1.Investigation of Effect of Different Drying Conditions on Appearance Characteristics and Internal Indicators of Pinelliae Rhizoma Based on Standardization

ObjectiveTo explore the effect of different drying conditions on the appearance and intrinsic quality indicators of Pinelliae Rhizoma for screening suitable drying conditions, so as to provide reference for its standardized production and quality evaluation. MethodsDifferent dried samples of Pinelliae Rhizoma were prepared by lime-assisted sweating method and intermittent drying method. Visual analysis was employed to measure the color brightness values(L*) of the surface, cross-section and powder of the samples, texture analyzer was used to determine the hardness of the samples under different drying conditions. The total starch content was calculated by measuring the contents of amylose and amylopectin in the samples with ultraviolet-visible spectroscopy. High performance liquid chromatography(HPLC) was used to determine the contents of seven nucleoside components(uracil, hypoxanthine, uridine, inosine, guanosine, β-thymidine and adenosine) in the samples. Pearson correlation analysis was conducted to explore the correlation between the external characteristics and intrinsic indicators of the different dried samples. Principal component analysis(PCA) was used to comprehensively rank the data of various indicators, and partial least squares-discriminant analysis(PLS-DA) was used to screen differential components with variable importance in the projection(VIP) value>1. Furthermore, the difference between the optimal drying condition for Pinelliae Rhizoma and the traditional sun-drying method was explored by independent samples t-test. ResultsWith the increase of temperature, the color of the intermittently dried samples gradually deepened, while their hardness gradually decreased. Concurrently, the contents of extract, total starch, uridine and adenosine exhibited an upward trend, whereas the contents of uracil, hypoxanthine and inosine displayed a downward trajectory. Compared with the intermittent drying group, the content of extract in the samples subjected to lime-assisted sweating increased. With the increase of lime dose, the hardness and the total content of nucleoside components in the samples showed a downward trend, while the total starch content showed an upward trend. Correlation analysis showed that the comprehensive score of L* was negatively correlated with the contents of uracil, hypoxanthine and inosine, and positively correlated with the contents of uridine, guanosine and adenosine. Hardness was negatively correlated with adenosine content, and positively correlated with the contents of inosine, uracil and hypoxanthine. Through comprehensive consideration and comprehensive score of principal components, the method of 5% lime-mixed sweating for 6 days emerged as the top-ranking approach. Except for the extract, the results of independent samples t-test showed that there was no significant difference between the 5% lime-mixed sweating for 6 days and the traditional sun-drying in terms of other content indicators. ConclusionThe whiteness and firmness of Pinelliae Rhizoma exhibit significant correlations with its chemical composition, while uridine, uracil, guanosine, adenosine and inosine are the key constituents responsible for the quality difference of Pinelliae Rhizoma under different drying conditions. The lime-assisted sweating method optimized in this study can be proposed as a viable alternative to the traditional sun-drying method. This method not only ensures the quality of the medicinal material but also effectively reduces the drying time and prevents mold contamination, which provides a valuable reference for the standardization of drying conditions and the establishment of quality evaluation criteria for Pinelliae Rhizoma.
2.Evidence that metformin promotes fibrosis resolution via activating alveolar epithelial stem cells and FGFR2b signaling.

Idiopathic pulmonary fibrosis (IPF) is a progressive disease lacking effective therapy. Metformin, an antidiabetic medication, has shown promising therapeutic properties in preclinical fibrosis models; however, its precise cellular targets and associated mechanisms in fibrosis resolution remain incompletely defined. Most research on metformin's effects has focused on mesenchymal and inflammatory responses with limited attention to epithelial cells. In this study, we utilized Sftpc lineage-traced and Fgfr2b conditional knockout mice, along with BMP2/PPARγ and AMPK inhibitors, to explore metformin's impact on alveolar epithelial cells in a bleomycin-induced pulmonary fibrosis model and cell culture. We found that metformin increased the proliferation and differentiation of alveolar type 2 (AT2) cells, particularly the recently identified injury-activated alveolar progenitors (IAAPs)-a subpopulation characterized by low SFTPC expression but enriched for PD-L1. Single-cell RNA sequencing revealed a reduction in apoptosis among mature AT2 cells. Interestingly, metformin's therapeutic effects were not significantly affected by BMP2 or PPARγ inhibition, which blocked the lipogenic differentiation of myofibroblasts. However, Fgfr2b deletion in Sftpc lineage cells significantly impaired metformin's ability to promote fibrosis resolution, a process linked to AMPK signaling. In conclusion, metformin alleviates fibrosis by directly activating AT2 cells, especially the IAAPs, through a mechanism that involves AMPK and FGFR2b signaling, but is largely independent of BMP2/PPARγ pathways.
3.Research on multiscale energy distribution of fetal heart rate signal based on fractal features

Objective To study the multifractal features of fetal heart rate(FHR)signals,and to investigate the multiscale energy distribution features of FHR variability.Methods The study focuses on FHR signals from an open database released by Czech Technical University-University Hospital in Brno(CTU-UHB).Daubechies second-order wavelets db2 was applied to decompose FHR signals into level d1-d7 detail components and approximation components.R/S analysis was performed on the decomposed components,and Hurst index was used to study the multifractal features of the decomposed signals.Hurst index<0.5 indicated that the signal had anti-persistence,Hurst index>0.5 indicated that the signal had positive persistence,and the decomposed signal was reconstructed by using fractal features.The distribution law of energy was analyzed.Results The Hurst index of d1-d7 approximation components was>0.5,which indicates that the approximation components of FHR signal have positive persistent single fractal characteristics.The Hurst index of d1-d3 detail components was>0.5,the Hurst index of d7 detail components was<0.5,and the Hurst index of d4-d6 detail components contained both>0.5 and<0.5,showing the bi-fractal feature.Based on the above multi-fractal analysis results,the FHR decomposed signals were reconstructed into three different subsequences,i.e.microscale sequence,mesoscale sequence,and macroscale sequence.The statistical results showed that the energy of these three sequences accounts for approximately 5%,70%,and 25%of the total energy,respectively(P<0.05).Conclusion The FHR signal was characterized by complex multifractal features,and the energy of the fetal heart rate signal was mainly concentrated in the mesoscale.
4.Visualization analysis of fetal health status assessment assisted by artificial intelligence technology

Objective Based on the knowledge graph to analyze the research status and application trends of artificial intelligence(AI)in the field of fetal health status assessment,and to explore the development context and research hotspots.Methods The relevant literature in the field of fetal health status assessment was retrieved in the Web of Science,PubMed,IEEE Xplore,CNKI and Wanfang Data Knowledge Service Platform,and the search time period was from January 2015 to December 2024.The CiteSpace software was used to generate visual maps of authors,institutions and key words,and bibliometric analysis was conducted on institutions,authors and key words,etc.Results Globally,the number of annual publications of research related to the application of AI to the assessment of fetal health status had been on the rise in the past 10 years.The research hotspots focused on influencing factors,prenatal diagnosis,feature extraction,diagnostic models,machine learning,deep learning,fetal heart rate,etc,indicating that AI had promoted the transformation of fetal condition monitoring and clinical decision-making towards digital diagnosis and treatment driven by deep learning through technological innovation.Conclusion At present,research on AI assisted intelligent monitoring of fetal health mainly focuses on signal processing,feature extraction,and innovative applications.The combination of medical and engineering is an important way to carry out related research and achieve application implementation.In the future,the intelligent,digital,and precise processes in this field will continue to accelerate.
5.Molecular epidemiological characteristics of clinical Haemophilus influenzae isolates from adults and children with community-acquired pneumonia

OBJECTIVE To investigate the molecular epidemiological characteristics of clinical Haemophilus influ-enzae(Hin)isolates from adults and children with community-acquired pneumonia(CAP)so as to provide theo-retical bases for effective prevention and treatment of Hin infection.METHODS The patients who were hospital-ized in respiratory and critical care medicine department and pediatrics department of Xinjiang Production and Con-struction Corps Hospital due to CAP from Jan.2023 to Dec.2024 were enrolled in the study,and the positive rates of Hin in sputum specimens were statistically analyzed.The clinical distribution and results of drug suscepti-bility testing for Hin strains were observed and compared between the adults with CAP and the children with CAP.The capsular types,drug resistance genes and multilocus sequencing typing(MLST)subtypes were detected for 72 strains of Hin.RESULTS The positive rate of Hin was higher among the children with CAP(9.46%)than among the adults with CAP(2.71%).The Hin was more prevalent among the children with CAP than among the adults,it was highly prevalent in autumn and winter,with the population dominated by the children aged between 13 and 17 years old(12.67%).The positive rate of β-lactamase-producing Hin strains was 74.71%among the a-dult patients and 90.17%among the children,respectively;the drug resistance rates to ampicillin were highest(73.86%and 92.57%),the drug resistance rates of the strains isolated from the children to the two types of β-lac-tams were higher than those of the strains isolated from the adults(P<0.05).The non-typeable Hin strains(94.45%)were the predominant type among the 72 strains of Hin,the production of β-lactamase mediated by blaTEM-1 was the major drug resistance mechanism.The result of MLST showed that CC155(ST-155),CC11(ST-103)and CC107(ST-1002)were the main subtypes.CONCLUSIONS The β-lactamase-producing non-typeable Hin strains are dominant among the Hin strains isolated from the adults and children with CAP in this area.The isolation rate of the strains is high among the children than among the adults.The strains are highly prevalent in autumn and winter,and ST-155 is the predominant clone type.The strains are highly resistant to ampicillin,which should be attached great importance to.
6.A prospective study of impact of spicy food intake on risk for cardio/cerebrovascular disease in residents aged 30-79 years

Objective:To explore the association between spicy food intake and the risk for cardio/cerebrovascular diseases.Methods:Data were collected from the China Kadoorie Biobank project conducted in Pengzhou, Sichuan Province. Using the Cox proportional hazards regression model, we analyzed the associations of the frequency of spicy food intake, spicy level, types of spicy food, and the age when regular intake of spicy food began (intake in 1 day/week), with the risk for cardio/cerebrovascular disease. Furthermore, the associations with the risks for ischemic heart disease (IHD) and cerebrovascular diseases, as well as the risk of ischemic stroke (IS) and hemorrhagic stroke (HS) were analyzed.Results:A total of 54 859 study participants were included in the study, in whom 49 320 had spicy food intake (89.90%). In these participants, 37 680 (68.69%) had spicy food intake in 6-7 days/week, 5 036 (9.18%) had spicy food intake in 1-5 days/week, and 6 604 (12.03%) had spicy food intake once a week; 5 539 (10.10%) had never/almost never had spicy food intake. After adjusting for multiple confounding factors, compared with those who never/almost never had spicy food intake, intake of spicy food was associated with reduced risks for IHD (intake in 6-7 days/week: HR=0.86, 95% CI: 0.78-0.95), cerebrovascular diseases (intake in 6-7 days/week: HR=0.88, 95% CI: 0.81-0.96), and IS (intak in 6-7 days/week: HR=0.85, 95% CI: 0.76-0.95). With the increase of spicy food intake frequency, the risk for cardio/cerebrovascular disease decreased (intake in 1-5 days/week: HR=0.91, 95% CI: 0.85-0.98; intake in 6-7 days/week: HR=0.89, 95% CI: 0.84-0.94) (trend test P<0.001). However, no statistical association was found between spicy food intake and the risk for HS. In terms of spicy level, after adjusting for multiple confounding factors, compared with those who never/almost never had spicy food intake, intake of spicy food was associated with reduced risk for cardio/cerebrovascular disease (moderate: HR=0.86, 95% CI: 0.82-0.90) and cerebrovascular disease (moderate: HR=0.90, 95% CI: 0.84-0.97). With the increase of spicy level, the risk for IHD decreased (moderate: HR=0.86, 95% CI: 0.79-0.93; strong: HR=0.84, 95% CI: 0.74-0.95) (trend test P<0.001). After adjusting for multiple confounding factors, compared with those who never/almost never had spicy food intake, intake of any type of spicy food was associated with reduced risk for cardio/cerebrovascular disease, IHD, and cerebrovascular disease. Regulat intake of spicy food from age 0-10 years was associated with reduced risk for cardio/cerebrovascular disease, IHD, and cerebrovascular disease. Regular intake of spicy food from age 11-20 years reduced the risk for cardio/cerebrovascular disease and IHD. There was no significant association between the regular intake of spicy food from age 21-79 years and the risks for cardio/cerebrovascular disease, IHD and cerebrovascular disease. Conclusion:The intake of spicy food could reduced the risk for cardio/cerebrovascular diseases, IHD, cerebrovascular diseases and IS in residents aged 30-79 years in Sichuan.
7.Research progress of anti-integrin αvβ6 autoantibody in ulcerative colitis

Ulcerative colitis (UC) is a chronic inflammatory disease of unknown etiology, primarily involving the colon and rectum. Characterized by recurrent episodes and prolonged disease course, UC requires dynamic monitoring and evaluation. Current commonly used methods for disease monitoring and assessment include C-reactive protein (CRP), fecal calprotectin (FC), and colonoscopy. However, CRP lacks specificity, FC fails to effectively differentiate UC from Crohn's disease, while colonoscopy involves complex bowel preparation, is invasive, and suffers from poor patient compliance. Therefore, there is an urgent clinical need to identify non-invasive biological markers with high sensitivity and specificity for the diagnosis and evaluation of UC. Recent studies have demonstrated that anti-integrin αvβ6 autoantibodies hold significant value in the diagnosis, differential diagnosis, and disease assessment of UC, potentially emerging as an important clinical biomarker. This article reviews the research progress of anti-integrin αvβ6 autoantibodies in UC for reference.
8.Intelligent design of nucleic acid elements in biomanufacturing.

Nucleic acid elements are essential functional sequences that play critical roles in regulating gene expression, optimizing pathways, and enabling gene editing to enhance the production of target products in biomanufacturing. Therefore, the design and optimization of these elements are crucial in constructing efficient cell factories. Artificial intelligence (AI) provides robust support for biomanufacturing by accurately predicting functional nucleic acid elements, designing and optimizing sequences with quantified functions, and elucidating the operating mechanisms of these elements. In recent years, AI has significantly accelerated the progress in biomanufacturing by reducing experimental workloads through the design and optimization of promoters, ribosome-binding sites, terminators, and their combinations. Despite these advancements, the application of AI in biomanufacturing remains limited due to the complexity of biological systems and the lack of highly quantified training data. This review summarizes the various nucleic acid elements utilized in biomanufacturing, the tools developed for predicting and designing these elements based on AI algorithms, and the case studies showcasing the applications of AI in biomanufacturing. By integrating AI with synthetic biology and high-throughput techniques, we anticipate the development of more efficient tools for designing nucleic acid elements and accelerating the application of AI in biomanufacturing.

9.Urban-Rural Disparities in Activities of Daily Living Among Older Adults in Sichuan Province and the Influencing Factors

Objective To investigate the disparities in the activities of daily living(ADL)among older adults living in urban and rural areas of Sichuan Province,China and the influencing factors,and to provide a basis for promoting the health of older adults and formulating relevant policies.Methods Using data from the Seventh Health Services Survey of Sichuan Province,we enrolled 7 369 older adults from urban and rural areas and analyzed their ability to perform ADL and the relevant influencing factors.All participants were aged 60 years or older.Chi-square tests and logistic regression were performed to identify the influencing factors.Results The ADL impairment rate among older adults living in urban areas of Sichuan Province was 17.7%,which was higher than the rate of 13.8%among older adults living in rural areas.According to the results of logistic regression analysis,age≥80 years(odds ratio[OR]=3.725;95%CI,2.460-5.639),accidental injuries(OR=2.375;95%CI,1.597-3.532),and good sleep quality(OR=0.420,95%CI:0.289-0.612)were factors influencing ADL among older adults living in urban areas,while age ≥ 80 years(OR=1.867;95%CI,1.459-2.390),being married(OR=0.805;95%CI,0.664-0.976),accidental injuries(OR=1.936;95%CI,1.564-2.397),and participation in social activities(OR=0.417;95%CI,0.352-0.495)were factors influencing ADL among older adults living in rural areas.Conclusion There are differences in ADL impairment rates and the relevant influencing factors between urban and rural older adults in Sichuan Province.These findings underscore the need to consider urban-rural differences when adopting relevant policies and measures.
10.Effect of the Otago Exercise Program on Activities of Daily Living in Older Adults With Sarcopenia

Objective To explore the effects of the Otago Exercise Program(OEP)on activities of daily living,muscle strength,balance,and physical function in older adults with sarcopenia,to compare OEP with conventional exercise training,and to provide a basis for clinical rehabilitation programs for older adults with sarcopenia.Methods In this randomized controlled trial,120 older adults clinically diagnosed with sarcopenia were enrolled.The participants were randomly assigned to the OEP intervention group(experimental group)and the conventional exercise intervention group(control group),with 60 in each group.The experimental group underwent 12 weeks of OEP training,three times a week,with each session lasting 45 minutes.The control group underwent conventional exercise training following the same schedule.The Modified Barthel Index was used as the primary outcome measure to assess activities of daily living.Secondary outcome measures included muscle strength,gait stability,dynamic balance,and physical function status,evaluated using grip strength,6-meter walking speed,the Timed Up and Go Test(TUGT),and the Short Physical Performance Battery(SPPB).Results A total of 120 older adults with sarcopenia were included.The mean age of the participants was(80.17±8.48)years.Baseline data before treatment showed no statistically significant differences between the two groups.Both groups completed the treatment within 12 weeks without experiencing any adverse events.The baseline data for the experimental group were as follows,MBI at(67.00±22.76)points,hand grip strength at(15.29±4.94)kg,gait speed at(0.61±0.26)m/s,TUGT time at(15.05±6.74)s,and SPPB score at(6.17±1.40)points,while the corresponding post-intervention findings were as follows,(78.72±15.83)points,(17.67±5.83)kg,(0.77±0.28)m/s,(13.49±6.16)s,and(9.25±1.71)points,respectively.The experimental group showed improvements in all measures from baseline to post-intervention(P<0.05 for all measures).As for the control group,the baseline data for the corresponding measures were as follows,(67.20±22.12)points,(15.00±5.35)kg,(0.58±0.23)m/s,(17.29±6.90)s,and(6.00±1.24)points,respectively.The post-intervention findings increased to(71.13±20.28)points,(15.47±5.72)kg,(0.64±0.28)m/s,(16.50±6.99)s,and(6.73±1.61)points,respectively,but the changes were not statistically significant(P>0.05).Furthermore,an intergroup comparison of intervention effects(post-intervention minus preintervention)revealed significant differences in mean changes from baseline.The experimental group demonstrated improvements of(+11.72±6.32)points in modified Barthel Index,(+11.72±6.32)kg in grip strength,(+0.16±0.09)m/s in gait speed,(—1.56±1.32)s in TUGT time,and(—1.56±1.32)points in SPPB score.In contrast,the control group showed smaller changes of(+3.93±5.65)points,(+0.47±1.37)kg,(+0.06±0.07)m/s,(—0.79±1.54)s,and(+0.73±1.12)points,respectively(all P<0.05).Intergroup comparisons revealed superior outcomes in the experimental group across all measures.Conclusion OEP significantly enhances activities of daily living,improves muscle strength,balance,and physical function in older adults,and is more effective than conventional rehabilitation exercise programs,making it suitable for extensive clinical application.
