Ferrets offer an advantage in nonclinical studies of anti-infective drugs because of their ability to be infected with and spread pathogenic microorganisms, especially viral strains, without the need for host adaptation.Additionally, the clinical symptoms exhibited by infected ferrets are very similar to those of humans.Although ferrets play a very important role in the research and development of antiviral drugs, the scope of their application remains limited. This may be related to the lack of corresponding national standards for laboratory animal feeding and application of ferrets as well as the lack of specific diagnostic and detection reagents. This paper summarizes the characteristics of ferrets as infectious disease models with a summary and analysis of the application direction of ferrets in anti-infective drug research. Our aim is to promote further standardization of the use of ferrets.

Objective:To investigate the effect of internal fixation of medial support and lateral wall on initial weight-bearing stability of femoral intertrochanteric fracture after reconstruction of intertrochanteric fracture. Methods:Based on the digital model of Sawbones femur, intertrochanteric fracture models with different medial support and lateral wall states were constructed. Combined with various intramedullary and extramedullary fixation methods, the mechanical models after fixation were simplified, and seven mechanical simplified models were obtained. The individual body weight load (70 kg) was simulated by applying concentrated force (700 N) to the end nodes of the femoral head. Through the analysis and calculation of each model in StrucMaster software, the axial force, shear force, bending moment, displacement deformation, and support reaction force of each bar in each mechanical model were obtained, and the results were output through the supporting SMConsole. Results:The median bending moment of model 1 was 11.55(5.57, 18.00) N·m, and the median bending moment was 2.89(2.81, 2.94) rad. The median bending moment of model 2 was 13.72(9.30, 20.05) N·m, and the median bending moment was 2.93(2.77, 3.05) rad. The median bending moment of model 3 was 19.65(10.23, 26.11) N·m, and the median bending moment was 2.93 (2.77, 3.05) rad. The median bending moment of model 4 was 25.95(25.14, 36.70) N·m, and the median bending moment was 4.38(3.16, 5.15) rad. The median bending moment of model 5 was 13.66(7.99, 25.62) N·m, and the median bending moment was 3.50(3.32, 3.56) rad. The median bending moment of model 6 was 24.60(9.33, 29.37) N·m, and the median bending moment was 3.90(3.69, 4.10) rad. The median bending moment of model 7 was 41.14(0.16, 41.30) N·m, and the median bending moment was 5.50(5.50, 7.05) rad. The mechanical properties of model 1 were better than those of the other six models. Conclusion:The simplified mechanical model and its mechanical calculation analysis based on the lever reconstruction balance theory can be used to preliminarily reveal the mechanical characteristics of femoral intertrochanteric fractures after internal fixation. The internal fixation methods of the medial support and lateral wall of femoral intertrochanteric fracture reconstruction can improve the initial stability of the weight bearing after internal fixation of femoral intertrochanteric fractures and have guiding significance for the functional rehabilitation of early postoperative weight bearing.

Lung squamous cell carcinoma with intratracheal diffuse leukoplakia as the main manifestation is very rare in clinic. The clinical data of a patient with pulmonary squamous cell carcinoma with intratracheal diffuse leukoplakia admitted to the Affiliated Hospital of Jining Medical University in December 25, 2021 were retrospectively analyzed in order to improve the understanding of this special manifestation. The patient was a 73-year-old male with clinical manifestations of cough, sputum, and blood-stained sputum. Chest CT indicated patchy high-density shadow on the upper right lung, whole-course thickening of trachea and bronchial walls, and bronchoscopy showed diffuse trachea-bronchial mucosa congestion and edema, with a large number of leukoplakia on the surface. The clinical effect was stable after 2 cycles of chemotherapy. Lung squamous cell carcinoma accompanied by diffuse leukoplakia in the trachea is a rare presentation. Chest CT can show thickening of the tracheal and bronchial wall, while the lesion and tumor signs of the primary lesion are not obvious. Electronic bronchoscopy as soon as possible can avoid missed diagnosis.

OBJECTIVE To study the triterpenoid saponins from Anemone rivularis var. flore-minore and their antitumor activities. METHODS The n-butanol extract of 70% ethanol extract from rhizome of the plant was separated. The triterpenoid saponins were separated and purified by normal silica gel column chromatography ,reversed phase ODS column chromatography , Sephadex LH- 20 gel column chromatography and semi-preparation high performance liquid chromatography. The structures of these saponins were identified by spectral analysis (NMR and MS )and physical and chemical properties. MTT assay was used to test the proliferation inhibitory activity of the compounds against five kinds of human tumor cells (HL-60 cells,A549 cells,HepG2 cells,HeLa cells and U 87MG cells ). The apoptosis inducing effect of compound 7 on U 87MG cells was evaluated by flow cytometric Annexin V-FITC/PI staining test. RESULTS:Sixteen triterpenoid saponins were obtained and identified as 3 β-O-β-D-xylopyranosyl-(1→2)-α-L-arabinopyranosyl-oleanolic acid-28-O-α-L-rhamnopyranosyl- (1→4) -β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside(1),3β-O-L-arabinopyranosyl oleanolic acid- 28-O-β-D-glucopyranoside(2),saponin B (3), 163.com oleanolic acid- 3β-O-β-D-glucopyranosyl-(1→2)-α-L-arabino- pyranoside(4),HN-saponin F (5),clematoside S (6),prosapogenin CP 4(7),cussonside B (8),pulsatilla saponin C (9), clemastanoside D (10),3 β-O-β-D-glucopyranosyl-(1→2)-β-L-arabinopyranosyl-hederagenin-28-O-β-D-glucopyranoside(11), ciwujianoside C 3(12),ciwujianoside A 1(13),huzhangoside D (14),kalopanaxsaponin B (15)and hederacolchiside E (16). Compounds 3,4,6-9 displayed inhibitory activities on the proliferation of tumor cells to different extent ,and compound 7 had the strongest activity ;compound 7 induced the apoptosis of U 87MG cell so as to inhibit the proliferation of cancer cells in a time-dependent manner. CONCLUSIONS The obtained 16 saponins are all identified as oleanolane-type ,among which compound 1 is a new compound. The monodesmosidic saponins ,the sugar chain of which attached at C- 3 and a free carboxyl at C- 28, possess stronger antitumor activity than others.

Chinese Medical Journal 2022;135(5):571-583
Chronic hepatitis B virus (HBV) infection remains a global health burden. Timely and effective antiviral therapy is beneficial for patients with HBV infection. With existing antiviral drugs, including nucleos(t)ide analogs and interferon-alfa, patients can achieve viral suppression with improved prognosis. However, the rate of hepatitis B surface antigen loss is low. To achieve a functional cure and even complete cure in chronic hepatitis B patients, new antivirals need to be developed. In this review, we summarized the advantages and disadvantages of existing antiviral drugs and focused on new antivirals including direct-acting antiviral drugs and immunotherapeutic approaches.

The purpose of this study was to understand the effect of gender on the traits of wool. One-year-old male and female Chinese merino sheep (JunKen type) were used to screen differentially expressed proteins in skin tissues by quantitative proteomics using liquid chromatography-tandem mass spectrometry (LC-MS/MS) and data independent acquisition (DIA) strategy. This was followed by GO function annotation, KEGG metabolic pathway and protein network interaction analysis on the differential proteins obtained. The result showed that there were 674 differentially expressed proteins between male and female groups, whereas 280 proteins were up-regulated and 394 proteins were down-regulated in the male group. Through further comparison and analysis, there were 43 differentially expressed proteins related to skin hair follicle development and wool phenotype, 30 up-regulated and 13 down-regulated. The GO annotation analysis of differentially expressed proteins were mainly enriched in 37 processes of molecular function such as oxygen binding, chondroitin sulfate binding, heme binding, glutathione peroxidase activity and glutathione transferase activity; 120 biological processes such as cellular oxidant detoxification, regulation of muscle contraction, notch signaling pathway, calcium ion transmembrane transport and glutathione metabolism; and 31 processes related to cellular components such as mast cell granule, nucleus, sarcoplasmic reticulum and endoplasmic reticulum lumen. KEGG analysis showed that the differentially expressed proteins were involved sixteen signaling pathways, among which MAPK and p53 signaling pathways were closely related to wool growth and development. The protein network interaction analysis discovered that COL1A1 was closely related to MMP2, SPARC, THBS1 and other differentially expressed proteins, which might play a key role in wool growth and development. Thus, this study obtained basic data for revealing the molecular mechanism of sheep wool traits of different gender.

Objective:To prospectively guide the change of chemotherapy regimen in mouse 5-fluorouracil (5-FU) resistance subcutaneous xenograft tumor model derived from gastric cancer patients by the early changes of MRI apparent diffusion coefficient (ADC), and to compare the difference of tumor load between ADC guided dressing change group and volume guided dressing change group. Methods:From January to June 2020, thirty patient-derived xenografts mouse models were established using 5-FU resistant gastric cancer cells coming from patients, and were randomly divided into experimental group and control group by AdaBoost algorithm, with 15 mice in each group. On the 26th day after transplantation, all mice began chemotherapy with 5-FU as the first-line chemotherapy drug, and underwent MR examination once every two days, including T 2WI and diffusion weighted imaging (DWI). Volumes of tumors were measured using an open-source software ITK-SNAP and values of ADC were measured on ADC maps. According to the change rate of tumor ADC value in the experimental group and the tumor volume growth rate in the control group, the replacement time of chemotherapy drugs was determined, and 5-FU was replaced by paclitaxel. The end point of the experiment was the day that the mice entered the cachexia state. Independent-sample t test was used to compare the difference of tumor load between the two groups. Results:After 5-FU treatment, the ADC value of the two groups both increased. The ADC value began to decline on the 4th day after chemotherapy, and the experimental group continued chemotherapy with paclitaxel instead of 5-FU at this time point. The tumor volume growth rate of the control group increased significantly on the 6th day after chemotherapy (from 8.6% to 16.1%), and the control group used paclitaxel instead of 5-FU chemotherapy at this time point. The observed end point was on the 18th day after chemotherapy. The tumor load of the experimental group [(1.82±0.09) cm 3] was lower than that of the control group [(2.01±0.09) cm 3], and the difference was statistically significant ( t=2.25, P=0.033). On the 16th day after chemotherapy in the experimental group and the 18th day after chemotherapy in the control group, the time of paclitaxel administration in both groups was 12 days. The tumor load in the experimental group [(1.61±0.12) cm 3] was also lower than that in the control group [(2.01±0.09) cm 3], and the difference was statistically significant ( t=2.03, P=0.040). Conclusions:For the subcutaneous transplantation model of 5-FU resistant gastric cancer mice, according to the early changes of tumor ADC value after chemotherapy, the replacement of chemotherapy drugs can obtain a lower tumor load, suggesting that it is a feasible method to optimize the chemotherapy regimen.
