use of CYSC, β2-MG, and TRF as biomarkers could significantly improve the early diagnosis and monitoring of PGN and may lead to better patient outcomes by facilitating earlier intervention and treatment strategies.
2.Establishment of HPLC Fingerprint ,Cluster Analysis and PLS-DA of Ligusticum chuanxiong Decoction Pieces

OBJECTIVE: To establish HPLC fingerprints of Ligusticum chuanxiong decoction pieces, and to conduct cluster analysis and PLS-DA analysis. METHODS: HPLC method was adopted. The determination was performed on Waters Symmetry C18 column with mobile phase consisted of acetonitrile-0.5% acetic acid solution (gradient elution) at the flow rate of 1 mL/min. The detection wavelength was set at 254 nm, and the column temperature was 30 ℃. The sample size was 10 μL. Using ligustilide as control, HPLC chromatograms of 21 batches of samples (S1-S20) were determined. The similarity evaluation was conducted by using Similarity Evaluation System for Chromatographic Fingerprint of TCM (2012 edition) to determine common peak. Cluster analysis was conducted by using SPSS 19.0 software and PLS-DA was used to distinguish the samples. RESULTS: There were 25 common peaks in HPLC chromatograms for 21 batches of samples, and 9 common peaks were identified. The similarity of samples was between 0.768-0.989, and the similarity of base and traditional medicinal part samples (S1-S10) were more than 0.970. The 21 batches of samples were clustered into 3 categories, in which S1-S10 were category Ⅰ; S15-S16, S19-S20 were category Ⅱ; other were category Ⅲ. By PLS-DA analysis, 11 classification markers were identified as well as 5 chromatogram peaks were identified, such as ferulic acid, pine cyperyl ferulate, n-butyl phthalide, ligustilide, ligustilide A, which could be used to distinguish base and non-markted samples (S1-S10) from marketed and non-base samples (S11-S21), which were consistent with the results of cluster analysis. CONCLUSIONS: Established fingerprint, cluster analysis and PLS-DA analysis can provide reference for quality evaluation of L. chuanxiong decoction pieces.
5.Determination of Schisandrin and Schisandrin B in Compound Wurenchun Capsule by RP-HPLC

OBJECTIVE:To establish a method for content determination of schisandrin and schisandrin B in compound wurenchun capsule by RP-HPLC.METHODS:Column lichrosphere C18 was used,mobile phase of water and MeOH(gradient elution)was set up,the flow rate was 1.0mL?min-1,the column temperature was 30℃and the detection wavelength was 254nm.RESULTS:The linear ranges of schizandrin and schisandrin B were within 0.182 4~1.641 6?g(r=1.000 3)and 0.189 6~ 1.706 4?g(r=0 .999 9),respectively.The average recoveries were 98.32%(RSD=1.02%)and 97.22 %(RSD=0.94%),respectively.CONCLUSION:The established method is convenient,accurate and highly specific,and it can be used for the quality control of compound wurenchun capsule.
