1.Differential analysis of biogas production in simulated experiments of aquitard layers in coal seam fire zones.

To explore the differences in biological gas production in the waterlogged zone of a coal seam fire-affected area, in this study the in-situ gas production experiment was conducted with the mine water from aquitard layers in coal seam fire zones in Xinjiang. The results showed that the biogas production first increased and then decreased with the increase in distance, and the highest gas production reached 216.55 mL. The changes in key metabolic pathways during the anaerobic fermentation of coal were analyzed, which showed that as the distance from the aquitard layer in the coal seam fire zone increased, the methanogenesis pathways gradually shifted from acetic acid decarboxylation and carbon dioxide reduction to acetic acid decarboxylation and methylamine methanogenesis. The significant variability in the in-situ mine water reservoir conditions contributed to the differences. In addition, the reservoir pressure and temperature increased as the distance from the fire zone became longer, and the salinity of the farthest mine water in the reverse fault was the highest due to the lack of groundwater supply. Pearson correlation analysis revealed significant correlations of microbial communities with key functional genes and the types and concentrations of ions. The ions significantly influencing microbial enzymatic metabolic activities included Al3+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+, Mg2+, PO43-, and Mo6+. The differences in metabolic pathways were attributed to the integrated effects of a co-occurring environment with multiple ions. The gas production simulation experiments and metagenomic analyses provide data support for the practical application of in-situ biogas experiments, laying a foundation for engineering applications.

2.A new variant of respiratory syncytial virus glycoprotein gene and clinical features of infected children

Objective:To analyze the epidemiological features of respiratory syncytial virus (RSV) infection in Beijing, and monitor the sequence variations in RSV glycoprotein (G) gene and clinical features of infected children.Methods:Respiratory tract specimens were collected from children with acute respiratory infection in the Children′s Hospital Affiliated to Capital Institute of Pediatrics from January 1, 2023 to December 31, 2023. RSV-positive specimens screened by multiple nucleic acid testing were subjected to PCR to amplify the full-length RSV G gene. A phylogenetic tree was constructed after gene sequencing to analyze RSV subtypes and trace G gene variants. Clinical data were retrieved from the medical record system to analyze the clinical features of children with RSV infection in Beijing.Results:A total of 5 489 respiratory specimens were collected from 3 046 male patients and 2 443 female patients. The average age of the patients was 4.36 years. A total of 589 RSV-positive specimens (10.7%, 589/5 489) were detected with 349 from male patients and 240 from female patients. The average age of children with RSV infection was (2.51±2.78) years and the median age was 0.48 years. RSV had been circulating among children in Beijing since March 2023 with two epidemic peaks in May (24.6%, 122/496) and December (18.2%, 126/693). The predominant subtype of RSV in the first half of 2023 was subtype A, but it was replaced by subtype B from November 2023. Phylogenetic analysis revealed a novel G gene of RSV subtype B (RSV-B-BA9-954bp) with a length of 954 bp, which belonged to a new cluster in the phylogenetic tree. The percentage of patients admitted to the Intensive Care Unit (ICU) was higher in children with new variant of RSV subtype B infection than in those with common RSV subtype B infection [44.1% (15/34) vs 25.2% (31/123), χ 2=4.600, P=0.032], while the counts of white blood cells and the levels of C-reactive protein were lower in the children with new variant infection ( P<0.05). Conclusions:RSV has been prevalent among children in Beijing since March 2023 with two epidemic peaks. The predominant A subtype is gradually replaced by to B subtype. A new variant of RSV B G gene (RSV-B-BA9-954bp) is detected among the children.
3.Effects of grain-sized moxibustion on Th1/Th2 balance and transcription factors T-bet and GATA3 in immunosuppressed mice induced by cyclophosphamide

Objective To observe the effects of grain-sized moxibustion on Th1 cell/Th2 cell(Th1/Th2)balance and transcription factors T-box transcription factor(T-bet)and GATA binding protein 3(GATA3)in immunosuppressive mice induced by chemotherapy.Methods According to the random number table method,32 SPF male CD-1(ICR)mice were randomly divided into the normal group,model group,levamisole hydrochloride group,and grain-sized moxibustion group,with eight mice per group.Except for the normal group,the immunosuppressive model was established by intraperitoneal injection of cyclophosphamide(80 mg/kg,once daily for three consecutive days).Mice in the levamisole hydrochloride group were given levamisole hydrochloride solution(10 mg/kg)by gavage.Mice in the grain-sized moxibustion group was given grain-sized moxibustion at"Guanyuan"(CV4),bilateral"Zusanli"(ST36),and bilateral"Sanyinjiao"(SP6),with three Zhuang at each acupoint for approximately 30 s each.The intervention was administered once daily for seven consecutive days.The general condition of mice was observed.The spleen mass and spleen index were detected.The pathological changes of spleen tissue were observed by HE staining.The protein and mRNA expressions of T-bet,GATA3,interferon-γ(IFN-γ),and interleukin(IL)-4 in spleen tissue of mice were detected by Western blotting and real-time PCR.The contents of IFN-γ,IL-2,and IL-4 in serum of mice were detected by enzyme-linked immunosorbent assay.Results Compared with the normal group,the mice in the model group were slow in response,unstable in gait;the spleen weight and spleen index were increased(P<0.05);the structure of spleen tissue was disordered,the mRNA and protein expressions of T-bet and IFN-γ in spleen tissue were decreased,and the mRNA and protein expressions of GATA3 and IL-4 were increased(P<0.05);the contents of IFN-γ and IL-2 in serum were decreased,and the content of IL-4 was increased(P<0.05).Compared with the model group,the general condition of mice in the levamisole hydrochloride group and the grain-sized moxibustion group was improved,the structure of spleen tissue was improved,the mRNA and protein expressions of T-bet and IFN-γ in spleen tissue were decreased,and the mRNA and protein expressions of GATA3 and IL-4 were increased(P<0.05);the contents of IFN-γ and IL-2 in serum were decreased,and the content of IL-4 was increased(P<0.05).Conclusion Grain-sized moxibustion can significantly improve the immunosuppressive symptoms induced by chemotherapy.The mechanism may be through regulating the expressions of transcription factors T-bet and GATA3,regulating Th1/Th2 balance,and thus restoring the immune balance.
4.Application of susceptibility-weighted imaging in intracranial dural arteriovenous fistula

Objective:To investigate the application value of susceptibility-weighted imaging (SWI) in the diagnosis of intracranial dural arteriovenous fistula (DAVF).Methods:Patients with DAVF confirmed by digital subtraction angiography (DSA) in Weihai Municipal Hospital from January 2014 to January 2021 were retrospectively included. All patients underwent conventional T 1-weighted imaging (T 1WI), T 2-weighted imaging (T 2WI) and SWI, and some patients also underwent 3D time-of-flight magnetic resonance angiography (3D-TOF-MRA). Results:A total of 36 patients with DAVF were enrolled, 29 of them received 3D-TOF-MRA. The fistula location of 24 patients (24/36, 66.7%) underwent SWI and 26 patients (26/29, 89.7%) underwent 3D-TOF-MRA were correctly judged, and the difference was statistically significant (Fisher's Exact Test, P=0.039). SWI showed that the proportion of patients with thickened supply arteries (7/36, 19.4%) was significantly lower than that on 3D-TOF-MRA (14/29, 48.3%; χ2=6.105, P=0.013). T 2WI, 3D-TOF-MRA and SWI showed no cerebral venous abnormalities in all 7 patients with DAVF without cortical venous reflux; in 29 patients with DAVF with cortical venous reflux revealed by DSA, SWI and T 2WI showed all patients (100%) and 26 patients (89.7%) had superficial venous dilatation respectively, but there was no significant difference ( χ2=0.693, P=0.405). SWI showed medullary vein dilation in 17 patients (47.2%), and only 2 patients (5.6%) had medullary vein thickening on T 2WI, and the difference was statistically significant ( P<0.001). The proportion of patients with venous cerebral infarction on T 2WI was significantly higher than that on SWI (22.2% vs. 0%; Fisher’s Exact Test P=0.005), and the proportion of patients with intracerebral hemorrhage on SWI was significantly higher than that on T 2WI (61.1% vs. 25.0%; χ2=9.574, P=0.004). Conclusion:SWI is helpful to evaluate the abnormal drainage vein of DAVF and the secondary changes in brain, especially intracerebral hemorrhage.
5.Genetic characterization analysis of the whole genome sequence of Coxsackievirus A8 associated with hand, foot and mouth disease in China

Objective:To study the genomic sequence of Coxsackievirus A8 (CV-A8) associated with hand, foot and mouth disease (HFMD) from 2013 to 2018 in China and to analyze the genetic evolution of each coding region of the full-length genome.Methods:The genome sequences of 11 CV-A8 strains isolated from patients with HFMD in different regions of China from 2013 to 2018 were determined. Sequence alignment and genetic evolution analysis were performed by Sequencher 5.0 and MEGA 7.0 software, etc.Results:Sequence alignment showed that the genome length of 11 CV-A8 strains ranged from 7 393 bp to 7 400 bp. There was no base insertion or deletion in the coding region compared with the prototype strain, but there were individual base insertion or deletion in the non-coding region. The nucleotide and amino acid similarities in the VP1 region of 11 CV-A8 strains were 78.3%-98.6% and 92.6%-99.7%, respectively, and the nucleotide and amino acid sequences identities with the CV-A8 prototype strain were 78.3%-98.2% and 92.6%-99.7%, respectively. Based on the phylogenetic analysis of VP1 region sequences, the CV-A8 can be divided into five genotypes: A, B, C, D and E. The 11 CV-A8 strains in this study belonged to genotypes C (1 strain), D (2 strains) and E (8 strains). The nucleotide and amino acid similarities of 11 CV-A8 full-length genomes were 81.3%-98.8% and 95.9%-99.5%, respectively. The phylogenetic tree of the P2 region showed that the eight E genotypes CV-A8 had the closest evolutionary distance with CV-A4, CV-A14, and CV-A16. The phylogenetic tree of the P3 region showed that the eight E genotypes CV-A8 had a close evolutionary distance with CV-A5, CV-A16, CV-A14 and CV-A4.Conclusions:The 11 CV-A8 stains in this study showed significant intra-genotype diversity in capsid region and recombinant diversity in non-capsid region which indicated that CV-A8 quasispecies were still undergoing dynamics variation. CV-A8 may become an important pathogen of HFMD and the monitoring of CV-A8 needs to be further strengthened.
6. Clinical effect of traditional Chinese medicine directing therapy in the treatment of pelvic inflammatory diseases sequelae with syndrome of Qi stagnation and blood stasis

Objective:
To observe the clinical efficacy of traditional Chinese medicine(TCM) directing therapy in the treatment of pelvic inflammatory disease sequelae (SPID) with the syndrome of Qi stagnation and blood stasis.
Methods:
From April 2016 to April 2018, 60 patients of SPID with the syndrome of Qi stagnation and blood stasis in the Traditional Chinese Medicine Hospital of Pudong New Area of Shanghai were randomly divided into the treatment group and the control group according to the digital table, with 30 cases in each group.The control group was treated with FukeQianjin capsules, and the treatment group was treated with TCM directing therapy, with a course of 1 month.The clinical efficacy was observed, and the TCM syndrome score and gynecological examination score were compared.
Results:
The total effective rate of the treatment group was 96.67%(29/30), which was significantly higher than 80.00%(24/30) of the control group (χ2=4.043,

ObjectiveTo summarizeand the law of lymph node metastases from adenocareinoma of esophagogastric junction ( AEG),analyses the extent of lymphadenectomy for AEG.Methods198 cases of AEG had been retrospectively analyzed from 2006.6.6 to 2010.12.31,and kept detailed record of the operation type、Siewert's type、TNM stage and group of metastatic lymph nodes.ResultsThe thoraco-abdominal two field lymph node dissection had been performed for 198 AEG,the total number of lymph nodes had been dissected was 3069,the average number was 15.50,the number of metastases lymph nodes was 859.In the 198 AEG,132 cases with lymph node positive,The metastatic rate and incidence was 66.67% and 27.99%,respectively.The manner of the metastasis about 130 cases was station by station and 2 cases was skipping over station,accout for 98.48% and 1.52% of all nodes-positive cases,1 case skiped to left gastric and celiac axis and another case skiped to tracheal bifurcation.The location of positive lymph nodes about Siewert Ⅰ and Ⅱ were recorded:tracheal bifurcation(6.25%,3.33% ) 、lower posterior mediastinum and paraoesophageai(25.00%,14.67% ),right paracardiac (47.92%,52.00% ),left paracardiac (31.25%,36.67% ) 、lesser curvatura(43.75%,43.33% ),greater curvatura( 27.08%,22.00% ) 、left gastric and celiac axis( 27.08%,30.00% ).The lymph nodes metastastic rate of stage T1 + T2 and T3 + T4 were 40.43%,74.83%,respectively,the difference were significantly.The lymph nodes metastastic rate of different tumor length were analyzed,≤ 3cm group were 40.74%,3 ~5cm group were 70.71% and ≥5cm group were 88.89%,the difference were significantly;but there were no significant difference related to different differentiation grades and different Siewert types.The mediastinal lymph node metastase rates between Siewert Ⅰ (27.08%) and Siewert Ⅱ (14.67%) were significant difference,but the difference were not found in pericardiac lymph nodes.ConclusionSiewert Ⅰ 、Ⅱ AEG trend to metastasized to middle and low mediastinal and pericardiac,the modified left thansthoracic route and two field lymph node dissection maybe suitable to the lymphadenectomy for AEG.

9.Correlation analysis between cerebral microbleed and stroke subtype

Objective To investigate the influence of microbleeds on the onset and development of different types of stroke. Methods According to stroke subtypes, 163 patients were classified into lacunar infarction (n= 56), transient iscbemia attach (n=31), atherothrombotic infarction (n=37), intracerebral hemorrhage (n=39), and control groups (n=43). Suaceptibility-weighted imaging , T1WI, T2WI and diffusion-weighted imaging were performed with 3.0 T system to observe cerebral microbleed and infarction. Results The incidence and the number of micrehleeds were significantly greater in patients with intracerebral hemorrhage (75.6% and 14.5±11.6, re-spectively),then lacunar infarction and therothrombetic infarction atherothrombotic infarction [(55.3% and 8.1± 3.4), (37.8% and 4.5±2.6)] and the incidence and the number of microhleeds in above groups are higher than transient ischemia attach (9.7% and 0.3±0.1), and controls (9.3% and 0.2±0.1) (P<0.01). There was a correlation between the number of microbleeds and the severity of lacunar infarction, aslo a correlation between the number of microbleeds and the number of intracerebral hemorrahages (r=0.402, P<0.001). Conclusion Microb-leeds are closely associated with stroke, which may indicate more advanced microangiopathy and higher risk of bleeding.
10.Effects of Bushen Huayu Qutan Recipe on local ovarian factors in androgen-sterilized rats

OBJECTIVE: To explore the effects of Bushen Huayu Qutan Recipe (BHQR), a compound traditional Chinese herbal medicine, on local ovarian factors in androgen-sterilized rats (ASR). METHODS: One hundred and five 9-day-old SD rats were randomly divided into normal control group (15 rats) and ASR group (90 rats). ASR was induced by subcutaneous injection of testosterone propionate, and the rat model of ASR was established successfully in 30 rats in the ASR group. Thirty ASR rats were randomly divided into untreated group and BHQR-treated group. There were 15 rats in each group. The rats in BHQR-treated group were intragastrically administered with BHQR. The rats in the normal control and untreated groups were intragastrically administered with distilled water. They were all treated for 30 days. The levels of serum testosterone and insulin (INS) were measured by radioimmunoassay. The inhibin (INH) level, and the expressions of insulin-like growth factor-I(IGF-I) and vascular endothelial growth factor (VEGF) in ovary were determined by immunohistochemical method. RESULTS: None of the ASR ovulated and all the rats in the normal control group ovulated. The rate of ovulation in BHQR-treated group was 66.67%. The serum concentrations of testosterone and INS in the untreated group were significantly higher than those in the normal control group (P<0.05), and the serum concentrations of testosterone and INS in BHQR-treated group were decreased as compared with the untreated group (P<0.05). The levels of ovarian INH, IGF-I and VEGF in the untreated group were significantly higher than those in the normal control group (P<0.01), and the levels of ovarian INH, IGF-I and VEGF in BHQR-treated group were decreased as compared with the untreated group (P<0.01). CONCLUSION: BHQR is an effective recipe in promoting ovulation, and the effects of BHQR on balancing the internal environment of ovary may be due to the reduction of serum INS level and decrease in the expressions of ovarian INH, IGF-I and VEGF.
