1.Protective effect of catalpol on liver injury in acute hepatitis B rats by regulating TLR4/NF-κB signaling pathway

Objective:To investigate the protective effect of catalpol on the liver of rats with acute hepatitis caused by hepatitis B virus(HBV)infection and its possible mechanism.Methods:SD rats were randomly divided into Sham group,Model group,acyclo-vir group(ACY group),low-dose catalpol group(L-CAT group),medium-dose catalpol group(M-CAT group)and high-dose catalpol group(H-CAT group).The levels of serum inflammatory cytokines were detected by ELISA.Flow cytometry was used to detect the per-centage of peripheral blood Th17 cells.Hematoxylin-eosin staining and TUNEL assay were used to detect liver tissue injury and apopto-sis.Western blot analysis was used to detect the expression of related proteins in liver tissue.Results:Compared with the Sham group,the levels of serum HBsAg,HBeAg,ALT,AST,Th17 cell ratio,IL-17 and IL-23,and the expression of TLR4 and p-p65 protein in liver tissues were significantly increased in the Model group,while the levels of serum IL-13 and IL-4 were significantly decreased in the Model group,the differences were statistically significant(P<0.05).Compared with the Model group,the levels of serum HBsAg,HBeAg,ALT,AST,Th17 cell ratio,IL-17 and IL-23,as well as the expressions of TLR4 and p-p65 protein in liver tissues of rats in the ACY,M-CAT and H-CAT groups were significantly decreased,while the levels of serum IL-13 and IL-4 were significantly in-creased,the differences were statistically significant(P<0.05).Compared with the L-CAT and M-CAT groups,the levels of serum HB-sAg,HBeAg,ALT,AST,Th17 cell ratio,IL-17 and IL-23,the expression of liver TLR4 and p-p65 protein were significantly de-creased,while the levels of serum IL-13 and IL-4 were significantly increased in the H-CAT group,the differences were statistically significant(P<0.05).Conclusion:Catalpol improves HBV infection-induced liver injury in rats by reducing Th17 cell proportion and pro-inflammatory cytokine levels,and increasing anti-inflammatory cytokine levels,possibly by inhibiting TLR4/NF-κB p65 signaling pathway activation.
2.Changes in voltage-dependent anion channel 3 in an animal model of sepsis-induced myocardial injury

Objective To observe changes in voltage-dependent anion channel 3(VDAC3)in a mouse model of sepsis-induced myocardial injury and to explore its potential mechanism.Methods Twenty male C57BL/6J mice were divided randomly into a Sham group and Sepsis group,respectively(n=10 mice per group).Sepsis was induced by the cecal ligation and puncture(CLP).Serum levels of interleukin(IL)-6,tumor necrosis factor(TNF)-α,creatine kinase MB(CK-MB),and cardiac troponin T(cTnT)were detected by enzyme-linked immunosorbent assay.Pathological changes in heart tissue were observed by hematoxylin and eosin staining.Structural and functional changes in the heart were evaluated by echocardiography.Changes in total glutathione,reduced glutathione(GSH),oxidized glutathione,and malondialdehyde(MDA)in heart tissue were detected by spectrophotometry.The morphological structure of mitochondria in mouse cardiomyocytes was observed by transmission electron microscopy.Expression levels of IL-6,IL-1β,VDAC3,glutathione peroxidase 4(GPX4),solute carrier family 7 member 11(SLC7A11),lipocalin-2(LCN2),and prostaglandin-endoperoxide synthase 2(PTGS2)mRNA were detected by real-time quantitative polymerase chain reaction and the localization and expression of VDAC3 and GPX4 proteins in mouse heart tissue were detected by immunofluorescence staining.The correlations between VDAC3 mRNA and GPX4,SLC7A11,PTGS2,LCN2,IL-6,and IL-1β mRNA were analyzed.Expression levels of VDAC3,GPX4,and SLC7A11 proteins were detected by Western blot.Results IL-6,TNF-α,CK-MB,and cTnT levels were significantly higher in the Sepsis group compared with the Sham group(P<0.05).In the Sepsis group,myocardial fibers were torn,the ventricular wall was thickened and edematous,the mitochondrial membrane was ruptured,and mitochondrial cristae were broken or absent.GSH levels were significantly reduced in the Sepsis group(P<0.05)and the lipid peroxide MDA was increased in the Sepsis group(P<0.05)compared with the Sham group.VDAC3,GPX4 and SLC7A11 mRNA and protein levels were all lower in the Sepsis group compared with the Sham group(P<0.05),while expression levels of IL-6,IL-1β,LCN2,and PTGS2 mRNA were increased(P<0.05).VDAC3 mRNA was positively correlated with GPX4 and SLC7A11 mRNA levels,and negatively correlated with LCN2,PTGS2,IL-6,and IL-1β.Conclusions VDAC3 expression decreases in myocardial injury,and it may participate in the occurrence of sepsis-induced myocardial injury by regulating ferroptosis.

5.Analysis of issues in quality control test results of linear accelerators in China

Objective To study the unqualified items in the reported quality control tests of linear accelerators, analyze the issues in quality control tests, and propose the key points and development directions for accelerator quality control test in China. Methods A literature review was conducted using the CNKI database to analyze the qualified rates of test items and the issues in quality control tests. Results In the literature on the quality control tests of linear accelerators, except for a few provinces where the qualified rates of all test items were 100%, unqualified items were reported in most of the literature. There were unqualified items related to X-ray and electron beam in different reports. Error of dose indication was the unqualified item with the highest occurrence rate in X-ray test, and the item with the lowest qualified rates in X-ray and electron beam tests. The lowest qualified rate of X-ray dose indication error was 73.5% in 2016, and the lowest qualified rate of electron beam dose indication error was 46.2% in 2017. Conclusion Tests should be carried out strictly according to the items and intervals specified by the quality control test standards. Hospitals, radiation health technology service institutions, and health administrative departments should each fulfill their respective responsibilities, work together, and place emphasis on ensuring effective quality control tests of linear accelerators to further enhance the overall quality control standards for these devices.
6.Current status of central vascular access devices in pediatric patients in 31 hospitals nationwide

Objective:To explore the currents status of central vascular access devices (CVADs) in children's medical center in China and to provide a reference for the standardized use of CVADs and specialized nursing as well as a basis for pediatric intravenous therapy management.Methods:Totally 31 hospitals of Futang Research Center of Pediatric Development, children's hospitals of the medical treatment alliance as well as women and children care centers and general hospitals with pediatric departments were selected by convenient sampling from July 23rd to 31st, 2018. All the pediatric patients using CVADs were investigated with the Central Vascular Access Device Questionnaire which was designed by the research group, and the incidence of complications was also observed. Totally 1 333 questionnaires were collected, among which 1 288 were valid, accounting for an effective recovery rate of 96.62%.Results:There were 22 ClassⅢ and 9 ClassⅡ hospitals among the 31 hospitals. Among the 1 288 pediatric patients who used CVADs, 860 used peripherally inserted central catheters (PICC) , accounting for 66.77%; 342 used central venous catheters (CVC) , accounting for 26.55%; 77 used implantable venous access port (PORT) , accounting for 5.98%; and 9 used umbilical venous catheters, accounting for 0.70%. CVADs were mainly used in the Departments of Hematological Neoplasms, ICU, Intensive Care Unit Pediatric Surgery, Vasculocardiology and Neonatals. PICC, CVC and PORT were mainly applied to upper extremities (90.00%, 74 cases) , jugular vein (73.39%, 251 cases) and the breast (96.10%, 74 cases) respectively. In the maintenance of CVADs, there was the problem of co-existing dressings.Conclusions:CVADs have been widely used in pediatric patients, and the choice of CVADs is relatively standardized. However, the maintenance of the catheters, the use of dressings, the selection of infusion connectors and the control of related complications still need to be strengthened. It is recommended that nursing professionals and nursing managers should further strengthen their knowledge about the placement and maintenance of CVADs, and intensify their management and supervision in accordance with international and national guidelines and standards for intravenous infusion therapy to further reduce the incidence of vascular access complications in pediatric patients.
7.MiR-183-5p Promotes Proliferation, Metastasis and Angiogenesis in Breast Cancer Cells through Negatively Regulating Four and a Half LIM Protein 1

Purpose:
Four and a half LIM protein 1 (FHL1) is involved in breast cancer (BC) development, but the regulatory mechanism involved remain unclear. In the present study, we examined the role of FHL1 in BC development.
Methods:
The expression of FHL1, miR-183-5p, and miR-96-5p in BC tissues was analyzed using StarBase analysis. FHL1 expression in BC tissues, a normal human breast epithelial cell line, and BC cell lines was detected using quantitative reverse transcription polymerase chain reaction (qRT-PCR). The relationship between FHL1 and miR-183-5p/miR-96-5p was analyzed via Pearson's rank correlation, TargetScan, and a dual-luciferase reporter assay. BT549 and MDA-MB-231 cells were transfected with either FHL1 and miR-183-5p mimics, or siFHL1 and a miR-183-5p inhibitor, respectively. The viability, colony number, migration, invasion, and tube length of BT549 and MDA-MB-231 cells were examined using cell counting kit-8, colony formation, wound-healing, Transwell, and tube formation assays, respectively. The levels of FHL1, vascular endothelial growth factor (VEGF), p53, E-cadherin, N-cadherin, and vimentin were quantified using western blotting and qRT-PCR.
Results:
FHL1 expression was downregulated in BC tissues and cells, whereas miR-183-5p and miR-96-5p were upregulated in BC tissues (negative correlation with FHL1 expression).FHL1 overexpression inhibited the viability, colony number, migration, and invasion of BC cells and the expression of VEGF, N-cadherin, and vimentin, and increased the expression of FHL1, p53, and E-cadherin in BT549 cells. Furthermore, a miR-183-5p mimic reversed these effects of FHL1 overexpression, whereas FHL1 silencing caused opposite results to those observed in MDA-MB-231 cells; however, this was reversed by a miR-183-5p inhibitor.
Conclusion
Our study suggests that miR-183-5p promotes cell proliferation, metastasis, and angiogenesis by negatively regulating FHL1 in BC.
9.Composition and drug resistance analysis of pathogens causing genital tracts infection during premature rupture of membranes

Objective To investigate the composing characteristics and drug resistances of pathogens isolated from genital tracts in premature rupture of membranes of obstetrics from 2012 to 2014 ,for instructing clinical application of antibiotics reasonably . Methods A retrospective investigation analysis was made for all the isolated bacteria from genital tracts specimens as well as their drug resistances from 2012 to 2014 .Results The results shows that 598 strains of bacterial pathogens were isolated from 2000 detected samples ,The infection rate of bacterial pathogens was 29 .90% .The top four bacteria pathogens were E .coli(31 .44% ) ,Can . albicans(20 .90% ) ,Str .agalactiae(19 .23% ) and Sta .aureus(10 .03% ) .E .coli and Sta .aureus were highly resistant to commonly used antibiotics and demonstrated multi‐drug resistance .Str .agalactiae and Can .albicans were lowly resistant to commonly used antibiotics .Conclusion Inspecting pathogens and studying the composition of pathogens and the trend of their drug resistance are im ‐ portant to rationally select antibiotics ,decrease the occurrence of drug resistant strains in perinatal period ,and control the outbreak and prevalence of nosocomial infection .
10.Application of Integrative Chinese and Western Medicine Fast-track Rehabilitation in Surgery for Patients During Peri-operation Period of Gynecological Laparoscopy

Objective To observe the effect of integrative Chinese and western medicine fast-track rehabilitation in surgery (FTS) for patients during peri-operation period of gynecological laparoscopy. Methods A total of 226 patients receiving gynecological laparoscopic surgery were randomized into observation group and control group, 113 cases in each group. The observation group received integrative Chinese and western FTS treatment, and the control group received routine non-surgery treatment. After operation, time for the first post-operation anal gas evacuation, time for out-of-bed activity, hospitalization time, and incidence of post-operation complications were compared between the two groups. Results In the observation group, time for the first post-operation anal gas evacuation and time for out-of-bed activity were earlier, and hospitalization time was shorter than those in the control group (P<0.05). The incidence of post-operation complications was 0.9%, lower than 4.4% in the control group ( P<0.05). Conclusion Integrative Chinese and western medicine fast-track rehabilitation in surgery shows certain effect on promoting enterokinesia, shortening time for the anal gas evacuation, reducing post-operation complica-tions, promoting post-operation recovery of gastrointestinal function of patients receiving gynecological la-paroscopy .
