ObjectiveTo investigate the molecular mechanisms by which Acan-Gal-1 regulates macrophage immune phenotypes and Annexin A2/S100A10 membrane translocation in Angiostrongylus cantonensis. MethodsImmunofluorescence assay (IFA) was performed to determine the subcellular localization of Annexin A2 and S100A10 in macrophages. Flow cytometry and ELISA were used to assess functional changes in macrophage. siRNA interference of Annexin A2 was used to detect the effect of Acan-Gal-1 on various indicators. The expression levels of related genes and proteins were analyzed by Western blot, qRT-PCR, and ELISA methods. Bioinformatics analyses were conducted to characterize the structural features of Acan-Gal-1, and sequence alignment and molecular interaction prediction were performed to explore its potential interaction with Annexin A2. ResultsThis study found that Acan-Gal-1 can reverse LPS-induced promotion of macrophage migration and phagocytic function; it significantly upregulated M2-type markers in macrophages (Arg-1, IL-10, TGF-β) while inhibiting the expression of M1-type markers (iNOS, IL-6, IL-1β) induced by LPS. Acan-Gal-1 reduced the protein levels of Annexin A2 in macrophages under an immunologically activated state; after interfering with Annexin A2 expression, Acan-Gal-1 not only fails to induce macrophages to transition to an immunologically inhibitory phenotype but also fails to inhibit LPS-induced transition of macrophages to an immunologically activated phenotype. Although Acan-Gal-1 did not affect the expression of Annexin A2 or its chaperone protein S100A10 in macrophages, it markedly promoted their translocation to the plasma membrane. ConclusionsAcan-Gal-1 facilitates the enrichment of the Annexin A2/S100A10 complex at the macrophage membrane, thereby exerting immunomodulatory effects on macrophage polarization.

ObjectiveTo observe the effect of enriched environment (EE) combined with acupuncture at head point (HA) on behavior in rats with autism spectrum disorder. MethodsHealthy female Wistar rats were given peritoneal injection of sodium valproate at 12.5 days of gestation. Twenty-four male offspring rats were randomly selected and then randomly divided into model group (n = 6), EE group (n = 6), HA group (n = 6) and EE combined with HA group (the combined group, n = 6). Six male offspring rats born from female mice injected with the same amount of saline intraperitoneally were as control group. After four weeks of treatment, all the five groups were tested with three-chamber test and marble burying test, and the sociability index, the social novelty index and the number of buried marbles were recorded. The levels of interleukin (IL)-1β and IL-6 in peripheral blood were determined by enzyme-linked immunosorbent assay (ELISA). ResultsAfter treatment, compared with the model group, the sociability index and the social novelty index improved (P < 0.05), the number of buried marbles reduced (P < 0.05), and the levels of IL-6 and IL-1β in peripheral blood decreased in EE group, HA group and the combined group (P < 0.05); while the combined group was the best (P < 0.01). ConclusionBoth EE or acupuncture at HA could improve behavioral symptoms, and reduce the expression of inflammatory factors in rats with autism spectrum disorder. The combination of the two methods showed the best result.

Objective:To explore the feasibility and clinical effect of the infrared vascular imager in the transplantation of free arterialized venous flaps. Methods:A retrospective analysis was conducted with patient data of hand microsurgery using free arterialized venous flaps to repair hand wounds in the Second Affiliated Hospital of Wenzhou Medical University from March 2019 to November 2020. An arterialized venous flap was designed according to the Goldschlager’s Type Ⅲ flap design with the aid of a magnifying glass following the venous course. The flap was transfer to cover the wound of the recipient area. The reconstructive effect was observed postoperatively, including the blood supply, color, texture, survival of the flap, the flexion and extension of the affected finger, and the appearance of the forearm donor site. The hand function was evaluated by the Chinese Medical Association Hand Surgery Society’s upper limb function evaluation trial standard. Results:A total of 17 patients were included, including 11 males and 6 females, ranging in age from 19 to 68 years old, with an average age of 34.6 years. A total of 18 skin flaps were designed and elevated, and the flap size ranged from 2.0 cm × 3.0 cm to 8.0 cm × 9.0 cm. The patients were followed up 3 to 12 months after the operation, with an average follow-up of 6 months. Blisters appeared on the second day after the operation in 8 flaps, and the affected limbs were raised to maintain the integrity of the blisters. The blisters of the flaps subsided one week after the operation. Congestion occurred on the second day after operation in 7 flaps, which subsided after two weeks after routine treatment, such as raising the affected limb and removing some sutures. One flap developed progressive hypoperfusion one week after the operation, and eventually necrosis. Seventeen flaps survived, and one flap failure was observe. All the surviving skin flaps were free of bloat and had a good appearance. In two cases, the skin graft area was pigmented, and the flexion and extension activities of the affected fingers were suitable. Using the Chinese Medical Association Society of Hand Surgery’s upper limb function evaluation trial standard, 16 patients (excluding one flap failure) were evaluated for hand function, showing 11 cases were excellent and 5 cases were good. Conclusions:Infrared vascular imaging technology enables free arterialized venous flap transplantation to realize non-contact, non-invasive, simple operation, intuitive and accurate, and truly realizes rapid flap design under direct vision. Infrared vascular imaging technology is an effective auxiliary tool for free arterialized venous flap transplantation.
