OBJECTIVE To investigate the effects of galangin on rheumatoid arthritis (RA) in rats by regulating the Janus kinase 3 (JAK3)/signal transducer and activator of transcription 3 (STAT3) pathway. METHODS Fifty male SD rats were taken, and an emulsion composed of bovine type Ⅱ collagen and Freund’s complete adjuvant was injected subcutaneously to establish an induced arthritis model. The rats that were successfully modeled were randomly divided into model group, low, medium and high dose groups of galangin (1, 5, 15 mg/kg), and methotrexate group (positive control, 2 mg/kg), with 10 rats in each group. Another 10 normal rats were taken as the normal group. Starting from the 15th day of modeling, each group of rats was gavaged with the corresponding drug solution or normal saline containing 0.5% Tween 80 once a day for 28 consecutive days. The arthritis index (AI) scores and paw volume of rats were compared before and after gavage administration. Twenty-four hours after the last administration, the serum levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), IL-4 and IL-10 were determined, the pathological changes in ankle joint synovial tissue were observed, and the protein expressions of UNC-51 like kinase 1 (ULK1), Beclin-1, microtubule-associated protein 1 light chain 3 (LC3), B cell lymphoma 2 (Bcl-2), Bcl-2-associated X protein (Bax), caspase-3, JAK3, phosphorylated JAK3 (p-JAK3), STAT3 and phosphorylated STAT3 (p-STAT3) in the synovial tissue of the ankle joint were detected, as well as the fluorescence intensity of LC3-positive areas. RESULTS Compared with the model group, the pathological changes such as cellular proliferation of ankle joint synovial tissue and infiltration of inflammatory cells in rats of each administration group showed improvement. Moreover, their AI scores and paw pad volumes (on day 28 after gavage), the levels of IL-6 and TNF-α, the protein expression of Bcl-2, and the phosphorylation levels of JAK3 and STAT3 were all significantly reduced ( P <0.05). The levels of IL-4 and IL-10, the protein expressions of ULK1, Beclin-1, Bax, caspase-3 and LC3, as well as the fluorescence intensity of LC3-positive areas, were all significantly increased ( P <0.05). Moreover, the effect of galangin was in a dose-dependent manner ( P <0.05). CONCLUSIONS Galangin can induce sustained autophagy in synovial tissue cells of RA rats, promote cell apoptosis, inhibit synovial cell proliferation, and alleviate persistent inflammatory responses. The above anti-RA effects may be related to the inhibition of the JAK3/STAT3 pathway.

Objective To understand the current status of animal experimental research on comorbidities of depression, and to provide method for preparing and evaluating relevant animal models of comorbidities of depression. Methods We searched the CNKI and PubMed databases to identify research literature on comorbidities of depression in animals. We summarized and analyzed the data regarding animal species, comorbidity types, modeling method, and model evaluation indicators. Results This study included 100 Chinese and 143 English studies. The total number of papers published in China and elsewhere has shown an upward trend since 2011.The top three comorbidity models in terms of the number of papers published were depression pain(97,39.91%),depression epilepsy(37,15.23%),and depression diabetes(20,8.23%).The depression neuropathic pain model was the most common comorbid type of depression and pain, and the most commonly used preparation method was selective injury of the sciatic nerve branch, often using C57BL/6J mice. The depression acute epilepsy model was the most common comorbid type of depression epilepsy, and was usually prepared using the lithium chloride pilocarpine model, often using SD rats. The most commonly used method for preparing the depression diabetes model was by intraperitoneal injection of streptozotocin, using C57BL/6J mice.We systematically summarized the evaluation method for the above comorbidity models. Conclusions Increasing research is being carried out using animal models of comorbidities of depression. While a few animal models for the preparation and evaluation of comorbidities of depression are gradually progressing, most animal experimental studies on comorbidities of depression are still in their infancy, and further studies are required to explore these.

Oropharyngeal squamous cell carcinoma(OPSCC)is a malignant tumor originating from the squamous epithelium of the oro-pharyngeal mucosa, accounting for more than 90%of oropharyngeal malignancies. In recent years, human papillomavirus(HPV)infec-tion has become one of the primary etiological factors of oropharyngeal squamous carcinoma. The incidence of HPV-associated oropharyn-geal squamous carcinoma has been rising annually, with a noticeable trend toward younger populations, posing a significant threat to hu-man health.Due to the distinct biological behavior and clinical characteristics of HPV-associated oropharyngeal squamous carcinoma com-pared to its non-HPV-related counterpart, the diagnostic and treatment strategies for oropharyngeal squamous carcinoma have undergone substantial changes.Prevention and screening for oropharyngeal squamous carcinoma are of critical importance. The diagnostic and treat-ment process involves multi-disciplinary collaboration, including oral and maxillofacial surgery, otolaryngology, head and neck surgery, oncology, radiology and pathology.Based on evidence from clinical practice,a comprehensive, integrated diagnostic and therapeutic ap-proach has been established, centered around the concept of"prevention, screening, diagnosis, treatment, and rehabilitation",covering the entire patient lifecycle and providing a valuable reference for clinical practice.

In recent decades, the incidence of human papillomavirus(HPV)-associated oropharyngeal squamous cell carcinoma(OPSCC)has shown a marked increase.Significant changes have also occurred in the OPSCC diagnosis and treatment paradigm.Deter-mining HPV status prior to treatment is now essential, and radiotherapy/chemotherapy,immunotherapy,and minimally invasive surgical techniques have progressively emerged as key modalities for managing OPSCC.However, alongside these paradigm shifts,a comprehen-sive technical consensus guiding the entire diagnostic and therapeutic process for OPSCC patients is currently lacking.Given China's large population base and the rising incidence of OPSCC,an expert panel convened to develop a clinical technical consensus on OPSCC diagno-sis and management tailored to China's specific context. This consensus aims to further enhance and standardize understanding of OPSCC management techniques among relevant healthcare professionals.

Oral squamous cell carcinoma(OSCC)is a malignant lesion originating from the oral mucosal squamous epithelium,account-ing for over 80%of oral and maxillofacial malignancies.Key etiological factors include tobacco, alcohol abuse, and betel quid chewing. In China, its incidence has shown an overall upward trend, posing a significant threat to public health.OSCC exhibits high local invasive-ness,making early diagnosis critical for improving prognosis. Its clinical management requires close multidisciplinary collaboration among oral and maxillofacial surgery, head and neck surgery, radiation oncology, medical oncology, reconstructive surgery,radiology,patholo-gy,and nutritional support teams.Given the increasing disease burden of OSCC and rapid development of multidisciplinary collaborative models,an expert panel has formulated this integrated management consensus based on evidence-based medicine and extensive deliber-ation.Centered on the'Prevention-Screening-Diagnosis-Treatment-Rehabilitation'framework,the consensus provides comprehensive guidance for the entire disease course of OSCC patients, aiming to standardize clinical practice.
