1.Overview of construction of effector platforms and behavioral assessments of acupuncture intervention in animal models of chronic pain accompanied by pain anxiety

Chronic pain has emerged as a prevalent medical challenge in contemporary society.Patients suffering from chronic pain frequently develop comorbid psychological disorders,including anxiety,depression,post-traumatic stress disorder,and various psychiatric syndromes.These psychological complications not only affect patients' pain perception and responses,but may also constitute critical obstacles during pain management interventions.Acupuncture is a long-established clinical practice that has demonstrated remarkable efficacy in alleviating diverse pain types and has shown favorable therapeutic outcomes in ameliorating emotional disturbances such as anxiety and depression.The precise mechanisms underlying acupuncture-induced analgesia and anxiolytic effects,however,remain to be fully elucidated.In this context,it is essential to establish suitable and stable animal models to allow in-depth investigations into the pathogenesis of pain-related emotional disorders and the mechanistic foundations of acupuncture.This article presents a comprehensive review of recent literature regarding the selection of experimental animals,model-establishment method ologies,and behavioral-assessment paradigms pertaining to animal model platforms of chronic pain with comorbid anxiety.We also provide an in-depth discussion of research advancements regarding acupuncture intervention parameters,including needling techniques,acupoint selection,treatment duration,and efficacy evaluation within these animal models.This review proposes comprehensive and reference strategies for constructing preclinical animal models to investigate the mechanisms of acupuncture in managing chronic pain with comorbid anxiety,thus supporting scientific advancements in related research fields.
2.Mechanisms of acupuncture regulating ADORA1 in the caudate puta-men to improve neuroplasticity and alleviate inflammatory pain

AIM:This study investigated the potential mechanism of acupuncture regulating adenosine A1 re-ceptor(ADORA1)in the mouse caudate putamen(CPu)to improve neuroplasticity and alleviate inflammatory pain.METHODS:Male C57BL/6 mice were randomly divided into five groups,namely,saline,complete Freund's adjuvant(CFA),acupuncture(MA),acupuncture+ADORA1 shRNA(MA+shRNA),and acupuncture+negative shRNA(MA+NCshRNA)groups.Twenty-one days before modeling,the mice in the MA+shRNA and MA+NCshRNA groups were in-jected with ADORA1 shRNA and control virus into the CPu.Modeling was performed 21 d later by injection of CFA into the right plantar skin of mice in the model and acupuncture groups to induce adjuvant-mediated arthritis.On day 2 after modeling,mice in the acupuncture groups received acupuncture at bilateral"Zusanli"points,30 min per session,once a day,for a total of 7 days.The paw thermal radiation pain threshold was used as an indicator of the effects on pain in the mice.Changes in the protein levels of ADORA1,synaptophysin(SYP),synapsin(SYN)I,SYN II,and brain-derived neurotrophic factor(BDNF)in the CPu were assessed by Western blot,and transmission electron microscopy was used to examine the dendritic structure and synaptic ultrastructure of neurons within the CPu.RESULTS:(1)Compared with the saline group,CFA modeling significantly reduced the thermal radiation pain threshold in mice(P<0.01),as well as the protein levels of ADORA1(P<0.01).Compared with the CFA group,the thermal pain threshold in the MA group in-creased between days 1 and 7 of acupuncture(P<0.05 or P<0.01),and ADORA1 protein levels increased(P<0.01).(2)Compared with the saline group,SYN I,SYN II,and BDNF protein levels were increased in the CFA group(P<0.05 or P<0.01),while relative to the CFA group,the levels of SYN I,SYN II,and BDNF were reduced in the acupuncture group(P<0.05 or P<0.01).No significant changes in SYP protein levels were observed in any of the groups.(3)Golgi staining and Sholl analysis showed that compared with the saline group,there were reductions in the total dendritic length and number of intersection points in the CFA group,while the dendritic spine density increased(P<0.05).Relative to the CFA group,the total dendritic length and the number of intersection points were increased in the MA group,while the dendritic spine density decreased(P<0.05).(4)Transmission electron microscopy revealed that compared with the Sa-line group,the synaptic clefts were narrower in the CFA group,while the density of synaptic vesicles in the presynaptic membrane was increased.Compared with the CFA group,synaptic clefts in the MA group were wider,and synaptic vesi-cle densities in the presynaptic membrane were reduced.(5)Twenty-one days after viral transfection,the thermal pain threshold in the MA and MA+NCshRNA groups increased from day 1 to day 7 of acupuncture relative to the CFA and MA+shRNA groups(P<0.05 or P<0.01),while the expression of ADORA1 was increased in both the MA and MA+NCshRNA groups(P<0.05 or P<0.01).(6)Compared with the CFA and MA+shRNA groups,the protein expression of SYN II and BDNF in the MA and MA+NCshRNA groups was reduced(P<0.05 or P<0.01).No significant changes in SYN I protein were observed in any of the groups.CONCLUSION:Acupuncture on Zusanli point can alleviate chronic pain in CFA-treated mice,potentially mediated by up-regulation of ADORA1 expression in the CPu brain region,thereby improving neuroplasticity.

Objective To understand and evaluate the knowledge, attitude and practice of female human papillomavirus and HPV vaccine in Chinese mainland. Methods We searched the relevant studies about the knowledge, attitude and behavior factors of HPV infection and HPV vaccine published in PubMed, MEDLINE, CNKI and Wanfang databases about Chinese mainland women from January 1, 1995 to January 31, 2021.Two researchers independently screened the literatures, extracted data and the literatures with quality score≥5 points.Chi square test was used to evaluate its heterogeneity.Begg' s test was used to evaluate publication bias. Results A total of 33 literatures were included, with a total number of 46013 people.The results showed that the total awareness rate of HPV in the population was 65.9%, the lowest rates were 16.0%, 41.2% and 14.4% in Northeast, rural areas and middle school students, respectively; while the highest rates of HPV were 77.4%, 56.3% and 71.0% in Central China, cities and towns and college students, respectively.The main source of HPV knowledge was the network/WeChat official account number (38.9%), and the lowest was family/friend (4.5%).The population's awareness rate of HPV vaccine related knowledge was 41.4%, the highest rates were 51.8% and 69.7% in East China and medical staff, respectively, while the lowest were 23.6% and 12.7% in Southwest and middle school students. Conclusion In order to achieve the total elimination of cervical cancer in China by 2030 as soon as possible, people should strengthen their awareness of HPV and HPV vaccine related knowledge.
7.Knowledge and attitudes toward prophylactic human papillomavirus vaccines among the mainland Chinese population

To assess knowledge of and attitudes toward prophylactic HPV vaccines among the mainland Chinese population and to determine possible influencing factors in order to provide reference data for policymakers to develop HPV vaccination strate-gies in China. Methods: This large-scale national cross-sectional epidemiological study was conducted from May 2009 to May 2012. The general population, medical personnel, and school students participated in the investigation. Participants’basic information, as well as their knowledge of and attitudes toward HPV vaccines were collected through questionnaires. Chi-square tests and Logistic re-gression were performed for the statistical analysis. Results: A total of 18,677 people responded effectively, with a median age of 32 (ranging from 15 to 79 years). In terms of vaccine awareness, only 24.5% had heard of prophylactic HPV vaccines and 14.1% knew that HPV vaccines can prevent diseases such as cervical cancer. The good news was that 83.5% of respondents were willing to vaccinate themselves, their partners, or their children. People who knew more about vaccines, the rural population, and female participants were more likely to hold positive attitudes toward vaccination (adjusted ORs were 2.81, 2.14, and 1.25, respectively). Those partici-pants who were reluctant to accept HPV vaccination were concerned mainly about the safety of the vaccines (64.7%). Furthermore, 66.3% of the respondents expected the maximum price of the HPV vaccine to be less than 300 RMB. Conclusions: Mainland Chinese people generally know little about HPV vaccines. However, most of them think positively about the vaccinations after learning about them. It is high time for the government to launch HPV and cervical cancer health education or promotional campaigns to eliminate concerns about HPV vaccine safety. Furthermore, discounted or free vaccines are needed.
8.Association of serum γ-glutamyl transpeptidase level with carotid intima media thickness in essential hypertension

Objective To analyze the association of serum γ-glutamyl transpeptidase(γ-GT)level with carotid intima media thickness(IMT) in essential hypertension.Methods All the 300 essential hypertension patients and 70 healthy controls were from Ganzhou People′s Hospital.They were divided into healthy control group(70 cases),hypertension with out arteriosclerosis group(141 cases) and hypertension with arteriosclerosis group(159 cases) based on carotid IMT.Fasting blood glucose,serum lipid,routine laboratory tests,hepatic and renal function were detected by routine methods.The one-way analysis of variance and χ2-test were used to compared among the three groups,and the relationship of γ-GT with IMT was assessed by using the Spearman correlation coefficient,multiple linear regressions and Logistic regression analysis.Results The diastolic blood pressure,body mass index,fasting blood glucose,glomerular filtration rate and glycosylated hemoglobin had no differences among the three groups(P>0.05),but systolic pressure,triacylglycerol,total cholesterol,high density lipoprotein cholesterol,aspartate transaminase(AST),low density lipoprotein cholesterol alanine,transaminase(ALT) and γ-GT had significant differences among the three groups(P<0.05).Multiple linear regression analysis showed that the serum γ-GT(β=-0.345,95%CI was-0.005——0.511,P=0.036) was also negatively correlated with IMT,and Logistic regression analysis showed that γ-GT(OR=0.569,95%CI was 0.237-0.867,P=0.029)was a protective factor for atherosclerosis.Conclusion Our study demonstrated the level of γ-GT is negative with IMT among essential hypertension patients,and monitoring the changes of serum liver enzyme might has significant effect on the early detection of arteriosclerosis.
9.Relationship between left atrial diameter with serum glutamyl transpeptidase level in patients with essential hypertension

Objective To analyze the relationship and clinical meaning of serumglutamyl transpeptidase (GGT) level with left atrial diameter.Methods All the 320 essential hypertension patients from Ganzhou People′s Hospital were divided into normal left atrial group and enlarged left atrial group based on the size of left atrial diameter.Fasting blood glucose,serum lipid,routine laboratory tests,hepatic and renal function and blood pressure were measured by routine methods,and used for analyzing between groups.Results Among essential hypertension patients,the GGT level in enlarged left atrial group [(24.78±10.93)U/L] was lower than normal left atrial group[(35.23±12.46)U/L] (P=0.004).Multiple linear regression analysis showed that the serum GGT was negatively correlated with left atrial diameter (P=0.032),and logistic regression analysis showed that GGT was a protective factor for enlarged left atrial (OR=0.424,P=0.026).Conclusion Our study demonstrated the level of GGT was negatively with left atrial diameter among essential hypertension patients,and monitoring the changes of serum liver enzyme may has significant effect on the early detection of enlarged left atrial diameter.
