Displaying Candida antarctica lipase B (CALB) on the cell surface of Aspergillus niger is effectively applied for the industries of food, cosmetics, pharmaceutical and so on. Displaying CALB using induced promoter of glucoamylase on the cell surface of A. niger SH-1 has some problems such as inhibiting its expression under high concentration of glucose, mycelium cleavage and decreasing enzyme activity in the later period of fermentation process. Displaying CALB manipulated by constitutive promoter from glyceraldehyde-3-phosphate dehydrogenase instead of glucoamylase on the cell surface of A. niger SH-1, called AN-GpdA, could solve the above problems effectively. Furthermore, it can not only use glucose, but also xylose as a sole carbon source. Enzyme activity of AN-GpdA using xylose for fermentation reached 1 100.28 U/g of dry cell. We also used lignocellulose such as the hydrolysate of bagasse for fermentation with good performance. The result would provide a novel strategy for the utilization of bagasse.

This study was aimed to evaluate the hemostatic effect and mechanism of action for water decoction of Blumea megacephala (Randeria) Chang et Tseng in order to understand its influence to the liver function. The glass slides method and capillary tube method were used in the measurement of the coagulation time (CT). And the tail-cutting method was used to measure the bleeding time (BT), prothrombin time (PT), activated part clotting live en-zyme time (APTT), thrombin time (TT), content of plasma fibrinogen (FIB), platelet count (PLC), plasma complex cal-cium time (PRT), alanine aminotransferase (ALT) and aspartate transaminase (AST). The results showed that intragastric administration with different doses of water decoction of Blumea megacephala (Randeria) Chang et Tseng (6.7 g·kg-1, 13.4 g·kg-1, 26.8 g·kg-1) can reduce CT and BT of mice. And intragastric administration with different doses of wa-ter decoction of Blumea megacephala (Randeria) Chang et Tseng (4.7 g·kg-1, 9.4 g·kg-1, 18.9 g·kg-1) can produce different degrees of impact on PT, APTT, TT and PRT of rats. Certain dose of water decoction of Blumea megacepha-la (Randeria) Chang et Tseng can reduce ALT and AST. It was concluded that Blumea megacephala (Randeria) Chang et Tseng had the hemostatic effect and its mechanism of action may be through the activation of the intrinsic and extrinsic coagulation system. ObjectiveTo investigate the expression and activity of histone deacetylase (HDAC) in prostate cancer.Methodshe pathological samples of 37 cases of PCa were collected. The mean age of the patients was 73 (53 - 88 ) years, the preoperative t-PSA was 81.69 ( 3.13 - 2000 ) ng/ml, Gleason score: 13 cases were ≤7, 24 cases were >7. Twenty-seven cases of BPH were set as controls. The mean age of the BPH patients was 69 (52 - 84) years, the preoperative t-PSA was 10.93 ( 1.11 - 55.07 ) ng/ml.Western blotting and colorimetric Assay kits were used to determine the HDAC expression and activity. The difference of HDAC activity in benign prostatic hyperplasia and prostate cancer was statistically analyzed. The correlation of the HDAC expression level and values of PSA and Gleason score was also assessed.ResultsHDACs were over-expressed in most cases of prostate cancer, the expression rates were HDAC1 :57%, HDAC2: 68%, HDAC3: 84% and HDAC4: 73%, respectively. The HDAC activity (P <0.05)was significantly different between the prostate cancer and benign prostatic hyperplasia groups. The expression level of HDAC did not correlate with the values of PSA and Gleason score.ConclusionsHDAC was highly expressed and strongly active in prostate cancer. The results suggest that HDAC might be a potential target for the management of prostate cancer patients.

Acta Pharmaceutica Sinica 2011;46(9):1150-5
The drug release characteristics ofDa Chuanxiong Fang multiunit drug delivery system (DCXFMDDS) in vivo and in vitro were evaluated. Ferulic acid (FA) and senkyunolide I (SI) were used as marker components, which were two of the effective components of Da Chuanxiong Fang. And their contents were determined by HPLC. Drug release characteristics in vitro of DCXFMDDS and Da Chuanxiong pills and pharmacokinetics characteristics of DCXFMDDS and Da Chuanxiong Fang active fraction (DCXFAF) in rats were compared. It was obvious that FA released from the DCXFMDDS in a sustained fashion but SI in a fast fashion both in vitro and in vivo. The releasing process and the releasing mechanism of FA and SI from DCXFMDDS were different, but the AUC value indicated that compared with DCXFAF the extent of absorption of FA and SI from DCXFMDDS was increased. Though from the same multiunit drug delivery system, FA an SI had different drug release characteristics both in vitro and in vivo, and that may be one of the reason why DCXFMDDS has the good properties such as rapid and long-lasting effect and high efficiency.
