1.The effect of bone morphogenetic protein 2 intervening in iron metabolism related protein expression on rats osteoporosis by regulating hepcidin level

Objective:To explore the effect of bone morphogenetic with protein 2 (BMP2) intervention in iron metabolism related protein expression on rats osteoporosis (OP) by regulating hepcidin level.Methods:Thirty-six 8-week-old SPF grade female SD rats with a body weight of 200-220 g were divided into sham group (Removal of adipose tissue around ovaries), model group (ovariectomy was used to prepare postmenopausal OP model rats), adenovirus group (OP model rats injected with adenovirus into the femoral cavity), and overexpression BMP2 adenovirus group (OP model rats injected with BMP2 overexpressing adenovirus into the femoral cavity) using a random number table method, with 9 rats in each group. Rats were euthanized after 2 months of injection, and femoral tissue was taken for micro computed tomography (micro-CT) three-dimensional reconstruction to observe the microstructural characteristics of bone trabeculae, and calculate bone mineral density (BMD), trabecular number (Tb.N), trabecular thickness (Tb.Th), bone volume/total volume (BV/TV), and trabecular separation (Tb.Sp) values. Hematoxylin eosin staining, Masson staining, and tartrate-resistant acid phosphatase (TRAP) staining were used to observe the pathological damage, collagen content, and osteoclast count of femoral tissues in four group rats. Malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), ferritin, hepcidin levels in serum were detected. Real-time quantitative PCR was used to detect BMP2 mRNA expression in femoral tissue. Western blotting were used to detect BMP2, ALP, OCN, OPN, RANKL, TRAP, CTX-I, hepcidin, FPN1, TfR1, DMT1 protein expression in femoral tissue.Results:The differences of BMD, Tb.N, Tb.Th, BV/TV, and Tb.Sp in sham group, model group, adenovirus group, and BMP2 overexpressing adenovirus group were statistically significant ( P<0.05), among them, BMD, Tb.N, Tb.Th, and BV/TV in model group were lower than those in sham group, Tb.Sp was higher than that in sham group, BMD, Tb.N, Tb.Th, and BV/TV in BMP2 overexpressing adenovirus group were higher than those in model group and adenovirus group, Tb.Sp was lower than that in model group and adenovirus group ( P<0.05). Sham group had intact bone trabeculae, abundant collagen fibers, and fewer osteoclasts in femoral tissues; Model group and adenovirus group had sparse bone trabeculae, severe loss of collagen fibers, and more osteoclasts; Bone trabeculae and collagen fibers in overexpression BMP2 adenovirus group were greater than those in model group and adenovirus group, osteoclasts were fewer than those in model group and adenovirus group. The differences of MDA, GSH, SOD, ferritin, and hepcidin in serum in four groups were statistically significant ( P<0.05), among them, MDA and ferritin in model group were higher than those in sham group, GSH, SOD, hepcidin were lower those in sham group, MDA and ferritin levels in overexpression BMP2 adenovirus group were lower those in model group and adenovirus group, GSH, SOD, hepcidin were higher than those in model group and adenovirus group ( P<0.05). BMP2 mRNA expression of femoral tissues in four groups were 1.00±0.10, 0.39±0.12, 0.39±0.08, and 3.46±0.41, respectively, BMP2 protein expression were 0.61±0.06, 0.17±0.06, 0.20±0.05, and 1.03±0.07, respectively, the differences were statistically significant ( P<0.05), among them, model group were lower those in sham group, overexpression BMP2 adenovirus group were higher than those in model group and adenovirus group ( P<0.05). The differences of ALP, OCN, OPN, RANKL, TRAP, CTX-I, hepcidin, FPN1, TfR1, and DMT1 expression of femoral tissues in sham group, model group, adenovirus group, and BMP2 overexpressing adenovirus group were statistically significant ( P<0.05), among them, ALP, OCN, OPN, and hepcidin in model group were lower than those in sham group, RANKL, TRAP, CTX-I, FPN1, TfR1, and DMT1 were higher than those in sham group, ALP, OCN, OPN, and hepcidin in BMP2 overexpressing adenovirus group were higher than those in model group and adenovirus group, RANKL, TRAP, CTX-I, FPN1, TfR1, and DMT1 were lower than those in model group and adenovirus group ( P<0.05). Conclusion:BMP2 intervention iron metabolism related protein expression promotes bone formation in OP rats by increasing hepcidin level.

3.Role of mitomycin C in interventional therapy for tuberculous cicatricial stenosis of the central airway

Objective To investigate the clinical efficacy of bronchoscopic MMC topical spraying for the treatment of tuberculous cicatricial stenosis of the central airway. Methods 45 patients with t tuberculous cicatricial stenosis of the central airway were randomly divided into a control group (14 patients), treatment group 1 (group1, 15 patients), or treatment group 2 (group 2, 16 patients), who received bronchial balloon dilatation alone, bronchial balloon dilatation combined with topical MMC spraying for one time, and for twice, respectively . The clinical efficacy was observed by using the MRC score and measuring airway diameter at the time points before treatment, end of treatment, and 3, 6, and 12 months after treatment, respectively. Results For the MRC scores at different time points, the MRC scores in group 2 (0.06 ± 0.25) and group 1 (0.33 ± 0.617) were significantly lower than those in the control group at 3 months after treatment (P < 0.05 for all comparisons);there were nosignificant differences at the other time points among the three groups. For the airway mean diameters at the different time points, the airway mean diameter was higher in group 2 than in the control group at 3 and 6 months after treatment (P < 0.01), and in group 1 at 3, 6, and 12 months (P < 0.05). No statistical differences were found in the other time points among three groups (P > 0.05). Conclusions Bronchial balloon dilatation combined with topical MMC spraying has certain short-term and long-term efficacy for improving dyspnea and maintaining the airway diameter after dilatation.
4.Cerebrovascular reserve capacity of patients with ischemic cerebral white matter lesions

Objective To investigate the changes of cerebrovascular reserve capacity (CVR) in patients with ischemic white matter lesions (WML). Methods 126 patients with WML were divided into mild lesion, moderate lesion, severe WML groups and a normal control group by brain MRI. Blood pressure, blood sugar, blood fat were measured and past medical histories were recorded in details. All the patients were routinely examined using TCD to evaluate CVR. Hypercapnia was induced by inhaling the CO2 they breathed themselves and hypocapnia was done by voluntary hyperventilation. Results Age, hypertension, diabetes mellitus and Apo-A were the independent risk factors for WML. Compared with the controls , CVR decreased significantly in the severe and moderate WML groups (P < 0.05). The extent of WML negatively correlated to the cerebrovascular reserve capacity (rs = -0.273, -0.392). Conclusions Age, hypertension, diabetes mellitus and Apo-A are the independent risk factors for WML. CVR is significantly decreased in the WML.
5.Effect of fluvastatin on the expressions of caspase-12, CHOP and JNK in ischemia-reperfusion brain injury in rats

Objective To investigate the effect of fluvastatin on the expressions of caspase-12,CCAAT/enhancer-binding protein homologous protein(CHOP), and c-Jun N-terminal kinases (JNK) in ischemia-reperfusion brain injury in rats.Methods Forty two rats were randomly divided into sham operation group (6 rats), ischemia-reperfusion (I/R) group (18 rats), and fluvastatin (Flu) group (18 rats).The rats of I/R and Flu groups were molded by modified Longa intraluminal thread, then put to death at 2 h occlusion and 24 h reperfusion point.Expressions of caspase-12, CHOP, and JNK were detected with immunohistochemistry and Western blot.Results Immunohistochemistry and Western blot showed that the expressions of caspase-12, CHOP, and JNK were increased at 24 h reperfusion.Compared to I/R group, the expressions of caspase-12 and CHOP in Flu group were decreased significantly (all P <0.01);and the expression of JNK had no difference between I/R and Flu groups(P > 0.05).Conclusions The increased expression of caspase-12, CHOP, and JNK showed that endoplasmic reticulum stress was involved in the pathological process of ischemia-reperfusion brain injury.Fluvastatin could inhibit the expression of caspase12 and CHOP, and could delete endoplasmic reticulum stress (ERS) in ischemia-reperfusion brain injury.
6.Study of cerebrovascular reserve capacity in obstructive sleep apnea-hypopnea syndrome

Objective To assess cerebrovascular reserve capacity in patients with obstructive sleep apnea-hypopnea syndrome(OSAHS).Methods One hundred and fourteen patients with OSAHS and 43 normal persons were enrolled in this study.The patients were divided into mild,moderate,severe according to apnea hypopnea index (AHI) and LSaO2 (lowest arterial oxygen saturation).All the patients and normal persons were routinely examined using transcranial Doppler (TCD) and end-tidal carbon dioxide partial pressure(ETCO2) to evaluate cerebrovascular reserve.Hypercapnia was induced by inhaling the CO2 which produced by the patients themselves,and hypocapnia was elicited by voluntary hyperventilation.Results CVR in the severe and moderate OSAHS were significantly lower than that in the control group [ (1.80 ± 1.34) %/mm Hg and (1.43 ±1.05)%/mm Hg vs (2.93 ±0.93)%/mm Hg,P <0.05] when patients in the condition of hypocapnia.And there was no significant difference on CRV between the mild OSAHS group and control group [ (2.53 ±1.83 ) %/mm Hg vs ( 2.93 ± 0.93 ) %/mm Hg,P > 0.05 ].When patients in the condition of Hypercapnia,CRV in the severe and moderate OSAHS were also significantly lower than that in the control group [ ( 1.83 ±1.32) %/mm Hg and (1.08 ± 1.00)%/mm Hg vs (3.32 ± 1.53)%/mm Hg,P < 0.05),AHI was negatively correlated with the cerebrovascular reserve at the condition of hypercapnia and hypocapnia (r=-0.665,-0.721; P < 0.05 ).Conclusion Inhaling CO2 is a effective method for assessing CVR.Cerebrovascular reserve capacity is associated with AHI.Reduced CVR causes hemodynamics change being severe hypoxia in the moderate and severe OSAHS.
7.The clinical feature and gene mutation of three pedigrees with spinocerebellar ataxia MA

Objective To investigate the clinical features and the gene mutation of patients with spinocerebellar ataxia type3 and type7.Methods The trinucleotide repeat mutations were detected by polymerase chain reaction (PCR),agarose gel electrophoresis method,and DNA sequencing in 13 patients,4 related members and 4 common members from 3 spinocerebellar ataxia families.Results Among the 13 patients,four patients had SCA3/MJD(CAG) n expansion mutation(n = 65 ~ 74),nine patients had SCA7 allele expansion for 40 ~ 52 times.Patients with type 3 or 7 showed significant difference in nervous system injury.Conclusion The difference of clinical feature might be used in diagnosis of SCA3/MJD and SCA7,but genotype determination would be the only method of definite diagnosis.
8.The changes of PECAM-1,Bcl-2 and Bax after focal cerebral ischemia in rats

Objective To observe the changes of PECAM-1,Bcl-2 and Bax expressed by ischemic cerebrum of adult rats after focal cerebral ischemia. Methods Middle cerebral artery occlusion (MCAO) models were made by operation with Longa suture method in Wistar rats. The expression levels of PECAM-1,Bcl-2 and Bax at the 6th,12th,18th,24th,48th,72nd hour after MCAO were detected by immunohistochemistry staining. Results The expression levels of PECAM-1,Bcl-2 and Bax in cerebrum were significantly increased after MCAO. The expression levels of Bcl-2 we、re up to the peak at the 12th hour after MCAO, while the levels of Bax and PECAM-1 were up to the peak at the 24th and 48th hour after MCAO. At the 72nd hour after MCAO, the expression levels of PECAM.1.Bcl-2 and Bax were still higher than that in the control group(all P<0.001).Conclusions PECAM-1,Bcl-2 and Bax participate in the different pathological stages of focal cerebral ischemia.

Objective To study the effect of intratumor angiogenesis and vascular growth factor(VEGF), basic fibroblase growth factor(bFGF), Platelet-derived growth factor (PDGF), and their receptors(flt-1 bFGFR.PDGFR) on the invasion, metaslasis of pancreatic carcimoma(PC) and the relationship between the expressions of three kinds of angiogenic factors, their receptors and microvessel count(MVC).
