1.Evaluation of Chinese and English Reports Quality of Randomized Controlled Trial of Acupuncture in the Treatment of Smoking Cessation Based on CONSORT Statment and STRICTA Checklist

Objective To evaluate the reporting quality of randomized controlled trials on acupuncture cessation by using CONSORT statement and STRICTA checklist.Methods According to inclusion and exclusion criteria,9 Chinese and English databases such as CNKI and PubMed were searched by computer,and the retrieved literatures were evaluated according to the CONSORT statement 2010 and STRICTA checklist 2010.Results After exclusion,34 Chinese and English literatures were included,including 27 in Chinese and 7 in English.In a risk review using ROB2,the Cochrane Collaboration's bias risk assessment tool,29 reviews were judged to be high risk overall,2 to be at some risk,and 3 to be at low risk.In the CONSORT statement evaluation,the reporting rate of 23 items was less than 76.4%,including the reporting rate of 17 items was less than 47%.In the STRICTA checklist evaluation,the reporting rate of 9 items was less than 76.4%,including the reporting rate of 4 items was less than 29.4%.Conclusion There are still shortcomings in the methodological design and item reporting standards of acupuncture smoking cessation RCT.It is suggested that researchers should strictly follow the CONSORT declaration and STRICTA list in designing RCT schemes and reporting RCT results in the future.
2.Effect of miR-92a-3p on the progression of ischemic stroke via SIRT1 suppression

Objective To examine the molecular mechanisms of action of miR-92a-3p in ischemic stroke(IS).Methods Database analysis was performed to evaluate miR-92a-3p expression in patients with IS.Oxygen-glucose deprivation/reoxygenation(OGD/R)of HT22 hippocampal neurons was used as an in vitro IS model.PCR was performed to analyze miR-92a-3p expression in HT22 cells fol-lowing 2,4,6,and 8 h of OGD/R.Western blotting was used to analyze the protein expression levels of silent information regulator 1(SIRT1),an miR-92a-3p target gene,and the apoptosis-related protein caspase-3 in HT22 cells after 4 h of OGD/R.Middle cerebral artery occlusion(MCAO)was used as an animal model of IS.Western blotting and PCR were used to analyze the mRNA and protein expression levels of SIRT1 and caspase-3 in the brain tissues of mice following MCAO.Results Database analysis showed increased miR-92a-3p expression in fetal rat cortical neurons after 12 h of OGD and in the brain tissues of mice 3 days post-MCAO.Following 2,4,6,and 8 h of OGD/R in HT22 cells,the expression of miR-92a-3p increased,accompanied by decreased expression of SIRT1 and increased expres-sion of caspase-3.In the brain tissues of mice model of IS,SIRT1 expression decreased and caspase-3 expression increased.Conclusion miR-92a-3p is involved in IS by promoting neuronal apoptosis via SIRT1 inhibition.
3.The development and validation of the quick self-assessment questionnaire for cochlear implant out-come

Objective To develop and validate a"quick self-assessment questionnaire for cochlear implant out-come(QSACI)".Methods A research team,composed of audiologists,otolaryngologists,data analysis experts,and cochlear implant(CI)recipients,was formed to establish objectives,research subject criteria,and framework of the QSACI.An item pool was creaed through literature review and brainstorming.Question items were evaluated and screened,and the framework and answer options of the questionnaire were established.The comprehensibility,etc.,was analyzed and refined through pilot test,interviews,and expert consultation,leading to the development of the final version.A total of 39 post-lingually deafened adults with known stable outcomes completed the question-naire.The split-half and test-retest reliabilty of the questionnaire was analyzed,and the validity was quantitatively analyzed by comparing scores with the categories of auditory performance(CAP)scores.Results The initial item pool of the questionnaire had 18 items,and the final questionnaire consisted of 12 questions in four dimensions:com-munication status,audiological status,medical factors,and other factors.The average score of 39 recipients was 88.81±6.17 and CAP was 6.19±0.94.The questionnaire showed good reliability and validity,with a Cronbach's alpha coefficient of 0.71 and a test-retest reliability of 0.824(P<0.05).The criterion-related validity,assessed by the correlation between the self-assessment questionnaire scores and CAP scores,showed a significant moderate pos-itive correlation(r=0.512,P<0.05).The correlation coefficient between self-assessment and professional assess-ment was 0.720(P<0.05),indicating a significant correlation.The area under the receiver operating characterstic(ROC)curve was 0.82(P<0.05),the cutoff values corresponding to the maximal Youden index were 82.5 and 88.6,therefore score of 85 was taken as the median threshold score of judgement.Conclusion The QSACI reflects the post-imlplant outcomes,and it can serve as a tool for people with postlingually deafness and their families to un-derstand the eligbility of CI and the expected outcomes,helping to establish realistic expectations before CI surgery.
4.Riboflavin reduces the range of ischemic stroke infarction by inhibiting the neuronal apoptosis in mice

6.Identification and activity analysis of ubiquitin ligase TP12446 gene from Trichinella spiralis

Based on the previous transcriptomic experimental data of Trichinella spiralis(T.spira-lis)in this study,the larval stage specific gene TP12446 was screened and its identity in the ubiq-uitin ligase RNF family was predicted.In the study,bioinformatics methods were used to analyze its physicochemical properties and its activity to lay the foundation for further exploring the func-tion of TP12446 gene.The physicochemical properties and protein structure of TP12446 protein were predicted by bioinformatics.Its ubiquitin ligase activity was also verified by ubiquitination re-actions in vitro.The expression characteristics of TP12446 protein in different stage of T.spiralis infection were analyzed by qPCR and Western blot.Bioinformatics analysis showed that TP12446 protein was composed of 453 amino acids and its molecular weight was 51.48 kDa.The protein had a transmembrane structure and contained signal peptides.The results indicated that it was a secre-tory protein and mainly located in the cytoplasmic membrane.The protein structure analysis re-vealed that the protein contained RING and PA domain,its secondary structure was mainly com-posed of α-helix and irregular crimp and there were 10 B cell epitopes on TP12446 protein.The prediction of glycosylation and phosphorylation sites indicated that TP12446 protein contained 38 potential phosphorylation sites.Results of PPI interaction protein prediction showed that TP12446 protein had strong interaction with Usp8,Tmem37,Otub1,Otub2,Ubox5 and CD151.The results of qPCR and Western blot showed that TP12446 gene expression was the highest in the larva stage of T.spiralis,the activity of ubiquitin ligase was verified by ubiquitination reaction in vitro.TP12446 protein was a secretory hydrophobic protein with E3 ubiquitin ligase activity,which was involved in regulating cell cycle and apoptosis.
7.High resolution magnetic resonance vessel wall imaging and hemodynamic characterization of different subtypes of middle cerebral artery infarction

Objective The aim of this study is to investigate the plaque characteristics of different subtypes of middle cerebral artery(MCA)infarction,lenticulostriate artery(LSA)characteristics,and hemodynamic characteristics through high-resolution magnetic resonance vessel wall imaging(HR-MRI VWI)and perfusion weighted imaging(PWI).Methods A retrospective analysis was conducted on patients with unilateral acute or subacute MCA infarction who underwent HR-MRI VWI and PWI examinations at Qinhuangdao First Hospital from January 2022 to April 2024.Patients were divided into three groups according to the type of infarction:artery-to-artery embolism(AA),branch occlusive disease(BOD),branch occlusive disease and artery-to-artery embolism(BOD-AA).Clinical data,plaque characteristics,lenticulostriate artery characteristics,and hemodynamic characteristics were compared and analyzed among the three groups.To determine factors associated with BOD,the BOD and BOD-AA groups were merged into a BOD+group which was then compared with the AA group.To determine factors associated with the AA group,the BOD-AA and AA groups were combined into an AA+group,followed by comparison with the BOD group.Results A total of 42 patients with unilateral acute or subacute MCA infarction were included according to the inclusion and exclusion criteria.Among them,9 patients were classified as having AA,14 patients as having BOD,and 19 as having BOD-AA.The results showed that the degree of stenosis was higher in the AA+group than in the BOD group,and the difference was statistically significant(Z=-2.536,P<0.05);The number of LSA on the affected side was higher in the BOD+group than in the AA group[2.00(1.50,3.00)vs.3.00(3.00,4.00)],and the difference was statistically significant(Z=-2.889,P<0.05);The relative cerebral blood flow(rCBF)of the centrum semiovale was lower in the AA+group than in the BOD group[0.90(0.84,0.93)vs.0.99(0.97,1.03)],and the difference was statistically significant(Z=-4.555,P<0.05);The rCBF in the basal ganglia was lower in the BOD+group than in the AA group[0.89(0.83,0.93)vs.1.01(0.98,1.04)],and the difference was statistically significant(Z=-4.280,P<0.05).Conclusion Patients in BOD+have a lower degree of stenosis compared to AA group.The spatial relationship between plaques and LSA varies across different subtypes of MCA infarction.Plaques in BOD+patients are positioned closer to the LSA orifice and thus have a higher tendency to occlude the LSA orifice.The affected side has few LSAs and the rCBF in the LSA perfusion area.is lower.

10.Antiviral mechanism of Euphorbia helioscopia diterpenoids against Zika virus in vitro

Aim To investigate the anti-Zika virus(ZIKV)mechanism of diterpenoid compound 9 from Euphorbia helioscopia in vitro.Methods The cytotox-icity of compound 9 was evaluated using the CCK-8 as-say.A ZIKV-infected Vero cell model was established,and the antiviral activity was assessed through RT-qPCR,plaque assay,Western blot,and immunofluores-cence.Furthermore,the mechanism of action was elu-cidated using multi-cell line validation,nanoparticle tracking analysis,cellular thermal shift assay,and mo-lecular docking.Results In Vero cells,compound 9 exhibited an EC50 of(3.95±0.15)μmol·L-1 and a CC50 of(272.12±8.56)μmol·L-1,demonstrating significantly higher antiviral efficacy than the positive control drug ribavirin(RBV).Its virus inactivation effect was time-dependent and could significantly re-duce viral load and plaque formation.Studies revealed that compound 9 altered the physicochemical properties of ZIKV particles,including reducing surface charge and increasing particle size distribution.Additionally,it significantly enhanced the thermal stability of the prM protein.Molecular docking analysis indicated that compound 9 formed a high-affinity interaction with the prM protein(binding energy:-38.52 kJ·mol-1)and stabilized its structure through hydrophobic interac-tions.Conclusion Compound 9 exerts in vitro anti-ZIKV activity by directly inactivating the virus,disrup-ting viral particle integrity,and targeting the prM pro-tein.
