Objective To explore the independent risk factors for complicated coronary heart disease (CHD) in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS) and to construct a risk prediction model. Methods A retrospective study was performed to analyze the clinical data of 308 OSAHS patients who visited Jiangsu Provincial People's Hospital from January 2021 to June 2025. The patients were divided into the CHD group (n=80) and non-CHD group (n=228) according to the presence or absence of complicated CHD. Independent risk factors were identified using univariate analysis and multivariate logistic regression, and a nomogram prediction model was constructed. The model was evaluated using the calibration curve, Hosmer-Lemeshow test, Decision Curve Analysis (DCA) curve, and Receiver Operating Characteristic (ROC) curve. Results The incidence of CHD in OSAHS patients was 25.97%. Age, history of hypertension, oxygen desaturation index (ODI), severe polysomnography (PSG) grade, decreased high-density lipoprotein cholesterol, elevated homocysteine, and elevated troponin I were independent risk factors for OSAHS complicated with CHD (P<0.05). The nomogram model demonstrated good calibration (Hosmer-Lemeshow test, P=0.995), and the DCA curve showed high clinical net benefit. The area under the ROC curve was 0.940 (95% CI: 0.908~0.972), with a sensitivity of 0.842 and a specificity of 0.900. Conclusion Age, history of hypertension, ODI, PSG grade, high-density lipoprotein cholesterol, homocysteine, and troponin I are independent risk factors for CHD in OSAHS patients. The prediction model constructed based on these factors can provide a reliable tool for clinical nurses to early identify high-risk individuals and formulate targeted intervention strategies.
2.The research on immune therapeutic strategy of dual-function cationic nano delivery system for adjunctive treatment of periodontitis
Acta Universitatis Medicinalis Anhui 2026;61(9):1548-1557
ObjectiveTo investigate the effects of a dual-functional cationic nucleic acid nano delivery system (DCNA) loaded with small interfering RNA targeting Bruton’s tyrosine kinase (siBTK) on the immune-inflammatory microenvironment of periodontitis by regulating BTK expression. MethodsDCNAsiBTK was prepared using a double-emulsion solvent evaporation method, and its physicochemical properties and sustained-release performance were characterized. Cytotoxicity was evaluated using the cell counting kit-8 (CCK-8) assay. Antibacterial activity against periodontal pathogens was assessed by colony-forming assays. BTK mRNA and protein expression levels were determined by reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR) and Western blot, respectively. Enzyme-linked immunosorbent assay (ELISA) was performed to quantify the levels of inflammatory cytokines, including IL-1β, IL-6, and TNF-α. A murine periodontitis model was established and treated with DCNAsiBTK. Therapeutic efficacy and biosafety were evaluated using quantitative polymerase chain reaction (qPCR), micro-computed tomography (micro-CT), hematoxylin-eosin (HE) staining, and immunohistochemistry (IHC). ResultsStable DCNAsiBTK nanoparticles with an average particle size of (168.8±2.2) nm and a zeta potential of (17.9±0.8) mV were successfully prepared. Transmission electron microscopy (TEM) revealed uniformly spherical morphology with favorable sustained-release properties. CCK-8 assays demonstrated no significant cytotoxicity of DCNAsiBTK within the concentration range of 0-100 nmol/L. In vitro studies showed that DCNAsiBTK effectively inhibited the growth of Porphyromonas gingivalis (Pg), Aggregatibacter actinomycetemcomitans (Aa), and Fusobacterium nucleatum (Fn), indicating broad-spectrum antibacterial activity. RT-qPCR and Western blot analyses demonstrated that DCNAsiBTK downregulated BTK mRNA and protein expression in a dose-dependent manner. ELISA results showed that DCNAsiBTK significantly reduced the secretion of IL-1β, IL-6, and TNF-α, thereby attenuating inflammatory responses. In vivo experiments further demonstrated that DCNAsiBTK suppressed anaerobic bacterial proliferation within periodontal pockets, alleviated alveolar bone resorption and periodontal inflammation, and caused no obvious pathological damage in major organs, including heart, liver, spleen, lungs, and kidneys. ConclusionDCNAsiBTK efficiently encapsulated and released siBTK, and exhibited dual activities including antibacterial and anti-inflammatory. By suppressing BTK expression, DCNAsiBTK effectively attenuated the progression of periodontitis, providing a promising novel strategy for adjunctive periodontal therapy.

Objective:To explore the mechanism of Coicis Semen in treating non -small cell lung cancer combined with network pharmacology and in vitro cell experiment. Methods:The components and potential targets of Coicis Semen were obtained from TCMSP, and NSCLC related targets were collected from Genecards and Drugbank. They were intersected to get common targets. An intersection target PPI network was constructed using STRING database. GO function and KEGG pathway enrichment analysis were performed using DAVID database. A "disease-drug component-target" network was constructed using Cytoscape 3.8.0. The CCK-8 method was used to screen the concentration of Coicis Semen, and the cells were randomly divided into control group, Coicis Semen low-concentration group (0.125 mg/ml), Coicis Semen high-concentration group (0.25 mg/ml), Coicis Semen low concentration+FoxO1 inhibitor group, and FoxO1 inhibitor group (50 μmol/L) using a random number table. Cell proliferation ability was determined using CCK-8 method, and cell migration and invasion ability were detected using cell scratch assay and Transwell cell invasion assay. Western blot was used to detect the expression of FoxO1, cofilin-1, and p-cofilin-1 proteins. Results:A total of 9 active components and 246 targets were screened from Coicis Semen, 1 074 targets related to non-small cell lung cancer, and 11 intersecting targets were identified. The key targets of Coicis Semen in the treatment of non-small cell lung cancer were MAPK1, MAPK8, MAPK14, CDK2, INSR, AR, FGFR2, PGR, DHFR, and PLAU; GO functional enrichment analysis showed that Coicis Semen had an impact on the cellular, molecular, and biological functions of A549 cells; KEGG pathway enrichment analysis showed that the pathways of Coicis Semen in treating non-small cell lung cancer mainly focus on FoxO pathway, MAPK pathway, prostate cancer pathway, Ras pathway, etc. Compared with the control group, the proliferation rate, migration rate, and invasion number of A549 cells in the low and high concentration groups of Coicis Semen decreased ( P<0.05), p-cofilin-1 expression decreased ( P<0.05), and FoxO1 expression increased ( P<0.05). Conclusion:Coicis Semen can treat non-small cell lung cancer by regulating MAPK1, MAPK8, and MAPK14 targets, and acting on pathways such as the FoxO and MAPK pathways.

T-cell acute lymphoblastic leukemia (T-ALL) is a highly aggressive hematologic malignancy with a poor prognosis, despite advancements in treatment. Many patients struggle with relapse or refractory disease. Investigating the role of the super-enhancer (SE) regulated gene ubiquitin-specific protease 20 (USP20) in T-ALL could enhance targeted therapies and improve clinical outcomes. Analysis of histone H3 lysine 27 acetylation (H3K27ac) chromatin immunoprecipitation sequencing (ChIP-seq) data from six T-ALL cell lines and seven pediatric samples identified USP20 as an SE-regulated driver gene. Utilizing the Cancer Cell Line Encyclopedia (CCLE) and BloodSpot databases, it was found that USP20 is specifically highly expressed in T-ALL. Knocking down USP20 with short hairpin RNA (shRNA) increased apoptosis and inhibited proliferation in T-ALL cells. In vivo studies showed that USP20 knockdown reduced tumor growth and improved survival. The USP20 inhibitor GSK2643943A demonstrated similar anti-tumor effects. Mass spectrometry, RNA-Seq, and immunoprecipitation revealed that USP20 interacted with hypoxia-inducible factor 1 subunit alpha (HIF1A) and stabilized it by deubiquitination. Cleavage under targets and tagmentation (CUT&Tag) results indicated that USP20 co-localized with HIF1A, jointly modulating target genes in T-ALL. This study identifies USP20 as a therapeutic target in T-ALL and suggests GSK2643943A as a potential treatment strategy.

This study aims to determine the nephrotoxic effects of 3-monochloropropanel-1,2-diol(3-MCPD)on female and male SD rats.A nephrotoxicity model was established by gavage of dif-ferent doses of 3-MCPD,and 80 SD rats were randomly divided into four groups:the control group(0 mg/kg 3-MCPD),low-dose group(15 mg/kg 3-MCPD),medium-dose group(30 mg/kg 3-MCPD),high-dose group(60 mg/kg 3-MCPD),with half female and half male. The body mass and food intake of the rats were recorded weekly, and the urine and blood and kidney tissues were col-lected after 28 consecutive days of gavage, and the blood erythrocyte count(WBC),white blood cell count(RBC),hemoglobin(HGB),erythrocyte-compaction-transfer-value(HCT),creatinine(CREA),serum urea nitrogen(BUN),and the indexes of blood phosphorus(P)and calcium(Ca)were detected;the level of kidney injury molecule(KIM-1)was measured by ELISA kit;the renal pathological changes was observed by histopathology method;and transcriptome sequencing was used to analyze differential genes in female and male rats. The results showed that 3-MCPD did not significantly affect the growth of male rats, but high doses significantly reduced the weight and food intake of female rats. The high-dose group of 3-MCPD caused a significant decrease in RBC,HGB,and HCT levels in both male and female rats, resulting in a significant increase in KIM-1 and P,and a significant decrease in Ca,but only a significant increase in CREA and BUN in female rats.Histopathology showed that in the high-dose group of male rats, only mild renal tubular dilation, epithelial cell edema, and clear tubular type were observed in the kidneys, while in female rats,a large number of renal sacs, clear tubular type, and interstitial inflammatory cells with fibrosis were observed in the kidneys.Transcriptome sequencing showed 1 712 differentially expressed genes in the high-dose group of female rats and 1 153 differentially expressed genes in the high-dose group of male rats.KEGG enrichment analysis showed that both male and female rats in the high-dose group experienced oxidative stress and cell apoptosis, but 3-MCPD may also participate in the process of kidney damage in females by inhibiting autophagy and inducing iron death pathways. The above results indicate that high-dose 3-MCPD has a more significant nephrotoxic effect on fe-male rats.
