Objective:To design a simulated navigation scheme of cranial puncture based on three dimensional (3D) infrared optical locator,so as to achieve accurately puncture of trigeminal nerve semilunar node and ensure maximization of the effect of clinical surgery. Methods:The experiment used computed tomography (CT) to scan skull phantom, and imported its data into 3D Slicer system. Spatial coordinates of skull phantom, surgical tool, surgical needle and needle tips were converted to 3D Slicer through the 3D optical locator and 3D Slicer's image guided therapy (IGT) module,so as to display the relationship of spatial position between needle tip and skull model in real-time,and to verify the accuracy of actual measurement of 3D optical locator, and the located accuracy of navigation scheme of skull puncture. Results:After the verification of 10 verification tests, the defined accuracy of 3D optical locator reached 0.2 mm. The operation of virtual world of 3D Slicer system can real-timely and accurately correspond with that of real world. The measured result of simulated navigation and position was (0.87±0.49) mm. Conclusion:The testing results verify the accuracy of navigation of simulated cranial puncture, which has favorable effect, and which can provide a reference for planning navigation scheme before percutaneous puncture balloon compression (PBC) surgery.

Objective:To examine the effects of preimplantation genetic testing for aneuploidies (PGT-A) using blastocyst biopsy on maternal and perinatal outcomes for women of advanced age. Methods:A retrospective cohort study was conducted during January 2016 to December 2018 at Center for Reproductive Medicine, Department of Obstetrics and Gynecology, Peking University Third Hospital. Women who were aged ≥35 years, underwent intracytoplasmic sperm injection (ICSI, control group) or PGT-A after ICSI (PGT-A group) with single frozen-thawed blastocyst transferred were eligible in this study. They were further divided into 35-37 years old subgroup and ≥38 years old subgroup according to age. The primary outcome was live birth, and the secondary outcomes were human chorionic gonadotropin (hCG) positivity, clinical pregnancy, pregnancy loss, hypertension in pregnancy, gestational diabetes mellitus, gestational age, preterm birth, caesarean section, low birth weight, small gestational age, and large gestational age. Results:For women aged ≥35 years, the live birth rate in PGT-A group was significantly higher than that in control group [38.0% (89/234) vs. 26.8% (237/885), OR(95% CI)=1.49(1.13-1.97), P=0.047], the miscarriage rate was significantly lower than that in control group [17.6% (19/108) vs. 29.0% (116/443), OR(95% CI)=0.45(0.24-0.85), P=0.013]. We found that for women who aged ≥38 years, the live birth rate [ OR(95% CI)=3.01(1.67-5.44), P<0.001], hCG positivity rate [ OR(95% CI)=2.08(1.25-3.47), P=0.005], clinical pregnancy rate [ OR(95% CI)=2.39(1.40-4.07), P=0.001] in PGT-A group were significantly higher than those in control group, and the miscarriage rate in PGT-A group was significantly lower than that in control group [ OR(95% CI)=0.34(0.13-0.85), P=0.022]; for women aged 35-37 years, there were no statistically significant differences in pregnancy outcomes between the two groups (all P>0.05). Moreover, there were no statistically significant differences in the rates of obstetric complications and perinatal outcomes for women aged ≥35 years between PGT-A group and control group (all P>0.05), and similar results were found in the subgroup analyses for women who aged 35-37 years or ≥38 years. Conclusion:PGT-A using blastocyst stage biopsy strategy in single blastocyst thaw transferred cycles could significantly improve the rates of hCG positivity, clinical pregnancy, and live birth, and significantly reduce the miscarriage rate for women aged ≥38 years, but could not improve the pregnancy outcomes for women aged 35-37 years. In addition, the use of PGT-A does not increase the risk of obstetric complications and perinatal outcomes for women of advanced age.

Journal of Gynecologic Oncology 2019;30(5):e77-
OBJECTIVES: Pseudolaric acid B (PAB) has been shown to inhibit the growth of various tumor cells, but the molecular details of its function are still unknown. This study investigated the molecular mechanisms by which PAB induces apoptosis in HeLa cells. METHODS: The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays were performed to investigate the effect of PAB treatment in various cervical cancer cell lines. Annexin V/propidium iodide staining combined with flow cytometry and Hoechst 33258 staining were used to assess PAB-induced apoptosis. Additionally, we performed bioinformatics analyses and identified a paired box 2 (PAX2) binding site on the BAX promoter. We then validated the binding using luciferase and chromatin immunoprecipitation assays. Finally, western blotting assays were used to investigate PAB effect on the Wnt signaling and the involved signaling molecules. RESULTS: PAB promotes apoptosis and downregulates PAX2 expression in HeLa cells in a time- and concentration-dependent manner. PAX2 binds to the promoter of BAX and inhibits its expression; therefore, PAX2 inhibition is associated with increased levels of BAX, which induces apoptosis of HeLa cells via the mitochondrial pathway. Additionally, PAB inhibits classical Wnt signaling. CONCLUSION: PAB effectively inhibits Wnt signaling and PAX2 expression, and increases BAX levels, which induce apoptosis in HeLa cells. Therefore, PAB is a promising natural molecule for the treatment of cervical cancer.
