Chinese Journal of Biologicals 2026;39(04):468-474
Objective To establish and verify a pseudovirus detection method for respiratory syncytial virus(RSV) neutralizing antibodies, in order to provide a rapid, high-throughput and safe method for RSV vaccine development and antiviral drug screening.MethodsThe RSV A2 pseudovirus was prepared, and the pseudovirus detection method for RSV neutralizing antibody was established by optimizing the pseudovirus concentration factor(0, 2, 4, 8 and 16 times), infected cell type(HEK293T, Hep-2, Huh7.5.1, HeLa and Vero cells), cell inoculation density(0. 5 × 10~4, 1. 0 × 10~4, 1. 5 × 10~4, 2. 0 × 10~4 and2. 5 × 10~4 cells/well), cell generation(1, 5, 10, 15, 20 and 25 generations), detection time(6, 12, 24, 36, 48, 60 and 72 h) and virus inoculation amount(5. 0 × 10~3, 2. 5 × 10~3, 1. 25 × 10~3, 6. 25 × 10~2, 3. 125 × 10~2, 1. 562 5 × 10~2 TCID50/mL). In addition, the precision, specificity, accuracy and edge effects of the method were verified. Finally, the established method and plaque-reduction neutralization test(PRNT) were used to detect 30 mouse positive sera simultaneously, and the correlation between the detection results was analyzed.ResultsThe optimal pseudovirus concentration factor was 8, the infected cells were HEK293T cells of 1-10 passages after resuscitation with the inoculation density of 2. 0 × 10~4 cells/well, the detection duration was 48 h, and the virus inoculation amount was 3. 125 × 10~2-2. 5 × 10~3 TCID50/mL. The coefficients of variation(CVs) of repeatability and intermediate precision verification were both less than 15%. No RSV neutralizing activity was detected in normal mouse serum, and high RSV neutralizing activity was detected in positive mouse serum and Nirsevimab monoclonal antibody. The recovery rates of accuracy verification were within the range of 80% to 120%. The ratios of the detection values of edge holes to non-edge holes were 0. 5-0. 7, indicating that there was edge hole effects, and non-edge holes should be used for detection. The results of 30 mouse positive sera detected by the two methods showed a good positive correlation(r = 0. 914 5, P < 0. 000 1).ConclusionThe established RSV neutralizing antibody titer pseudovirus detection method has good precision, specificity, accuracy, with simple and fast operation, which can be used for the evaluation of RSV vaccine immunogenicity and the high-throughput detection of serum neutralizing antibody potency.

The incidence of apoplexy remains persistently high among the older population.Based on the traditional Chinese medicine principle of"treating the elderly by focusing on the fu",this paper explores the holistic connotation of"fu".It proposes that the onset of apoplexy in the elderly is characterized by obstruction, stagnation, depression, and sluggishness, which should be treated from six fu viscera, xuanfu, and collaterals. The interconnected hierarchical network of these three systems, serving as macro-and micro-channels for qi and fluid metabolism, plays a central role in both disease development and treatment. The failure of the six fu viscera to descend and the upward invasion of turbid yin are identified as prerequisites for apoplexy onset, whereas yang qi stagnation and xuanfu blockage act as key pathogenic drivers, and the core mechanisms involve phlegm-stasis-toxin accumulation, and dysfunction of the collaterals and fu.In the treatment, acute phase requires unblocking fu viscera and restoring xuanfu patency;chronic phase focuses on dredging collaterals and opening xuanfu;and recovery phase emphasizes tonifying combined with regulating xuanfu. The understanding of"treating the elderly by focusing on the fu"emphasizes that the pathogenesis of the disease should be changed to individual conditions;"six fu viscera-xuanfu-collaterals"exhibits a pathological mechanism characterized by the transmission and reception of pathogenic factors,as well as progressive mutual involvement;in clinical practice, treatment should meticulously assess the severity and nuances of the condition, prioritizing method that emphasizes unobstructed flow;additionally, therapy should be essential to protect stomach qi and integrate therapeutic attacks within a framework of supplementation. These principles offer valuable reference for the differentiation and treatment of apoplexy.

Xiang thinking is a cognitive approach that reflects the relationships between phenomena and their underlying principles by analyzing their external manifestations through methods such as analogy, reasoning, deduction, and symbolism. This article applied xiang thinking to analyze the etiology and pathogenesis of "wind, fire, phlegm, and blood stasis" in stroke, thereby exploring its impact on the principles of syndrome differentiation and treatment of this condition. Meanwhile, the article traced the construction process of xiang thinking, and interpreted the concept of "toxin pathogen" in traditional Chinese medicine from four perspectives, state, attribute, origin, and law. Furthermore, the relationship between the process of constructing xiang thinking and the origin of etiology, identification methods, pathogenesis evolution, and treatment strategies for stroke with syndrome of combined blood stasis and toxin was explored, so as to provide insights into research on the etiology and pathogenesis of stroke, as well as clinical diagnosis and treatment approaches.

ObjectiveTo observe and compare the electrocardiogram index, myocardial morphology, and connexin 43 (Cx43) expression of two rat models of acute cerebral infarction (ACI) due to stasis combined with toxin complicated with cerebral-cardiac syndrome (CCS), and to provide experimental evidence for the research on the occurrence mechanism of cardiac diseases induced by ACI and the clinical diagnosis and treatment of CCS. MethodSixty SPF-grade male SD rats were randomized into six groups (n=10): normal , syndrome of stasis combined with toxin induced by carrageenin combined with dry yeast (CA/Y), multi-infarct induced by micro-embolism (ME), middle cerebral artery occlusion (MCAO), CA/Y+ME, and CA/Y+MCAO groups. The model of syndrome of stasis combined with toxin was established by intraperitoneal injection with carrageenan (CA) at 10 mg·kg-1 on the first day and subcutaneous injection with dry yeast (Y) suspension (2 mg·kg-1) on the second day of modeling. Twenty-four hours after the modeling of ACI, the electrocardiograms (ECGs) of rats in each group were collected and the number/percentage (%) of abnormal ECG was calculated. The infarct area of the brain was evaluated by 2,3,5-triphenyltetrazolium chloride (TTC) staining, and myocardial injury was assessed by hematoxylin-eosin (HE) staining. Immumohistochemical staining and Western blot were employed to determine the expression of Cx43 in the myocardium. ResultA certain number of rats in each model group presented abnormal ECG. Compared with the normal group and CA/Y group, CA/Y+MCAO group had the highest rate of abnormal ECG (P<0.01). Compared with the normal, CA/Y, ME, and CA/Y+ME groups, the CA/Y+ME and CA/Y+MCAO groups showed decreased amplitudes of P-wave and T-wave, shortened P-R interval, and extended Q-T interval, which were particularly obvious in the CA/Y+MCAO group (P<0.05, P<0.01) and in accordance with the cerebral infarction area and pathological changes. The expression of Cx43 was up-regulated in both CA/Y+ME and CA/Y+MCAO groups, especially in the CA/Y+MCAO group (P<0.01). ConclusionThe two rat models of ACI due to stasis combined with toxin complicated with CCS can be used to study the mechanism of heart diseases caused by cerebrovascular diseases and the therapeutic effects of Chinese medicines with the functions of resolving stasis and detoxifying. Moreover, the CA/Y+MCAO method has higher abnormal electrocardiogram rate, severer myocardial pathological injury, and higher expression of Cx43 protein. The models can be chosen according to specific experimental purpose.

Xiang thinking is the key way of thinking to construct the life model of human body in traditional Chinese medicine (TCM), and the theory of ascending and descending of qi movement is an important manifestation of xiang thinking in the theory of TCM. Based on the theory of qi movement, this paper interpreted the mechanism of ischemic stroke through the perspective of xiang thinking "earth weakness - wood constraint - fire hyperactivity", as "earth weakness in the central and dampness accumulated to phlegm" "wood constraint and stirring wind led to blood stasis" and "fire hyperactivity and fire toxin showed flaming upward" due to disorder of qi movement. Combined with the "xiang of medicinal properties and therapy methods" to discuss the treatment and prescriptions of ischaemic stroke, applying wind medicinals to elevate ji-earth (己土) and yi-wood (乙木), so that phlegm and stasis can be eliminated, and cold medicinals to descend jia-wood (甲木) and wu-earth (戊土) so that fire toxin can be cleared, with a view to restore ascending and descending of qi movement for ischaemic stroke.
