1.Effect of LncRNA RMRP on ferroptosis induced by oxygen glucose deprivation/reperfusion in mouse HL-1 cardiomyocytes by regulating miR-766-5p

Objective:
To investigate the effect and mechanism of long non-coding RNA RMRP(LncRNA RMRP) on oxygen-glucose deprivation/reperfusion(OGD/R)-induced ferroptosis in mouse HL-1 cardiomyocytes by regulating miR-766-5p.
Methods:
HL-1 cells were cultured in vitro, and OGD/R models were established. The expression levels of LncRNA RMRP in HL-1 cells at various reperfusion time points were subsequently quantified using qRT-PCR. The LncRNA RMRP small RNA interference fragment(si-RMRP) and its corresponding negative control(si-NC), as well as the miR-766-5p inhibitor and its respective negative control(inhibitor-NC), were transfected into HL-1 cells. Subsequently, the cells were subjected to OGD/R treatment. CCK-8 assay was employed to evaluate cell viability. Assay kits were employed to measure the levels of lactate dehydrogenase(LDH) in the cell supernatant, as well as the intracellular levels of malondialdehyde(MDA), superoxide dismutase(SOD), glutathione(GSH), and ferrous ion(Fe2+). qRT-PCR analysis was conducted to assess the expression levels of LncRNA RMRP and miR-766-5p. Western blot analysis was conducted to assess the expression levels of proteins associated with ferroptosis including GPX4, SLC7A11, and FTH1. Dual-luciferase reporter assays were performed to investigate the sponge adsorption relationship between LncRNA RMRP and miR-766-5p.
Results:
As reperfusion time extended, the expression level of LncRNA RMRP in cells progressively increased(P2+ levels within the cells, and decreased the activities of SOD and GSH in cells(P2+ levels within the cells, and promoting SOD and GSH activities in cells(PP < 0. 01) . Furthermore , silencing LncRNA RMRP upregulated the protein expression levels of GPX4 , SLC7A11 , and FTH1(P < 0. 01) . The dual-luciferase reporter assay confirmed that LncRNA RMRP could regulate the expression of miR-766- 5p through a sponge adsorption mechanism. Partial inhibition of miR-766-5p inhibitor expression could mitigate the improvement effect caused by LncRNA RMRP silencing on OGD/R-induced ferroptosis in HL-1 cells.
Conclusion
Silencing LncRNA RMRP inhibits OGD/R-induced ferroptosis in HL-1 cells , potentially through the sponge-mediated regulation of miR-766-5p expression.

3.Correlation of caries susceptibility with different means of tooth surface and patients' oral hygiene habits after interproximal enamel reduction in clinical practice

Objective To explore the correlation of caries susceptibility of patients with the immediate tooth polish,application of fluoride,and the patients' oral hygiene habits after interproximal enamel reduction (IER) in clinical practice.Methods The present study retrospectively collected 45 cases of IER (18 males and 27 females) in 196 tooth sites.Each sample was examined clinically and radiologically to obtain the incidence of caries,and the outcome was correlated with amount of IER,tooth polish,application of fluoride,and the brushing method and frequency for statistical analysis.Results The present study showed that there was no statistical significance among the amount of IER,the site of IER,the patients' brushing method,and the gender with the susceptibility of caries.However,there was statistical significance among the application of fluoride,tooth polish and brushing frequency with the susceptibility of caries.Conclusions The current research suggests that it is desirable to polish tooth and apply fluoride for patients with IER in clinical practices.In the meanwhile,the clinician should pay attention to patients' oral hygiene habits,especially brushing frequency,due to the positive correlation between the brushing frequency and incidence of caries.
4.Relationship between P-gp expression and clinical efficacy of transcatheter arterial embolization chemotherapy (TACE) in primary liver cancer and clinical significance

Objective To explore the relationship of MDR1 and its encoded product P-gp expressions with clinical efficacy of transcatheter arterial chemoembolization (TACE)in primary liver cancer and their clinical significance.Methods We selected 108 patients with primary liver cancer who came to our hospital between June 2010 and June 2013 as observation subjects.Meanwhile 50 healthy people in our hospital for liver biopsy were selected as controls.MDR1 mRNA level in observation group and control group was determined by real-time quantitative PCR.P-gp protein level was analyzed by immunohistochemistry.According to P-gp level,the 108 patients were divided into drug-resistance groups and non-resistance group;the relationship between P-gp expression level and clinical efficacy was analyzed.Results MDR1 mRNA level in liver tissues significantly enhanced in observation group compared with that in control group (P <0.05).In observation group 32 patients had the ratio of MDR1 mRNA level-normal level of more than 2 and 76 patients had the ratio of MDR1 mRNA level-normal level of less than 2. Immunohistochemistry revealed that MDR1 encoded product P-gp was brownish yellow, mainly expressed in the cell surface of liver cancer cells.There were 35 P-gp protein-negative patients (non-resistance group)and 73 positive patients (resistance group).Clinical efficacy was significantly higher in non-resistance group (74.28%)than in resistance group (43.28%)(P <0.05).The 1 year and 2-year cumulative survival rates were 54. 12% and 27.40% in resistance group and 77.14% and 42.86%% in non-resistance group.They were significantly higher in the latter group (P <0.05 ).Conclusion The overexpressed MDR1 encoded product P-gp in primary liver cancer is associated with multidrug resistance in tumor chemotherapy,suggesting that P-gp can be used as one of the guiding clinical markers of chemotherapy.
