1.Regulatory effect of Jiedu Huayu granules on liver injury in mice with acute liver failure and its mechanism

ObjectiveTo investigate the mechanism of action of Jiedu Huayu granules in improving liver injury in mice with acute liver failure (ALF) by observing its effect on a mouse model of ALF after prophylactic administration, and to provide a basis for clinical medication. MethodsA total of 60 specific pathogen-free male C57BL/6J mice were divided into normal group, model group, Jiedu Huayu granules group (JDHY group), and farnesoid X receptor (FXR) agonist (GW4064) group using a random number table, with 15 mice in each group. The model of ALF was induced by a single intraperitoneal injection of D-galactosamine combined with lipopolysaccharide. The mice in the JDHY group were given prophylactic administration of 0.3 g/mL drug solution of Jiedu Huayu granules by gavage for 3 days before modeling, those in the normal group and the model group were given 0.9% NaCl solution by gavage, and those in the GW4064 group were given intraperitoneal injection of GW4064 for 3 consecutive days before modeling. The mice were sacrificed after modeling, and serum and liver tissue samples were collected. A veterinary automatic biochemical analyzer was used to measure the serum levels of total bilirubin (TBil), total bile acids (TBA), gamma-glutamyl transferase (GGT), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) in mice from each group; HE staining was used to observe liver pathological changes; RT-PCR was used to measure the mRNA expression levels of FXR, fibroblast growth factor 15 (FGF15), fibroblast growth factor receptor 4 (FGFR4), small heterodimer partner (SHP), and bile salt export pump (BSEP) in mice, and Western blot was used to measure the protein expression levels of FXR, FGF15, FGFR4, SHP, and BSEP. A one-way analysis of variance was used for comparison between groups, and the Dunett method was used for further comparison between two groups. ResultsCompared with the normal group, the model group had significant increases in the serum levels of TBil, ALT, AST, TBA, and GGT (all P<0.01), and compared with the model group, the JDHY group and the GW4064 group had significant reductions in the serum levels of TBil, ALT, AST, TBA, and GGT (all P <0.01). HE staining showed that compared with the model group, the JDHY group and the GW4064 group had milder pathological injury, a reduction in the area of hepatocyte necrosis, and alleviation of cellular swelling and edema. Compared with the normal group, the model group had significant reductions in the mRNA and protein expression levels of FXR, FGF15, FGFR4, SHP, and BSEP in liver tissue (all P <0.01), and compared with the model group, the JDHY group and the GW4064 group had significant increases in the mRNA and protein expression levels of FXR, FGF15, FGFR4, SHP, and BSEP in liver tissue (all P <0.05). ConclusionJiedu Huayu granules may alleviate liver injury in mice with ALF through the FXR/SHP axis.
2.Construction of a smart elderly education system based on the AHP method under the Healthy China strategy

Objective:To construct a smart elderly education system that in line with the current situation under the Healthy China strategy and explore the constraints on its institutional implementation,in order to provide a basis for scientifically evaluating the implementation effect of this system.Methods:A questionnaire survey was conducted on 34 experts in related fields.The analytic hierarchy process(AHP)was used to perform quantitative and qualitative analyses on the construction of the smart elderly education system,and the weights of the importance of each scheme were obtained.Results:The constructed model of the smart elderly education system included 5 first-level indicators and 15 second-level indicators.Calculations of the judgment matrix for the 5 first-level indicators showed that the maximum eigenvalue(λmax)=5.071 4,the random consistency index(RI)=1.12,the consistency index(CI)=0.017 8,and the consistency ratio(CR)=0.015 9.With CR<0.1,the result met the consistency requirement.For the second-level indicators under each first-level indicator,pairwise comparison judgment matrices were constructed.The CR values of all 5 judgment matrices were<0.1.In summary,all judgment matrices passed the consistency test and were applicable.Among the first-level indicators,the top three by weights were guarantee mechanisms,demand orientation,and implementation effectiveness.Among the second-level indicators,the top three by combined weights were funding support capacity,health literacy improvement rate,and health demand identification mechanism.Conclusions:The indicator evaluation system for the smart elderly education system constructed based on AHP includes core dimensions such as guarantee mechanisms and demand orientation,responding to the key issues of institutional implementation.The selected indicators are both objective and practical,and can serve as reference tools for actual evaluation.
3.Quality Evaluation on Xuanmai Ganjie Preparation Based on National Drug Sampling and Testing

Objective Based on the national drug sampling inspection program,this study aims to comprehensively and systematically evaluate the quality of Xuanmai Ganjie preparations,analyze existing quality issues,and provide references and suggestions for quality control of this variety.Methods A total of 237 batches of Xuanmai Ganjie preparations were tested using legal standards,and methods were established for detecting adulteration of Ophiopogon japonicus with counterfeit varieties,paclobutrazol residue levels,and determining the content of platycodin D in Xuanmai Ganjie preparations.These methods were applied to the quality control and evaluation of Xuanmai Ganjie preparations.Results Through statutory inspection,one batch of Xuanmai Ganjie granules was found non-compliant.Specific batches were identified to contain the following irregularities:adulteration of Ophiopogon japonicus with counterfeit varieties,paclobutrazol residue levels exceeding proposed limits,and platycodin D content below the established threshold.Conclusion The overall quality of Xuanmai Ganjie granules was average,while the overall quality of Xuanmai Ganjie capsules and lozenges was relatively good.Manufacturing enterprises should strengthen their sense of primary responsibility and enhance control over the entire drug production process.
4.Important factors affecting depression:modulatory effects of Cx43 on neuroinflammation

Numerous studies have shown that depression is main-ly associated with the abnormal expression of connexin 43(Cx43)in astrocytes(Astro)and its mediated dysfunction of gap junction(GJ).However,the molecular mechanism of post-translational modifications targeting Cx43 to regulate neuroin-flammation-associated depression is still unclear.Post-transla-tional modifications of Cx43 mainly include phosphorylation of specific amino acid sites by PKC,PKA,PKG,MAPK and PTK,and protein degradation of Cx43 through the K48/K63 polyubiq-uitylation and deubiquitination pathways,which ultimately lead to protein degradation through K48/K63 polyubiquitination and deubiquitination.These modifications are ultimately involved in the regulation of neuroinflammatory responses through the associ-ation of GJ function.In this paper,we systematically review the role of Cx43 post-translational modifications in neuroinflamma-tion,with the aim of further exploring the potential application of targeting these modifications to modulate the inflammatory re-sponse mechanism in improving depressive symptoms.
5.Liver and Kidney Safety Assessment of Chinese Herbal Compound in Treatment of Gastric Cancer Based on Real-World Data

[Purpose]To evaluate the liver and kidney safety of traditional Chinese medicine(TCM)compounds in the treatment of gastric cancer.[Methods]The clinical data of gastric cancer patients who attended in Longhua Hospital Affiliated to Shanghai University of Traditional Chinese Medicine from January 2012 to February 2022 were collected.The results of liver and renal func-tion tests were rated according to National Cancer Institute-Common Terminology Criteria for Ad-verse Events(NCI-CTCAE)V5.0.The patients were divided into 4 groups:TCM group,TCM+chemotherapy/targeted/antivascular therapy group,TCM+TCM preparation group and TCM+chemotherapy/targeted/antivascular therapy+TCM preparation group,and the effects of TCM on liver and renal functions were analyzed.[Results]A total of 7 943 patients were included in the analysis,of which 2 941 cases receiving TCM ≥12 months,1 468 months ≥36 months,and 687 months≥60 months.The highest incidence rate of liver function abnormality was 13.71%,the highest in-cidence rate of grade 3/4 abnormality was 2.58%;the highest incidence of creatinine abnormality was 2.32%,the highest incidence rate of grade 3/4 abnormality was 0.37%in patients with differ-ent duration of taking TCM.Most of liver and renal function abnormalities occurred in the early stage of drug taking.The incidence of liver and renal function abnormalities in the TCM group was lower than that of the other three groups.The incidence of grade 3/4 abnormality for direct biliru-bin(DBIL)in the TCM group was 0.14%,for aspartate transaminase(AST)was 0.11%,for alka-line phosphatase(ALP)was 0.16%,for alanine aminotransferase(ALT)was 0.06%and for total bilirubin(TBIL)was 0.07%,and there was no grade 3/4 creatinine abnormality observed.The ab-normal liver and renal function indexes were not increased with the increase of the length of herbal medicine taking.[Conclusion]The study shows that long term taking TCM drugs and stan-dardized prescriptions for gastric cancer patients are safe.

To construct a recombinant fowl adenovirus 4(FAdV-4)expressing the Cap protein of goose astrovirus genotype 2(GoAstV-2),the expression cassette of Cap gene was inserted into the natural 1 966 bp deletion region of the FAdV-4 genome in the infectious clone p15A-cm-FAdV4-HNJZ.The resulted recombinant plasmid p15A-cm-FAdV4-HNJZ-Cap/GoAstV-2 was linearized with restriction enzyme and transfected into chicken hepatoma cell line(LMH)to rescue the recombinant FAdV-4 expressing the Cap protein of GoAstV-2,rF Ad V4-Cap/GoAstV-2.After 15 passages in LMH cells,the recombinant rFAdV4-Cap/GoAstV-2 was identified by PCR using primers flanking the insertion site of the Cap gene expression cassette and using viral genome DNA extracted from rFAdV4-Cap/GoAstV-2 infected LMH cells as template.LMH cells were in-fected with 15th passage rFAdV4-Cap/GoAstV-2 and indirect immunofluorescence was performed with a polyclonal antibody against Cap protein as the primary antibody.Western blot was carried out with lysates of rFAdV4-Cap/GoAstV-2 infected LMH cells.The in vitro replication dynamic of the 15th passage of the rFAdV4-Cap/GoAstV-2 was also investigated in LMH cells.The results demonstrated that the Cap gene of GoAstV-2 was presented in the genome of the recombinant vi-rus rF AdV4-Cap/Go Ast V-2,and could be expressed stably.The prepared recombinant virus in this study will lay a foundation for developing inactivated bivalent vaccine candidate against co-in-fection of FAdV-4 and GoAstV-2 in goose.
7.Pharmacological effects of linarin on Aβ deposition and neuroinflammation in APP/PS1 mice

Aim To investigate the effect of linarin on improving cognitive behavior of APP/PS1 mice,and to explore the therapeutic effect of linarin on A β deposi-tion and neuroinflammation and its correlation.Meth-ods APP/PS1 transgenic mice were randomly divid-ed into the model group,high-dose group,medium-dose group,low-dose group and positive control group.C57BL/6J mice were set as the normal group.Morris water maze was used to evaluate the learning and mem-ory abilities of mice.TUNEL staining was used to de-tect the apoptosis of neurons in the CA1 region of mice.IHC was used to detect the expression levels of Aβ42 and GFAP.Western blot was used to detect the expression levels of BACE1 and PS-1.Results Com-pared with the normal group,mice of the model group showed lower NCP,shorter target quadrant travel,less target quadrant residence time percentage(all P<0.01),higher apoptosis rate of neurons in the CA1 re-gion(P<0.01),significantly higher protein expres-sion levels of A β42 and GFAP(all P<0.01),and significantly higher protein expression levels of BACE1 and PS-1(all P<0.01).Compared with the model group,the medium-dose group,high-dose group and positive control group showed higher NCP,longer tar-get quadrant travel,more target quadrant residence time percentage(all P<0.05),lower apoptosis rate of neurons in the CA1 region(P<0.01),significantly lower protein expression levels of A β42 and GFAP(all P<0.01),and significantly lower protein expression levels of BACE1 and PS-1(all P<0.01).Conclu-sions Linarin can inhibit two key enzymes to reduce the decomposition of APP and the generation of A β42,thereby inhibiting the activation of astrocytes,allevia-ting neuroinflammation,improving the core pathologi-cal features of AD,and thus significantly improving learning and memory impairment in APP/PS1 mice.
8.Effects of aerobic exercise on adipogenesis and insulin resistance in mice by modulating lnc-ORA

Objective To explore the effect of aerobic exercise on adipogenesis and insulin resis-tance(IR)in mice on high-fat diet,and to explore the mechanism of aerobic exercise regulating obe-sity-related long noncoding RNA(lnc-ORA)in this process.Methods Thirty-six 8-week-old male C57BL/6J mice,weighing 21.4±0.92 g,were randomized into a normal control group(NC),a high-fat control group(HC)and the high-fat exercise group(HE),each of 12,after one-week acclimatiz-ing feeding.Group NC was fed on normal diet,while the high-fat groups were given high-fat food for 4 weeks.Then,group HE underwent daily 45-minute aerobic exercise on a 6%-incline treadmill at a speed of 18 m/min,6 days per week,for 8-week.After the intervention,the body weight,body length,and epididymal fat weight of all groups were measured,and the adipocyte size was observed.Moreover,lipid levels,glucose tolerance,insulin sensitivity,and expression of genes related to adipo-genesis(FASN,FABP4,and PPARγ)were examined,together with the expression levels of the rele-vant signaling axes(IRS-1/PI3K/AKT/mTOR/GLUT4)and related metrics(lnc-ORA,TNFα),and en-richment levels of peroxisome proliferator-activated receptor γ(PPARγ)promoter region for histone H3 acetylation(H3ac)and histone H3K4 trimethylation(H3K4me3).Results The body weight,epidid-ymal adipose tissue weight,and body mass index(Lee's index)of group HC were significantly higher(P<0.05)than group NC,and the above indexes of group HE were significantly lower(P<0.05)than group HC after 8 weeks of aerobic exercise intervention.The levels of total cholesterol(TC),low-den-sity lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)of the HC group were significantly higher than the NC group(P<0.05),and the lipid levels of the HE group im-proved significantly compared to the HC group(P<0.05).Moreover,the expression of adipogenesis-re-lated genes PPARγ,fatty acid synthase(FASN)and fatty acid binding protein 4(FABP4)up-regulat-ed significantly in group HC compared with group NC(P<0.05),while that of adipogenesis-related genes in group HE was significantly lower than group HC(P<0.05),with the adipocytes'size of HC group larger than the other two groups.What's more,the expression level of insulin receptor substrate 1(IRS-1)/phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamy-cin(mTOR)/glucose transporter type 4(GLUT4)pathway decreased significantly in group HC com-pared with group NC and HE(P<0.05;P<0.01,P<0.05).Meanwhile,both glucose tolerance and insu-lin sensitivity of group HC decreased significantly compared with the NC and HE groups(P<0.01,P<0.05;P<0.01,P<0.05).However,the expression of lnc-ORA and TNF-α increased significantly in group HC compared with group NC(P<0.01,P<0.05),but significantly down-regulated in group HE compared with group HC(P<0.05).The enriched levels of H3ac and H3K4me3 in the PPARγ promot-er region of group HC increased significantly compared with group NC(P<0.05),but decreased signifi-cantly in group HE compared with group HC(P<0.05).Conclusions Aerobic exercise reduces adipogen-esis via lnc-ORA/PPARγ and improves IR by modulating the IRS-1/PI3K/AKT/GLUT4 axis.

Game-based psychological assessment utilizes game mechanisms and elements to enhance partic-ipants'motivation and improve measurement accuracy,but similar to traditional methods,it mainly focuses on static results.By integrating game-based psychological assessment with artificial intelligence technology,this paper propo-ses the paradigm of intelligent psychological assessment based on interactive environment.With analyzing dynamic process data within the game,this paradigm can accurately depict the participants'actions and results in the interac-tive context,more effectively representing individual psychological states and behavioral characteristics.This new paradigm demonstrates advancements and significant potential for development in terms of demand,data collection,and analysis techniques.
10.Brain-computer interface combined with different therapies for limb dysfunction in stroke patients:effectiveness and mechanism analysis

BACKGROUND:In recent years,brain-computer interface technology has shown significant promise for rehabilitating limb dysfunction in stroke patients.With ongoing research deepening and its broader clinical application,combining brain-computer interface with other rehabilitation therapies to improve limb function has become a focal point of study.OBJECTIVE:To analyze and summarize the efficacy of brain-computer interface combined with various therapies in treating limb dysfunction in stroke patients and to explore the clinical value of these combined strategies.METHODS:The search terms used for the literature review in Chinese databases were"brain-computer interface,BCI,stroke,"while the terms"brain-computer interface,BCI,brain-computer interaction,brain-machine interface,BMI,stroke"were used for English databases.Literature searches were conducted in CNKI,WanFang,VIP,PubMed,Embase,and Web of Science,from the time of database construction to September 2024.Finally,a total of 3 054 articles were retrieved,and 75 articles were included after screening for summarization.RESULTS AND CONCLUSION:Currently,brain-computer interfaces,used alone or in combination with other treatments such as Chinese medical treatment,conventional rehabilitation therapy,or physical factor therapy,are achieving better outcomes in treating limb dysfunction in stroke patients.However,the efficacy of brain-computer interfaces combined with transcranial direct current stimulation for treating upper and lower limb dysfunctions is still debated.Researchers are increasingly recognizing the feasibility of these combined therapies.Yet,challenges such as limited exploration of therapeutic mechanisms,absence of standardized protocols,and small sample sizes hinder their broad application.Future research should therefore focus on understanding the mechanisms by which brain-computer interfaces can enhance effects when combined with other therapies and on standardizing criteria for clinical trials to enable widespread clinical adoption.
