1.Neuropeptide Y receptor Y8b(npy8br)regulates feeding and digestion in Japanese medaka(Oryzias latipes)larvae:evidence from gene knockout

Neuropeptide Y receptor Y8(NPY8R)is a fish-specific receptor with two subtypes,NPY8AR and NPY8BR.Changes in expression levels during physiological processes or in vivo regulation after ventricular injection suggest that NPY8BR plays an important role in feeding regulation;this has been found in only a few fish,at present.In order to better understand the physiological function of npy8br,especially in digestion,we used clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)technology to generate npy8br-/-Japanese medaka(Oryzias latipes).We found that the deletion of npy8br in medaka larvae affected their feeding and digestion ability,ultimately affecting their growth.Specifically,npy8br deficiency in medaka larvae resulted in decreased feed intake and decreased expression levels of orexigenic genes(npy and agrp).npy8br-/-medaka larvae fed for 10 d(10th day of feeding)still had incompletely digested brine shrimp(Artemia nauplii)in the digestive tract 8 h after feeding,the messenger RNA(mRNA)expression levels of digestion-related genes(amy,lpl,ctra,and ctrb)were significantly decreased,and the activity of amylase,trypsin,and lipase also significantly decreased.The deletion of npy8br in medaka larvae inhibited the growth and significantly decreased the expression of growth-related genes(gh and igf1).Hematoxylin and eosin(H&E)sections of intestinal tissue showed that npy8br-/-medaka larvae had damaged intestine,thinned intestinal wall,and shortened intestinal villi.So far,this is the first npy8br gene knockout model established in fish and the first demonstration that npy8br plays an important role in digestion.
2.Nursing care of a patient before operation with mechanical circulation-assisted bridging heart transplantation

To summarize the nursing care of a patient with acute myocardial infarction complicated with cardiogenic shock before operation.The main nursing points are as follows:in the acute stage,the integrated rescue was implemented with rapid on-machine coordination of external cardiopulmonary resuscitation,multidisciplinary collaboration to ensure safe patient transport;during the bridging period,the combined operation care of extracorporeal membrane oxygenation,intra-aortic balloon counter pulsation and continuous kidney replacement therapy was carried out with goal-oriented anticoagulation care,prevention of catheter-related infection with the assistance of mechanical circulation,and neurological function monitoring.The patient successfully passed the waiting period for heart transplantation and underwent heart transplantation 21 days after admission.With the follow-up for 1 year,the patient recovered well.

4.Studies on the synthesis and anti-fatigue activity of the salidroside derivatives

Objective A series of 4 substituted salidroside derivatives were designed and synthesized .Their anti-fatigue effects were investigated .Methods With five-acetyl glucose and different 4-substituted benzyl tyrosols as the starting materi-als ,salidroside derivatives were synthesized through glycosidation and deacetylation reactions .The exercise exhaustive mice model was used to study the anti-fatigue effects of those synthesized derivatives by comparing the loading swimming time of mice .Results 10 novel salidroside derivatives were synthesized .The loading swimming tests showed that the swimming time of the mice in the positive group (salidroside) and 3a-1 group (phenethyl-β-D-glucoside) was longer than that in the control group with statistically significant difference(P<0 .05) .The swimming times for other groups were similar to control group with no statistically significant difference .Conclusion This synthetic method for salidroside derivatives was convenient and feasible for large production .The 4-hydroxyl groups on the benzene ring of salidroside and its derivatives may be the active site responsible for their anti-fatigue activity .
5.Analysis of the factors associated with treatment outcomes in spinal cord decompression sickness

Objective To analyze the factors associated with treatment outcomes of dive-related spinal cord decompression sickness (DCS).Methods A retrospective analysis was made in 95 patients with dive-related spinal cord DCS (with an age range of 43 ± 11).Such medical data as dive data,clinical medical data,spinal cord magnetic resonance imaging (MRI),electrophysiological data,latency of DCS symptoms upon completion of diving,the relationship between symptom onset and the interval from the start of HBO therapy to the termination of HBO therapy were collected for analysis.The severity and treatment outcomes of spinal cord DCS were assessed by the Boussuges grade system during the acute stage and 3 months after treatment.The treatment outeomes of spinal cord DCS were classified into complete recovery and incomplete recovery.All the patients were treated with the widely-recognized treatment protocol.Results Fifty-six patients (59%) had complete recovery,3 months after DCS seizure,and 39 patients (41%) had incomplete recovery following 3 months of treatment.Complete recovery rate in the young patients was higher than that in the old ones.The complete recovery rate of the patients with normal spinal cord MRI detection results was higher than that of the patients with abnormal MRI detection results,and the complete recovery rate of the patients with normal electrophysiological detection results was higher than that of the patients with abnormal electrophysiological detection results.The complete recovery rate of the patients with Boussuges scores ≤7 at onset time was higher than that of the patients with Boussuges scores of over 7.The complete recovery rate of the patients without back pain was higher than that of the patients with acute back pain.The complete recovery rate of the patients with over 50-minute symptom latency upon surfacing was higher than that of the patients with less than 50-minute symptom latency.The complete recovery rate of the patients with less than 4.5-hour time interval between symptom onset and HBO therapy was higher than that of the patients with over 4.5-hour time interval.Conclusions The patients with younger age,normal MRI detection results,normal electrophysiological data,Boussuges scores ≤7,no complaint of back pain after surfacing,longer symptom latency upon dive completion,and shorter time interval between symptom onset and HBO therapy would have better treatment outcomes.

7.Investigation on the sleep status and mental stress of the commissioned and enlisted onboard the large naval vessel

Objective To investigate the sleep status and mental stress of the commissioned and enlisted onboard the large naval vessel.Methods Sample surveys and statistical analyses were made on the sleep status and mental stress of 80 commissioned and enlisted personnel onboard the large naval vessel by using Pittsburgh Sleep Quality Index (PQSI) and Acute Stress Response Scale (ASRS).Results (1) The average sleep time at night was (5.61 ± 1.84) hours,and total scores of PSQI were (8.05 ± 3.55).Survey results indicated that 61.9% of the subjects had sleep disorder.(2) The most common problem of sleep disorder among the commissioned and enlisted personnel was difficult in falling asleep,and other problems were reduced sleep time,wake-up during sleep,early awakening,having dreams or nightmares.(3) Total PSQI scores and various factors such as sleep efficiency,sleep disorder,and daytime dysfunction were all positively correlated with emotional response and changes in behavior.Conclusions There existed certain problems in the overall sleep status and mental stress among the commissioned and enlisted onboard the large naval vessel.Such intervention measures as mental consultation and further improvement of shipboard environment should be taken to improve their sleep status and enhance their mental health.

Malachite green (MG) is a dye which has been widely used as a topical fungicide and antiseptic in aquaculture throughout the world. Now,MG has become a highly controversial compound due to its potential highly residual and toxic characteristics in recent years. In the paper,the advances in toxicity,toxicological mechanism,detection methods,metabolism and removal of malachite green and its metabolite were reviewed,and the problems in aquaculture and the safety of aquaculture technology were discussed,in order to make us be aware of potential hazard of malachite green and provide a dependable evidences of safely breeding in aquatic products.
