1.Comprehensive analysis of the antibacterial activity of 5,8-dihydroxy-1,4-naphthoquinone derivatives against methicillin-resistant Staphylococcus aureus.

Given the increasing concern regarding antibacterial resistance, the antimicrobial properties of naphthoquinones have recently attracted significant attention. While 1,4-naphthoquinone and its derivatives have been extensively studied, the antibacterial properties of 5,8-dihydroxy-1,4-naphthoquinone derivatives remain relatively unexplored. This study presents a comprehensive in vitro and in vivo analysis of the antibacterial activity of 35 naturally sourced and chemically synthesized derivatives of 5,8-dihydroxy-1,4-naphthoquinone. Kirby-Bauer antibiotic testing identified three compounds with activity against methicillin-resistant Staphylococcus aureus (MRSA), with one compound (PNP-02) demonstrating activity comparable to vancomycin in minimum inhibitory concentration, minimum bactericidal concentration (MBC), and time-kill assays. Microscopic and biochemical analyses revealed that PNP-02 adversely affects the cell wall and cell membrane of MRSA. Mechanistic investigations, including proteomic sequencing analyses, Western blotting, and RT-qPCR assays, indicated that PNP-02 compromises cell membrane integrity by inhibiting arginine biosynthesis and pyrimidine metabolism pathways, thereby increasing membrane permeability and inducing bacterial death. In an in vivo mouse model of skin wound healing, PNP-02 exhibited antibacterial efficacy similar to vancomycin. The compound demonstrated low toxicity to cultured human cells and in hemolysis assays and remained stable during serum incubation. These findings suggest that PNP-02 possesses promising bioactivity against MRSA and represents a potential novel antibacterial agent.

2.Research on Construction of Performance Evaluation lndicators System of Traditional Chinese Medicine Doctors

Objective To clarify the connotation of TCM doctors'work performance,explore the evaluation indicators system of TCM doctors'work performance conforming to the characteristics of TCM,and provide a basis for reasonable measurement of TCM doctors'work performance.Methods The connotation of TCM doctors'work performance was sorted out by literature data method,the TCM doctors'work performance evaluation index was constructed in line with the characteristics of TCM by grounded theory method,and 412 TCM doctors from 1 1 hospitals in Zhejiang Province were investigated by questionnaire method,and the scientificity of the evaluation index was tested by project analysis and reliability and validity analysis.Results The connotation of TCM doctors'work performance was clarified,and three first-level indicators,14 second-level indicators and 23 third-level indicators,including diagnosis and treatment workload,work quality and characteristics,teaching and scientific research,were established.Project analysis showed that all indicators had good discrimination and homogeneity,Cronbach's coefficient showed that the constructed indicators had high internal consistency,and the 3 common factors extracted by exploratory factor analysis were consistent with the preset first-level indicators.Conclusion It scientifically constructs the performance evaluation index of TCM doctors,which can provide reference for optimizing the setting of performance evaluation index of TCM doctors.
3.Meta-analysis of the Clinical Efficacy of Acupuncture in the Treatment of Chronic Cough and the Study of Acupoint Selection Rules

Objective To systematically evaluate the effectiveness of acupuncture in treating chronic cough and analyze its acupoint selection patterns.Methods Randomized controlled trials(RCTs)on acupuncture for chronic cough were retrieved from databases including CNKI,VIP,Wan Fang Data,CBM,PubMed,and Web of Science,Cochrane Library and Embase from their inception to April 2025.Original data from the included studies were extracted.Meta-analysis was performed to compare the acupuncture treatment group with the western medicine control group[using leukotriene receptor antagonists(LTRAs)and/or glucocorticoids]on outcomes including cough symptom scores,serum tumor necrosis factor-α(TNF-α)levels,pulmonary function indicators[forced vital capacity(FVC)and forced expiratory volume in the first second(FEV1)],overall clinical effectiveness,and incidence of adverse reactions.Publication bias was assessed using funnel plots,and descriptive analysis of the acupoint prescriptions from the included studies was conducted using Rstudio software to summarize the patterns of acupoint selection for chronic cough.Results A total of 10 studies involving 806 patients were included.Meta-analysis results showed that acupuncture was superior to the western medicine control group in improving cough symptom scores,reducing serum TNF-α levels,enhancing pulmonary function(FVC and FEV1),increasing overall clinical effectiveness,and reducing adverse reactions,with statistically significant differences.Further analysis of the 10 included studies revealed that the most frequently used acupoints were Feishu(BL13)(n=9 times),Zusanli(ST36)(n=7 times),Lieque(LU7)(n=6 times),and Pishu(BL20)(n=5 times).The most frequently utilized meridians were the Bladder Meridian of Foot-Taiyang(BL)(n=10 times),the Stomach Meridian of Foot-Yangming(ST)(n=7 times),and the Lung Meridian of Hand-Taiyin(LU)(n=7 times).Acupoints were primarily located on the lower back(36.92%),upper limbs(26.15%),and lower limbs(21.53%).Conclusion Acupuncture is effective in treating chronic cough.The acupoint prescriptions for chronic cough should follow the principles of symptomatic point selection and anterior-posterior point pairing,emphasizing the back-shu and front-mu point pairing method,as well as the upper-lower point pairing method.Points are predominantly selected from the Bladder meridian(BL),lung meridian(LU),and stomach meridian(ST).

Spinal surgical site infection (SSI), especially deep SSI after internal fixation is difficult in treatment, with long course of disease and poor prognosis. At present, there are many controversies in the diagnosis and treatment of spinal SSI, with unsatisfactory overall efficacy of its diagnosis and treatment. Besides, no diagnosis and treatment guideline based on evidence-based medicine has been in existence. To this end, the Spinal Infection Group of the Orthopedic Branch of the Chinese Medical Doctor Association and the Spinal Infection Group of the Spinal Surgery Branch of the Chinese Rehabilitation Medicine Association jointly organized relevant experts to formulate Evidence-based clinical guideline for the diagnosis and treatment of surgical site infection in spinal trauma ( version 2024) based on an evidence-based approach. A total of 10 recommendations were proposed on the diagnosis and treatment of spinal SSI, so as to provide a clinical reference for the diagnosis and treatment of spinal SSI.

8.Effects of progesterone on the proliferation and expressions of TNF-α and IL-6 genes of human decidual stromal cells in late pregnancy in vitro

Objective:To study the effects of progesterone on the morphology, proliferation, and secretion of cytokines of human decidual stromal cells (DSCs) in late pregnancy, and to explore the mechanism of progesterone in preventing spontaneous preterm birth.Methods:Human decidual stromal cells in late pregnancy were cultured and treated with different concentrations of progesterone (in the experimental groups, 10 -6 mol/L, 10 -5 mol/L and 10 -4 mol/L progesterone was added to the culture medium respectively, and no progesterone was added to the culture medium of control group). The morphology of DSCs was observed under the microscope, the cell length/width ratio was measured, the proliferation was detected by methyl thiazolyl tetrazolium (MTT) method, and the expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) mRNA were detected by RT-qPCR. Results:The length/width ratios of DSCs in progesterone 10 -4 mol/L (5.87±0.19) and 10 -5 mol/L (5.98±0.27) groups were lower than that in control group (6.42±0.19), the differences were statistically significant ( P<0.001, P=0.002). The length/width ratio in the 10 -4 mol/L group was lower than that in the 10 -6 mol/L group (6.28±0.32, P=0.005). The proliferation of DSCs in the 10 -5 mol/L (0.70±0.04) and 10 -4 mol/L (0.78±0.04) groups was higher than that in control group (0.59±0.05; P=0.027, P=0.002), and proliferation of DSCs in 10 -4 mol/L group was higher than that in 10 -6 mol/L group (0.61±0.01, P=0.004). The expression of TNF-α mRNA in each progesterone group was lower than that in control group (all P<0.001) and the expression of TNF-α mRNA in the 10 -5 mol/L and 10 -4 mol/L groups was lower than that in the 10 -6 mol/L group (all P<0.001) . The expressions of IL-6 mRNA decreased gradually in control group, 10 -6 mol/L, 10 -5 mol/L and 10 -4 mol/L groups, the differences were statistically significant (all P<0.001). Conclusion:Progesterone can make the decidual stromal cells wider, promote proliferation, and decrease the expressions of TNF-α and IL-6 mRNA, which may play an important role in the mechanism of progesterone preventing spontaneous preterm birth.

10.Spindle assembly checkpoint complex-related genes TTK and MAD2L1 are over-expressed in lung adenocarcinoma: a big data and bioinformatics analysis.

OBJECTIVE:
To screen the key genes related to the prognosis of lung adenocarcinoma through big data analysis and explore their clinical value and potential mechanism.
METHODS:
We analyzed GSE18842, GSE27262, and GSE33532 gene expression profile data obtained from the Gene Expression Omnibus (GEO). Bioinformatics methods were used to screen the differentially expressed genes in lung adenocarcinoma tissues and KEGG and GO enrichment analysis was performed, followed by PPI interaction network analysis, module analysis, differential expression analysis, and prognosis analysis. The expressions of MAD2L1 and TTK by immunohistochemistry were verified in 35 non-small cell lung cancer specimens and paired adjacent tissues.
RESULTS:
We identified a total of 256 genes that showed significant differential expressions in lung adenocarcinoma, including 66 up-regulated and 190 down-regulated genes. Thirty-two up-regulated core genes were screened by functional analysis, and among them 29 were shown to significantly correlate with a poor prognosis of patients with lung adenocarcinoma. All the 29 genes were highly expressed in lung adenocarcinoma tissues compared with normal lung tissues and were mainly enriched in cell cycle pathways. Seven of these key genes were closely related to the spindle assembly checkpoint (SAC) complex and responsible for regulating cell behavior in G2/M phase. We selected SAC-related proteins TTK and MAD2L1 to test their expressions in clinical tumor samples, and detected their overexpression in lung adenocarcinoma tissues as compared with the adjacent tissues.
CONCLUSIONS
Seven SAC complex-related genes, including TTK and MAD2L1, are overexpressed in lung adenocarcinoma tissues with close correlation with the prognosis of the patients.
