Objective:To explore the impact of plant and animal dietary behaviors on type 2 diabetes mellitus (T2DM) in older adults aged ≥65 in 18 longevity areas of China.Methods:The subjects were 5 223 older adults over 65 years old from the Healthy Ageing and Biomarkers Cohort Study (HABCS) in 18 longevity areas in China. Through a questionnaire survey and physical examination, information about their demographic characteristics, lifestyles, daily activities, self-health status, current diseases, and fasting venous blood were collected. Food Frequency and Questionnaire (FFQ) was used to collect data on food intake frequency. Based on the prior method, the plant-based diet index (PDI) and animal-based diet index (ADI) of 5 223 older adults were calculated. Subjects were divided into three groups (low-level group: PDI<39 or ADI<31, middle-level group: 39≤PDI≤42 or 31≤ADI≤34, high-level group: PDI>42 or ADI>34) by tertiles of PDI and ADI. Multivariate logistic regression was used to analyze the association between PDI and ADI and the risk of T2DM.Results:The average age of 5 223 subjects was (84.8±11.5) years, with the median ( Q1, Q3) of PDI about 41(38, 43) and the median ( Q1, Q3) of ADI about 33 (30, 35). The prevalence rate of T2DM was 16.41% (857/5 223). After adjusting for covariates, multivariate logistic regression showed that PDI was negatively associated with T2DM. Compared with the low-level group, the OR (95% CI) for T2DM in the high-level group was 0.83 (0.69-0.99). ADI was positively associated with T2DM, and compared with the low-level group, the OR (95% CI) for T2DM in the high-level group was 1.28 (1.06-1.55). For every one-point increase in PDI and ADI, the risk of T2DM decreased by 2% and increased by 3%, respectively, with the OR (95% CI) of 0.98 (0.96-1.00) and 1.03 (1.01-1.06), respectively. Conclusion:In Chinese older adults ≥65 years in 18 longevity areas, higher adherence to the plant-based behavior may be negatively associated with the risk of T2DM, while higher adherence to the animal-based behavior may be positively associated with the risk of T2DM.

Objective:To explore the effect of early mother to child separation on neonatal nervous system and its related mechanism.Methods:Randomly selected during January 2015 to September 2015,was born in 120 cases of newborn mice as the research object,the newborn mice was born after the implemented of mother-to-child separation as observation group (60),not the implemented of separation of mother and baby after birth as the control group (60),in view of the two groups of newborn mice of the nervous system,the change of the nerve cells were compared and researched.Results:The apoptotic rate of neurons in the neonatal mice was significantly higher than that in the control group (P ＜0.05).The neurons of caspase-3 protein expression was significantly higher than the control group (P ＜0.05).The expression of Caveo-1 protein in the glial cells of 14 days and 21 days was observed in the neonatal mice.The expression of Caveo-1 protein in the glial cells of the control group For comparison,(P ＜0.05),which were statistically significant.Conclusion:The early implementation of matemal and infant birth separation of neonatal nervous system will produce great influence,and influence the expression of the nervous system in the development of newborn,which affect the behavior of the neonatal adult dysplasia.

Objective To investigate the relationship between hereditary deafness and SLC26A4 gene IVS16+10C>T mutation.Methods One hundred and two patients with hereditary deafness admitted to the Third Affiliated Hospital of Qiqihar Medical College from January 2014 to May 2016 were enrolled and assigned as the observation group, and another 102 cases with normal hearing were selected as the control group. The gene mutation types and hearing thresholds were detected in the two groups and compared between them, the mutations of alleles 1 and 2 situations of patients with GJB2 and SLC26A4 mutations were analyzed, Ab initio software was used to predict whether there was obstacle preventing the recognition on slice sites, and polymerase chain reaction (PCR) was adopted to detect the common mutation types of SLC26A4 gene.Results In 102 patients with hereditary deafness, the cases caused by SLC26A4 gene mutations were more than those caused by GJB2 gene mutations (30 cases vs. 15 cases). Compared with the normal hearing control group, the mutation rates of GJB2 and SLC26A4 genes were significantly increased in the observation group [GJB2: 14.71% (15/102) vs. 2.94% (3/102), SLC26A4: 29.41% (30/102) vs. 1.96%(2/102), bothP <0.01], and there was a tendency that the percentages of GJB2 and SLC26A4 mutations were increased (GJB2: 0.98%, 1.96%, 4.90%, 6.86%, SLC26A4: 4.90%, 6.86%, 7.84%, 9.80%) with the increase of the severity of deafness (mild—moderate—severe—extreme severe) in the observation group. Compared with the control group, the hearing threshold was significantly increased (dB: 67.83±8.96 vs. 10.43±2.89,P < 0.01), and along with the increase of deafness severity (mild—moderate—severe—extreme severe), the hearing threshold (dB) was increased (34.96±4.98, 58.42±10.61, 83.96±12.17, 96.77±11.42, respectively) in the observation group. Thirty patients with SLC26A4 gene did not show any IVS16+10C>T mutation, indicating that IVS16+10C>T gene mutation was not the cause of genetic deafness.Conclusion There is no obvious relationship between the IVS16+10C>T mutation of SLC26A4 gene and patients with hereditary deafness, which may provide a basis clinically for the prediction of deafness occurrence in the patient's next generation.

Objective To observe the clinical effect of Danggui Sini decoction for treatment of patients with chronic non-atrophic gastritis and spleen-stomach deficiency cold syndrome.Methods A prospective randomized control trial was conducted, including 126 in- and out-patients with chronic non-atrophic gastritis diagnosed by gastroscopy and spleen-stomach deficiency cold syndrome differentiated by traditional Chinese medicine (TCM) method admitted to Chinese Medicine Hospital of Haikou City. They were randomly assigned to an observation group (65 cases) and a control group (61 cases). The patients in observation group were given Danggui Sini decoction with addition or/with subtraction of ingredients according to individual situation (ingredients: angelica 15 g, ramulus cinnamomi 15 g, paeonia 15 g, asari 5 g, glycyrrhizae 10 g, Chinese jujube 12, medulla tetrapanacis 10 g ). The warm 200 mL (one dose) decoction was divided into two parts, one part was taken orally before a meal, twice daily, the therapeutic course was 4 weeks and in one week only 6 doses were applied; while the patients in control group, omeprazole capsules (dissolved in intestine), each time 20 mg, once a day combined with domperidone (bite phenanthroline) 10 mg each time, 3 times a day were given. After 4 weeks of treatment, the clinical efficacy, the total integral of symptoms, the therapeutic effect of TCM symptoms score, the improvement of the therapeutic effect of mucosal histological and changes of the number of patients with the patholoyical changes of mucosa under gastroscopy were observed.Results The total effective rate in the observation group was significantly higher than that in the control group [96.9% (63/65) vs. 86.9% (53/61),P < 0.05]. After treatment, the total integral of symptoms and the integrals of main TCM symptoms after treatment were lower than those before treatment, the degree of decrease in the observation group being more marked (the total integral of symptoms score: 12.36±2.11 vs. 15.94±2.37, epigastric distention integral score: 1.23±0.72 vs. 2.12±0.78, epigastric pain score: 1.18±0.67 vs. 2.25±0.94, thermophilic and like pressure integral score: 1.03±0.46 vs. 1.62±0.79, anorexia: 1.06±0.57 vs. 1.43±0.53, integral of loose stools score: 1.36±0.43 vs. 1.67±0.58, lassitude integral score: 1.24±0.32 vs. 1.65±0.42, allP < 0.05). Under gastroscope, the efficacy of mucosal histological change was significantly higher in observation group than that in control group [90.77% (59/65) vs. 81.97% (50/61),P < 0.05]. After treatment, the numbers of patients with chronic inflammatory cell infiltration (47 cases vs. 54 cases), erythema exudation (28 cases vs. 30 cases), flat erosion (8 cases vs. 15 cases), swelling erosion (5 cases vs. 6 cases), bile reflux (2 cases vs. 3 cases) and positive pylorus spirobacteria (20 cases vs. 29 cases) were significantly lower in the observation group than those in the control group (allP < 0.05).Conclusion The therapeutic effects of Danggui Sini decoction for treatment of patients with chronic non-atrophic gastritis and TCM syndrome of spleen-stomach deficiency-cold are remarkable.

Objective To investigate changes in number of endothelial progenitor cells(EPCs)from bone marrow and circulation in mice with acute pancreatitis.Methods BALB/c mice were assigned randomly to saline group and cerulein group.Animals were sacrificed at 12, 24 and 48 hours after injection.Bone marrow and circulating EPCs were detected by flow cyzometric analysis.Plasma VEGF, TNF-α and ET-1 were determined by enzyme-linked immunosorbent assay.The expression of VEGF in the pancreas was assessed by Western blotting.Apoptosis in situ was detected by TUNEL.Results The amounts of EPCs in bone marrow and circulation increased remarkably after cerulein injection(P < 0.05), also the levels of plasma VEGF TNF-α and ET-1(P < 0.05), the EPCs levels in bone marrow and circulation seen in the study closely mirrors the levels of VEGF detected in the circulation(r = 0.77, 0.67 individually).VEGF expression in pancreas was up-regulated after 12 h of cerulein injection compared with that of control group.Apoptosis of endothelial cells also increased in the cerulein group.Conclusion EPCs were mobilized by acute pancreatitis, which may be due to the mobilizing effect of increased levels of VEGF, EPCs may participate in the repair process of injured endothelium induced by acute pancreatitis.

Pulmonary surfactant ( PS ) compromises lipids and surfactant proteins (SP) and lines on the alveolar air-liquid interface. It can reduce surface tension, prevent alveoli from collapse and reduce alveoli edema by disaturated dipalmitoylphosphatidylcholine. It also modulates the pulmonary immunology by SP-A and SP-D. In this study,we established a rat model of immunocompromised host (ICH) with pulmonary infection of Pseudomonas aeruginosa (P. aeruginosa), then studied its pulmonary inflammatory reaction and analyzed the concentration of lipids and SP-A in bronchoalveolar lavage fluid (BALF) during infection.

Objective To understand the interaction between surfactant proteins and pneumocystis carinii pneumonia (PCP),and the impact of corticosteriods on surfactant proteins.Methods We established rat models of PCP and bacterial pneumonia induced by subcutaneous injection of 25mg cortisone acetate.At 8- 12 wk,the bronchoalveolar lavage fluid(BALF)of rats was collected.Total nucleated cells of BALF were counted and differentiated,and the concentrations of surfactant protein A(SP-A)and surfactant protein D(SP-D)were measured by immunoblotting assay.The rats were divided into three immunosuppressive groups and a normal control group.Group I,normal control(n = 6),consisted of healthy SD rats;group Ⅱ,negative control(n = 6),consisted of rats with cortisone acetate injection for over 8 wk without lung infection;group Ⅲ,bacterial pneumonia(n = 11),rats were injected with cortisone acetate over 8 wk that resulted in bacterial pneumonia without other pathogens isolated;and group Ⅳ,PCP(n = 14),rats with injected cortisone acetate for 8 - 12 wk and developed PCP without other pathogens isolated.Results Our results indicated that the total cell count in BALF in the negative control group was lower than that in the normal control group(P ＜ 0.001).During PCP infection,the total cell count and the percentage of polymorphonuclearcytes(PMNs)in BALF were significantly increased(P ＜ 0.01),but were lower than those in the bacterial pneumonia group.The concentration of SP-A of BALF in PCP(45.1 ± 22.1 μg/ml)was significantly increased in comparison with that in the negative control(16.2 ± 9.9 μg/ml,P ＜ 0.05)and bacterial pneumonia groups(6.2 ± 5.6 μg/ml,P ＜ 0.001).We also found that the relative content of SP-D was significantly higher in PCP(24249 ±4780 grey values)than that in the negative control (13 384 ± 2887 grey values,P ＜ 0.001)and that in bacterial pneumonia(11 989 ± 2750 grey values,P＜0.001).SP-A and SP-D were also higher in the moderate to heavy group of PCP than those seen in the mild group(P ＜ 0.01,P ＜ 0.001).SP-A and SP-D were higher in the negative control group than those in the normal control group,but there was no significant difference between the 2 groups.Conclusion These results suggest that the concentrations of SP-A and SP-D in BALF are increased by pneumocystis carinii specific stimulation,but the alteration is not related to the corticosteriod usage.

Objective To observe the effect of inhibition of polo like kinasel (Plk-1) gene expression on 5-Fu and CDDP induced apoptosis of gastric cancer cell line-MKN45 in vitro. Methods The Plk-1 expression was inhibited by chemically synthesized siRNA, the Plk-1 mRNA and protein level were measured by real-time PCR and Western blotting, MKN45 cells proliferation was measured by MTT method, the apoptosis rate was detected by flow-cytometry. Results After treatment by siRNA targeting Plk-1, Plk-1 mRNA level decreased by 51.91% .89.45%.91.03% at 24 h.48 h and 72 h, and Plk-1 protein level decreased by 38.43% and 60.66% at 48 h and 72 h, compared with control group. The proliferation of MKN45 cells treated by a combination of 5-Fu, CDDP and Plk-1 siRNA significantly slowed down when compared with treatment of 5-Fu, CDDP or Plk-1 siRNA(P
