Anti-neutrophil cytoplasmic antibody-associated vasculitides (AAV) are rare autoimmune disorders typically involving the lungs and kidneys. A few case reports have described an association with hematopoietic stem cell transplant (HSCT). We present a case of myeloperoxidase-positive AAV with predominant renal involvement in a 49-year-old male with a history of acute myeloid leukemia treated with allogenic HSCT and complicated by chronic graft-versus-host disease (GVHD). He presented with rapidly progressive glomerulonephritis, and kidney biopsy revealed pauci-immune necrotizing crescentic glomerulonephritis. Treatment with methylprednisolone pulses followed by oral prednisolone, rituximab, and plasmapheresis led to improved kidney function and a tailored rituximab maintenance regimen was followed. However, a renal-limited relapse occurred at 15 months, requiring repeat induction therapy. Dialysis was avoided, but renal function only partially recovered. This case suggests that AAV can develop after HSCT and GVHD and highlights the importance of long-term follow-up and further research into underlying mechanisms.
2.Biofabricated 3D Intestinal Models as an Alternative to AnimalBased Approaches for Drug Toxicity Assays

BACKGROUND:
The main challenge in new drug development is accurately predicting the human response in preclinical models.
METHODS:
In this study, we developed three different intestinal barrier models using advanced biofabrication techniques: (i) a manual model containing Caco-2 and HT-29 cells on a collagen bed, (ii) a manual model with a Caco-2/HT-29 layer on a HDFn-laden collagen layer, and (iii) a 3D bioprinted model incorporating both cellular layers. Each model was rigorously tested for its ability to simulate a functional intestinal membrane.
RESULTS:
All models successfully replicated the structural and functional aspects of the intestinal barrier. The 3D bioprinted intestinal model, however, demonstrated superior epithelial barrier integrity enhanced tight junction formation, microvilli development, and increased mucus production. When subjected to Ibuprofen, the 3D bioprinted model provided a more predictive response, underscoring its potential as a reliable in vitro tool for drug toxicity testing.
CONCLUSION
Our 3D bioprinted intestinal model presents a robust and predictive platform for drug toxicity assessments, significantly reducing the need for animal testing. This model not only aligns with ethical testing protocols but also offers enhanced accuracy in predicting human responses, thereby advancing the field of drug development.

7.Radiomorphometric indices and bone findings on panoramic images of childhood cancer survivors

Purpose:
The aim of this study was to assess radiomorphometric indices in panoramic radiographs of pediatric cancer survivors.
Materials and Methods:
This population-based case-control study included 31 patients who were 18 years or older at the time of the radiograph and had a history of cancer diagnosed during childhood or adolescence (between 0 and 19 years of age), treated with chemotherapy and/or radiotherapy. The radiomorphometric indices mandibular cortical width (MCW) and mandibular cortical index (MCI) were assessed in panoramic radiographs to determine whether there was evidence of a reduction in bone mineral density (BMD). A 95% confidence level (P<0.05) was used for all statistical tests.
Results:
In the MCW analysis, the case group showed a variation between 2.2 mm and 4.6 mm, while the control group showed a variation between 2.9 mm and 5.2 mm (P<0.05). For MCI, most (54.8%) of the images in the casegroup were classified as C2, and 16.1% as C3, whereas none in the control group were classified as C3, demonstrating a statistically significant difference (P<0. 05).
Conclusion
Childhood cancer survivors are at an increased risk of long-term reduction in BMD, and panoramic radiomorphometric indices represent an accessible and reliable screening tool for predicting this risk.

The main challenge in new drug development is accurately predicting the human response in preclinical models.
METHODS:
In this study, we developed three different intestinal barrier models using advanced biofabrication techniques: (i) a manual model containing Caco-2 and HT-29 cells on a collagen bed, (ii) a manual model with a Caco-2/HT-29 layer on a HDFn-laden collagen layer, and (iii) a 3D bioprinted model incorporating both cellular layers. Each model was rigorously tested for its ability to simulate a functional intestinal membrane.
RESULTS:
All models successfully replicated the structural and functional aspects of the intestinal barrier. The 3D bioprinted intestinal model, however, demonstrated superior epithelial barrier integrity enhanced tight junction formation, microvilli development, and increased mucus production. When subjected to Ibuprofen, the 3D bioprinted model provided a more predictive response, underscoring its potential as a reliable in vitro tool for drug toxicity testing.
CONCLUSION
Our 3D bioprinted intestinal model presents a robust and predictive platform for drug toxicity assessments, significantly reducing the need for animal testing. This model not only aligns with ethical testing protocols but also offers enhanced accuracy in predicting human responses, thereby advancing the field of drug development.
