1.Correlation between serum total bile acid level and cognitive function in patients with stable schizophrenia and its predictive value for cognitive impairment

BackgroundPersistent cognitive impairment is prevalent among patients with stable schizophrenia. While serum total bile acid (TBA) level in acute-phase patients are known to be associated with cognitive dysfunction, the relationship between serum TBA and multi-dimensional cognitive functions in stable phase patients remains unclear. ObjectiveTo investigate the correlation between serum TBA level and cognitive function in patients with stable schizophrenia, and to evaluate its predictive value for cognitive impairment, thereby providing a serological biomarker for the timely identification and objective assessment of cognitive dysfunction. MethodsA cross-sectional study was conducted on 137 inpatients with stable schizophrenia at The Fourth People's Hospital of Yancheng from March to December 2024. All participants met the diagnostic criteria of the Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5). Cognitive function was evaluated using the Chinese Brief Cognitive Test (C-BCT), patients were categorized into four groups: normal cognition (n=28), mild impairment (n=28), moderate impairment (n=47), and severe impairment (n=34). Fasting venous blood samples were collected, and serum TBA level was quantified using an enzymatic cycle assay. Spearman correlation analysis was ultilized to determine the relationship between serum TBA level, overall cognitive function, and specific cognitive domains. Binary Logistic regression model was used (adjusting for covariates such as age, gender, and disease duration) to analyze the impact of serum TBA level on overall and individual cognitive functions. The predictive value of serum TBA level for overall cognitive impairment was evaluated using receiver operating characteristic (ROC) curve. ResultsSerum TBA levels differed significantly among the four groups (H=18.677, P<0.01). Specifically, serum TBA levels in both the moderate and severe cognitive impairment groups were significantly higher than those in the normal cognitive group (adjusted P<0.01). Serum TBA level was positively correlated with the severity grading of overall cognitive impairment (rs=0.354, P<0.05), and negatively correlated with T-scores on the trail making test (rs=-0.328, P<0.05), continuous performance test (rs=-0.247, P<0.05), digit span (rs=-0.265, P<0.05), and symbol coding (rs=-0.221, P<0.05). Binary Logistic regression analysis identified serum TBA level as an independent risk factor for overall cognitive impairment (OR=1.322, 95% CI: 1.021 - 1.713, P=0.034), with a particularly robust predictive ability for impaired information processing speed (OR=1.325, 95% CI: 1.057 - 1.661, P=0.015). The area under ROC curve (AUC) for serum TBA level in predicting overall cognitive impairment was 0.738, with a sensitivity of 60.61% and a specificity of 78.64%. ConclusionIn patients with stable schizophrenia, elevated serum TBA levels are associated with worse overall cognitive function, as well as deficits in information processing speed, attention, working memory, and executive function. Serum TBA serves as an independent risk factor and exhibits moderate predictive value for overall cognitive impairmen,particularly in the domain of information processing speed. Objective: To compare the biological characteristics of human induced pluripotent stem cell-derived platelet exosomes (hiPSC-Plt-Exos) with those of conventional apheresis platelet exosomes (Plt-Exos), specifically focusing on their differential abilities to enhance the proliferation and migration of human umbilical cord mesenchymal stem cells (hUC-MSCs). Methods: Exosomes were isolated from hiPSC-derived Plt and apheresis Plt concentrate using size exclusion chromatography. These exosomes were then characterized through nanoparticle tracking analysis (NTA), transmission electron microscopy (TEM), and Western blotting. Co-culture experiments into hUC-MSCs were conducted with hiPSC-Plt-Exos and apheresis Plt-Exos, respectively. Their effects on the proliferation and migration of hUC-MSCs were assessed via cell proliferation assays and scratch tests. Results: hiPSC-Plt-Exos and apheresis Plt-Exos exhibited comparable particle sizes, morphological features (such as the characteristic cup-shaped structure), and surface markers (including CD9 and HSP70). Notably, hiPSC-Plt-Exos demonstrated a significantly greater ability to enhance the proliferation and migration of hUC-MSCs compared to apheresis Plt-Exos (P<0.05). These differences provide critical comparative data for their application in various clinical contexts. Conclusion: This study establishes a theoretical foundation for developing precise therapeutic strategies based on hiPSC-Plt-Exos. Furthermore, it underscores the necessity of selecting the appropriate type of exosomes according to the specific disease microenvironment to achieve optimal therapeutic outcomes.
4.An investigation of diagnostic X-ray resources in Jinan in 2023

Objective:To investigate the current status of diagnostic X-ray resources in Jinan in order to provide theoretical basis for continuously optimizing the allocation of diagnostic resources and strengthening the management of radiological health and radiation protection.Methods:In accordance with the 2023 Jinan radiation protection monitoring program of medical and health institutions, a survey was carried out for medical institutions involved in diagnostic X-ray examinations (excluding military hospitals and dental clinics) in Jinan by using uniform questionnaires. The survey included the basic situation of diagnostic X-ray institutions, the allocation of diagnostic X-ray equipment, the number of diagnostic staff, and the frequency of diagnositic X-ray examinations.Results:In 2023, there were 298 diagnostic X-ray institutions in Jinan, with 3 494 diagnostic workers and 1 387 items of diagnostic X-ray equipment. The included were 41 tertiary hospitals, 57 secondary hospitals, 110 primary hospitals and 90 unrated hospitals. The tertiary hospitals possessed the largest number of the diagnostic workers and equipment, accounting for 60.99% and 45.35% of the total, respectively. Among all the items of diagnostic X-ray equipment, the number of items of digital radiography equipment ranked top (34.37%), followed by CT machines (24.44%). There were 147.32 items of diagnostic X-ray equipment per million population on average. A total of 8 180 363 individuals received various types of diagnostic X-ray examinations, with CT diagnosis accounting for 46.93% and X-ray for 34.41%. The frequency of diagnostic X-ray examinations was 868.86/1 000 population, including the frequency of CT examinations of 407.76 per 1 000 population.Conclusions:The per capita possession of medical X-ray equipment in Jinan is at a high level. The frequency of medical exposure has increased significantly, especially CT examinations. There is a big gap in diagnostic X-ray resource allocation between different levels of hospitals, so it is necessary to control the resource allocation and strengthen the protection of CT examination on the scientific basis.
5.Analysis on the epidemic and genetic characteristics of varicella in Fengtai District, Beijing City, 2024-2025

This study analyzed the epidemiological characteristics and strain genotypic distribution of varicella in Fengtai District, Beijing, utilizing population-based surveillance data from March 2024 to February 2025 combined with laboratory nucleic acid detection and genotyping.We reported 522 varicella cases with male predominance (282 cases, 54.41%) and a majority aged >15 years (358 cases, 68.58%).A bimodal incidence pattern peaked in May and November, while 161 breakthrough infections (30.8%) occurred predominantly among students aged 6-20 years.The multivariate logistic regression model analysis showed that, compared to the 0-5 years age group, individuals aged 6-14 years ( OR: 2.729, 95% CI: 1.083-6.88), 15-20 years ( OR: 2.495, 95% CI: 1.158-5.378), and>20 years ( OR: 5.382, 95% CI: 2.478-11.689) exhibited progressively higher odds of oderate-to-severe rash; females demonstrated a lower risk compared to males ( OR: 0.485, 95% CI: 0.286-0.822); regarding vaccination status, recipients of one vaccine dose ( OR: 0.301, 95% CI: 0.161-0.564) and two doses ( OR: 0.203, 95% CI: 0.086-0.48) showed significantly reduced risks relative to unvaccinated individuals. Genotyping of 174 specimens identified 161 VZV-positive samples, with successful ORF22/38/62 sequencing in 142 samples confirming Clade 2 predominance (141 strains, 99.3%) and one Clade 5 strain; local isolates exhibited high vaccine-strain homology (ORF22 nucleotide:99.5%-100%, amino acid:99.3%-100%) with mutation sites partially overlapping other Chinese regions.
6.CT-assisted 3D printing template-guided implantation of 125I particles for the treatment of head and neck malignancies:a clinical study

Objective To evaluate the short-term clinical effect(including curative efficacy,pain relief,and quality of life)of CT-assisted 3D printing template-guided implantation of 125I particles in treating head and neck malignancies.Methods A total of 12 patients with head and neck malignant tumors complicated by moderate to severe cancerous pain,who were admitted to Yichang Municipal second People's Hospital to receive treatment from September 2019 to July 2024,were enrolled in this study.All patients received CT-assisted 3D individualized template-guided implantation of 125I particles therapy.The short-term curative efficacy,pain relief,and quality of life of patients were evaluated.Results Three months after implantation of 125I particles,complete response(CR)was obtained in 3 patients,partial remission(PR)in 5 patients,and stable disease(SD)in 4 patients,with an objective response rate(ORR)of 66.67%and a disease control rate(DCR)of 100%.The preoperative mean NRS score was(5.58±0.79)points,and the postoperative 3-month mean NRS score was(2.08±0.67)points.The postoperative 3-month physiological well-being(PWB),emotional well-being(EWB),functional well-being(FWB),and the total score were significantly better than their preoperative values,the differences were statistically significant(P<0.05).No statistically significant difference in social/family well-being(SWB)existed between its postoperative 3-month value and its preoperative value.Adverse reactions occurred in 2 patients,including local mucosal ulceration(n=1)and skin redness with swelling(n=1),which were improved after treatment.No serious complications occurred.Conclusion For the treatment of patients with head and neck malignant tumors complicated by moderate to severe cancerous pain,radioactive 125I particle implantation has a significant short-term effect,it can effectively relieve cancerous pain and improve quality of life of patients with reliable clinical safety.
7.Administration of Psoralea corylifolia L. (Buguzhi) during pregnancy causes mild liver injury in mouse mothers and weaned offspring

Background: Psoralea corylifolia L. (Buguzhi, BGZ), known for its efficacy in supporting pregnancy and preventing miscarriage, has been used in China for over 1000 years. Recently, BGZ has been identified as a potential cause of drug-induced liver injury. However, its safety during pregnancy remains unclear, which significantly hinders its routine clinical application. Objective: To investigate the effects of BGZ administration during pregnancy on the liver of mouse mothers and their weaned 21-day-old offspring. Methods: Mice were orally administered BGZ at doses of 2.5 and 10 g/kg during pregnancy, with BGZ withdrawal during the lactation period. Liver histopathology (hematoxylin-eosin staining), biochemical analysis, and evaluation of liver bile acid metabolism were performed after the lactation period. Results: BGZ administration at doses of 2.5 and 10 g/kg during pregnancy, followed by withdrawal during the lactation period, caused mild liver damage in both mothers and their 21-day-old offspring. Serum total bile acid (TBA) levels were elevated compared with those in the control group. Additionally, changes were observed in the levels and proportions of various bile acids (BAs) in the liver, suggesting mild effects on BA metabolism. Conclusion: BGZ administration during pregnancy caused mild liver damage and increased serum TBA levels in both mouse mothers and their 21-day-old offspring. This phenomenon may be associated with imbalanced BA metabolism in the liver. Based on the present study and the limited toxicological research on BGZ, pregnant women should avoid prolonged use of BGZ. If BGZ is administered during pregnancy, serum TBA levels should be monitored, and if elevated, BGZ should be discontinued.
8.Stat1 inhibits Foxp3 expression and Treg production

Objective:To explore the influence of Stat1 on Foxp3 expression and production of Treg.Methods:C57BL/6 mice were used and separated into normal control group and Stat1 specific inhibitor Fludarabine(Flud)treatment group.Ratio of CD4+Foxp3+Treg and expression of Foxp3 in spleen,lymph nodes and peripheral blood of mice in each group were detected by flow cy-tometry.Human Stat1 overexpression plasmid was constructed and transfected into human breast cancer MCF-7 cells,and expression changes of Foxp3 was detected by RT-qPCR and Western blot.Results:Compared with mice in normal control group,proportion of Treg and expression of Foxp3 in lymph nodes and peripheral blood of mice in Flud treatment group were increased,while Stat1 overex-pression resulted in decreased Foxp3 mRNA and protein expression in MCF-7 cells.Conclusion:Stat1 inhibits expression of Foxp3 and production of Tregs.
