Objective To preliminarily investigate the safety and efficacy of donor pancreas needle biopsy in simultaneous pancreas-kidney transplantation. Methods Clinical data of 7 cases undergoing donor pancreas biopsy were collected retrospectively. All cases underwent donor pancreas biopsy before or during simultaneous pancreas-kidney transplantation. Frozen section or paraffin sectioning techniques were used for tissue preparation, and hematoxylin-eosin and Masson staining were performed to histologically evaluate the donor pancreas. The quality of donor pancreas was comprehensively assessed by combining histological findings with the donor's clinical data. Postoperative follow-up data of 5 simultaneous pancreas-kidney transplant recipients were collected to summarize the safety of donor pancreas biopsy and the prognosis of transplant recipients. Results The 7 pancreas donors were aged 28 to 62 years, with a body mass index ranging from 20.76 to 27.68 kg/m2. Liver ultrasound indicated fatty liver in 3 cases, while pancreatic ultrasound did not reveal any significant abnormalities. Among them, biopsy was performed on 2 donors after completion of pancreatic procurement and processing, and the frozen section histology showed moderate acute pancreatitis changes (edema of acinar cells, necrosis and inflammatory cell infiltration). Combined with a serum amylase level elevated more than 3 times the upper limit of normal value, these two donor pancreases were finally discarded. The remaining 5 cases underwent biopsy immediately after pancreatic vascular anastomosis during simultaneous pancreas-kidney transplantation, and histological evaluation was performed on paraffin-embedded sections. No biopsy-related complications (such as bleeding, pancreatic fistula, etc.) occurred after transplantation. One recipient died of severe infection 2 months after transplantation, while the other 4 recipients were followed up for more than 5 years, with well-functioning transplant kidneys and pancreases. Conclusions Donor pancreas biopsy is relatively safe, and the risk of biopsy-related complications after transplantation is controllable. Comprehensive assessment of donor pancreas quality by combining histological evaluation with the donor's clinical indicators is conducive to improving the accuracy of donor pancreas selection and organ utilization.

3.Progress and perspectives in exploring the biological mechanisms of early chondrocyte damage in Kashin-Beck disease based on iPSC-derived disease model
Chinese Journal of Endemiology 2025;44(10):775-779
To address the key issues of unclear cartilage damage mechanisms, lack of disease models, and targeted interventions in Kashin-Beck disease (KBD), which poses a serious threat to the health of the Chinese population, this article reviews the progress and application of constructing disease cell models that are highly homogeneous with KBD primary chondrocytes both domestically and internationally, providing new scientific basis for further study on the cartilage damage mechanisms of this disease. Firstly, China has successfully constructed an induced pluripotent stem cell (iPSC)-derived disease model that is highly homogeneous with KBD primary chondrocytes, providing stable disease cell resources for in-depth study on the mechanism of cartilage damage in this disease. Based on the KBD-iPSCs-derived disease model, it is found that T-2 toxin, HT-2 toxin, deoxynivalenol (DON), and/or low selenium have an early damaging effect on the KBD-iPSCs disease model. Among them, the combined environmental factors have a more significant effect on KBD-iPSCs chondrocytes damage than a single environmental factor. It is found that selenomethionine (Se-Met) and C-Jun N-terminal kinase 1 (JNK1) inhibitors have protective effects against early damage to KBD-iPSCs chondrocytes. In the future, the KBD-iPSCs-derived disease model can be further applied, combined with the establishment of animal models similar to human cartilage growth and development, to determine the molecular biological mechanisms, biomarkers in early disease stage, and targeted therapies of KBD chondrocytes damage, so as to provide scientific basis for advancing the study on new strategies to eliminate KBD.

This study was aimed at investigating the effectiveness of various host nucleic acid removal and non-specific amplifica-tion techniques in animal tissue samples,to increase the accuracy of pathogen identification in tissue samples.Simulated samples were prepared with a mixture of mouse lung tissue homogenates and Klebsiella pneumoniae fluids, and processed with six host nucleic acid removal kits and three non-specific amplification techniques. The effectiveness of each method in removing host DNA and enriching nucleic acids of pathogenic microorganisms was evaluated through real-time fluorescence quantitative PCR and high-throughput se-quencing.For host nucleic acid removal techniques, the method of selective cleavage and quantitative degradation of host DNA(Com-plete5 kit)effectively decreased the host nucleic acid content in tissue samples and increased the relative abundance of pathogen nucleic acids. In contrast, the magnetic bead method for host DNA removal(Next microbiome DNA enrichment Kit kit)was less effec-tive.At lower pathogen concentrations(77 CFU/mL),the Vazyme kit was more effective than the other kits in removing host nucleic acids.Non-specific amplification techniques(MALBAC whole genome amplification,MDA isothermal amplification, and random primer amplification)were not applicable to tissue samples and were not effective in increasing the relative abundance of pathogen nucleic acids.Selective lysis and quantitative degradation of host DNA were suitable for processing tissue samples with high host back-ground and low pathogenic microorganism levels, whereas non-specific amplification methods were not applicable to tissue samples for pre-processing of macro-genome high-throughput sequencing.
