Myelin is an essential structure that facilitates rapid saltatory conduction in the nervous system. Discrepancies in myelin microstructure are a hallmark of numerous neurological disorders, rendering the assessment of myelin integrity and content an indispensable tool in clinical diagnostics and neuroscience research. Extensive research has been dedicated to scrutinizing its biochemical makeup and morphology under normal, pathological, and experimental conditions over the years. In this review, we present an updated summary of the myelin sheath's structure, composition, and developmental trajectory. We systematically enumerate and contrast eight prevalent myelin staining techniques across dimensions of sensitivity, specificity, and resolution, delving into their underlying staining principles. With an initial application of myelin histology on the mouse demyelination model, our review accentuates the accurate delineation of myelination and the microstructural analysis of the myelin sheath. Such insights are anticipated to significantly contribute to the evaluation and understanding of white matter pathologies.

OBJECTIVE: This study aims to elucidate the therapeutic potential of osthole for the treatment of atopic dermatitis (AD), focusing on its ability to alleviate chronic pruritus (CP) and the underlying molecular mechanisms. METHODS: In this study, we investigated the anti-inflammatory effects of osthole in both a 2,4-dichloronitrobenzene (DNCB)-induced AD mouse model and tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) stimulated huma immortalized epidermal (HaCaT) cells. The anti-itch effect of osthole was specifically assessed in the AD mouse model. Using methods such as hematoxylin and eosin (HE) staining, enzyme-linked immunosorbent assay (ELISA), western blot (WB), quantitative real-time PCR (qRT-PCR), and immunofluorescence staining. RESULTS: Osthole improved skin damage and clinical dermatitis scores, reduced scratching bouts, and decreased epidermal thickness AD-like mice. It also reduced the levels of interleukin (IL)-31 and IL-31 receptor A (IL-31 RA) in both skin tissues and HaCaT cells. Furthermore, Osthole suppressed the protein expression levels of phosphor-p65 (p-p65) and phosphor-inhibitor of nuclear factor kappa-Bα (p-IκBα). Meanwhile, it increased the protein expression levels of peroxisome proliferator-activated receptor α (PPARα) and PPARγ in HaCaT cells. CONCLUSION These findings indicated that osthole effectively inhibited CP in AD by activating PPARα, PPARγ, repressing the NF-κB signaling pathway, as well as the expression of IL-31 and IL-31 RA.

Objective To explore the changes of interleukin -33 in serum of patients with chronic severe hepatitis B.Methods 25 patients with mild chronic hepatitis B ( MCHB group) and 25 patients with severe chronic hepatitis B(SCHB group) were collected.18 healthy people were collected as control (HC group).ELISA method was used to detect IL -33 level in peripheral blood .Results The IL-33 levels in MCHB and SCHB group were (11.7 ±6.9)ng/L and (16.5 ±9.1)ng/L,respectively,which were significantly higher than (5.1 ±2.9)ng/L in HC group(t=4.037,5.434,P=0.000,0.000).The IL-33 level in MCHB group was lower than that in SCHB group(t=2.108,P=0.040).The IL-33 level was positively correlated with ALT ,AST and HBV DNA(r=0.526, 0.568,0.285,P=0.000,0.000,0.045 ).Conclusion IL -33 is involved in the pathogenesis of hepatitis B . IL-33 plays an important role in HBV -relate injury.

Objective To explore the significance of interleukin -35 (IL -35)in serum of patients with severe hepatitis.Methods Collected 39 cases of chronic hepatitis B(CHB)patients and severe hepatitis,and col-lected 19 cases of healthy control's(HC)peripheral blood at the same time.ELISA method was used to detect IL -35 levels.Results IL -35 in CHB and severe hepatitis group [(483.5 ±280.7)ng/mL and (277.9 ±248.7)ng/mL] were higher than HC group (50.5 ±47.8)ng/mL(t =2.089,3.303,P =0.044,0.002).In severe hepatitis group, the IL -35 levels in survivors group (305.3 ±301.2)ng/mL was higher than death group (78.7 ±33.2)ng/mL (P =0.012).IL -35 was positively correlated with ALT and AST,the correlation coefficient were 0.649 and 0.599. Conclusion IL -35 is involved in the pathogenesis of hepatitis B process,the low serum IL -35 levels in severe hepatitis patients may herald a bad prognosis.

Objective To investigate the clinical value of combined detection of the serum levels of CA125,CA72-4 and tumor specific growth factor(TSGF)in the diagnosis of ovary cancer and the curative effect.Methods Serum levels of CA125,CA72-4 and TSGF in 68 patients with ovary cancer,53 patients with benign ovary rumor and 50 normal controls were measured by chemiluminescent immunoassay(CLIA)and chemical colorimetry.Results and clinical data were statisticaly analyzed.Results The levels of CA125,CA72-4 and TSGF in ovary cancer group were higher than that in the benign ovarian tumor group and healthy controls(all P ＜0.01).When use three tumor markers united detection,sensitivity,specificity and accuracy were 91.2％,83.0％ and 87.4％ respectively.The sensitivity and accuracy was higher than any single detection.The specificity with single CA125 testing was consistently.The levels of CA125,CA72-4 and TSGF in patients with ovary cancer were significantly different after surgical treatment for five days(all P ＜0.05).Conclusion The combined detection of the three markers may increase the positive rate in the early diagnosis of ovarian cancer,and help to differentiate the benign and malignant ovarian tumor,but also is valuable to observe the curative rate and postoperative monitor of the ovary cancer.
