Periodontitis is a common oral disease caused by bacteria coupled with an excessive host immune response. Stem cell therapy can be a promising treatment strategy for periodontitis, but the relevant mechanism is complicated. This study aimed to explore the therapeutic potential of mitochondria from human embryonic stem cell-derived mesenchymal stem cells (hESC-MSCs) for the treatment of periodontitis. The gingival tissues of periodontitis patients are characterized by abnormal mitochondrial structure. Human gingival fibroblasts (HGFs) were exposed to 5 μg/mL lipopolysaccharide (LPS) for 24 h to establish a cell injury model. When treated with hESC-MSCs or mitochondria derived from hESC-MSCs, HGFs showed reduced expression of inflammatory genes, increased adenosine triphosphate (ATP) level, decreased reactive oxygen species (ROS) production, and enhanced mitochondrial function compared to the control. The average efficiency of isolated mitochondrial transfer by hESC-MSCs was determined to be 8.93%. Besides, a therapy of local mitochondrial injection in mice with LPS-induced periodontitis showed a reduction in inflammatory gene expression, as well as an increase in both the mitochondrial number and the aspect ratio in gingival tissues. In conclusion, our results indicate that mitochondria derived from hESC-MSCs can reduce the inflammatory response and improve mitochondrial function in HGFs, suggesting that the transfer of mitochondria between hESC-MSCs and HGFs serves as a potential mechanism underlying the therapeutic effect of stem cells.

OBJECTIVE To investigate the effects and mechanism of Glycyrrhiza inflata polysaccharides （GiP） and GiP-B1 on the maturation and anti-tumor effect of dendritic cell （DC）. METHODS The immature DC （imDC） of hepatocellular carcinoma cell H22 tumor-bearing mice cultured in vitro were divided into control group， tumor necrosis factor-α （TNF-α） group， GiP group， and GiP-B1 group. The viability， positive expressions of surface markers （CD11c， CD80， CD86， MHC-Ⅱ）， the levels of interleukin-12p70 （IL-12p70） and IL-4 in mature DC （mDC） of tumor-bearing mice were detected. mDC and CD4+T lymphocytes were co-cultured to generate CD4-cytotoxic T cell （CD4-CTL）； stimulation index， the levels of IL-12p70， interferon-γ （IFN-γ）， IL-4 and IL-10， the killing activity of CD4-CTL to H22 cell were detected. mRNA expressions of IL-12， IL-12 receptor （IL-12R）， signal transducer and activator of transcription-4 （STAT-4）， as well as the protein expression of IL-12 receptor β2 （IL-12Rβ2）， phosphorylation levels of nuclear factor-kappa B （NF- κB） p65 and STAT-4 proteins in mDC were detected after co-culture. RESULTS Compared with the control group， the viability of mDC， the positive expressions of MHC-Ⅱ， and the levels of IL-12p70 and IL-4 were increased significantly in GiP group and GiP-B1 group （P＜0.05）. The positive expressions of CD11c， CD80 and CD86 showed an increasing trend， but the differences were not statistically significant （P＞0.05）. After co-culturing， the stimulation index， the levels of IL-12p70 and IFN-γ were significantly increased （P＜0.05）， while the levels of IL-4 and IL-10 （except for the GiP group） were significantly decreased （P＜0.05）； the cytotoxicity against H22 cells was significantly enhanced （P＜0.05）. mRNA expressions of IL-12 and IL-12R （except for GiP group） and STAT-4， protein expression of IL-12Rβ2 as well as phosphorylation levels of NF-κB p65 and STAT-4 protein were increased significantly in mDC （P＜0.05）. CONCLUSIONS GiP and GiP-B1 have a good promoting effect on the maturation of DC in tumor-bearing mice， effectively stimulate CD4+T cell proliferation， enhance the anti-tumor activity of CD4-CTL，and its mechanism may be related to activating IL-12/NF-κB/ STAT-4 signaling pathway.

Objective To investigate the current status,research hotspots,and emerging trends in the field of TCM treatment for tic disorder(TD);To provide references for relevant research.Methods Literature on the TCM treatment for TD was retrieved from CNKI,VIP,Wanfang Data,Web of Science Core Collection,and PubMed databases from January 1,2000 to May 1,2022.NoteExpress 3.7.0.9296 software was used for literature management.VOSviewer 1.6.18 software was utilized to conduct co-occurrence analysis on author,institution,and keyword information.R 4.1.3 was applied to statistically analyze keyword frequency and time distributions and generate visualizations.Results A total of 1 520 articles were included in the analysis,involving 107 core authors.High-frequency keywords included experience of famous doctors,clinical observation,acupuncture,and ear acupoints.Commonly used Chinese herbal medicines included Acori Tatarinowii Rhizoma and Bombyx Batryticatus.Frequently prescribed TCM formulae included Wendan Decoction,Changpu Yujin Decoction,Yinqiao Powder,and Tianma Gouteng Decoction.Conclusion Current research hotspots in this field focus on experience of famous doctors,the clinical efficacy of TCM interventions for TD,and molecular biological mechanism studies.Emerging research trends include external TCM treatment and comorbidity investigations.

Objective To explore the chain mediating effect of interpersonal promotion and calling on work autonomy and work engagement of medical staff.Methods From June to July 2023,a total of 310 medical staff from 5 tertiary public hos-pitals in Hangzhou were selected by convenience sampling.The questionnaire included work autonomy scale,interpersonal promo-tion scale,calling scale and work engagement scale for accurate and effective investigation,and data analysis was conducted using SPSS2 6.0 and AMOS 26.0.Results A total of 310 questionnaires were distributed and 257 were recovered with a recovery rate of 82.90％.There were statistically significant differences in work engagement among medical staff with different ages,marital status,working time,titles and positions(all P＜0.05).Calling partially mediated the effect of work autonomy on work engage-ment(95％CI[0.090,0.291],P＜0.001),and interpersonal promotion and calling played a partially chain mediating role be-tween work autonomy and work engagement(95％CI[0.016,0.103],P＜0.001).Conclusion Interpersonal promotion and calling play a partially chain mediating role between work autonomy and work engagement of medical staff.Work autonomy of medical staff not only has an indirect effect on work engagement through the mediating role of calling,but also enhances calling through interpersonal promotion and ultimately affects work engagement.

Attention deficit disorder(ADHD)is a common neurodevelopmental disorder in childhood,which seriously affects the physical and mental health of children.Up to now,the etiology and pathogenesis of ADHD are not clear,which is also the focus of global research.Animal model is an important carrier for basic research of ADHD,but the use of ADHD animal model is still controversial.This paper introduces the main emerging ADHD animal models in the world in recent years and analyzes their advantages and disadvantages.The genetic model,environmental induction model and neurodevelopmental disorder model are introduced and summarized from three aspects:surface validity,construct validity and predictive validity.The purpose of this study is to find a suitable animal model for the basic research of ADHD and to provide reference for the basic research of ADHD in China.
