Objective To analyze the application value of the American Society of Anesthesiologists (ASA) classification in treating patients with percutaneous nephrolithotomy (PCNL) under local anesthesia for upper urinary tract calculi. Methods A total of 80 patients with PCNL under local anesthesia for upper urinary tract calculi were divided into high-risk group (ASA Ⅲ to Ⅳ level) with 36 cases and low-risk group (ASA Ⅰ to Ⅱ level) with 44 cases according to ASA classification, and perioperative indicators (operation time, intraoperative blood loss and hospital stay), stone clearance rate, inflammatory factors[C reactive protein (CRP), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α)], score of the Visual Analogue Scale (VAS) for pain, and the incidence of complications were compared between the two groups. Objective:To explore the hospital-institute integration mode based on project cooperation.Methods:Focusing on the management innovation of clinical and basic scientific research cooperation mode, the method of hospital-institute integration mode based on project cooperation and the key points in practice were discussed, to develop better understandings of the construction and development such mode.Results:Several key points of developing hospital-institute integration mode based on project cooperation were proposed. Main measurements adopted including the integration of scientific research and clinic service, comprehensive construction of management concept, scientific research capacity, teaching organization and service concept, cultivation of high-level research talents, construction of research departments, according to which formed the main structure of the hospital. Furthermore, it is important to change the development mode in time to promote the healthy development of the hospital.Conclusions:It is a new direction of the clinical and basic research cooperation innovation to encourage the hospital-institute integration mode based on project cooperation. It is also an effective strategy to promote the improvement of diagnosis and treatment level under the new medical reform situation.

Objective:Explore the " project-talent-base" combining international science and technology cooperation management model and mechanism through summarizing the practical experience of a tertiary pediatric hospital in Beijing in recent years, to provide reference for strengthening the hospital′s international science and technology cooperation.Methods:Main problems existing in the management of international science and technology cooperation in domestic medical institutions are analyzed, and management methods of international science and technology cooperation adopted by the hospital are classified through policy sorting, literature research, summarization and classification.Results:The hospital has established an efficient and high-quality international science and technology cooperation management model mainly through four aspects: improving the international cooperation management system, enriching international cooperation and exchange projects, building different types of international science and technology cooperation platforms, and strengthening personnel training.Conclusions:Under the background of the globalization of science and technology and the accelerating pace of scientific research in China, improving the hospital′s international science and technology cooperation management model will effectively promote the introduction of international advanced technologies and concepts, and enhance the overall science and technology innovation capabilities and discipline construction.
4.Expression of CXC chemokine receptor 4 in clear cell renal cell carcinoma

Objective To investigate the expression of CXC chemokine receptor 4 (CXCR4) in clear cell renal cell carcinoma (ccRCC) and its relationship with the clinical pathological parameters of ccRCC.Methods The expression of CXCR4 was detected by immuno-histochemistry method in 63 cases of ccRCCs,20 cases of para-carcinoma tissues and 20 cases of normal renal tissues.The correlation between expression level of CXCR4 and clinical pathological parameters of ccRCC patients were analyzed,and the clinical significance of its expression in ccRCC was evaluated.Results The positive expression rate of CXCR4(49.2%) in ccRCCs was significantly higher than that in para-carcinoma tissues (15%) and normal renal tissue (10%),and the difference was statistically significant (P < 0.05).The expression level of CXCR4 and the clinical stage and pathological grade of ccRCC were correlated (P < 0.05),and was associated with lymph node transfer (P < 0.05).The CXCR4 negative group overall survival rate [55.2% (16/29)] and the average survival time(46 months) was significantly better than the positive group [38.5% (10/26),32 months;P < 0.05].Conclusions The expression level of CXCR4 in ccRCC is correlated with the clinicopathological parameters and prognosis.CXCR4 is expected to be an important marker for diagnosis and prognosis evaluation of renal cell carcinoma.
5.Study on Extraction of cpDNA from Endangered Medicinal Plant of Gentiana straminea Maxim . and Gentiana crassicaulis Duthie ex Burk

This study was aimed to show the extraction research of cpDNA in Gentiana straminea Maxim. and G. crassicaulis Duthie ex Burk., which will increase the knowledge about the chloroplast genome sequence characteris-tics, understand its genetic diversity and improve the efficiency of breeding schemes. G. straminea Maxim. and G. crassicaulis Duthie ex Burk. were taken as study objects. The improved sucrose-gradient centrifugation and purifica-tion methods were used to obtain the cpDNA, measure the concentration, detect the OD value, and amplify the ITS2 sequence to verify the nucleus pollution. The results showed that the high quality and purity cpDNA (0.1 - 0.4 μg/10 g) was demonstrated without other pollution after ITS2 sequence amplification, which met to the subsequent effi-cient sequencing of chloroplast genomes. The purity and mass have an important influence on the accuracy and cred-ibility of the cp genome sequencing. It was concluded that this study improved the sucrose-gradient centrifugation and purification with great effect of the purity and mass of cpDNA, and guaranteed the obtaining of complete cpDNA of Gentiana.
6.Regulative effect of substance P on proliferation of human skin fibroblast and expression of its monocyte chemoattractant protein-1 under high glucose condition

Objective To investigate effects of exogenous substance P (SP) on proliferation of humau skin fibroblast (HSF) and expression of its monocyte chemoattractant protein-1 (MCP-1) under high glucose condition.Methods HSF cultured in high glucose DMEM medium were treated with SP of various concentrations at different time points.Levels of MCP-1 in the supernatants were determined by ELISA and time-effect relationship of SP regulating expression of MCP-1 was analyzed.Expression of MCP-1 mRNA in HSF was detected by semi-quantitative RT-PCR and Real Time-PCR,Conclusion,while cxpression of MCP-1 by Western blot.Proliferation of HSF treated with SP of different concentrations was detected by cell counting Kit-8 (CCK-8).Results MCP-1 had a strong expression at 8 hours after SP disposal (P <0.05) and reached the peak at 24 hours (P < 0.01).Meanwhile,MCP-1 expressed the most at 10 nmol/L and 100 nmol/L SP (P <0.01).SP significantly enhanced the expression of MCP-1 mRNA and its synthesis under high glucose condition,especially at the concentration of 10 nmol/L (P <0.01).Also,SP induced obvious proliferation of HSF at concentrations of 10 nmol/L and 1 000 nmol/L(P < 0.05).Conclusion SP promotes proliferation of HSF and expression of MCP-1 in high glucose DMEM medium,which may be of significance in promoting diabetic wound healing.
7.A marine anthraquinone SZ-685C inhibits cell proliferation and induces apoptosis in GH3 cells

Objective To explore the effect of SZ-685C on rat pituitary adenoma GH3 cell line.Methods MTT method evaluated its effect of proliferation and inhibitory concentration 50 (IC5o) on GH3 cells,after treated by 0,2.5,5.0,7.5,10.0,12.5,15.0,17.5,20.0 and 30.0 μmol/L SZ-685C for 48 h,GH3 cells were changed to complete medium for 12 d,crystal violet staining was used to investigate colony formation; microscopy and Hoechst 33342 staining assay observed whether the changes of morphological as the result of apoptosis,then detected cell apoptosis by flow cytometry.Results SZ-685C had a dose-dependent inhibitory effect on GH3 cell proliferation and IC50 was (12.9 ± 0.47) μmol/L,MEF(considered as a control group) had little affect in cell proliferation on the concentration of IC50.Inhibitory effects persisted even on removal of SZ- 685C after 12 d,and SZ-685C blocked colony formation ability of pituitary tumor cells.Apoptotic morphological observation of microscope and Hoechst 33342 staining proved apoptosis during treatment of SZ-685C,flow cytometry detection showed that SZ-685C induces 36.4% of apoptosis,while only 2.0% in control group.Conclusion SZ-685C inhibits pituitary tumor cell proliferation and induces apoptosis.SZ-685C can be a new anti-patuitary tumor drug for a further study.
8.Studies of modifying effects of fullerene on ~(60)Co - γ irradiated zebrafish

Objective To evaluate the modification of C_(60) on the radiation effects of ~(60)Co-γ irradiation on zebrafish.Methods The adult and embryonic zebrafish were used as model organisms to examine the potential of C_(60) to elicit oxidative stress responses on the surviving rate,hatching rate and malformation occurrence,both upon exposure to light or in the dark.Reactive oxygen species (ROS) production and DNA damage were examined as the possible underlying mechanism.Results 500 × 10~(-9) nano-C_(60) waterborne exposure could enhance the γ-irradiation effects by decreasing adult fish survival upon light exposure,which resulted in ROS and DNA damage increasing.The hatching rates were also inhibited with higher malformation,though dark exposure did not make any enhancement,except that the 5000× 10~(-9) C_(60) would inhibit larvae hatching and induced more malformation.Conclusions Waterborne nano-C_(60) exposure may enhance the radiation effects on zebrafish,ROS production and DNA damage increasing may be the underlying mechanism.

The effect of interventional therapy combined with multifactorial intervention on critical limb ischemia in patients with diabetes mellitus was investigated. The patency rate and limb salvage rate were followed up. Interventional therapy is effective in treating diabetic foot with critical limb ischemia. Multifactorial intervention was helpful for reducing amputation.
10.The modified rhinotomy for treatment of tumors involving skull base.

OBJECTIVE:
To introduction the clinical application and surgical technique of the modified rhinotomy for treatment of skull base neoplasms.
METHOD:
The diagnosis and surgical outcome of 20 patients with skull base neoplasms were reviewed retrospectively.
RESULT:
All the cases were involvement with nasal cavity, ethmoidal sinus, sphenoidal sinus and medial wall of orbit. Two cases were involvement with pterygopalatine fossa, infratemporal fossa and intra calvarium separately. All cases were treated by modified rhinotomy. Among that 2 patient were with juvenal angiofibroma treated by combined with maxillary transposition and pterional and zygomatic approach, and 2 cases were with craniofacial approach. No complications were found relating to surgery. 6 patients was recurrent during follow-up among 6 to 60 months.
CONCLUSION
The modified rhinotomy is a effective and safe surgical approach to treat skull base neoplasms located in nasal cavity, ethmoidal sinus and sphenoidal sinus. Tumor involved with pterygopalatine fossa, inferotemporal fossa and intracranial extension, other surgical approach should be used at the same time.
