Objective To investigate the effect of eosinophil(EOS)/neutrophil(NER)in peripheral blood on asthma recurrence in children with bronchial asthma(BA)after treatment.Methods A prospective cohort study was conducted in the study.BA children admitted to the hospital from February 2020 to February 2022 were selected as the research objects.All children were treated regularly and their condition was under con-trol.After a follow-up of 6 months,the recurrence of the children was recorded.EOS/NER,interleukin(IL)-4 and IL-5 of children with and without relapse before treatment and at 1 week of treatment were compared.Lo-gistic regression analysis model was established to test the effect of EOS/NER on the recurrence of asthma in children with BA after treatment.The receiver operating characteristic(ROC)curve was drawn to analyze the predictive value of EOS/NER,IL-4 and IL-5 before treatment for the recurrence risk of BA children after treatment.Results A total of 128 children with BA were included.25 cases recurred after 6 months of follow-up,with a recurrence rate of 19.53%.EOS/NER,IL-4 and IL-5 of BA children after 1 week of treatment were lower than those before treatment(P<0.05).EOS/NER,IL-4 and IL-5 of recurrent children before treat-ment and 1 week after treatment were higher than those of non-recurrent children(P<0.05).There were no statistically significant differences in EOS/NER,IL-4,IL-5 levels between children of different genders,age groups,and treatment regimens before and after 1 week of treatment(P>0.05).Increased EOS/NER,in-creased level of IL-4 and increased level of IL-5 had effect on the recurrence of BA children after treatment and may be risk factors for recurrence(OR>1,P<0.05).The ROC curve showed that area under the curve of EOS/NER before treatment for predicting the recurrence was 0.810(95%CI:0.724-0.896,P<0.001)in BA patients after 6 months of treatment,with moderate predictive value.Conclusion EOS/NER before treat-ment in BA children may be an independent risk factor for recurrence after treatment,and has moderate pre-dictive value for recurrence risk.

Objective To analyze the differences in the expression of specific antibodies targeting different human immunodeficiency virus (HIV) antigens among patients at different clinical stages in Qiandongnan Prefecture of Guizhou Province, and to explore their association with the clinical staging of acquired immunodeficiency syndrome. Methods A total of 307 HIV-positive blood samples from Qiandongnan of Guizhou Province were selected for specific HIV antibody immunoblotting assays. CD3+CD4+T cell counts andHIV viral load nucleic acid testing were performed on the blood samples. Multivariate regression analysis was conducted on relevant indicators during HIV infection and AIDS stages. Results Among the 307 HIV-infected individuals, 218 were male and 89 were female, with a mean age of (48.53±16.03) years. The composition ratios of specific antibodies gp160, gp120, gp41, p66, p51, p31, p55, p24 and p17 were 98.7%, 90.9%, 92.2%, 74.3%, 66.4%, 86.6%, 3.9%, 97.4% and 73.9%, respectively. Multivariate binary Logistic regression model analysis showed that expression of p24-specific antibodies were more likely to be judged as influencing factors in the infection stage (OR=0.158, 95%CI, 0.032 to 0.768, P < 0.05). There was a certain correlation between p24-specific antibodies and the clinical staging of HIV infection (OR=0.217, 95%CI, 0.005 to 0.944, P < 0.05). Using the ratio of CD3⁺ CD4⁺ T cell count to viral load when p24 was specifically expressed as the test variable, and the clinical stage as the state variable (AIDS stage=1, infection stage=2), a receiver operating characteristic (ROC) curve was plotted. The area under the curve was 0.653 (95%CI, 0.529 to 0.774, P < 0.05). Conclusion Differential expression of p24-specific antibodies may indicate that patients are in the infection stage and could potentially serve as an early warning indicator of immune function for HIV-infected individuals.

Objective To compare the clinical features of drug induced hepatitis caused by traditional Chinese medicines (TCM) and western medicines. Methods A total of 181 patients hospitalized with drug in-duced hepatitis between January and December 2015 were enrolled. Among the patients ,75 cases were in TCM group,66 cases in western medicine group and 40 cases in combined group(accepted both TCM and western medi-cine treatment). Liver biopsies were performed and Roussed Uclaf Causality Assessment Method (RUCAM) scores showed that all patients were with scores higher than 3. The data including gender,age,clinical manifesta-tions,physical signs,laboratory tests and image characteristics were analyzed individually. Results There was no significant difference in age,the pattern of liver injury,and the course of liver damage among the three groups (P>0.05). Anorexia was common symptom in all the three groups without any significant difference(P>0.05). Jaundice was most frequently observed in TCM group while fever was most commonly found in western medicine group,which both had significant difference(P< 0.05). The results of routine coagulation tests and serum albu-min values were normal in 3 group with increasing level of aminotransferase observed ,and there was no significant differences among 3 groups(P>0.05). Compared with western medicine group ,the patients in TCM group had a higher level of platelet counts ,serum levels of total bilirubin ,total bile acids(TBA)and serum iron(P<0.05), and less proportion of eosinophils and lower level alkaline phosphatase(P< 0.05). Conclusions The age,the pattern of liver injury,the course of liver damage and aminotransferase levels in patients with drug induced hepati-tis caused by TCM and western medicines were similar;however,western medicines were more likely to cause al-lergic responses and hyperbilirubinemia occurred more frequently in patients with drug induced hepatitis caused by TCM.

Objective To evaluate the effect of therapeutic hypercapnia on cerebral oxygen metabo?lism in the patients undergoing thoracoscopic surgery in beach chair position ( BCP ) . Methods Sixty pa?tients of both sexes, aged 18-32 yr, with body mass index of 19-24 kg∕m2 , of American Society of Anes?thesiologists physical statusⅠorⅡ, scheduled for elective bilateral thoracic sympathectomy performed via a thoracoscope, were divided into control group ( group C ) and hypercapnia group ( group H ) , with 30 patients in each group using a random number table. After induction of anesthesia, all the patients in both groups were tracheally intubated and mechanically ventilated using the ventilation regimen low tidal vol?ume intermittent positive pressure ventilation combined with low level of positive end?expiratory pressure ( 5 cmH2 O) , maintaining arterial carbon dioxide partial pressure ( PaCO2 ) at 35-45 mmHg. PaCO2 was maintained at 45-55 mmHg by adjusting the respiratory rate after the patients were placed in BCP in group H. Anesthesia was maintained with target?controlled infusion of propofol and intermittent intravenous boluses of rocuronium and sufentanil. Bispectral index value was maintained at 45-55. Before anesthesia induction ( baseline) , at 5 min after intubation, and at 5, 10, 15 and 20 min after the patients were placed in BCP, blood samples were taken from the radial artery and jugular bulb for blood gas analysis, jugular ve?nous bulb oxygen saturation was measured, and arteriovenous blood O2 content difference, cerebral O2 ex?traction rate, and venous to arterial blood lactate concentration difference were calculated. Results Com?pared with group C, PaCO2 and jugular venous bulb oxygen saturation were significantly increased, and ar?teriovenous blood O2 content difference and cerebral O2 extraction rate were significantly decreased at at 5, 10, 15 and 20 min after the patients were placed in BCP in group H ( P<0?05) , and there was no signifi?cant change in venous to arterial blood lactate concentration difference at each time point between the two groups ( P>0?05) . Conclusion Therapeutic hypercapnia can improve the cerebral oxygen metabolism in the patients undergoing thoracoscopic surgery in BCP .

CD34+ cells were selected from umbilical cord blood mononuclear cells (MNC) by MiniMACS system, expanded under the same conditions as those for total MNC, coincubation of CD34+ and CD34- from the same donor for EPCs. In addition, the effects of vessel endothelial growth factor (VEGF) and passage on cell differentiation, expansion kinetics and apoptosis were examined. EPCs were determined and quantified by immunocytochemistry and flow cytometry. The results showed that both coculture of CD34+ and CD34- and total MNC led to a significant increase in the expansion of CD34+ cells as compared with CD34 enrichment (P < 0.05). There was a tendency toward decreased apoptosis in cultures when early passage was performed immediately after cord like structures appeared. VEGF had no significant effect on apoptosis (P > 0.05). These differentiated EPCs were positive for CD34+, von Willebrand factor (vWF), KDR, CD31 staining and phagocytized acetylated low-density lipoprotein (LDL). CD34+ cells accounted for (68.2 +/- 6.3)% of attaching (AT) cells at day 7 of culture. It was suggested the most efficient method to ex vivo expansion of EPCs was coculture of CD34+ and CD34- or total MNC. Early passage makes cell apoptosis rate decrease. VEGF had no significant effect on ex vivo expansion of EPCs.

Objective To determine the level of anticyclophilin A auto-antibodies (ACA) in sera of patients with psoriasis. Methods Cyclophilin A (CyPA) was extracted and purified from bovine thymus. The level of anticyclophilin A auto-antibodies (ACA) was detected by enzyme-linked immunosorbent assay (ELISA) and immunoblotting test (IBT) in sera of patients with psoriasis. Results It was shown that the level of ACA was significantly higher in the sera of patients with psoriasis than those in control group (IBT:P<0.01,ELISA:P< 0.01). ACA was found to be positive in 41.3％ of the patients by ELISA. In psoriatic group, the serum level of ACA was higher during the progressive stage than that in the stationary stage, however, no significant difference was observed (IBT:P>0.05,ELISA:P >0.05). Conclusion The results suggest that there is autoimmunological mechanism in the pathogenesis of psoriasis.

Objective To study the cytokines secreted by fibroblasts in the pathogenesis of psoriasis.Methods The healthy or psoriatic fibroblasts were co-cultured with keratinocytes in skin equivalents for15days.Changes in the expression pattern of268selected genes related to cytokines and their receptors were determined by comparing mRNA levels of fibroblasts under different culture conditions.Results Sixty-eight of268genes were found to be expressed on the fibroblasts.Compared with normal controls,a total of27genes was found to be differentially expressed at RNA level by a factor of twofold or greater,among which24genes were up-regulated,whereas3genes were down-regulated in psoriatic fibroblasts.These differentially expressed genes encoded interleukins,interferons,growth factors,chemokines,protein kinase receptors and chemokine receptors.Conclusions These results suggest that fibroblasts from psoriatic uninvolved skin strongly express cytokines which closely relate to the pathogenesis of psoriasis.It is indicated that dermal fibroblasts may play a potentially role in promoting keratinocyte proliferation and leading to the formation of psoriatic lesion.

Objective: To investigate the therapeutic effects of ets-1 antisense oligoxydeonucleotide(AsODN) on gastric carcinoma. Methods: Cultured SGC-7901 cells were devided into control group, sense oligoxydeonucleotide(sODN) group and AsODN group. After transfection for 24 h, expression of ets-1mRNA was detected by RT-PCR, growth inhibition was detected by colone formation assay, in vitro invasive ability assay was carried out in Transwell chamber,the animal model of xenotransplanted gastric carcinoma in nude mice was established to detect invasive ability in vivo. Results: ets-1 AsODN could under-regulate the expression of ets-1 mRNA, number of colone formation of AsODN group was significantly lower than the other two groups(24.2?4.8 vs 47.6?8.1 vs 44.3?7.6, P

0.05). Conclusion The results suggest that there is autoimmunological mechani sm in the pathogenesis of psoriasis.
