1.From "standardized patients" to "digital twin patients": Disciplinary logic and technical divergence of five patient modeling paradigms

With the rapid development of large language models and related technologies, various forms of patient modeling have emerged, including virtual patients, synthetic patients, artificial intelligence patients, and digital twin patients. Although these forms all share the core objective of patient simulation, they exhibit significant differences in theoretical ontology, data sources, modeling logic, and application orientation. Based on the abstraction levels of simulation objects and system mapping logic, this study systematically compares five paradigms—standardized patients, virtual patients, synthetic patients, AI patients, and digital twin patients—to clarify their conceptual boundaries and technical positioning. The findings indicate that these paradigms represent distinct modeling pathways: behavioral reproduction, scenario simulation, data generation, cognitive interaction, and individual system mapping, respectively. By constructing a multidimensional comparative framework, this study provides a theoretical foundation for the conceptual standardization of patient modeling and the strategic deployment of intelligent healthcare systems.
2.Prevention and treatment of monkeypox in pregnant women

Using China National Knowledge Infrastructure, Wanfang, and PubMed database, literature search was conducted with the keywords “pregnancy” and “monkeypox”, and 27 related research articles were selected for analysis. Through a comprehensive review of the related literature, we aim to improve our knowledge of this viral disease, better our prevention, treatment and responses to future monkeypox outbreaks in China, so as to better protect the safety of mothers and infants. Maternal monkeypox can be prevented and controlled, if active and effective measures are taken in time. Drawing on the experience and lessons from monkeypox outbreaks at home and abroad, it is suggested that hospitals and public health agencies at all levels should raise awareness, and establish an effective emergency preparedness system for the prevention and control of potential future outbreaks.
3.Butorphanol affects the proliferation, migration and invasion of osteosarcoma MG-63 cells via Hippo/YAP signaling pathway

[摘 要] 目的:探讨布托啡诺(BPH)对骨肉瘤(OS)细胞增殖、迁移和侵袭的影响及其相关的作用机制。方法:将MG-63细胞分为对照组、YAP抑制剂组(维替泊芬组)和BPH低、中、高浓度组,MTT法、克隆形成实验、FCM术、划痕愈合实验、Transwell实验、qPCR法、WB法和移植瘤实验分别检测处理后各组细胞的增殖活性、克隆形成数、细胞凋亡率、划痕愈合率,以及上皮钙黏蛋白(E-cadherin)、神经钙黏蛋白(N-cadherin)、波形蛋白(vimentin)mRNA的表达和YAP、TAZ蛋白的表达,同时观察BPH和维替泊芬对移植瘤生长的影响。结果:与对照组相比,维替泊芬组和BPH低、中、高浓度组细胞增殖活性、克隆数、划痕愈合率、侵袭细胞数,以及N-cadherin和vimentin mRNA水平、YAP和TAZ蛋白表达及移植瘤体积均显著降低(均P<0.05),细胞凋亡率、E-cadherin mRNA水平及对移植瘤的抑瘤率均升高(均P<0.05),且BPH高浓度组与维替泊芬组之间各项指标均无明显差异(均P>0.05)。结论:BPH可能通过抑制Hippo/YAP信号通路来抑制OS细胞MG-63增殖、迁移和侵袭。
4.Comprehensive analysis on expression and biological role of miR-224 in hepatocellular carcinoma

[Abstract] Objective: To analyze the expression level of miR-224 in cancer tissues and plasma of hepatocellular carcinoma (HCC)
patients, and its correlation with clinicopathological characteristics, diagnosis and prognosis of HCC patients, and to further analyze its
mechanism of action in the occurrence and development of liver cancer through bioinformatics analysis and in vitro experiments.
Methods: The expression level of miR-224 in HCC tissues and normal tissues was analyzed using large sample data from Gene
Expression Omnibus (GEO). qPCR method was used to verify the expression level of miR-224 in the tumor tissues and
corresponding adjacent tissues that surgically resected from 80 HCC patients in Hebei Provincial People ’s Hospital from January
2017 to January 2020; in addition, the miR-224 level was also examined in plasma samples from 30 HCC patients. The Kaplan-Meier
plotter database was used to analyze the correlation between the miR-224 expression and the overall survival time of HCC patients.
The biological processes and signal pathways involving miR-224 were analyzed using bioinformatics tools. Hepatocellular
carcinoma HepG2 cells were transfected with miR-224 inhibitor, and then Clone formation experiment, Transwell chamber
experiment, qPCR and WB methods were used to detect the effect of miR-224 knockdown on the proliferation and invasion of
HepG2 cells and the expression level of EMT-related molecules. Results: The results of GEO database analysis showed that the
expression level of miR-224 in HCC tissues was significantly higher than that in normal tissues. The results of clinical specimen verification showed that the expression level of miR-224 in the tumor tissues and plasma of HCC patients was significantly higher than
that in the corresponding adjacent tissues and plasma from healthy controls (all P<0.01). The expression level of miR-224 was
significantly correlated with the TNM stage, lymph node metastasis status and tumor size of HCC patients (P<0.05 or P<0.01). ROC
analysis indicated that miR-224 showed a prominent diagnostic value in liver cancer, and the increased expression level of miR-224 was
significantly related to the poor prognosis of HCC patients (P<0.05). Functional enrichment analysis revealed that miR-224 was mainly
involved in the mTOR signaling pathway, AGE-RAGE signaling pathway, Rap1 signaling pathway, Ras signaling pathway, ErbB
signaling pathway, HIF-1 signaling pathway and p53 signaling pathway and other signaling pathways related to tumor occurrence and
development. Knockdown of miR-224 could significantly inhibit the colony formation and invasion of HepG2 cells and affect the
expression of EMT-related markers (P<0.05 or P<0.01). Conclusion: miR-224 is highly expressed in HCC tissues and plasma and is
significantly related to the poor prognosis of HCC patients. Knockdown of miR-224 expression can inhibit the colony formation,
invasion and EMT process of liver cancer HepG2 cells.
5.Application of diabetic retinopathy diagnosis and treatment and follow-up system in clinical practice

AIM: To develop diabetic retinopathy treatment and follow-up system to screen and follow up the patients with diabetic retinopathy, and to reduce the incidence of diabetic retinopathy and blindness.
METHODS: Through the establishment of diabetic retinopathy diagnosis and follow-up system, we established diabetic retinopathy fundus image database, and to do early intervention treatment and regular follow-up of patients with diabetes.
RESULTS: Through more than 1a of follow-up and timely and effective medical intervention, significantly reduced the incidence of severe proliferative diabetic retinopathy, delayed the further vision damage.
CONCLUSION: “Diabetic retinopathy diagnosis and treatment and follow-up system” builds “health education-screening-treatment-follow-up-health management” as the main line of the “active” service system, reduces the blindness rate and the incidence of diabetic retinopathy.
