OBJECTIVE To study the effects of sophoranone （SOP） on the biological behavior of nasopharyngeal carcinoma CNE-1 cells and mitogen-activated protein kinase （MAPK） signaling pathway. METHODS CNE-1 cells were divided into blank group and SOP low-， medium- and high-concentration groups （SOP-L group， SOP-M group， SOP-H group， 25， 50 and 100 μmol/L）. The number of invasive cells， the number of migratory cells， and the apoptosis rate of cells were detected. The expression levels of mitogen-activated protein kinase kinase （MEK）， extracellular signal-regulated kinase 1 （ERK1）， ERK2， and c-Jun N-terminal kinase （JNK） mRNA， as well as phosphorylation levels of ERK， JNK， and p38 mitogen-activated protein kinase （abbreviated as “p38”） proteins in cells were all detected. Additionally， cells were divided into blank group， SOP high-concentration group （SOP- H group， 100 μmol/L）， SOP high-concentration combined with p38 inhibitor group （SOP-H+SB group， 100 μmol/L SOP+10 μmol/L SB）， and SOP high-concentration combined with JNK inhibitor group （SOP-H+SP group， 100 μmol/L SOP+10 μmol/L SP）. The number of invasive cells， cell migration rate， and the protein phosphorylation levels of JNK and p38 in cells， as well as the protein expression levels of matrix metalloproteinase-9（MMP-9）， proliferating cell nuclear antigen Ki67， and cleaved-caspase-3 were measured. RESULTS Compared with the blank group， SOP for each concentration could significantly decrease the number of invasive cells， the number of migratory cells， and mRNA expressions of MEK， ERK1， ERK2 （except for the SOP-L group） and JNK， but increase the apoptosis rate of cells and phosphorylation levels of ERK， JNK， and p38 proteins （P＜0.05）. Compared with the SOP-H group， the protein phosphorylation levels of p38 and JNK， and the protein expression of cleaved-caspase-3 were decreased significantly in SOP-H+SB group and SOP-H+SP group， while the number of invasive cells， cell migration rate， and the protein expression levels of MMP-9 and Ki67 were all increased significantly （P＜0.05）. CONCLUSIONS SOP can inhibit the proliferation， migration and invasion of CNE-1 cells， and induce the apoptosis， the mechanisms of which may be associated with promoting the phosphorylation of proteins related to the MAPK signaling pathway.

Objective:To understand trends and hotspots of TCM research papers published in journals with high IF basing on a bibliometric analysis.Methods:TCM research papers published between January 1, 2014 and December 31, 2024 from 80 journals with IF higher than 16.0 were retrieved from medical, life sciences and comprehensive journals in Journal Citation Reports of 2024. CiteSpace 6.3.R1 and Excel 2021 were used to analyze and visualize annual publication volume, research fields, features of clinical study, institutes, funds and keywords.Results:A total of 51 papers were included, showing an increasing trend in annual publication volume; the main research areas were pharmacology, acupuncture&moxibustion and internal medicine. Multi-center randomized controlled trials were the main clinical studies; China Academy of Chinese Medical Sciences was the leading institute in terms of publication volume; 84 funds were involved, including National Natural Science Foundation of China. Keywords that appeared most frequently were TCM, efficacy, double blind, electroacupuncture, stimulation and management.Conclusion:The number and quality of TCM research papers are improved simultaneously; future research needs to deepen international cooperation, and pay attention to the integration of TCM diagnosis and treatment characteristics and modern scientific research methods.

Objective: To explore the effects and mechanisms of interfering with transient receptor potential melastatin subfamily member 7(TRPM7) on biological behaviors(proliferation, apoptosis, invasion) of laryngeal carcinoma cells. Methods: Human laryngeal carcinoma cells TU212 were culturedin vitro. TRPM7-shRNA plasmid vectors(TRPM7-shRNA1 group, TRPM7-shRNA2 group, TRPM7-shRNA3 group) and negative control shRNA-NC(shRNA-NC group) were constructed. TU212 cells were transfected by liposome transfection method. The cells transfected with empty vector were enrolled as Control group. The level of lactic dehydrogenase(LDH) in TU212 supernatant was detected by ELISA. The level of malondialdehyde(MDA) and activity of superoxide dismutase(SOD) in supernatant were detected by colorimetry. The effects of knocking-down TRPM7 on proliferation and invasion of TU212 cells were detected by CCK-8, clone formation assay and Transwell assay. The expressions of invasion and apoptosis-related proteins were detected by Western blot. Results: After transfection, expression levels of TRPM7 mRNA and protein were down-regulated in TRPM7-shRNA1 group, TRPM7-shRNA2 group and TRPM7-shRNA3 group compared with Control group(P<0.05), and the decrease was the most significant in TRPM7-shRNA1 group(P<0.05). In functional experiments, SOD level in TRPM7-shRNA1 group decreased compared with Control group(P<0.05), while MDA and LDH levels increased(P<0.05). The cells proliferation rate and clone formation rate were decreased(P<0.05), the number of invasion cells was reduced(P<0.05), the expressions of N-cadherin and Vimentin proteins were down-regulated(P<0.05), mitochondrial membrane potentials were reduced(P<0.05), Bax/Bcl-2 and cleaved caspase-3/caspase-3 increased(P<0.05). Conclusion Knocking-down TRPM7 can increase oxidative stress level in laryngeal carcinoma cells TU212, inhibit their proliferation and invasion, and induce their apoptosis by mitochondrial pathways.

Objective To investigate the pollution levels of six indicator polychlorinated biphenyls (PCBs) in animal-derived foods in Hebei Province. Methods A total of 96 food samples (including beltfish, snakehead, egg, pork liver, pork and beef meat) were collected from Hebei Province in 2019. The contents of six indicator PCBs (PCB 28, PCB 52, PCB 101, PCB 138, PCB 153 and PCB180) in different food samples were determined using isotope dilution-high resolution gas chromatography-high resolution mass spectrometry (HRGC-HRMS). Results The contents of indicative PCBs in fish samples ranged from 0.12 to 0.32 ngg-1 fresh weight, while the content of PCBs in other samples ranged from 0.11 to 1.10 ng g-1 fat. PCB 138 and PCB 153 were the main compounds detected in fish samples, while PCB 28 and PCB 52 were the main compounds detected in other samples. The average estimated daily dietary intake of PCBs for residents in Hebei Province was 0.14 ng kg^-1·bw^-1·d^-1, and the highest contribution rate was beltfish (61.52%). Conclusion The pollution levels of indicator PCBs in animal-derived foods in Hebei Province are relatively low, and the residents has lower health risks through dietary intake of indicator PCBs.

Objective To investigate the effect of potassium channel blocker tetraethtylamine (TEA) on the proliferation and apoptosis of human laryngeal carcinoma cells (Hep-2) through the PI3K/Akt signaling pathway.Methods TEA acted on HEp-2.The methyl thiazolyl blue (MTT) method was used to determine the Hep-2 cell activity and the proliferation inhibition rate of TEA on Hep-2 was calculated;the Hoechest33258 staining was used to detect cell apoptosis,and the Hep-2 apoptosis rate was detected by flow cytometry (FCM) assay.Results After inoculation by 5,10,20 mmol/L TEA,the Hep-2 cell proliferation inhibition rate reached 12.573%,31.385% and 56.132%,respectively.After 96 h Hep-2 cell action,the cellular apoptosis rates were (41.64±2.67)%,(58.76±4.32)% and (72.65±6.54)% respectively,the inhibition rate and apoptosis rate in the TEA treating group were significantly higher than those in the non-TEA treating group,the differences were statistically significant (P<0.05).Conclusion TEA can inhibit the proliferation and in vivo growth of human laryngeal carcinoma cells and promotes the apoptosis of laryngeal cancer cells.

OBJECTIVE: To investigate the myeloid differentiation factor 88 (MyD88) expression in laryngeal carcinoma and its clinical significance. METHOD: Fifty-one patients with laryngeal carcinoma were collected, and all patients were confirmed by pathological diagnosis results. The expression of MyD88 protein was detected by immunohistochemical method in laryngeal cancer and its adjacent tissues to investigate the correlation among MyD88 expression, clinicopathological characteristics and prognosis of patients. RESULT: The positive expression rate of MyD88 in laryngeal cancer tissues was 68.6%, significantly higher than that in normal tissues adjacent to carcinoma of which positive expression rate was 11.8%; MyD88 positive rate had nothing to do with laryngeal cancer patients age, sex, differentiation and tumour location (all P > 0.05), but correlated with clinical stage (P < 0.01) and lymph node metastasis (P < 0.05). In addition, the study also found that the expression of MyD88 quantity was inversely proportional with the five-year survival rate. The survival rate of patients with higher expression of MyD88 was significantly lower than that of patients with lower expression (P < 0.05). CONCLUSION MyD88 may be an important participant in the pathogenesis of laryngeal carcinoma, MyD88 targeted therapy may improve the prognosis of patients with laryngeal cancer.

OBJECTIVE: To investigate the diagnosis applying effects of ocular vestibular evoked myogenic potentials(oVEMP) in peripheral BPPV disease. METHOD: During September 2012 to January 2015, we selected 80 healthy people in our hospital medical center as the control group, choose the same period of primary benign paroxysmal positional vertigo as the observation group of 80 patients. Two groups were carried out fully functional auditory evoked potential analysis, determination of oVEMP and cervical vestibular evoked myogenic potentials (cVEMP) anomaly amplitude threshold, P1 latencies, N1 incubation period. RESULT: The cVEMP abnormal rate in the observation group was 28.8%, the oVEMP abnormal rate was 38.8%, while cVEMP and oVEMP abnormal rates in the control group was 1.3% and 2.5% respectively that compared to significant differences between the two groups (P < 0.05). The oVEMP test amplitude in the observation group was (5.98 ± 2.15) µv, the N1 incubation period was (10.03 ± 0.76)ms, while the control group were (4.09 ± 2.11)µv and (11.67 ± 0.78) ms that compared difference were statistically significant (P < 0.05). The cVEMP test amplitude in the observation group was (154.8 ± 43.9)2 µv, while the control group was (180.49 ± 45.34)µv, compared the difference was statistically significant (P < 0.05). CONCLUSION Paroxysmal positional vertigo patients ocular vestibular evoked myogenic potentials abnormal rate is relatively high, the utricle dysfunction for more severe than the balloon can be the subject of an objective function of the ear stone judgment, judgment in favor of the disease.

Objective To investigate the characteristics and distribution of class teaching skill points in state high quality course pathology using Flanders interaction analysis system to analyze the teaching video of pathology course.Methods Teaching videos of state high quality course (pathology course) between 2006 and 2012 were analyzed by Flanders interaction analysis system.Frequencies of using teaching skill,presentation skill and interaction skill were statistically analyzed.Results The study found that frequencies of using teaching skill points in pathology course were decreased in the following order:presentation skill (53.94％),interaction skill (26.64％) and teaching skill (19.42％).Sensory focusing was most commonly used in presentation skill.Monitoring and expressing concern were most commonly used in interaction skill.Gestures and cases were most commonly used in teaching skill.Conclusions Teachers in pathology teaching should correctly handle the relationship among the presentation,interaction and teaching skills according to the teaching objectives and teaching content.
