1.Research progress on the relationship between frailty and depressive disorders and exercise interventions in older adults

Frailty and depressive disorders exhibit a high prevalence and comorbidity rate in the elderly population.Their coexistence significantly reduces patients'quality of life,increases the risk of disability and mortality,and substantially exacerbates the socioeconomic burden.Emerging evidence indicates a significant bidirectional causal relationship between frailty and depressive disorders.The underlying comorbid mechanisms may be related to elevated levels of pro-inflammatory cytokines such as C-reactive protein,neutrophils,and white blood cells.Grey matter volume reduces in specific brain regions including the bilateral thalamus and right precentral gyrus.And abnormal hormone secretion,such as cortisol,resulting from the overactivation of the hypothalamic-pituitary-adrenal axis.Exercise interventions demonstrate positive effects in preventing and managing both frailty and depressive disorders,indicating broad application prospects.However,the underlying mechanisms require further validation.In summary,the comorbidity of frailty and depressive disorders in the elderly requires greater attention.Current evidence supports exercise intervention as an effective therapeutic strategy for improving health outcomes in this population.
2.Deep progressive reconstruction algorithm applicated in reconstructing whole-body 18 F-FDG PET images

Objective To observe the value of deep progressive reconstruction(DPR)algorithm for reconstructing whole-body 18 F-FDG PET images.Methods Totally 67 patients who underwent whole-body 18 F-FDG PET/CT were retrospectively enrolled.PET data of 30 s,60 s,90 s and 120 s per bed in equipment list were reconstructed using ordered subset expectation maximization(OSEM)and DPR algorithms,respectively.Finally 7 groups of reconstructed images were obtained,including OSEM_30,OSEM_60 and OSEM_120,also DPR_30,DPR_60,DPR_90 and DPR_120 groups.The subjective scores,also objective evaluation indexes,i.e.the maximum and mean standard uptake values(SUV)of lesions and livers,namely SUVmax and SUVmean,were compared,and target-to-background ratio(TBR),signal-to-noise ratio(SNR),contrast-to-noise ratio(CNR)and coefficient of liver variation(CVliver)were calculated.Taken results based in OSEM_120 group as references,Bland-Altman plot was drawn to explore the consistency of SUV of lesions and livers obtained based on DPR_30,DPR_60 and DPR_90 groups with those in OSEM_120 group.Results Under the same acquisition time,subjective scores,SUVmax and SUVmean of lesions,TBR,SNR,CNR and CVliver in DPR_30,DPR_60 and DPR_120 groups were superior to those in corresponding OSEM_30,OSEM_60 and OSEM_120 groups(all P<0.001).Compared with OSEM_120 group,subjective scores and SNR decreased but TBR and CVliver increased in DPR_30 group,while subjective and objective evaluation results in DPR_60 group and DPR_90 group increased(all P<0.05)or being not significantly different from those in OSEM_120 group(all P>0.05).No significant difference of liver SUV mean was found among 7 groups(P=0.955).SUVmax and SUVmean of lesions and livers obtained based on DPR_30,DPR_60 and DPR_90 groups were in good agreement with those oibtained based on OSEM_120 group.Conclusion Using DPR algorithm to reconstruct whole-body 18 F-FDG PET image could shorten acquisition time under the premise of ensuring image quality.

5.Spatial and weighted gene co-expression analysis revealed the mechanism of cross-species ischemic heart failure

Aim To reveal the mechanism of cross-species ischemic heart failure from the perspective of spatial and gene co-expression networks.Methods GSE210374 and GSE57338 high-throughput sequencing datas were re-trieved from the national center for biotechnology information gene expression database(NCBI-GEO),and R language soft-ware packages was used to analyze and screen differentially expressed genes(DEG)in different myocardial regions of myo-cardial infarction rats,as well as DEG of myocardial samples from patients with ischemic heart failure and healthy controls,and the regional expression of common genes was analyzed.Weighted gene co-expression network analysis(WGCNA)was used to screen the genes related to myocardial infarction and to carry out enrichment analysis,protein-protein interac-tion network(PPI)was constructed to screen core genes(HG).Results A total of 4 835 differentially expressed genes were screened out in myocardial infarction rats and normal controls,and 51 differentially expressed genes were screened out in ischemic heart failure patients and normal control samples,which revealed representative gene sets in the left ventricular myocardial infarction area(I area),border area(BZ area),and remote area(R area)after myocardial in-farction.Spatial expression analysis revealed that there were 20 co-expressed genes in each myocardial region,16 of which were expressed in all three regions,the number of genes specifically expressed in I,BZ and R regions were 2,0 and 2,respectively.Enrichment analysis showed that the functions of co-expressed genes were different in different region.The I and BZ regions were related to collagen fiber assembly,stress-induced cardiomyocyte hypertrophy,down-regulation of c-Jun amino terminal kinase(JNK signal)and cell proliferation,and complement signaling pathways;The I and R regions were enriched in the binding of Wnt and collagen;As a non-ischemic distal R region,the co-expressed genes were signifi-cantly enriched in the extracellular matrix for functions such as compressive resistance,cytolysis and inhibition of T cell proliferation.Furthermore,it was worth noting that the products of co-expressed genes in the three regions were mostly lo-cated in the extracellular space and extracellular matrix,suggesting that there may be active cellular secretion and interac-tion regulation.Further PPI analysis suggested that asporin(ASPN),osteoglycin(OGN)and collagentype ⅩⅥ alpha chain(COL14A1)gene might be the core genes of the mechanism mentioned above.Conclusions The common mechanism of ischemic heart failure in rats and human involves multiple signaling pathways such as complement and coagu-lation cascade signaling and Wnt;which may be closely related to cell apoptosis mediated by extracellular matrix and exo-somes;ASPN,OGN,and COL14A1 may be the core genes.This work is expected to provide spatial and pathway refer-ence for the selection of intervention targets and pathway in the transformation research related to ischemic heart failure.
6.Effect of glycogen synthase kinase 3β overexpression in hippocampus on antidepressant and anxiolytic activity of total flavoids from Xiaobuxin Tang in mice

OBJECTIVE To study the influence of glycogen synthase kinase3β (GSK3β) over expres?sion in the hippocampus on the antidepressant and anxiolytic effects of total flavoids from Xiaobuxin Tang (XBXT-2). METHODS Adeno-associated virus containing GSK3β(S9A) mutation was microinjected into the hippocampus. After three weeks of recovery, GSK3βand p-GSK3βwere detected by Western blotting, and open field test (OFT) was used to evaluate the locomotor activity. Then, AAV group and GSK3β over expression group were divided into administration group and solvent group, respectively. XBXT-2 (100 mg · kg-1) and solvent were ig administered chronically. After 14 d and 16 d of administra?tion, the tail suspension test (TST) and forced swimming test (FST) were used to investigate the influence of GSK3βover expression on the antidepressant effect of XBXT-2, respectively. After 18 d and 20 d of administration, the elevated plus maze test (EPMT) and staircase test (ST) were used to investigate the influence of GSK3β over expression on the anxiolytic effects of XBXT-2, respectively. RESULTS Western blotting analysis showed that the protein level of GSK3βincreased significantly in GSK3βover expression group (P<0.01) compared with AAV group, but there was no significant difference in p-GSK3β. In OFT, the number of crossings and rearings showed no difference between AAV group and GSK3β over expression group. The results of TST and FST showed that compared with AAV group, the immobility time was significantly reduced in AAV+XBXT-2 group (P<0.05, P<0.01), but compared with GSK3β over expression group, the immobility time showed no difference in GSK3β over expression+XBXT-2 group. In EPMT, compared with AAV group, the percentage of entrances and time into open arms in AAV+XBXT-2 group was significantly increased (P<0.01, P<0.05), but compared with GSK3βover expression group, these indexes showed no difference in GSK3βover expression+XBXT-2 group. In ST, compared with AAV group, the number of rearings was significantly reduced in AAV+XBXT-2 group (P<0.05), but there was no difference between GSK3β over expression+XBXT-2 group and GSK3βover expression group. CONCLUSION GSK3βover expression in the hippocampus can reverse the antidepressant effects of XBXT-2 in the TST and FST, and the anxiolytic effects in the EPM and ST.
7.Analysis on influencing factors of concurrent free anterolateral femoral skin flap repair in radical operation for oral cancer

Objective To explore the curative effect of oral cancer radical surgery plus free anterolateral thigh flap repair and analyze the influence factors of skin flap necrosis.Methods The data of 98 patients who underwent oral cancer radical surgery with concurrent free anterolateral thigh flap repair in our hospital from January 2010 to June 2015 were retrospectively analyzed.The complete survival rate of skin flaps and incidence of complications in all patients were statistically analyzed.The age,diabetes,infection in implanted skin flap area,smoking and mental status,etc.of patients with survived or necrotic skin flaps were compared between patients with survival flaps and patients with flap necrosis,and multivariate Logistic regression analysis was performed.Results Among the 98 cases,86 cases of flaps survived completely and the survival rate was 87.8%.Fifteen cases with the skin flap necrosis,the incidence of complications was 15.3%,including 7 cases of surgi-cal site infection and 5 cases of skin flap necrosis with massive hemorrhage.The donor site incisions of all patients were primary healing and the patients were satisfied with the postoperative appearance.Among the patients with flap necrosis,the incidence rates of ≥60 years old,with diabetes,infection in flap implanted area,smoking and poor mental state (41.7%,41.7%,58.3%,75.0%,83.3%)were higher than those corresponding proportions in the patients with with survival flaps(P <0.05).Multivariate Logistic regression analysis showed that age,diabe-tes,infection in implanted skin flap area,smoking and poor mental status were the influencing factors for flap necrosis.Conclusion Oral cancer radical surgery with concurrent free anterolateral thigh flap repair has good curative effects.The age,combined with diabetes,smoking,infec-tion in flap implanted area and poor mental state are the risk factors for flap necrosis in patients with oral cancer after flap repair,which should be given intervention to improve the survival rate of skin flap in clinic.

Intravenous glucose tolerance test(IVGTT)and oral glucose-insulin releasing test(OGIRT) in 12 subjects with normal glucose tolerance were performed.The results showed that the peak of insulin secretion Was at 40 minutes during()GIRT.The ratio of the change of insulin-to-glucose at 40 minutes(△I40/△G40)is an index in reflecting insulin sensitivity and β-cell function.
10.Effects of investment materials on cast shrinkage of base metal alloys.

OBJECTIVETo compare the influence of different investment materials, methods and crown height on the cast shrinkage of full crowns made of dental base metal alloy.
METHODSAll of the 288 wax patterns were made and divided into three groups according to their crown height. In every group, wax patterns were invested in metal rings and paper rings with three types investment material and cast with base metal alloys. Then the inside diameters of castings were measured and analyzed by statistical package SPSS.
RESULTSDifferences between groups invested with the three types of material were significant(P < 0.01). There were significant difference between the groups investing with metal rings and paper rings(P < 0.01).
CONCLUSIONThe component of investment material and invested ring can affect the cast shrinkage of dental base metal alloy apparently.
