2.Aldehyde Dehydrogenase 2 Gene Mutation May Reduce the Risk of Rupture of Intracranial Aneurysm in Chinese Han Population

Background:
and Purpose Ruptured intracranial aneurysms (RIA) are associated with a mortality rate of up to 40% in the Chinese population, highlighting the critical need for targeted treatment interventions for at-risk individuals. Although the impact of aldehyde dehydrogenase 2 (ALDH2) gene mutations on susceptibility to intracranial aneurysms (IA) is well documented, the potential connection between ALDH2 rs671 single-nucleotide polymorphism (SNP) and RIA remains unexplored. Given the increased prevalence of ALDH2 gene mutations among Chinese Han individuals, it is clinically relevant to investigate the link between ALDH2 rs671 SNP and IA rupture.
Methods:
A prospective study was conducted on 546 patients diagnosed with IA to investigate the association between ALDH2 rs671 SNP and the risk of IA rupture.
Results:
The ALDH2 rs671 SNP (ALDH2*2) was significantly more prevalent in patients with unruptured IA (UIA) than in those with RIA (32.56% vs. 18.58%, P=0.004). Multivariate logistic regression analysis revealed that people with the ALDH2 mutation (ALDH2*1/*2 and ALDH2*2/*2 gene type) had a significantly reduced odds ratio (OR=0.49; 95% confidence level [CI] 0.27–0.88; P=0.018) for RIAs. Age-specific subgroup analysis indicated that the ALDH2 mutation provided a stronger protective effect in individuals aged 60 years and above with IA compared to those under 60 years old (OR=0.38 vs. OR=0.52, both P<0.05).
Conclusion
The incidence of RIA was significantly higher in individuals with a normal ALDH2 gene (ALDH2*1/*1) than in those with an ALDH2 rs671 SNP (ALDH2*1/*2 or ALDH2*2/*2). ALDH2 rs671 SNP may serve as a protective factor against RIA in the Chinese Han population.
3.Application of the back-calculation method for estimating new HIV infections in Dehong Dai and Jingpo Autonomous Prefecture, Yunnan Province, 2010-2023

Objective:New HIV infections serve as a crucial indicator for assessing the dynamic changes in the HIV epidemic. This study aims to estimate the number of new HIV infections in Dehong Dai and Jingpo Autonomous Prefecture of Yunnan Province (Dehong), using a back-calculation method that integrates diagnosis delay approaches and Bayesian theory. Additionally, it compares the differences between these two estimation methods.Methods:Data were obtained from the Chinese Information System for Disease Control and Prevention. Based on CD4 + T lymphocytes (CD4) counts depletion model, the first CD4 count prior to antiretroviral therapy of HIV-infected individuals diagnosed in Dehong from 2010 to 2023 was utilized to retroactively determine the infection date of HIV-infected individuals and ascertain the annual number of new HIV infections who had been diagnosed. Subsequently, the diagnosis delay distribution method and Bayesian theory were leveraged to assess the diagnosis probability of newly infected individuals, thereby projecting the number of new HIV infections in the region over the specified period. Results:During 2010-2023, a total of 5 693 individuals aged 15 and above, excluding mother-to-child transmission, were diagnosed with HIV in Dehong. After excluding 364 cases due to missing CD4 count results or abnormal first CD4 counts (≥2 000 cells/μl), 5 329 HIV-infected individuals were included in the final analysis. Through CD4 counts back-calculation from 2010 to 2023, the annual number of new infections diagnosed was 479, 427, 337, 305, 256, 219, 194, 193, 131, 166, 120, 71, 42 and 47. When using the diagnosis delay distribution method and life table analysis, the cumulative diagnosis probability rose from 0.301 within one year to 0.913 within 14 years, leading to a reduction in the number of estimated new infections from 577 in 2010 to 168 in 2023, with a total estimate of 4 412 (95% CI:4 350-4 480). Alternatively, based on Bayesian theory, the diagnosis probability increased from 0.413 within one year to 0.946 within 14 years, leading to a reduction in the number of estimated new infections from 557 in 2010 to 122 in 2023, with a total of 3 814 (95% CI: 3 787-3 837). Conclusions:Both methods yielded consistent results in estimating new HIV infections in Dehong from 2010 to 2023. Given the region's ongoing expansion of HIV testing, the estimates derived from Bayesian theory may more accurately reflect the actual situation. These findings provide a reference basis for formulating and optimizing HIV/AIDS prevention and control strategies in Dehong, facilitating progress toward the goal of eliminating AIDS by 2030 in the region.
4.Systematic review of assessment tools for maladaptive eating behaviors in bariatric surgery patients

Objective To systematically evaluate the methodological quality and measurement properties of assessment tools for maladaptive eating behaviors in bariatric surgery patients.Methods A systematic search of the CNKI,Wanfang database,VIP,CBM,PubMed,Web of Science,Embase,and Cochrane Library was conducted from inception to March 15,2025.Totally 2 researchers independently screened the literature and extracted the data.The consensus-based standards for the selection of health measurement instruments checklist and quality criteria were employed to evaluate the methodological quality and psychometric properties of the included assessment tools,and combined with a modified version of the GRADE to form recommendations.Results A total of 14 articles were included,involving 2 specific assessment tools for bariatric surgery patients and 10 universal assessment tools.The methodological quality of content validity was rated as"unclear"for all scales,with moderate certainty of evidence.Structural validity and internal consistency generally performed well(Cronbach's α>0.7).However,evidence for stability,criterion validity,and cross-cultural validity remains insufficient.A total of 9 instruments were recommended as Grade B,while 3 were recommended as Grade C.Conclusion The disordered eating after bariatric surgery and the food craving questionnaire-trait are recommended for clinical use,but further validation of their psychometric properties is still needed.

7.New targets for the treatment of acute graft-versus-host disease after allogeneic hematopoietic stem cell transplantation

Allogeneic haematopoietic stem cell transplantation(allo-HSCT)is the most effective curative treatment for hematologic malignancies.Its efficacy hinges on eliminating primary hematological disorders and restoring bone marrow hematopoiesis during conditioning,as well as leveraging the graft-versus-leukemia(GVL)effect.However,acute graft-versus-host disease(aGVHD)remains a significant complication following allo-HSCT,substantially affecting patient survival and quality of life.Current preclinical studies focus on strategies to mitigate aGVHD while preserving adequate GVL effects to improve transplant outcomes.This review summarizes recent preclinical research findings in this field,emphasizing the regulatory roles and specific molecular mechanisms of T cells,antigen-presenting cells,myeloid-derived suppressor cells,and mesenchymal stem cells in aGVHD.It further highlights the latest therapeutic strategies for aGVHD from preclinical studies,aiming to provide valuable insights for researchers and clinicians to develop more effective therapeutic targets and strategies.
8.Detection and analysis of pathogens associated with neoplastic diseases in breeders in Hebei Province

In order to determine the causative agents responsible for the lethality of tumor disease in breeders,liver,spleen and other tissues of dead chickens were collected,and pathogen detection,se-quencing and genetic evolution analysis were carried out by PCR.The results showed that all three samples were negative for reticuloendotheliosis virus(REV),samples 1 and 2 wereavian leukosis virus subgroup J(ALV-J)positive,and sample 3 was positive for ALV-J and Marek's disease virus(MDV),which preliminarily determined that the breeder died of ALV-J infection or ALV-J and MDV mixed infection.Among them,the ALV-J gp85 gene had the highest nucleotide homology with the reference strain of ALV-J,ranging from 87.70%to 99.30%,and was in the same branch with the reference strain of subgroup J.Amino acid homology ranged from 78.00%to 96.20%,with some mutations.The nucleotide homology between the MDV meq gene and the vv+strain was the highest,which was 98.00%-99.00%,and they were in the same branch.The homology of amino acids was 95.90%-98.20%,and there were multiple mutation sites,among which the 176th amino acid destroyed the original Pro repeat of the virus MDV meq due to the mutation of Pro to Arg or Ala,which may lead to the enhancement of its virulence.This study will provide a reference for the prevention and control of neoplastic diseases in breeders in this region.

The 2025 edition of the Chinese Pharmacopoeia Volume Ⅳ has added General Chapter 9211"Guidelines for the Application of Water Activity in Microbial Control of Non-sterile Pharmaceuticals".This guideline is rela-tively new in China's pharmaceutical industry,and it also serves as an important concept and measure to compre-hensively promote microbial process control of pharmaceuticals,conduct microbial quality risk management,and optimize microbial limit setting for non-sterile pharmaceuticals.This article elaborates and interprets the guidelines from four aspects:the background of establishing the guidelines,the necessity and significance of adding the guide-lines,the process of adding the guidelines,and the main framework and key content of the guidelines.It aims to guide enterprises to optimize formulations,formulate personalized microbial control strategies,and realize the trans-formation from terminal quality control to process control,so as to provide important tools and theoretical guidance for microbial quality risk management.
